[Familial occurrence of Elfin's face (Williams-Beurens Syndrome =wbs) and supravalvular aortic stenosis (= svas) (author's transl)].

Three siblings out of two families -- the mothers being sisters -- have signs of WBS: a girl shows the complete picture of elfin's face syndrome; a boy who needed surgical treatment for severe supravalvular aortic stenosis, presents only with minimal signs of the peculiar facies like his mother. His brother has slight supravalvular aortic stenosis. The three children described show trivial peripheral pulmonary stenosis. It is known that SVAS and WBS may show a dominant mode of inheritance with variable expressivity. The purpose of this study was to find signs which would prove a dominant inheritance in this kinship. Dental malpositions and coarse upperlip with shallow philtrum where the only symptomes found in the patients and their mothers. If we accept this traits as markers for the syndrome than we could assume a dominant way of inheritance.

Lymphocytes within glial cells ("emperipolesis") in a case of a granular cell tumor.

A case of a combined neurofibroma - granular cell tumor of the left middle cranial fossa with infiltration of the temporal lobe and expansion into the orbit is presented. The granular cell tumor shows big fibrillary astrocytes in its peripheral zone. Even in the tumor-free brain tissue adjacent to the tumor plenty of these obviously reactively proliferated astrocytes are found. Furthermore, lymphocytes within the cytoplasm of such astrocytes ("emperipolesis") strike the eye. Neither the lymphocytes nor the astrocytes show signs of damage. A review of the literature and the theories concerning the phenomenon of emperipolesis are given. The problem of its biological significance and the question whether emperipolesis is the result of invasion of engulfment must remain unsolved.

Granular cell tumor of the optic nerve.

A case of a combined neurofibroma-granular cell tumor is presented. The tumor had developed in the left middle cranial fossa after an intracranial trigeminal nerve operation 23 years before. It had displaced and infiltrated the temporal lobe and expanded probably via the fissura orbitalis into the orbit. The thickened optic nerve revealed a marked widening of the subdural fissure by typical granular cells. The cufflike tumor cell aggregation in the subdural fissure had compressed the optic fascicle, thereby inducing a demyelinisation and axonal degeneration. A reactive gliosis in the optic fascicle masks the typical pattern of the optic nerve atrophy. The gangliocytes of the third retinal neuron are atrophic and replaced by reactively proliferated fibrillary astrocytes. A short review of the literature and theories concerning histogenesis of the granular cell tumor (neurogenic or mesoderma) is given. An intermediary type of neurofibromatous and granular cells which we found motivates us to agree with a mesodermal origin.

Luxol-fast-blue-staining of nerve cell nucleoli as indicator for acidotic lesion in cases of intracranial hypertension?

The allochromatic behaviour of nerve cell nucleoli in the Luxol-fast-blue-method (blue nucleoli) in cases of raised brain pressure is probably due to a lactacidotic pH-shift basing on cerebral hypoxia. In 109 out of 134 cases with blue nucleoli signs of cerebral hypertension could be observed. The IEP of nerve cell nucleoli was determined in the nuclei pontis. Clinical and pathophysiological data were evaluated.

Chromosome analyses in man in the course of chemoprophylaxis against tuberculosis and of antituberculosis chemotherapy with isoniazid.

Cytogenetic analyses were carried out in lymphocytes of 37 persons before and during a primary chemoprophylaxis or a preventive chemotherapy with isoniazid as well as of 30 patients suffering from active pulmonary tuberculosis before and during a chemotherapy with isoniazid in combination with two other antimycobacterial drugs. The results of the four participating research groups consistently reveal no indication for a chromosome damaging activity of isoniazid, of its metabolites in the human organism and of the administered drug regimens.

Open chain crown-type polyethers and pyridinophane cryptands act as ionophores upon frog motor nerve and isolated rat heart cells.

Frog motor nerves and isolated heart cells from neonatal rats were incubated with solutions of open chain crown-type polyether or pyridinophane cryptand. The following alterations in membrane excitability and energy consumption were found: 1. The non-cyclic ligand stabilizes the resting potential of the frog nerve and reduces the pulsation rate of heart muscle cells. It is reversibly bound at the cell surface and does not affect the energy metabolism of the heart cells. (formula: see text) 2. The cryptand 1,12-dioxo-2,11-diaza-5,8,21,24-tetraoxa[12-8(2,11)] (2,6)-pyridinophane) ([2.2.1py]-diamide) is irreversibly bound by the tissues. It facilitates the depolarization of the nerve and shows a positively chronotropic effect upon the heart muscle cells. Single treatment of the cell cultures with 10 microgram [2.2.1py]-diamide per ml medium increased the activities of lactate dehydrogenase and of creatine kinase. When the cell cultures were treated three times at 24 h intervals with 10 microgram complexone/ml, the creatine kinase activity of the heart muscle cells decreased by about 40%. The physiological properties of the ligands are correlated with the stability of their alkali metal ion complexes and with the rate constants of complex formation. It is concluded that [2.2.1py]-diamide can act as a passive carrier for Na+ K+.

[Determination of trace elements in dietary fats and emulsifiers by flameless atomic absorption spectrophotometry. 1. Determination of the ionogenically bound heavy metals copper, iron, nickel, zinc, lead and cadmium in dietary fats].

The knowledge of the trace metal contents in dietary fats is of considerable interest from the viewpoints of food hygiene and and storage stability. In the framework of the present paper, it is dealt with the determination of the ionogenically bound heavy metals copper, iron, nickel, zinc, lead and cadmium by means of flameless atomic absorption spectrometry (AAS). Problems related to trace analysis in dietary fats are discussed. Problem-oriented surveys in the form of tables inform about:--analytical conditions of the AAS technique,--metal contents as compared to data with the approved standard method,--contents of the ionogenically bound toxic trace elements cadmium, zinc and lead in samples of various dietary fats currently used in the German Democratic Republic.

Differential inhibition of biphenyl hydroxylation in perfused rat liver.

A differential inhibition of biphenyl hydroxylation by alpha-naphthoflavone and metyrapone was observed in isolated perfused rat liver. alpha-Naphthoflavone inhibited 2- and 4-hydroxylation in livers from beta-naphthoflavone-pretreated animals but had no effect on both reactions in livers from phenobarbital-pretreated animals. Metyrapone inhibited 2- and 4-hydroxylation in phenobarbital-stimulated livers, but only insignificant inhibition of 2-hydroxylation and a slight enhancement of 4-hydroxylation by metyrapone was observed in beta-naphthoflavone-stimulated livers. Conjugation of 2-hydroxybiphenyl and 4-hydroxybiphenyl by isolated perfused livers was also studied. 4-Hydroxybiphenyl preferentially formed sulphates in livers from untreated animals but after induction glucuronidation was as effective as sulphation or even exceeded sulphation. Only glucuronic acid conjugates of 2-hydroxybiphenyl were detected.

A case of fatal cryptococcus meningitis with intraventricular granuloma.

Differential diagnostic difficulties in a case of probably spontaneous cryptococcus meningitis in a young girl are the reason for an extensive presentation of this case. The problems of CSF examination are discussed. In the course of the disease an intraventricular mycetoma had arisen, and caused the picture of a space-occupying process. Histological examination revealed an atypical reaction of the perifocal brain tissue, the possible causes of which are evaluated.

Supratentorial recurrences of gliomas. Results of reoperations on astrocytomas and oligodendrogliomas.

On the basis of our previous investigation regarding histological transformation of recurrent astrocytomas and oligodendrogliomas we report the clinical results of reoperations. The present observations deal with 121 cases: grade 1 astrocytomas (45), oligodendrogliomas (12), grade 2 astrocytomas (47), and oligodendrogliomas (17), respectively. In all these a second reoperation was performed in 14 cases. Operative mortality was relatively high, but without significant difference in the two groups of maligancy: 24.5% and 29.6%. Survival times in both groups showed striking individual differences. We achieved post-reoperation survivals exceeding one year in 18 and 14 cases, respectively. The results of second reoperations were generally poor. We find no remarkable differences in survival time following reoperations in primary grade 1 and grade 2 tumours. Similarly, malignant change proved to be not decisive for survival times. On the contrary, most of our patients with unchanged grade 1 astrocytomas, reoperated on after a longer interval, achieved a short second survival only. Prolongation of life expectancy by radiotherapy is doubtful. Irradiation seems to be necessary with malignantly transformed tumours if the patient did not have any treatment before reoperation.

The isolation of DNA from agarose gels by electrophoretic elution onto malachite green-polyacrylamide columns.

Electrophoretic elution of DNA coupled with direct adsorption onto malachite green-polyacrylamide columns was used to isolate double- and single-stranded DNA from agarose gels. Subsequently, DNA was eluted with a high salt buffer and filtered through Sephadex which permitted recovery of the DNA in a low salt buffer at concentrations suitable for heteroduplex analysis by electron microscopy. This method was tested by examining heteroduplexes formed from the isolated complementary single strands of T7 wild type DNA and a T7 deletion mutant. More than 80% of the reannealed molecules were intact heteroduplexes showing the deletion loop. Irradiation of single-stranded DNA with 254 nm light resulted in distorted, convoluted heteroduplexes while 366 nm light did not show this effect.
