[Study on the psychological status of video display terminal operator].

Occupational psychological test designed by the Labour Hygiene Department of West China University of Medical Sciences was applied to survey the mental health of 516 video display terminal (VDT) workers and 396 control workers in Chengdu. The results showed that mental health level of VDT workers was higher than that of controls. In the somatic disorder, anxious and hostile are increased obviously. With the prolonging of operating hours per week, somatic disorders, depression and obsession are increased, especially when VDT operating time is more than 30 hours per week and operating on VDT is more than ten years. This indicates that the operating hours should not be more than 30 hours per week. It is found by multiple factors analysis of data that the main adverse effects on mental health are age and VDT operating time.

[Distribution of morphine in acute morphine-treated rats].

The distribution of morphine in acute morphine-treated rats was studied by immunohistochemistry (PAP). Samples were examined for morphine distribution and localization in internal organs at 10-160 min after intraperitoneal injection of morphine. The results showed that morphine was distributed in the heart, hepatocytes, epithelial cells of the bile canalicular cells in liver, spleen, kidney, adrenal glands, stomach parietal cells, small intestinal villus, pancreas islet, sublingual glands and submandibular glands. Morphine was found in the cardiac tissue, liver, spleen, pancreas islet, small intestinal villus at 10 min after injection and in the cardiac tissue, stomach parietal cells at 160 min after injection. The cause of the postmortem the distribution of morphine in internal organs at different times after injection was discussed. The results of this study may be used as a basis for the choice of samples for the forensic toxicological analysis.

[Determination and significance of nitric oxide and tumor necrosis factor-alpha induced by peripheral blood monocyte in vitro in patients with malignant bone tumors].

Peripheral blood monocytes from 15 cases of malignant bone tumors and 12 cases of health volunteers were seperated and induced by lipopolysaccharide(LPS) for 48-hour culture. The supernatant was collected for determining the content of nitrite and tumor necrosis factor-alpha(TNF-alpha) (by Griess and ELISA methods, respectively). The results were as follows: The content of nitrite and TNF-alpha in patients with malignant bone tumors was significantly higher than that in normal control; the content of nitrite and TNF-alpha in the malignant bone tumor group presented the positive correlation (r = 0.8909, P < 0.01). The results suggest that NO and TNF-alpha produced by activated macrophages may be related to the pathogenesis of malignant bone tumors.

[Adenovirus-mediated p53 gene therapy of human laryngeal cancer].

OBJECTIVE: To explore the potential use of p53 in gene therapy for laryngeal cancer. METHODS: A human laryngeal cancer cell line Hep-2 was used. Recombinant cytomegalovirus-promoted adeno-viruses containing human wild-type p53 cDNA was transiently introduced into Hep-2 cells in vitro and injected into tumor nodules in vivo. The growth of Hep-2 cells in vitro and established s.c. squamous carcinoma nodules in nude mice was examined. RESULTS: The transduction efficiency of Hep-2 cell line was 100% at > or = 100 MOI. The p53 protein expression peaked on day 2 after infection and lasted far 5 days. Cell growth was greatly suppressed. In vivo studies, Ad5CMV-p53 transfestion in vitro inhibited tumorigenicity of Hep-2 cells in nude mice. Intra-tumoral injection of Ad5CMV-p53 significantly inhibited established s.c. implanted xenograft. CONCLUSION: Transfection of wild-type p53 gene via Ad5CMV-p53 is a potential approach to the therapy of laryngeal cancer.

[Preliminary studies of apoptotic cell-associated antigens on hepatoma].

OBJECTIVE: To analyse antigens transiently expressed on hepatoma cells during apoptotic response. METHODS: Hepatoma HCC9204 cells were treated with 6% ethanol for 6 hr to induce apoptosis. Apoptotic HCC9204 cells were isolated and homogenized. After centrifugation at 14,000 rpm, the supernatant was dialyzed against water with molecular weight cutoff at 12,000. The dialysate was used as hepatoma apoptotic cell-associated antigen (HACAA). Subtractive immunization technique was used to prepare polyclonal antibodies against apoptotic cell-associated antigens on hepatoma. The antibody response to HACAA was determined by ELISA and immunoblot assays. Localization of HACAA on hepatoma apoptotic cells was demonstrated immunocytochemically. RESULTS: This polyclonal antibody reacted more strongly with HACAA than with antigen likewise prepared from non-apoptotic hepatoma cell. The antigenic determinants were sensitive to trypsin and mainly located on the cytoplasmic membrane. CONCLUSION: Epitopes are expressed on hepatoma apoptotic cells which can be used to examine an apoptotic response. The antibody reported in this paper is useful to screen novel apoptosis associated genes.

[Analysis on the risk factors in patients with diabetes mellitus from population in mining districts--a population-based case-control study].

According to data from prevalence study on population from Pingdingshan coal mining districts in Henan province, we analysed 174 patients with diabetes mellitus(DM) and 3,066 control subjects with normal blood glucose(NGT) by a population-based case-control study. After the adjustment of other factors and controlled on confounding factors, the results of unconditional logistic multivariate regression analysis demonstrated that age, DM history of mother and sib, highest BMI through one's life, higher concurrent WHR, higher systolic blood pressure, frequently eating Chinese sorghum and legume may serve as independent risk factors of DM, their odds ratios(OR) were 2.04, 6.04, 2.24, 1.85, 2.57, 1.51, 2.22, 1.25 and their population attribution rates (PAR%) were 80.04%, 7.19%, 3.18%, 37.35%, 48.80%, 8.15%, 3.20%, 10.63% respectively. Higher occupational physical activity and frequently eating vegetables of light colour might serve as independent protective factors of DM, with ORs 0.89 and 0.50 and PAR% of -19.20% and -269.5% respectively. Confounding analysis showed that age was both a positive and negative confounding factor to other factors in the logistic regression model.

[The clinical meaning and determination of the superoxid dismutase activity and malondialehyde content in the serum of the patients suffering from chronic tonsillitis].

The activity of SOD and MDA content in the serum of patients suffering from chronic tonsillitis were determined one day before operation and one week after of operation. It was found that the activity of T-SOD Mn-SOD and CuZn-SOD were decreased in the serum of patients with chronic tonsillitis than that in the normal control group with statistical significance (P < 0.01). T-SOD activity was increased and MDA content was decreased significantly after operation than the time before operation (P < 0.001, P < 0.01). This study indicates that tonsil excision could decrease MDA content and increased SOD activity for the chronic tonsillitis patient.

[Immunohistochemical study of Cath-D and CD44 in laryngeal squamous cell carcinomas].

The expression of Cath-D and CD44 was examined immunohistochemically in 96 laryngeal squamous cell carcinomas and compared with clinic pathological feature. Expression of Cath-D and CD44 was detected in 52.1% and 41.7% respectively. The correlation was observed between Cath-D expression and metastasis (including lymph node metastasis and distant metastasis), but Cath-D expression was not associated with lymph node metastasis. Expression of CD44 was not associated with lymph node metastasis also. In addition, expression of Cath-D and CD44 did not correlated with clinical stage, histologic grade, size of tumor and postoperative 3 years survival rates. Our results indicate that Cath-D may play a role in metastasis.

24-hour gastroesophageal double pH monitoring acid and alkaline gastroesophageal and duodenogastric refluxes in pediatric patients.

OBJECTIVE: To study the pathophysiologic significance of gastroesophageal and duodenogastric-esophageal refluxes in pediatric patients. METHODS: Gastroesophageal double pH monitoring was performed on 68 children with gastroesophageal reflux (GER) diseases and 39 normal children. The pH shifts in the intra-gastric and esophageal lumina were recorded for a total of 24 hour period (P1) and for the period of gastric empty (P2) in supine and upright body postures. RESULTS: The following reflux types were identified: acid GER in 40 cases (58.8%), alkaline GER (AGER) in 8 cases (11.8%), mixed GER (MGER) in 14 cases (20.6%) and silent GER (SGER, i.e., reflux with normal pH values in P1) in 6 cases (8.8%) as well as duodenogastric reflux (DGR). The results showed a number of transient GER and DGR (19 of 39 children) in the control group, recorded mainly in the upright position at meal times and 2 hours postprandially. All pH variables for acid reflux were higher in the acid GER and MGER groups than those in the control group (P < 0.01). The alkaline episode was significantly noted in cases with AGER and MGER in the supine position during P2. There was no significant difference in terms of incidence of esophagitis between GER groups. The gastroesophageal double pH monitoring produces the higher positive diagnostic rate of 91.2% than single esophageal pH testing (79.4%). CONCLUSIONS: Transitory GER and DGR at meal times and 2 hours later might be a physiologic phenomenon. Acid and alkaline reflux occurring in the supine position during P2 should be considered of pathologic significance. This combined gastroesophageal pH monitoring is used not only to improve the diagnostic rate but also to guide clinicians to choose efficient anti-reflux therapy based on the type of reflux as well as to provide refined information for the further study of the pathophysiology of duodenogastroesophageal reflux.

[Observation on inhibitory effect of tetrandrine on the growth of human skin fibroblast in vitro].

The effect of tetrandrine on the growth of human skin fibroblasts was observed in vitro culture. The result revealed that tetrandrine significantly inhibited the growth of fibroblast. The higher the concentration of tetrandrine, the higher the inhibition rate, forming a time-dependent relationship. The possible value of tetrandrine in the treatment of hypertrophic scar is discussed.

[Efficient expression of multidrug resistance gene mdr1 in retroviral vector under control of an internal ribosome entry site].

OBJECTIVE: To test the efficiency of human mdr1 gene expression under the control of an internal ribosome entry site (IRES). METHODS: The expression retroviral vector Halmdr1 was constructed from pSXLC/pHa system which contains an IRES from encephalomyocarditis virus. The vector was introduced into ecotropic packaging cells GP + E86 by liposome-mediated transfection. The retrovirus producing cells were obtained by 50 micrograms/L vincristine selection. The success of transfer and expression of mdr1 gene were determined by polymerase chain reaction (PCR) and flow cytometry (FCM). RESULTS: Virus in the supernatant of the producer cells, in which the integration of mdr1 gene was confirmed by PCR, was titrated to 2.0 x 10(5) cfu/ml. The selected producer cells exhibited a 24-52-fold increase of resistance to vincristine, daunorubicin and taxol in comparison with control cells GP + E86. The expression of mdr1 gene was confirmed by both reverse transcription PCR at RNA level and FCM at protein level, although the HaImdr1 transfectants showed somewhat less expression of P-gp when compared to that with cap-dependent translation of Ha mdr1. CONCLUSION: mdr1 gene can be effectively translated and expressed under the control of IRES in cap-independent manner, it may be used as a dominant selectable marker in bicistronic vectors for gene therapy.

[Generation and prediction of hypoxemia in patients with chronic obstructive pulmonary disease].

OBJECTIVE: To study the pathogenesis of nocturnal hypoxemia in patients with chronic obstructive pulmonary disease (COPD). METHOD: Daytime pulmonary functions, arterial blood gases and nocturnal oxygen level were measured in 30 patients and compared with those of 8 normal subjects. RESULTS: The nocturnal oxygen level significantly decreased in patients with COPD compared with that in normal subjects, especially during rapid eye movement (REM) period. The decreased extents of nocturnal mean oxyhemoglobin saturation (MSaO2) and nocturnal minimum oxyhemoglobin saturation (MmSaO2) were also very high in COPD (P < 0.05). From the distribution of oxyhemoglobin, the percentage of the time that oxyhemoglobin saturation (SaO2) < 80% and < 90% during sleep (T80, T90) in patients were found higher than those in normal subjects (P < 0.05). The sleep time in which SaO2 below 90 percent obviously increased in COPD patients. The correlative analysis indicated that daytime SaO2 and FEV1% were related to the nocturnal mean oxyhemoglobin saturation (r = 0.79, 0.54). So predictive formula of nocturnal oxyhemoglobin level was derived as follows: MSaO2(%) = 1.03 x SaO2(%) + 0.19 x FEV1%(%) - 14.35(r = 0.83). Daytime SaO2 and PaCO2 were also found concerned with nocturnal oxygen distribution: T80(%) = -2.64 x SaO2(%) + 6.98 x PaCO2(kPa) + 211.31(r = 0.78), T90(%) = -4.49 x SaO2(%) + 437.18(r = 0.62). CONCLUSION: It was observered that hypoxemia became worse in patients with COPD than in normal subjects during sleep. The level and the distribution of nocturnal oxygen could be predicted by daytime SaO2 and PaCO2. All these indicated that nocturnal oxygen therapy could be useful to improve the outcome of the COPD patients.

[Low-concentration nitrous oxide inhalation in the treatment of high-altitude pulmonary edema].

OBJECTIVE: To investigate the therapeutic effect of low-concentration of nitroic oxide (NO) inhalation in high-altitude pulmonary edema. METHOD: Sixty-five male patients with high-altitude pulmonary edema were randomized into three groups. Patients in the conventional therapy group received oxygen, intravenous furosemide, aminophylline and dexamethasone; patients in the nifedipine group received oral nifedipine (10 mg, tid) in addition to conventional therapy; and patients in the NO group received NO (10 ppm) inhalation for 30 min, in addition to oral nifedipine. The time for which pulmonary rales on auscultation and shadows on chest radiograph lasted, and the course of disease, were compared. RESULT: In the NO group, pulmonary rales disappeared in 0.4 +/- 0.3 d, shadows on chest radiograph disappeared in 0.6 +/- 0.2 d, and the course of disease was 1.8 +/- 0.7 d, all of which were significantly different from those of the nifedipine group (2.4 +/- 1.4 d, 4.1 +/- 1.7 d, 6.8 +/- 1.8 d, respectively) and the conventional therapy group (3.7 +/- 1.2 d, 5.5 +/- 1.8 d, 9.6 +/- 3.1 d, respectively). CONCLUSION: Low-concentration NO inhalation on the basis of conventional and nifedipine therapies was very effective in the treatment of high-altitude pulmonary edema, which deserves further and larger scale investigation.
