OBJECTIVE: To investigate the histologic and histogenetic relationship between various types of rhabdomyosarcomas (RMS) and embryonal skeletal muscle (ESM) for further understanding of the histogenesis, classification and pattern of differentiation of RMS. METHODS: Fifty cases of variant types of RMS and 20 cases of ESM at different gestational ages were available. All specimens were stained with HE, PAS, Van Gieson, Masson, phosphotungstic acid hematoxilin and with antibodies for the demonstration of vimentin, desmin, HHF-35 and myoglobin by ABC method. RESULTS: The results showed that the order of positive expression and the intensity of positive reaction of the different immunohistochemical staining were consistent with the degree of differentiation of the tumor and the development of the ESM. It is obvious that each type of RMS is composed of tumor cells in different degree of differentiation and is derived from primitive mesodermal cells which are capable of potential differentiation towards mature skeletal muscles. CONCLUSIONS: Based on the results of this study, an ideal histologic classification of RMS should reflect not only the cell morphology and histologic structures but also the degree of differentiation of the tumor cells.
OBJECTIVE: To evaluate the magnesium and calcium concentration of peripheral serum and mononuclear in patients with pregnancy induced hypertension (PIH). METHODS: Twenty-six patients with PIH and 27 healthy women in their late pregnancy were studied. The serum and mononuclear cells magnesium and calcium contents were measured with atomic absorption spectrophotometry (fire atomization). RESULTS: When compared with the healthy women in their late pregnancy, (1) the peripheral serum magnesium and calcium contents in patients with PIH decreased significantly (P < 0.01 and P < 0.05), (2) the magnesium contents of the peripheral mononuclear cells in patients with moderate and severe degrees of PIH decreased significantly (P < 0.001 and P < 0.01), (3) the calcium contents of the peripheral mononuclear cells in patients with moderate and severe degrees of PIH decreased significantly (P < 0.01 and P < 0.05). CONCLUSION: The decrease of magnesium and calcium concentration may be one of the important factors responsible for the pathophysiologic changes of PIH.
OBJECTIVE: To evaluate the safety and efficacy of intravaginal misoprostol for cervical ripening in late trimester. METHOD: A randomized, double-blind, placebo-controlled trial was conducted. 85 patients with indication for induction of labor and unfavorable cervices were randomly assigned to receive either intravaginal misoprostol (100 micrograms) or placebo placed in the posterior vaginal fornix at one time. The Bishop score, fetal heart rate monitoring and Doppler blood flow velocity waveforms were measured before and 12 hours after drug insertion. Placenta and decidua were observed histopathologically in some cases. Among 85 patients enrolled, 43 received misoprostol and 42 received placebo. RESULTS: The mean initial Bishop scores were not significantly different between the two groups, while the mean Bishop score in the misoprostol group was significantly higher than that in the placebo group (4.4 for misoprostol versus 1.0 for placebo, P < 0.01). The prevalence of spontaneous labor within 12 hours after misoprostol insertion (67.4%, 29/43) was significantly higher than that in the placebo group (14.3%, 6/42) (P < 0.01). The average Doppler velocity systolic to diastolic (S/D) ratios of umbilical artery, middle cerebral artery, renal artery were not significantly different before and 12 hours after drug insertion between both groups. There was no significant difference in frequency of abnormal fetal heart rate tracings or fetal distress and in the mean Apgar score between the two groups. Except the presence of vasodilation in villi vessels in the misoprostol group, no significant difference existed between the two groups in the placental and decidual histopathological changes. CONCLUSION: Intravaginal misoprostol may be an effective and safe cervical ripening agent in late pregnancy.
OBJECTIVE: To study the expressions of angiotensin II 1A receptor (AT1A) both at the mRNA level and the protein level and their modulation by an angiotensin converting enzyme inhibitor, benazepril, in the renal tubulointerstitial injuries induced by unilateral ureteral obstruction (UUO) in rats. METHODS: Six SD rats (UUO-T) were administered benazepril in the drinking water (50mg/L). Additional 6 rats were used as untreated controls (UUO-C). Mean arterial Prossure (MAP) was measured at the 10th day after UUO, and then all animals were sacrificed. The expressions of AT1A were examined both at the mRNA level by in situ hybridization using a subtype-specific probe and at the protein level by an immunohistochemistry methods using AT1 receptor antibody in normal rat kidneys and the obstructed rat kidneys. RESULTS: MAP was 14.2 +/- 0. 6kPa in the UUO-T and 16.6 +/- 0.7kPa in the UUO-C (P < 0.05). Benazepril slowed the tubulointerstitial fibrosis (TIF) and reduced the expression of AT1A in the renal tubular epithelial cells, the interstitial areas, and the walls of renal arteriole in the UUO-T group. CONCLUSION: The increased expressions of AT1A were found in the acute renal tubulointerstitial pathogenesis induced by UUO. Benazepril may retard the progression of TIF and decrease the expression of AT1A in the obstructed kidneys. We conclude that the effects of Ang II on the obstructed kidneys may be due to its binding with AT1A.
OBJECTIVE: To investigate the impact of nitric oxide on endothelin-1 (ET-1) mRNA expression in pulmonary artery endothelial cells and pulmonary artery smooth muscle cells of chronic hypoxic rats. METHODS: In situ hybridization was performed on lung sections from 40 chronic hypoxic rats treated either with L-Arginine (L-Arg) or N omega-nitro-L-arginine methyl ester (L-NAME) by using cRNA probe for ET-1. RESULTS: Most intrapulmonary arteries had 1%-50% of the endothelial cells expressing ET-1 mRNA in both one-week and two-week hypoxic rats (75% +/- 3% and 71% +/- 6%, respectively), which was significantly inhibited by L-Arg but augmented by L-NAME administration. Most pulmonary artery smooth muscle cells showed no ET-1 mRNA signals in both one-week and two-week hypoxic rats (85% +/- 6% and 98% +/- 2%, respectively). However, L-NAME increased ET-1 mRNA expression in pulmonary artery smooth muscle cells of hypoxic rats. CONCLUSION: Nitric oxide inhibited ET gene expression in both pulmonary artery endothelial cells and smooth muscle cells of rats exposed to chronic hypoxia.
OBJECTIVE: To study the treatment results and prognostic factors of epithelial ovarian carcinoma. METHODS: Retrospective study of 155 cases of ovarian epithelial carcinoma admitted in our hospital since Jan. 1970 to Dec. 1992. All of these cases had postoperative pathological diagnosis and staging according to FIGO. 42 cases had second operations and 4 cases had the third. 149 cases had postoperative chemotherapies except for 6. Chemotherapies were also given to 32 cases after the second operation, while 9 for recurrence. RESULTS: The 2-year, 5-year, 10-year survival rates of stage I were 92.4%, 87.0%, 70.6%; and II 91.9%, 63.6% 47.8%; III 59.9%, 38.2%, 19.2% and IV 25.0%, 25.0%, 0.0% respectively. All 6 cases without chemotherapy after surgery died within 2 years. The results indicated that the prognosis of ovarian epithelial carcinoma was related to clinical stage, cellular grade and tumor residues. The 5-year survival rates for stage I was 87.0% and III 38.2% (P < 0.001); and cell G1 95.0% and G3 11.8% (P < 0.001), while no residual tumor 97.6% and with residual tumor > 2cm 21.2% (P < 0.001) respectively. CONCLUSION: Residual tumor of ovarian epithelial carcinoma < 2cm after primary surgery and chemotherapy right after surgery can improve survival rates.

OBJECTIVE: To investigate the nature of retinal capillary pericyte dropout in early human diabetic retinopathy. METHODS: In the present study, neural retinas of 12 postmortem eyes from 6 patients (3 diabetic and 3 nondiabetic) were first fixed in buffered formaldehyde and then digested with trypsin. The preparations of trypsin-digested retinal blood vessels were stained with terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) technique and then with perdiodic acid-Schiff technique (PAS), and counter-stained with hematoxylin to evaluate DNA fragmentation of cell nuclei and diabetic changes of retinal capillaries. RESULTS: TUNEL-positive apoptotic pericytes were observed in retinal capillaries in all three diabetic patients in this study. The TUNEL-positive pericytes were presented as darker stained cells with PAS-hematoxylin staining. A few TUNEL-positive retinal capillary endothelial cells were discovered in both eyes of one of the three diabetic patients. Neither TUNEL-positive pericytes nor positive endothelial cells were found in the three nondiabetic patients. A variety of typical diabetic microvascular changes were noticeable with PAS-hematoxylin staining in trypsin-digested retinal preparations from diabetic patients. These changes included thickened capillary walls, microaneurysms, "ghost-cell"-appearance of pericytes, acellular segments of capillaries, and a decreased ratio of retinal capillary pericytes to endothelial cells. All of these changes are characteristics of diabetic retinal microangiopathy. CONCLUSIONS: Retinal capillary pericyte apoptosis is a specific form of cell death which occurs during the early development of diabetic retinopathy. The significant decrease in the ratio of pericytes to endothelial cells and the predominant TUNEL-positive pericytes over endothelial cells suggest that a greater impact of apoptosis-related mechanisms occurs in pericytes than in endothelial cells and, thereby, a greater loss of pericytes takes place in early diabetic retinopathy. Apoptosis, therefore, is an important mechanism of pericyte dropout in the early diabetic retinopathy.
OBJECTIVE: To identify the astigmatism changes after excimer laser photorefractive keratectomy (PRK) and the visual influence of astigmatism. METHODS: 109 myopic eyes followed up for more than 1 year were analyzed by videokeratography, and their visual acuities were examined. Before the operation, the astigmatic errors with cycloplegic refractive examination were -1.00 approximately -2.00 D. RESULTS: The position of astigmatism axis was basically consistent with that in corneal topography, but in astigmatism diopter there were some differences between the cycloplegic examination and topographic analysis. A stigmatism was found in 62% of the eyes, asymmetrical in 33% and against-the-rule in 5% by using topographic analysis. There was little change observed in the position of astigmatism axis after PRK observed. Some changes of postoperative astigmatism diopter were seen in different periods. It increased in the postoperative 10 days or 1 month, and afterwards gradually decreased and became stable in 6 months to 1 year. The postoperative residual astigmatism was low in degree, and did not affect the visual acuity. The actually corrected diopter was within the anticipated corrected diopter range of +/- 1.00 D. accounting for 97.4%. CONCLUSION: By using spherical equivalent method of calculation in PRK, the refractive correction of the operation cases with myopia and low degree of astigmatism (< -2.00 D) was satisfactory.
OBJECTIVE: To investigate the effect of acoustic overstimulation on concentrations of cations in cochlear endolymph and analyze the relationship between the ionic changes in endolymph and the hearing loss. METHODS: The endocochlear potentials (EP), K+, Na+ and Ca2+ concentration in cochlear endolymph were examined in vivo for normal and 167 +/- 2 dB SPL impulse noise exposed groups of guinea pig bored on time course by means of double-barreled ion-selective microelectrodes. Brain stem auditory evoked potential (BAEP) was used to evaluate the auditory function. Ca(2+)-ATPase activity was demonstrated cytochemically in the lateral cochlear wall as induced by Ando et al with slight modification. RESULTS: The K+ and Ca2+ concentration exhibited significant changes in 8-hour groups (P < 0.05 for K+ and P < 0.01 for Ca2+). Then the K+ concentration was eventually resumed to the initial levels in accordance with the EP recovery in 7 days, while during the same period Ca2+ concentration was always significantly higher than that in control group (P < 0.01). The BAEP threshold shifts were correlated well with changes in ionic concentrations, especially Ca2+ (P < 0.001), in the endolymph induced by acoustic trauma. Although the normal positive EP was observed 7 days after noise exposure, the function of the vascular stria was not completely restored as revealed by the fact that the Ca(2+)-ATPase was diffused to the apical membrane surface. CONCLUSIONS: Endolymph compartment intrinsic mechanism for maintaining ionic composition is seriously deteriorated after high impulse noise stimulation. The changes of the unique environment of endolymph may play an important role in the mechanism of sensorineural hearing loss induced by acoustic trauma.
The infection rate and risk factors of CTRTI among 664 married women at reporductive age in rural areas in Chengjiang county, Yunnan province were investigated. Specimens of cervical swab were detected for CT antigen using Chlamydia EIA(Syva co., USA). The selection of major risk factors was through Logistic regression and chi 2-test. The results showed that the overall infection rate of CTRTI was 5.0%, with 60.6% of these cases asymptomatic. The major risk factors include: spouse who has symptoms of RTIs; history of others vaginitis; divorce; habit of washing face, feet and vulva with the same wash basin; age of menarche; amenorrhoea during lactation; low education and living in poverty.
Population-based survival and relative survival of primary liver-cancer (PLC) were analyzed for 11,200 cases in an area with high-incidence, Qidong, Jiangsu Province, during 1972 to 1991. Results showed that observed survival was 2.1% and 2.0% in the first (1972-1981) and the second ten years (1982-1991), respectively, and relative survival 2.2% and 2.3%, respectively. Females had more favorable survival than did males, with five-year survival of 3.5% and 1.7% in the first ten years, respectively, and 2.6% and 1.8% in the second ten years, respectively, but without statistical significance in their difference. No significant difference in age groups was found during 1982 to 1991, with five-year survival of 2.2%, 1.9%, 1.3%, 2.2% and 1.5% for males, and 3.7%, 3.1%, 2.8%, 2.1% and 2.6% for females, and with five-year relative survival of 2.2%, 1.95%, 1.4%, 2.4% and 1.9% for males, and 3.7%, 3.1%, 2.9%, 2.2%, and 3.0% for females, in 0-34, 35-44, 45-54, 55-64 and 65-over age groups, respectively. It indicated that prognosis of PLC was poor in Qidong, and it is urgent to screen for PLC in high risk population, to detect it earlier, to manage it with adequate measures, and to improve its survival.

Cellular interleukin-10 (cIL-10) has been shown to inhibit cytokine production by T helper type 1 (Th1) cells by blocking antigen presenting cell function. This activity has suggested that IL-10 might be useful in the treatment of transplant rejection. Stimulatory effects of IL-10 however, have also been observed both on T and B cell differentiation. In this study, we examined the influence of recombinant (r) mouse (m) IL-10 on heterotopic vascularized heart allograft survival in the B10(H2b)-->C3H(H2k) strain combination that crosses both major histocompatibility complex (MHC) and non-MHC-histocompatibility antigen (non-MHC-HA) barriers. The influence of IL-10 was also examined in the B10.BR (H2k)--> C3H combination with disparity at only non-MHC-HA loci. Postoperative intraperitoneal administration of IL-10 (100 microg/d, days 0-6) significantly accelerated heart graft rejection both in the B10-->C3H (mean survival time [MST] 7.8+/-0.2 days; control MST 10.6+/-0.6 days; P<0.05) and the B10.BR-->C3H combination (MST 14.3+/-0.5 days; control MST 77.7+/-14.4 days). Ex vivo IL-10 perfusion of donor hearts for either 15 min or 2 hr did not affect subsequent graft survival. Immunologic monitoring of transplanted mice revealed that IL-10 treatment (100 microg/d, i.p., days 0-6) increased both the circulating complement-dependent cytotoxic (CDC) antibody titer and splenic anti-donor cytotoxic T lymphocyte (CTL) activity measured up to 3 weeks posttransplant. These findings indicate that post transplant systemic administration of cIL-10 can promote vascularized allograft rejection, and that this may reflect stimulation both of B and T cell alloimmune responses.

Protein kinase C-delta (PKC-delta) has been demonstrated to be phosphorylated on tyrosine residue(s) in many different biological systems (Li, W., Yu, J.-C., Michieli, P., Beeler, J. F., Ellmore, N., Heidaran, M. A., and Pierce, J. H. (1994) Mol. Cell. Biol. 14, 6727-6735; Li, W., Mischak, H., Yu, J.-C., Wang, L.-M., Mushinski, J. F., Heidaran, M. A., and Pierce, J. H. (1994) J. Biol. Chem. 269, 2349-2352; Denning, M. F., Dlugosz, A. A., Howett, M. A., and Yuspa, S. H. (1993) J. Biol. Chem. 268, 26079-26081). Tyrosine phosphorylation of PKC-delta has also been shown to occur in vitro when purified PKC-delta is coincubated with different tyrosine kinase sources. However, the tyrosine phosphorylation site(s) is currently unknown and the exact effect of this phosphorylation on its serine/threonine kinase activity and biological functions is still controversial. To directly investigate the potential role of PKC-delta tyrosine phosphorylation, tyrosine 187 was converted to phenylalanine (PKC-deltaY187F) by site-directed mutagenesis, and expression vectors containing PKC-deltaY187F cDNAs were transfected into both 32D myeloid progenitor cells and NIH 3T3 fibroblasts. The results showed that tyrosine 187 of PKC-delta became phosphorylated in vivo in response to 12-O-tetradecanoylphorbol-13-acetate stimulation or platelet-derived growth factor receptor activation. In vivo labeling and subsequent two-dimensional phosphopeptide analysis demonstrated that one phosphopeptide was absent in PKC-deltaY187F when compared to wild type PKC-delta, further substantiating that tyrosine 187 of PKC-delta is phosphorylated in vivo. Although the phosphotyrosine content of PKC-deltaY187F was reduced compared with PKC-deltaWT, the kinase activity of PKC-deltaY187F toward a PKC-delta substrate was not altered. Moreover, 12-O-tetradecanoylphorbol-13-acetate-mediated monocytic differentiation of 32D cells was not affected by expression of the PKC-deltaY187F mutant. Taken together, these results suggest that tyrosine phosphorylation of PKC-delta on 187 may not influence PKC-delta activation and known functions.
