[Infection of white spot baculo-like virus(WSBV) in Penaeus chinensis: evidence from electron microscopy and DNA hybridization].

Severe epizootic causing high mortalities in cultivated Penaeus chinensis has occurred every year in mainland China since 1993 with great economic loss. Naturally diseased P. chinensis sampled in Qingdao shrimp farming regions between 1993 and 1998 were examined by electron microscopy. A similar non-occluded nuclear bacilliform virus like the white spot baculo-like virus (WSBV) reported in Taiwan was mostly found in the gill, cuticular epidermis under the exoskeleton, stomach, lymphoid organ and other tissues of ectodermal and mesodermal origin from these diseased samples. The viron was enveloped with a mean size of (125 +/- 7.6) nm x (345 +/- 16) nm. Sometimes, a nipple-like appendage could be found protruding from one extremity of the viron. Envelopes were all lost after purification from cesium chloride gradient ultracentrifugation. The cylindrical nucleocapsid in negatively staining preparations was (80 +/- 13) nm x (380 +/- 24) nm with 13-16 stripes perpendicular to the longitudinal axis of the nucleocapsid. Based on its pathogenic and morphological characteristics, this virus should be related to WSBV. A pair of primers created from information of WSBV genome DNA SalI fragment produced a 355 bp band using the WSBV isolate from P. chinensis in mainland China, as the DNA template. The specific PCR product was cloned, sequenced and labeled with digoxigenin (DIG) DNA labeling kit (Boehringer Mannherm). The sequence of the probe is identical to that of WSBV. All of the sampled diseased shrimps between 1993 and 1998 reacted with the DIG-labeled probe by dot-blot hybridization and no hybridization was observed using DNA from health shrimp as templates. These results based on electron microscopy and DNA hybridization indicated that the shrimp epizootic occurred in mainland China was related to the WSBV infection previously occurred in Taiwan early 1992.

[Cloning of a new catechol 1,2-dioxygenase gene (tfd C) from Plesiomonas and its expression in the E. coli].

A new catechol-1,2-dioxygenase gene (tfd C) was cloned from the Plesiomonas using the PCR method. Primers were designed according to the reported sequence of Catechol-1,2-dioxygenase (C120) gene from Alcaligenes eutroplus. The amplified fragment contained a 765 bp open reading frame (ORF), encoding a protein of 255 amino acids. The new tfd C gene shared a high homology with the one cloned from Alcaligenes eutroplus, showing only one base difference at 693 site (C-->A) and consequently one amino acid difference at 228 site (P-->T). The ORF was cloned to the plasmid pBluescriptII KS, which was transferred to E. coli JM109 and a positive clone, pBt2G, was then selected. A significant activity of C120 was detected in the positive clone. When the ORF was cloned to the plasmid pET-30a, which was transferred to E. coli BL21(DE3) plysS, the expected 33 kD protein was detected from a positive clone, pET30A, by SDS-PAGE. The C120 is a key enzyme in degrading aromatic pollutants in the environment. In order to use plants to degrade aromatic pollutants, the gene will be introduced into the turfgrass. To express the gene properly in plants, its translation initiation codon was modified from GTG to ATG. A similar activity of C120 was obtained following the modification.

[The clinical value of laryngeal image analysis with two methods].

OBJECTIVE: To analyze normal and pathological glottal image quantitatively with computed image technique, and try to calculate the absolute value of the laryngeal parameter. METHOD: The glottal image parameters of 60 normal adults, 60 cases of vocal polyps and 40 unilateral vocal fold paralysis(UVFP) patients were measured and analyzed, including the relative and absolute maximum or minimum glottal area, that is Amax(r), Amin(r), Amax(a) and Amin(a) respectively. RESULT: Amin(r) of females was larger than that of males; and both were larger than zero. He and she were equal in Amin(a) and Amax(r), while Amax(a) of males was larger than that of females. Compared with normal adults, Amin(r) of the patients with vocal polyp increased, and Amax(r) of UVFP patients diminished, while their Amin(r) increased. CONCLUSION: Laryngeal image could be analyzed quantitatively with computer assisted fibrolaryngoscopy. Normal adults have incomplete glottal closure to certain degree, and the level is different between sexes. The patients with vocal polyp and UVFP have relevant pathological glottal image.

[The multicolour three-photon resonant ionization spectrum studies in uranium atom].

The multicolour three-photon resonant ionization spectra of 238U were measured by using the pulsed dye lasers system synchronously pumped by a frequency doubled Nd:YAG-laser 532 nm output(operated at 10 Hz), a device for atomic beam of U, time-of-flight mass spectrometer and boxcar integrator. The dye laser pulses have a 6 ns duration. Beams from the dye lasers, which have the same polarization direction and are focused by lenses, entered an interaction chamber through opposite windows on a common axis and spatially overlapped the U atomic beam. The optical pulse from dye laser DL2 was delayed to arrive at the interaction region 8 ns after the pulse from dye laser DL1; in the same way, the pulse from DL3 was delayed 8 ns after from DL2. The atomic beam device was made from stainless steel. We generated the U vapor by heating solid U in a graphite crucible by e-type electron-field on first excited states were studied in uranium atom. The question how to determine single-colour, two-colour and three-colour three-photon resonant ionization peak in the three-colour three-photon resonant ionization spectra diagram were solved.

[Photoluminescence characteristics of mixed aggregates in Langmuir-Blodgett multilayers].

Effects of the mixed aggregates of different active molecules and UV illumination on photoluminescence(PL) spectra of Langmuir-Blodgett multilayers were investigated. The strong interaction between hemicyanine and stilbazolium salt molecules in the mixed films caused a blue or red shift of the fluorescence band with respect to that of interleaving hemicyanine or stilbazolium salt/arachidic acid multilayers. UV illumination would dissociate the aggregates even more destroy the molecular structure, leading to a significant reduction of fluorescence intensity.

The effect of pH on the structure, binding and model membrane lysis by cecropin B and analogs.

Cecropins are a group of anti-bacterial, cationic peptides that have an amphipathic N-terminal segment, and a largely hydrophobic C-terminal segment and normally form a helix-hinge-helix structure. In this study, the ability of cecropin B (CB) and two analogs to lyse phospholipid bilayers, which have two levels of anionic content, has been examined by dye-leakage measurements over the pH range 2. 0-12.0. The two analogs differ from the natural peptide by having either two amphipathic segments (CB1) or two hydrophobic segments (CB3). All these peptides (except CB3 on low anionic content bilayers where it is not active) have maximal lytic activity on both types of bilayers at high pH. However, the pattern of secondary structure formation on these bilayers by the peptides, as measured by circular dichroism (CD), and the pattern of their ability to bind lipid monolayers, as measured using a biosensor, do not directly correlate with the pattern of their lytic ability. CB and CB1 with low anionic content bilayers have secondary structures as measured by CD with a similar pattern to membrane lysis, but binding is maximal near neutral, not high, pH. CB3 has some secondary structures on low anionic content bilayers at low pH and this becomes maximal over the basic range, but CB3 neither binds to nor lyses with these lipid layers. On high anionic content lipid layers, all peptides show high levels of secondary structures over most of the pH range and maximal binding at neutral pH (except for CB3, which does not bind). All three peptides lyse with high anionic content bilayers, but show no activity at neutral pH and reach maximal activity at very high pH. This work shows that pH is a major factor in the capability of antibacterial peptides to lyse with liposomes and that secondary structure and binding ability may not be the main determinants.

Peritransplant tolerance induction in macaques: early events reflecting the unique synergy between immunotoxin and deoxyspergualin.

BACKGROUND: Day of transplant T cell depletion with anti-CD3 immunotoxin or F(Ab)2 immunotoxin induces stable tolerance to renal allografts in rhesus monkeys given 15-deoxyspergualin (DSG), a NF-kappaB inhibitor that suppresses proinflammatory cytokine (PC) production. Because PC and NF-kappaB are involved in dendritic cell (DC) maturation, we asked if impaired DC maturation and Th2-type cytokine deviation might be related to the synergistic effect of DSG in this novel model. METHODS: Immunosuppression was initiated 4 hr before transplanting a major histocompatibility complex mismatched renal allograft. Some groups received a supplemental 5-day course of cyclosporine A or DSG or a 15-day course of DSG. Peripheral lymph nodes were sequentially examined for presence of mature DC. In vitro effects of DSG on PC-induced maturation of DC were also examined. RESULTS: Allografts survived without rejection in 87% of recipients given immunotoxin or F(Ab)2 immunotoxin with DSG x 15 days, in 50% with DSG x 5 days, and 0% with cyclosporine A. The longest DSG survivors are >1000 days with normal graft function and tolerance validated, including acceptance of challenge second donor kidneys without treatment. DSG-treated recipients were unique in developing polarized Th2-type plasma cytokines. In DSG recipients, mature DC were significantly reduced in day +5 lymph node biopsies, with complete repopulation by 30 days. In vitro studies verified an inhibitory effect of DSG on DC maturation. CONCLUSIONS: The study suggests DSG arrests DC maturation. The unusual synergy of immunotoxin and DSG apparently involves coincidental reduction in lymph node T cell mass and mature DC, a transient circumstance favoring development of stable tolerance.

Cutting edge: the HLA-A*0101-restricted HY minor histocompatibility antigen originates from DFFRY and contains a cysteinylated cysteine residue as identified by a novel mass spectrometric technique.

In this report, we describe the use of novel mass spectrometry instrumentation to identify a male-specific minor histocompatibility Ag restricted by HLA-A*0101 (A1-HY). This Ag has the sequence IVDC*LTEMY, where C* represents a cysteine disulfide bonded to a second cysteine residue. The core peptide sequence is found in the protein product of DFFRY, a Y chromosome gene not previously identified as the source of an HY Ag. The male-specific form of the peptide differs from its X chromosomal counterpart by the substitution of serine for the C* residue. Both peptides are expressed on the cell surface at 30 or fewer copies per cell. However, A1-HY-specific CTL recognize the DFFRY-derived peptide at a 1500-fold lower dose than the female homologue. Thus, these studies have identified a new source of HY epitopes and provide additional information about the influence of posttranslational modifications of class I-associated peptides on T cell recognition.

Alteration of hSNF5/INI1/BAF47 detected in rhabdoid cell lines and primary rhabdomyosarcomas but not Wilms' tumors.

The organization of genomic DNA into chromatin aids in the regulation of gene expression by limiting the access of transcriptional binding domains. The SWI/SNF family of chromatin-remodeling complexes, which are conserved from yeast to humans, open the chromatin to facilitate the transcriptional machinery to access their targets. The gene encoding the BAF47/hSNF5 subunit of the complex has been found mutated in both rhabdoid cell lines and in primary rhabdoid tumors. Since the pediatric tumors rhabdomyosarcoma (RMS) and Wilms' tumor (WT) share a similar genetic link with rhabdoid tumors, it was hypothesized that they may also show alterations of the BAF47 gene. Using primary tumors, the BAF47 protein was detected in all WT but less than 75% of the RMS tested. In cell lines, the BAF47 protein was missing in all rhabdoid cell lines and one RMS cell line. Analysis of sample DNA displayed either a mutation or deletion of the BAF47 gene in all samples negative for the protein. Several other subunits of the human SWI/SNF complex, including BRG1 which is the subunit directly interacting with the Rb tumor suppressor gene, were detected in all tumor samples. Alteration of BAF47 may be a genetic marker associated with the poor prognosis seen in all rhabdoid tumors but only some RMS.
