Neutrophil-derived eosinophil chemotactic factor production induced by immune complexes.

Immune complexes have previously been shown to be chemotactic for eosinophils in vivo. In this investigation, the in vitro chemotactic activity of human or guinea pig eosinophils was tested towards several types of immune complexes. Neither the immune complexes nor their individual components induced eosinophil migration. On incubation with purified human neutrophils, the immune complexes caused the release of a highly selective, low molecular weight (< 500 daltons) eosinophil chemotactic factor (ECF). Size and type of immune complexes influenced the amount of ECF released from the cells. The findings suggest that in vivo accumulation of eosinophils is not due to the immune complexes, but to the production of factors induced by them.

[Changes in diameters of small pulmonary venous and arterial vessels after nitroglycerin (author's transl)].

Diameters of the small arterial and venous pulmonary vessels were studied in 17 patients, using selective pulmonary angiography. The majority had coronary heart disease. Pulmonary and systemic hemodynamics were also evaluated. In 11 patients, these procedures were repeated 3 minutes after the sublingual administration of 1.6 mg of nitroglycerin (Group I). The remaining 6 patients served as controls (Group II). In Group I, pulmonary circulatory pressures, left ventricular filling pressure, and pulmonary vascular resistance declined significantly compared with Group II. The diameters of the small pulmonary veins declined by more than 10 percent in Group I while remaining constant in Group II. Differences between the two groups in terms of the response of the small arterial vessels was less evident. Increases in vessel cross-sectional area were observed despite considerably reduced intravascular pressure. The increase in elasticity and capacitance of the pulmonary vessels following nitroglycerin is more pronounced on the venous than on the arterial side. These findings suggest that the pulmonary vascular bed contributes significantly to the therapeutically important pooling effect caused by nitroglycerin in the treatment of angina pectoris and pulmonary edema.

[Microsurgical anastomoses between extra- and intracranial arteries (author's transl)].

86 patients underwent microanastomoses to branches of the anterior-, medial-and posterior cerebral arteries and the inferior cerebellar artery. Intraoperative electromagnetic measurement yielded a mean flow of 33 ml/min with a velocity of 11 to 54 cm/s. The brain circulation increased by a percentage of 38. In a percentage of 90 postoperative angiographic controls turned out anastomoses to be patent. By this surgical technique unilateral brain circulation can be improved significantly.

[Diagnosis and treatment of chylopericardium (author's transl)].

In a 22-year-old patient without clinical symptoms, a chylous pericardial effusion was found as cause of pericardial enlargement persisting over 7 years. Ultrasonic echocardiography, diagnostic pericardial puncture and the demonstration of chylomicrons enabled the diagnosis to be made. Lymphography showed lymphangiectasis of the thoracic duct. At operation two fistulous connections between pericardium and lymphatic system were found. Surgical ligature of the thoracic duct and the lymphatic fistulae and pericardial fenestration resulted in rapid normalisation of all findings.

Role of purified serum components in polymorphonuclear leukocyte chemotaxis.

Three highly purified serum proteins are examined for their role during leukocyte chemotaxis. In the absence of added proteins, migration of eosinophils towards eosinophil chemotactic factor and of neutrophils towards bacterial factor does not occur at all. Dose-response curves with human serum albumin (HSA) show that chemotaxis proceeds well after addition of HSA, with an optimum at 1 mg/ml. SHA has to be present in the buffer during chemotaxis since preincubation of cells followed by washing is ineffective. In contrast, alpha 2-macroglobulin and alpha 1-antitrypsin enhance leukocyte migration at doses close to physiological serum levels and affect cells irreversibly. The data suggest a complex and powerful modulating influence of serum proteins on the influx of eosinophils and neutrophils to tissue sites.

The action of 1,25 (OH)2D3 on turnover kinetic, remodelling surfaces and structure of trabecular bone in chronic renal failure.

Fibroosteoclasia as well as osteoidosis are reduced by 1,25(OH)2D3 treatment if secondary hyperparathyroidism preexists. Fibrosteoclasia completely disappeared after 6 months therapy in some cases. 1,25(OH)2D3 has no or only a very slight effect on the disturbed mineralization in type II of renal bone disorder (osteoidosis only, no signs of secondary hyperparathyroidism). The appositional rate of the osteoblasts increases under 1,25(OH)2D3 treatment if serum PTH values are raised. 1,25(OH)2D3 seemed to have, in the chosen dosage of this study, a self-limiting effect by reducing the bone-forming cells, i.e. the osteoblasts. This state already represents an overtreatment of the underlying bone disease.

The synthesis of glycosaminoglycans in aging rat liver. A brief note.

The synthesis of glycosaminoglycans (GAG) was studied in liver slices from postnatal (9 days), young (140 days), adult (490 days) and senescent (940 days) rats. It was found that the rate of synthesis was highest in postnatal rat liver and decreased to about half in young rats with no further reduction in adult and senescent age groups. The specific radioactivity of the precursors of GAG synthesis did not change with age. The synthesis pattern of specific types of GAG in postnatal liver was characterized by a significant higher percentage of chondroitin sulfate and hyaluronic acid. In the following age classes the profile of specific GAG synthesis did not change significantly (heparin sulfate: chondroitin sulfate" hyaluronic acid: "keratin sulfate" = 84%:8.3%:1.5%:1.6%).

HZI systems for EEG parametrization and classification of psychotropic drugs.

The EEG effects of twenty, clinically most frequently used psychotropic drugs and five placebos were studied in 75 male volunteers in five simultaneously designed basic studies. In each of the five studies single oral dosages of five drugs (well known representatives of neuroleptics, antidepressants, anxiolytics and psychostimulants, as well as placebos) were investigated in 15 subjects in a double-blind latin-square research design using the methods of the Quantitative Pharmaco-EEG. The results demonstrated that the therapeutically equivalent effective compounds also have similar effects on human EEG. With a classification rule, based on discriminant function 20, and with a classification rule, based on correlation statistics 19 of 25 compounds could be reclassified into correct clinical-therapeutic psychotropic drug groups. It is suggested that CEEG is an important tool in predicting and describing psychotropic properties of compounds, and should routinely be used in psychotropic drug development.
