[An experimental study on transplantation of frozen autogenous tooth-mandible compound].

OBJECTIVE: To study the feasibility of reserving the teeth in the transplantation of frozen autogenous mandible. METHODS: Forty adult Wistar rats were used for these operations of transplantation of frozen autogenous tooth-mandible compound (ATMC) and fresh ATMC. Then the healing process after transplantation of frozen ATMC and fresh ATMC was compared by clinical, radiographical, histological and microangiographical observation. RESULTS: There were no significant differences between the healing process of frozen ATMC and that of fresh ATMC, and the fibres of regenerated periodontal membrane which maintained the dental root in normal position could be imbedded in the newly formed cementum. CONCLUSION: It is feasible to transplant the frozen ATMC for reconstructing the mandible and improving its function in clinic.

[Study and application on the device of the drop volume of amniotic fluid in estimating fetal lung maturity].

A novel device of drop volume of amniotic fluid has been made to estimate fetal lung maturity. The design of the device is based on the principle that the surface tension of amniotic fluid is inversely proportional to the quantity of surfactant in the solution. The device measures the falling time of fluid drops from drop-tube and estimates the fetal lung maturity by using the method of comparison and electric-optic timing-counting. 148 samples of amniotic fluid were measured by the device and the data were compared with those obtained by biochemical method. The results demonstrate that this device has high stability and accuracy, and the rate of agreement between the two methods is as high as 93%.

[The effects of reactive oxidants on Bacillus thuringiensis parasporal crystals].

The effects of hydrogen peroxides(H2O2) and hydroxyl radicals(.OH) on the purified parasporal crystals from Bacillus thuringiensis var. Kurstaki HD-1 were studied by SDS-PAGE and bioassay. The results showed that to some extent H2O2 and .OH produced a direct and deleterious effect. Hydroxyl radicals from the system Fe(II)-EDTA-H2O2 indicated more serious damage than hydrogen peroxides. There was a 44.5% loss in activity against 12-h-old larvae of Heliothis armigera when parasporal crystals were treated with 0.3% H2O2 only. However, 85.2% activity lost when parasporal crystals were treated with 0.3% H2O2 in the presence of iron(II) and EDTA.

[Quantitative analysis of vocal fold vibration].

OBJECTIVE: To measure vocal fold vibration quantitatively and study the effects of gender and phonation mode. METHOD: The authors adopted a new computerized image analysis technique and EGG in 60 normal speakers, and the parameters of vocal fold vibration were measured and analyzed according to the loudness, pitch and gender. RESULT: With frequency increasing, CQ got smaller and OQ larger which expressed longer open phase. As intensity increased, the duration of open phase became shorter and the closing phase got relatively much shorter. Males had a longer closed phase than females. The closing duration appeared relatively shorter than opening duration in phonation. CONCLUSION: The quantitative measures of vocal fold vibration become feasible with modern computers and commercially available image analysis software. Certain features of vocal fold vibration vary with intensity variations, while others vary with gender or frequency variation.

[Efficient expression of human papillomavirus type 16 (HPV16) capsid proteins L1 and L2 in E. coli].

OBJECTIVE: To study the expression of HPV16 capsid proteins L1 and L2 in E. coli. METHODS: The plasmids pET-30a-L1 and pET-30a-L2 were constructed from the pET-30a vector, and transducted into the BL21. The fusion proteins 6 x His-L1 and 6 x His-L2 were expressed when induced by adding of IPTG. SDS-PAGE and Western blot analyses were used to detect the expressed proteins. RESULTS: Fusion proteins 6 x His-L1 and 6 x His-L2 were expressed efficiently in E. coli, the Mr were about 60,000 and 97,000 respectively, 6 x His-L1 degraded more than 6 x His-L2. CONCLUSION: L1 and L2 proteins were expressed at a high level in prokaryotic expression system, and 12 protein were more stable than L1 protein.

[Time-resolved fluorescence investigations of aggregation in Langmuir-Blodgett multilayers of hemicyanine dye].

UV-visible absorption and steady-state and time-resolved fluorescence techniques were used to investigate the aggregation behavior in Y-type hemicyanine Langmuir-Blodgett (LB) multilayers. Hemicyanine molecules formed H aggregates in LB films leading to blue-shifts of the absorption and fluorescence bands with respect to those of monomeric hemicyanine in chloroform solutions. The aggregates in LB films were dissociated by mixing with arachidic acid. The measurement of fluorescence decay profiles could be used to quantitatively characterize the lifetime, weights and size of aggregates.

[Study on laser flash spectroscopy of processes of photoionization of tyrosine: pH effects].

The processes of photoionization of tyrosine on three different pH value solutions were investigated by 248 nm laser flash photolysis. The results showed that the processes of photoionization of tyrosine were affected by pH value of the solution. With pH value increasing, the molecule number of photoionization increases via a monophotonic process, and decreases via a biphotonic process. Furthermore, the pH effect of photoionization of tyrosine resulting from deprotonation of phenolic hydroxyl group in alkaline solution was demonstrated.

The nuclear factor-kappa B RelA transcription factor is constitutively activated in human pancreatic adenocarcinoma cells.

Pancreatic adenocarcinoma is a leading cause of adult cancer mortality in the United States. Recent studies have revealed that point mutation of the K-ras oncogene is a common event in pancreatic cancer, and oncogenesis mediated by Ras may also involve activation of Rel/nuclear factor (NF)-kappa B transcription factors. Furthermore, the c-rel member of Rel/NF-kappa B transcription factor family was first identified as a cellular homologue of the v-rel oncogene, suggesting that other members of the Rel/NF-kappa B family are potentially oncogenes. We therefore investigated the possibility that Rel/NF-kappa B transcription factors are activated in pancreatic cancer. Immunohistochemical analysis, Western blot and Northern blot analysis, electrophoretic mobility shift assays, and chloramphenicol acetyltransferase assays were performed to determine RelA activity in human pancreatic adenocarcinomas and normal tissues and nontumorigenic or tumorigenic cell lines. RelA, the p65 subunit of NF-kappa B, was constitutively activated in approximately 67% (16 of 24) of pancreatic adenocarcinomas but not in normal pancreatic tissues. Constitutive RelA activity was also detected in 9 of 11 human pancreatic tumor cell lines but not in nontumorigenic Syrian golden hamster cell lines. I kappa B alpha, a previously identified NF-kappa B-inducible gene, was overexpressed in human pancreatic tumor tissues and cell lines, and RelA activation could be inhibited by curcumin and dominant-negative mutants of I kappa B alpha, raf, and MEKK1. This is the first report demonstrating constitutive activation of RelA in nonlymphoid human cancer. These data are consistent with the possibility that RelA is constitutively activated by the upstream signaling pathway involving Ras and mitogen-activated protein kinases in pancreatic tumor cells. Constitutive RelA activity may play a key role in pancreatic tumorigenesis through activation of its downstream target genes.
