[In vitro study of radiation sensitizing effect of hydroxycamptothecin].

OBJECTIVE: To study the radiation sensitizing effect of hydroxycamptothecin (HCPT) on human stomach adenocacinoma and nasopharyngeal carcinoma. METHODS: Human stomach cancer cell line BGC823 and nasopharyngeal cancer cell line CNE-1 were treated in vitro with different doses of radiation alone or radiation followed by HCPT (2 micrograms/ml) for 4 h. Colony forming assay was used for survival fraction analysis. Single-hit multitarget model was used to plot survival curve. RESULTS: The value of D0, Dq and N of BGC823 cells received radiation treatment alone was 1.17 Gy, 1.91 Gy and 5.14 respectively. That of BGC 823 cells received radiation plus HCPT was 0.95 Gy, 0.01 Gy and 1.01 respectively. The sensitization enhanecment (SER) ratio was 1.23 (1.17/0.95). Similar results were obtained for CNE-1 cells treated with radiation alone or in combination with HCPT, with an SER of 1.68. CONCLUSION: HCPT has been shown a radiosensitizing effect in vitro on gastric and nasopharyngeal cancer cells.

[The clinical analysis of 38 cases with amniotic fluid embolism].

OBJECTIVE: To analyze the clinical course of amniotic fluid embolism (AFE) and identify the high risk factors. METHODS: Thirty-eight cases diagnosed as AFE in Suzhou region during period of past 15 years were analyzed retrospectively. Fifteen years were divided into 5 stages with 3 years each. RESULTS: Of 38 cases, 30 (78.9%) were primigravida and one twins. There were 34 maternal deaths. Among them, 31 died in the first four stages and 3 died in the last stage. There were 4 cases survived from AFE in the last stage. AFE accounted for 15% of total maternal deaths in the past 15 years, which is the second cause of maternal death. Of 38 cases, 2 (5.2%) occurred before 28 gestational weeks. 20 (52.6%) suffered from AFE during labour, 13 (34.2%) after delivery, and 3 (7.8%) before labour. All cases presented respiratory distress, cyanosis, chest discomfort and/or shock, cardiopulmonary collapse. Sixteen cases had postpartum hemorrhage and/or laboratory evidence of DIC. Of 38 cases, 15 (39.5%) died within one hour after onset of AFE. The predisposing factors for AFE included strong uterine contractions due to oxytocin or PEG augmented 17 cases (44.7%), pregnancy induced hypertension 11 cases (28.9%), multigravida and/or elder gravida 9 cases (23.7%) and cesarean section 8 cases (21.1%). CONCLUSIONS: AFE must be suspected in maternal cases with sudden collapse, especially the clinical symptoms of allergic reaction. Prevention of hypertonic uterine contraction and prompt resuscitation, and emergency surgical delivery can improve the prognosis of AFE.

[Suppressing effect of isoforskolin and forskolin on ocular hypertension in rabbits].

OBJECTIVE: To examine the effects of isoforskolin (isoF) and forskolin (FSK) suspensions administered topically on eyes with ocular hypertension (OHT) and observe the local irritation of the drugs. METHODS: The intraocular pressure (IOP) was measured with pneumotonometer in screened spontaneous or water-loading induced OHT rabbits for evaluation of drug effects, and the irritation indices were recorded as well. RESULTS: In screened OHT rabbits, isoF significantly lowered IOP from 0.5 to at least 4.5 h after the administration (P < 0.05, paired t-test vs. contralateral control eyes), with the maximum decreases of 0.25% and 1% isoF reaching 8.1 mm Hg and 7.0 mm Hg (1 mm Hg = 0.133 kPa) at about 1.5 h later, respectively. In water-loaded OHT rabbits, both 0.06%, 0.1%, 0.25%, 1% isoF and 1% FSK concentration-dependently suppressed the increments of IOP (P < 0.05 with the exception of 0.06% isoF) induced by water-loading, and the peak effects of the optimal concentration of 0.25% isoF and 1% FSK were 7.1 and 6.9 mm Hg (difference of IOP between bilateral eyes) respectively. The results also indicated that 0.25% isoF or FSK suspension had basically no irritation, but 1% induced slight irritating responses during the experiment. CONCLUSION: Both isoF and FSK had suppressing effects on OHT in rabbits with the optimal concentration being 0.25% and 1% respectively; at concentration of 0.25%, topically both drugs had basically no irritation, while 1% of the drugs may induce mild irritating responses.

[A pharmacological study on suppository for anti-prostatis].

OBJECTIVE: To approach the pharmacological mechanism of the suppository for anti-prostatis (SAP). METHOD: Investigating the inflammation of prostate in rat models with bacterial and nonbacterial prostatis induced by colonbacillus and "Xiao Zhi Ling", as well as the change of rheology in the rat model with acute blood stasis. Several tests on anti-inflammation, analgesia and bacteriostasis were conducted. RESULT AND CONCLUSION: SAP could restrain the inflammation of prostate and resume its secretion obviously. It is very effective for anti-inflammation, analgesia and bacteriostasis, and also helps improve the change of theology in rats with blood stasis.

[Supercritical extraction of Alpinia officinarum and GC-MS analysis of its components].

In order to find the effective components and new pharmaceutical effect of Alpinia officinarum, and further exploit it, we extracted its essential oils with the technique of Supercritical-CO2 fluid, then confirmed there were 27 components in Pot I And 111 components in Pot II by using the chemical component analysis and the computer information retrieval in the GC-MS system.

[The study on superoxide dismutase and trace element in patients with senile cataract].

PURPOSE: To investigate the relation of senile cataract with superoxide dismutase(SOD) and trace elements. METHODS: Superoxide dismutase of red blood cells and trace elements(Cu, Zn, Ca, Fe) of serum in 61 patients with senile cataract and 121 normal persons were investigated. RESULTS: The activity of superoxide dismutase in patients with senile cataract decrease dramatically in comparison with normal control, and there was no statistical difference about trace elements of Cu, Zn, Ca between senile cataract and control group. But Fe element in patients is higher than that of the normal group. CONCLUSION: The results support the hypothesis that the formation of senile cataract is capable of antioxidative scavenger system of lens decreases with age.

[Surgical techniques to deal with lens nucleus' fragment of posterior capsular rupture during the phacoemulsification].

OBJECTIVE: To investigate some surgical techniques in dealing with a impending dropped nucleus when posterior capsular rupture during the phacoemulsification. METHODS: The postoperative results of 18 cases (18 eyes) whose posterior capsular of lens ruptured with a impending dropped nucleus during the phacoemulsification were studied retrospectively. Three surgical techniques to prevent lens nucleus' fragments dropping into the vitreous cavity were applied. They are consecutive phacoemulsification with lower flowing rate and lower irrigation, injection of viscoelastic substance with high elasticity and anterior segment vitrectomy with no irrigation. The follow-up was 12-24 months (average, 18.2 months). RESULTS: Except for a special case, no lens nucleus' fragments dropped into the vitreous cavity. Posterior chamber intraocular lens(IOL) was implanted primarily in all cases (18 eyes). Best corrected visual acuity was above 0.5 in 15 eyes (83.3%). The main complications included corneal edema and cystoid macular edema. CONCLUSION: With safety, effectiveness and less complications, these three surgical techniques are suitable to deal with different types of impending dropped lens nucleus' fragments during the phacoemulsification. It is advisable to apply these techniques in clinical practice.

[A novel method for screening anti-inner ear autoantibody in patients with autoimmune diseases].

OBJECTIVE: To appraise the clinical value of a newly established method, rapid electric field immobilizing liquid phase molecule dot blot analysis (REILMD), for screening anti-inner ear autoantibody in patients with autoimmune diseases. METHODS: Seventy-one patients with 11 kinds of autoimmune diseases were chosen for the study. Both the general immunity and autoantibodies were tested. In the processes of detection of anti-inner ear autoantibody, REILMD was used for screening, and then the Western blot was used to define the molecular weight of inner ear antigen recognized by the positive autoantibody. RESULTS: Acceleration of erythrocyte sedimentation rate (ES), positive rheumatoid factor (RF), increases in C reactive protein (CRP), IgG and circulating immune complex (CIC) were found in most cases with rheumatoid arthritis (RA) and systemic lupus erythematodes (SLE). Some of these patients had increased IgA, IgM and C4. Two of 16 RA had anti-double-stranded DNA (dsDNA) and anti-mitochondria and 4/16 had anti-nucleus antibodies. Eleven of 16 SLE had antinucleus, 7/13 had anti-ribonucleoprotein (RNP), anti-Sjögren syndrome A (SSA) and anti-dsDNA, 3/13 had anti-smooth muscle (Sm) and 1/13 had anti-DNA topoisomerase I (Scl)-70, striated muscle and stomach acid cell antibodies. No autoantibody was detected in AS. Anti-inner ear autoantibody existed in 9 out of 71 patients (13%) with autoimmune diseases, in 2 of 21 patients (10%) with sudden deafness and only in 1 of 48 control subjects (2%, coronary heart disease). The anti-inner ear autoantibody was positive in 5 of 16 (31%) patients with SLE and 1 each in RA, AS, Behset's disease and streptococcus infection syndrome. In patients with positive anti-inner ear antibody, 67% had anti-nucleus antibody, 50% had anti-RNP and dsDNA antibody. The molecules recognized by the positive anti-inner ear antibody were defined as 52,000, 36,000, 31,000 and 15,000 molecules of inner ear antigen. CONCLUSION: REILMD is a feasible and easy method for screening anti-inner ear autoantibody. Several autoimmune diseases, particularly SLE, may be implicated in damage to the inner ear.

[Giant cell reparative granuloma of the skull].

OBJECTIVE: To investigate the clinical course, histologic feature, diagnosis, differential diagnosis and treatment of giant cell reparative granuloma (GCRG). METHODS: Three cases of GCRG, which arose in the skull, were collected from 1987 to 1999 and analyzed clinicopathologically. RESULTS: Two patients had the lesion in the temporal bone and one in the orbit. One patient with GCRG had a recurrence after incomplete surgical resection. Histologically, all of three cases were characterized by fibroblastic proliferation with osteoclast-like giant cell rich areas, hemorrage, and newly formed bone or osteoid trabeculae. GCRG appeared as a typical bony lytic lesion on computed tomography (CT) and as low signal intensity on both T1 and T2 weighted images on magnetic resonance imaging (MRI). CONCLUSION: GCRG is an uncommon nonneoplastic lesion that typically arises in mandible and maxilla and its involvement in skull is rare. Although behaves benignly, GCRG may be locally aggresive. It is usually misdiagnosed for a giant cell tumor (GCT). Diagnosis of GCRG is based on typical histologic features as well as clinical information. Complete surgical resection is believed to be the most suitable treatment for GCRG and long-term follow-up by CT scanning is essential.

[Sodium salicylate induced intracellular Ca2+ concentration change in guinea pig outer hair cells].

OBJECTIVE: In order to explore the mechanism of sodium salicylate ototoxicity the effect of sodium salicylate on intracellular Ca2+ ([Ca2+]i) of the outer hair cells (OHCs) and the effects of calcium channel antagonist on sodium salicylate-induced [Ca2+]i changes of the OHCs were examined. METHODS: The OHCs of guinea pig cochlear were isolated using an enzyme-machine methods and loaded with 10 mumol/L Fluo-3/AM for 30 min at 37 degrees C. Individual Fluo-3 loaded OHC was examined with a confocal microscope (ACAS Ultima, USA) using a 20 x objective lens and linear scan mean. The fluorescent images, collected each 5-sec for 300 sec, were stored in a computer. The fluorescent intensity of the OHCs were analyzed by the software cooperated with the confocal microscope, and a curve of fluorescent intensity changes trend against time was obtained. RESULTS: The [Ca2+]i of OHCs were steady under normal extracellular liquid perfusion. Salicylate increased [Ca2+]i in OHCs in calcium-free medium(10/10) and standard medium (8/10). 3 mmol/L lidocaine inhibited [Ca2+]i increase in OHCs induced by salicylate(0/10). The [Ca2+]i increase in OHCs induced by salicylate could be blocked by flunarizine(3/12). Nimodipin failed to block the [Ca2+]i increase of OHCs induced by salicylate (7/7). CONCLUSION: Sodium salicylate can result in [Ca2+]i increase in OHC markedly, which may be due to the Ca2+ release from intracellular calcium stores. The effects of salicylate on [Ca2+]i in OHCs might be one of the mechanisms of salicylate ototoxicity.

[Evaluating effects of some medicine on tinnitus with animal behavioral model in rats].

OBJECTIVE: To develop a valid behavioral animal model of tinnitus and to determine the animals whether have the perception of tinnitus and to evaluate effects of nimodipine and Chinese herbs on tinnitus using the behavioral model. METHODS: Adult male albino rats (n = 60) were fell evenly and randomly into ten groups. Animals in Group I and II were administrated sodium salicylate 350 mg/kg/d (i.p.). Instead, animals in control groups (Group III and IV) were saline injected. Pure tone (8 kHz, 65 dBSPL) was given to rats in Group V and VI instead of sodium salicylate. Nimodipine (1 mg/kg) were administrated to animals in Group VII and VIII. Gushenshuerfang, one kind of Chinese herbs were administrated to animals in Group IX and X. Medicine or pure tone were being given to all animals in Group I, III, V, VII, IX before conditioned reflex. Animals in Group II, IV, VI, VIII, X began to have medicine or pure tone after conditioned reflex. Thirsty animals (deprived of water for several days) were breeded in sound-proofed room and listened a persistent white noise (55 dB SPL) and trained daily with our instrument system, according to the procedure based on Pavlovian conditioned suppression paradigm. Subjects were permitted to lick water at an automatic dispenser during daily training sessions and the number of licks was recorded automatically. Noise offset (silence) was the conditioned stimulus and the electrical shock was the unconditioned stimulus. After two or three days training, subjects learnt rapidly to associate noise offset with shock and then established a behavioral conditioned reflex. Because of shock fear they decrease or even stop licking with the presentations of conditioned stimulus. However, subjects gradually forgot or ignored the fear without any shock present in extinction test sessions. RESULTS: Animals' licking recovered to their pre-training levels respectively at the following days: Group I--5 days, Group II--2 days, both Group III and IV--4 days, Group V--5 days, Group VI--2 days, Group VII--5 days, Group VIII--4 days, Group IX--5 days, Group X--4 days. There were much significant differences among all groups and all training days (F test, P < 0.001). Above results provided sufficient evidence for that tinnitus was induced by salicylate and tinnitus presented with noise offset. In group I, tinnitus became additional conditioned stimulus. Therefore, the fear of animals was much severe and the numbers of licking were much fewer. Extinction lasted longer. In group II, animals ignored the noise offset much easily and the extinction lasted much shorter. Moreover, results from group V and VI, pure tone instead of salicylate, indicated that sensation (tinnitus) sounded like pure tone. The groups VII, VIII, IX, X demonstrated that nimodipine and Gushenshuerfang, one kind of Chinese herbs can cure the tinnitus induced by salicylate. CONCLUSION: All behavioral evidence indicated that animals perceived tinnitus induced by salicylate and this animal model could be used in evaluating curative effects of some medicine on tinnitus.

[Thiol reagent thimerosal-induced Ca2+ mobilization in isolated guinea pig cochlear outer hair cells].

OBJECTIVE: To understand mechanism of cochlear outer hair cells (OHCs) intracellular Ca2+ mobilization further. METHODS: Intracellular calcium of isolated guinea pig was investigated using thimerosal, a--SH group oxidizing agent, and fura-2 fluorescence ratio imaging microscopy. RESULTS: In the presence of thimerosal, intracellular Ca2+ concentrations ([Ca2+]i) of OHCs were elevated in a dose-dependent manner. Even in Ca(2+)-free medium, Ca2+ response was still induced. The effects of thimerosal on [Ca2+]i were completely blocked and reversed by (DTT). Neither 1-100 mumol/L ryanodine nor 5-20 mmol/L caffeine altered the effects of thimerosal. Pretreatment with pertussis toxin (PTX) for 30 min did not affect the thimerosal-induced increase in [Ca2+]i The increase in [Ca2+]i when Ca2+ was added during thimerosal application in Ca(2+)-free medium was almost completely blocked by 500 mol/L LaCl3, while nifedipine did not inhibit further increase in [Ca2+]i caused by thimerosal. CONCLUSION: Oxidation of the -SH group of the OHC membrane can induce a Ca2+ release from intracellular Ca2+ stores, which are ryanodine- and caffeine-insensitive, and Ca2+ influx through non-specific Ca2+ channels, but not the nifedipine-sensitive Ca2+ Channels. The possible oxidation of--SH group gated Ca2+ channels in OHCs are worthy of further study.

[An observation on long-term influence of middle ear bacterial infection on inner ear function and systemic immune reaction].

OBJECTIVE: To understand whether long-term inner ear heat shock response related to heat shock protein(HSP70) caused by middle ear bacterial infection and the potential influence on inner ear function. METHODS: Sixty BALB/c mice were randomly classified into 6 groups including Klebsiella pneumoniae (KP), Staphylococcus aureus, Bacillus pyocyaneus, Bacillus coli, Bacillus proteus and physiological saline control groups. On 135 days after injection, distortion product otoacoustic emissions(DPOAE) was tested and all the samples were collected, which were examined with light and electronic microscopes. HSP70 related molecule expression in inner ear, nuclear factor (NF) kappa Bp65 characterization in mononuclear cell, anti-KP antibody and anti-membranous labyrinth proteins (MLP) were examined. RESULTS: No nuclear transfer of NF kappa Bp65 was observed in any animal. Anti-KP antibody was detected in 30% (3/10) of Staphylococcus aureus group, 29% (2/7) of KP group, 33% (3/9) of Bacillus pyocyaneus group and 10% (1/10) of control group. Anti-MLP antibody was created in 20% (2/10) of Staphylococcus aureus group, 20% (2/10) of KP group, one each in Bacillus pyocyaneus group and control group respectively. Double positive antibody against KP and MLP were found in Staphylococcus aureus group and KP group. When analyzed with Western blot, all the positive bands were small molecules including strongest 26,000-30,000, medium degree 38,000-41,000 and weak 46,000-50,000 except for 68,000 in one case. There was only one significant DPOAE amplification decrease at 1,625 Hz (2f1-f2) in left ear of Bacillus pyocyaneus group and right ear of Bacillus proteus group. No abnormal phenomenon was found in inner ear both under light microscope and electronic microscope. No significant expression of HSP70 was observed in inner ears. CONCLUSION: No long-term heat shock response related to HSP70 existed in the inner ear and the immune inner ear damage may be caused by multiplefactors.

[IR and Raman studies of inorganic nano giant cluster compounds].

The syntheses, IR and Raman spectra of four nanosize giant cluster compounds were reported in this paper. These compounds show similar IR and Raman spectra because they were assembled by the similar building blocks. The bands with frequencies in IR spectra in the range from 980 to 550 cm-1 and those absorptions under 970 cm-1 in Raman spectra have to be attributed to the absorptions of Mo-O bonds including Mo = O, Mo-O-Mo and mu 3-O-Mo bonds.

[Study on the third-order nonlinear optical effect of fused-cyclic aromatic hydrocabon].

The third-order nonlinear optical susceptibility X(3), nonlinear refractive index n2 and time response property of 1,2,5,6,dibenzathracene (C22H14) and dibenzopentacene (C30H18) in fused-cyclic aromatic hydrocarbon were studied with forward degenerate four-wave mixing (DFWM). The mechanism for their larger nonlinear effect and faster time response was discussed.

Selective epimerization of rapamycin via a retroaldol/aldol mechanism mediated by titanium tetraisopropoxide.

We describe the efficient and selective epimerization of the immunosuppressant rapamycin to 28-epirapamycin under mild conditions. The mechanism of epimerization involves an equilibrium of the four C28/C29 diastereomers through a two-step retroaldol/aldol (macrocycle ring-opening/ring-closing) sequence. This retroaldol/aldol equilibration is not restricted to rapamycin but is also applicable to acyclic beta-hydroxyketones. A potentially useful extension of the method--the use of beta-hydroxyketones as enolate synthons for effecting inter- or intramolecular aldol reactions under neutral conditions--is demonstrated.
