Morphological studies on the muscle cysts of Sarcocystis dirumpens (Hoare 1933) Häfner and Matuschka 1984 in several host species revealing endopolygeny in metrocytes.

Muscle cysts from rodents (Mastomys natalensis, Mus musculus, Rattus norvegicus, Meriones unguiculatus, Phodopus sungorus) experimentally infected with Sarcocystis dirumpens were examined by light and electron microscopy. The corrugated primary wall showed a pattern of densely arranged invaginations surrounded by minute bleb-like evaginations. True protrusions were absent. The length of the blebs and the thickness of the primary wall varied insignificantly but not remarkable host-dependent morphological differences could be noticed between the cyst wall structures 60-418 days p.i. In the mature parts of the cysts merozoites and metrocytes showed typical apicomplexan features of fine structure and mode of multiplication (endodyogeny). In the tips of the cysts large metrocytes simultaneously formed more than two daughter cells (at least up to 12) by endopolygeny.

[The accuracy of ultrasound in the diagnosis of intra-abdominal abscess formations].

During the last two years 73 patients with a total number of 84 abdominal abscesses have been investigated by ultrasound. Two thirds of the patients (67%) had postoperative abscesses. Spontaneous abscesses occurred in the remaining 33%. Multiple abscesses occurred in the postoperative group only (7 patients). The overall sensitivity in detecting an abscess was 84%. The sensitivity was much better with spontaneous abscesses (92%) than with postoperative abscesses (80%). One third of the patients have also been investigated by computed tomography. The diagnosis was confirmed either by needle aspiration or by operative drainage. Ultrasound was most useful in detecting abscesses in the right and left upper quadrant and in the pelvis, while CT was superior in the mid-abdomen and when dealing with multiple abscesses.

[Hygienic problems of keeping pets in the Federal Republic of Germany].

Pet animals i.e. vertebrates from fish to mammals are often suspected to transmit germs of various systematic position (viruses, bacteria, fungi and parasites) to human beings. A critical analysis shows that only a very limited number of such organisms can be transmitted from animals to man. Zooanthroponoses with pet animal origin is a very rare phenomenon. Bacteria like salmonellae which were incriminated to be the cause of about 250,000 infections of man per year, particularly of children, in the USA in the early 70th have really no importance in the Federal Republic of Germany. The statistical dates published every year by the "Statistisches Bundesamt" show clearly that only about 1 case in a year can be traced backwards to reptiles in spite of about 50,000 registered cases of human salmonellosis per year. If we compare the number of about 1 million of reptiles in captivity the incidence is very low. Similar relations exist for other germs, too. As an example the choriomeningitis of golden hamsters is often mentioned. In newspapers and other popular publications this infection is said to be extremely dangerous for children but in reality only a few cases were registered years ago. Nowadays precautions in the breeding farms etc. make it very unlikely that such virus transmissions have any practical importance. This situation is comparable with mycosis caused by fungi like Trichophyton which have their origin in certain mammals. Also psittacosis--in form r times not infrequently the cause of sickness and death in man--nowadays is only found sporadically in man, in spite of about the same number of cases in parakeets and other birds. The terrible risk of the disease can be prevented since tetracycline therapy is a successful treatment. It can be stated that zooanthroponoses which have their origin in pet animals is a very rare manifestation, but often misinterpretations are given either ignoring the knowledge of a certain infection or fishing for publicity! One of such examples is toxoplasmosis. About 15 years after the discovery of the life cycle of the protozoon Toxoplasma gondii and the experimentally proven fact that oocysts can be produced only in cats, and therefore transmission to humans is only possible with these stages other animals like rabbits or guinea pigs are still incriminated as the cause of human infections. Apart from the discussed problems and the possibility of transmission of infectious agents from animals to man we must also take into consideration the positive psychological aspects of animal keeping for the hobbyist, and particularly for children.(ABSTRACT TRUNCATED AT 400 WORDS)

[CT of the sella following hypophyseal surgery].

A CT control scan of 32 patients after transsphenoidal resection of a pituitary tumour revealed sellar contents of different density and varying extent. Additional criteria were used to differentiate the intrasellar structures (size, extent, shape, structure, contrast enhancement, bone changes). CT made the definite diagnosis in 6 patients with pituitary tumour recurrence. Hormonal measurements were superior to CT in identifying recurrent tumours in another 3 patients. It is concluded that hormonal methods should be used in evaluating patients after surgery of a hormone-producing tumour. However, CT is the most useful method in detecting tumour recurrence in patients with non-functioning adenomas, and, if an adequate technique is used (coronal scanning, iv-contrast), even clinical asymptomatic tumours can be identified.

[Otologic manifestation of Wegener granulomatosis].

Ever since Wegener first described the granulomatosis named after him, involvement of the middle ear tracts as a consequence of inflammatory changes in the nose and the nasopharynx has been known. In recent years there have been a remarkable number of reports on a primary affection of the mucosa of the middle ear with Wegener's granulomatosis. The corresponding symptoms clearly precede the affection of the typical parts of predilection in the upper respiratory tract. A 37-year old woman patient is reported to have fallen ill with a primary left-sided serous otitis media. Only several months later the changes typical of Wegener's granulomatosis concerning the nasal mucosa, trachea and especially the kidneys set in. The differential diagnostic difficulties appear with respect to the classification of inflammatory granulomatosis of the mucosa of the middle ear. The now valid concept of chemotherapy regarding treatment with cyclophosphamide and corticosteroids. Premature therapy can achieve encouragingly long remission of a relatively high number of cases of this disease.

Host specificity and host range of the genus Sarcocystis in three snake-rodent life cycles.

Three Sarcocystis species with snake-rodent life cycles were studied for their host range and host specificity in systematically related intermediate and definitive hosts. While S. singaporensis and S. villivillosi developed only in murids closely related to the genus Rattus, a third Sarcocystis species (natural definitive host: Bitis nasicornis - syn. Isospora dirumpens ?) showed a broad intermediate host range. This species was found to use several Bitis species as definitive hosts. S. singaporensis and S. villivillosi in contrary produced sporocysts only in three Python species and in Aspidites melanocephalus .

The independence of insulin release and ambient insulin in vitro.

The effect of insulin on its own secretion was tested in three independent experimental models using insulin concentrations that approached physiologic values. The collected secretions from glucose-stimulated islet tissue had no effect on insulin release from other islets. Perifused insulin had no effect on the release of endogenous insulin even when the assay was completely controlled for dilutional effects. Perifused insulin had no effect on the release of prelabeled insulin from glucose-stimulated islets. Similarly, proinsulin did not affect insulin release. Porcine insulin did not affect the function of porcine or canine islets. These studies strongly support the independence of glucose-stimulated insulin secretion and ambient insulin.

Ultrastructural modifications in Heterakis spumosa after treatment with febantel or mebendazole.

The effects of the anthelmintics febantel and mebendazole on Heterakis spumosa were investigated by means of transmission and scanning electron microscopy. Ultramorphological changes were only seen in the intestinal cells. Microtubules disappeared and granules were absent from the terminal web 12 h after treatment with either febantel or mebendazole, and up to 72 h after treatment, the number and size of autophagic vacuoles increased. Moreover the parasites became sluggish and were frequently rolled up like spiral springs. Destruction to microtubules and neurotoxic influences are discussed as possible modes of action.

Besnoitia in a palaearctic lizard (Lacerta dugesii) from Madeira.

Besnoitia cysts in the heart of a lizard (Lacerta dugesii) from the islands of Madeira are the first record of besnoitiosis in a poikilothermic animal in the Old World. The size of the cysts corresponds to those found in lizard genera (Basiliscus and Ameiva) from Panama which belong to B. darlingi. Up to now nothing is known concerning the life cycle of this species from Madeira but it seems possible that cats function as definitive hosts as well as in the other species.

Increased phosphatidylinositol synthesis in rat embryo fibroblasts after growth stimulation and its inhibition by delta-hexachlorocyclohexane.

When resting rat embryo fibroblasts are stimulated to grow, a substantial increase in phosphatidylinositol synthesis can be observed. This increase cannot be explained by increased glucose uptake or glycolysis. delta-Hexachlorocyclohexane having the same configuration as myo-inositol, inhibits phosphatidyl inositol synthesis as well as DNA synthesis and mitosis, but has no effect on phosphatidyl choline synthesis. When delta-hexachlorocyclohexane is added to fibroblast cultures during the first hours after stimulation, a delay of DNA synthesis and mitosis compared to uninhibited cultures can be observed. Since delta-hexachlorocyclohexane also inhibits the uptake of nucleotides, hexoses and amino acids, it is suggested that phosphatidylinositol is necessary for the proper functioning of those receptors and carriers which are an essential part of the early cellular processes after growth stimulation, and this role of phosphatidyl-inositol may explain its increased turnover in growing cells. The increased phosphatidylinositol synthesis could not be associated to one of the subcellular fractions. When cells were labeled with [32P]orthophosphate during the first 10 min after growth stimulation and were subsequently separated into cellular fractions such as nuclei, mitochondria, plasma membranes and microsomes, no significant differences in radioactivity of phosphatidylinositol among those fractions could be observed.
