[A case report of welder's lung].

The case of a 57-year-old man is reported, who worked as a plasma welder about 15 years. Chronic recurrent bronchitis was diagnosed 12 years after beginning of this work. The patient changed the place of his employment but the course of disease was uncommonly progressive. He died of right ventricular heart failure 10 years later. One year before death, the chronic obstructive pulmonary lesions were acknowledged as occupational disease. The post-mortem histological and chemical analysis of lung tissue yielded a mixed dust pneumoconiosis. These findings supported additionally the diagnosis of occupational disease.

[Urine flow measurement in pregnancy and the puerperium].

To elucidate effects of gestation on micturition, 337 standardised urine flow measurements with weight transducers were performed on 296 women in normal pregnancy (n = 128), under oral or intravenous tocolysis (n = 103), after spontaneous delivery (n = 20) or obstetric surgery (n = 26) as well as on non-pregnant probands (n = 19) with clinically intact urinary tract for comparison. Average micturition frequency was calculated, as well (n = 86). However, only general trends were discernible from mean value calculations because of wide scatter of uroflow parameters: In normal pregnancy, significant increase in flow rate was observed only in the second trimenon. Major opposite effects were recordable from beta-adrenergics (Fenoterol) only in intravenous tocolysis. Micturition frequency was at its highest level towards the end of pregnancy. Post-partum, only flow rates after forceps delivery were worse than those in the non-pregnant control group, on the other hand after spontaneous delivery flow rates were even better than in the controls. Yet, even significant differences stayed within normal range, so that in general severe disadvantages should not be expected from voiding alterations during pregnancy. Uroflow measurements and frequency/volume charts are not required in the care of pregnant women unless conspicuous findings are derived from analyses of micturition habits, types, and frequencies.

[Examination of cattle with respiratory diseases for Mycoplasma and bacterial bronchopneumonia agents].

A total of 247 mycoplasma strains was isolated from 435 lungs, tracheobronchial secretions and nasal swabs originating from cattle with symptoms of bronchopneumonia. Mycoplasma (M.) bovis was found 89 times (36%) and was the most common mycoplasma species in the lungs. M. bovirhinis, M. bovigenitalium, M. spec. and Acholeplasma (A.) laidlawii were isolated 158 times (64%). Among these mycoplasmas M. bovirhinis was the most widespread species (114 isolations). In 55 cases (62%) M. bovis was associated with Pasteurella or Actinomyces (A.) pyogenes. The other mycoplasma species were found in 67 cases (42%) together with these bacteria. Without mycoplasmas Pasteurella and A. pyogenes occurred in 33 of the probes investigated (21%). Beside mycoplasmas Haemophilus (H.) somnus was isolated from 16 of 162 tracheobronchial secretions investigated. The results confirm earlier suppositions that in most of the cases bronchopneumonia of cattle is a multifactorial event, frequently associated with mycoplasmas--especially M. bovis.

Distribution analyses of chain substituents of lipoteichoic acids by chemical degradation.

The lipoteichoic acid from Lactococcus lactis Kiel 48337 was analyzed. It had 61% of its glycerophosphate residues substituted with alpha-D-galactopyranosyl residues. Non-substituted glycerophosphate residues were split off by two alkaline hydrolyses and an intermediate enzymatic phosphomonoester cleavage. The resulting (GalGroP)nGroGal and (GalGroP)nGlc2Gro oligomers were separated by chromatography on DEAE-Sephadex into 10 pairs of molecular species with n from 1 to 10. The relative frequencies of GalGro and these oligomers were close to the values calculated by computer simulation for a random distribution of chain substituents. A similar series of oligomers was obtained in one step by hydrolysis of the lipoteichoic acid with 98% (by vol.) acetic acid. Due to side reactions, the picture was less precise but nevertheless indicative of the same distribution pattern. The data provide indirect evidence that the alanine ester substituents of the native lipoteichoic acid (Ala/P = 0.38) occupy the free positions between the galactosylated oligomers and are therefore themselves distributed randomly.

Rapid myosin phosphorylation transients in phasic contractions in chicken gizzard smooth muscle.

In intact smooth muscle strips from chicken gizzard, electrical stimulation and carbachol elicited brief, phasic contractions which were associated with a very rapid, transient phosphorylation of the 20 kDa myosin light chains. The phosphorylation transients reached their peak after 3 s and 6 s and preceded that of force. Phosphorylation was not significantly different from basal levels after 10 s and 30 s while force still amounted to 50% of the peak value. The rate of tension decline could be increased by cessation of stimulation or by addition of atropine, even at apparently basal phosphorylation levels suggesting a phosphorylation independent regulation.

NGF receptor reexpression and NGF-mediated cholinergic neuronal hypertrophy in the damaged adult neostriatum.

Adult cholinergic interneurons of the neostriatum are not immunoreactive for monoclonal antibody to NGF receptor, whereas the developing neostriatum is immunoreactive for this same antibody. Chronic NGF infusion into the adult neostriatum resulted in reexpression of the NGF receptor such that many cholinergic interneurons became immunoreactive for NGF receptor. NGF infusion dramatically increased the size and choline acetyltransferase immunoreactivity of these same cholinergic neurons. Additionally, in situ hybridization demonstrated an increase in the number of cells expressing NGF receptor mRNA in the NGF-infused striatum. These findings indicate that central cholinergic neurons which lose their NGF receptors during postnatal development will resume their NGF responsiveness when the tissue is damaged. Such a damage-induced mechanism may act to enhance the action of trophic factors, including NGF, released at the site of injury and enhance the responsiveness of damaged CNS neurons to exogenously administered trophic factors.

[Conservative treatment of calcaneus fracture versus repositioning and percutaneous bore wire fixation. A comparison of 2 methods].

From 1976 to 1985 we treated 236 patients with fractures of the calcaneum without additional injuries to the legs or pelvis. Among 129 patients, 22 female/107 male, with an average age of 50 and 42 years, respectively, we performed a total of 141 fractures of the calcaneum. The average post-traumatic follow-up was 6 years. For 62 fractures we performed conservative therapy with immobilization in plaster and for 79 fractures, reduction and percutaneous drill wire fixation. The fractures were classified according to Boehler, Watson Jones, and Essex Lopresti, and on the basis of the tuber joint angle (Boehler's angle). The resulting groups, however, were not homogeneously structured. Of 82 intraarticular fractures with fragment dislocation, 19 were treated conservatively and 63 surgically. Boehler's angle at trauma was 14 degrees in the conservative group and 2 degrees in the surgical group. The more severe types of fractures were thus treated by surgery. There were two Sudeck's dystrophies with conservative and three with surgical treatment. Inadequate surgical technique (drill wires extending greater than 2 mm over the bone and insufficient compression) led to the following complications: 10 drill wire perforations without subsequent complications, 8 drill wire migrations, 2 deep infections. In four patients drill wires had to be shortened, and in 1 patient drill wires had to be shortened, and in 1 patient drill wires were misplaced and had to be corrected. Surgically treated fractures were followed by arthrodesis in 2 patients, medialization of the lateral wall of the calcaneum in 1 patient, and peroneal tendon revision in 1 patient.(ABSTRACT TRUNCATED AT 250 WORDS)

[Morphological and immunohistochemical investigation of serotonergic neurons in the dorsal raphe nucleus of Wistar rats].

The serotonergic neurons of the nucleus raphe dorsalis (NRD) of the adult male Wistar rat were characterized morphologically using Nissl and Golgi-Kopsch techniques, fluorescence histochemistry as well as a peroxidase-antiperoxidase (PAP) immunocytochemical method employing specific antibodies to serotonin. 1. The numerous serotonin-positive neurons within (and around) the NRD exhibit--in agreement with data of the literature--characteristic heterogeneous distribution patterns both in rostro-caudal direction and also dorsoventral particularly in the mesencephalic part, the zone of the largest extension of the nucleus. Several groups with high cell concentration could be divided. The neurons differ in their immunoreactivity. 2. Based on different criteria, especially the number and arrangement of dendrites as well as the cell shape, three types of serotonergic neurons could be identified: multipolar, fusiform (bipolar) and small ovoid ones. A parallel analysis in Golgi-stained material offers the possibility for more detailed studies and further characterization. 3. With respect to functional interpretations some morphological peculiarities seem to be remarkable, e.g. the close relation between several neurons, the tight dendritic contact to many blood vessels or the formation of a dense serotonin-positive fiber plexus along the floor of the aqueductus and fourth ventricle. Studies, following stereotactic application of the neurotoxin 5.7-dihydroxytryptamine into the NRD, show furthermore that the majority of serotonergic neurons--independently of the cell type--reveals signs of cell degeneration. The findings presented confirm previous studies and should provide also further insights into the morphology of the 5-HT neurons of rat NRD as well as for the explanation of functional aspects.

Granulocyte-related bioincompatibility of hemodialysis: inhibition of oxidative metabolism, degranulation reaction, enzyme release and leukocyte sequestration in the lung.

Hemodialysis (HD) induced contact of granulocytes with cuprophan and modified cuprophan, i.e. hemophan, membranes leads to over-stimulation of their oxidative metabolism. This is shown by the fact that after starting HD the peripheral granulocytes are less stimulable to chemiluminescence (CL) and peroxide production (cytochrome C reduction). Such alterations are likely to be connected with a granular staining loss, enzyme release and sequestration of granulocytes in the lung, which coincides with reported clinical features like dyspnoea and drop in arterial oxygen pressure. HD with polysulfone membranes causes a degranulation reaction too, but does not produce granulocyte inhibition and pulmonary sequestration of leukocytes, clearly indicating that these phenomena strongly depend on the different dialyzer membranes.

[A simple method for the isolation of GFAP and its use for the study of brain tumors].

A simple method is described in this paper for the production of a polyclonal antiserum against GFAP. The antiserum was tested on 212 primary brain tumours which had been selected from biopsy and autopsy material of the Institute of Pathological Anatomy at the Medical Academy of Erfurt, GDR. 52 of 81 astrocytomas (64%) and 26 of 47 glioblastomas (55%) gave GFAP-positive results. GFAP-negative responses were primarily recorded from tumours with severe anaplasia. GFAP was found in all 22 ependymomas tested. Epithelioid ependymomas, however, exhibited lower immunological reactions than tanycytic variants. Isomorphic oligodendrogliomas, meningiomas, medulloblastomas, and brain metastases of carcinomas were GFAP-negative. The possibility is discussed in some detail of falsely negative results on account of too little biopsy material or insufficient fixation of tumour tissue.

Alterations of cellular lipids in rat thymocytes during cell cycle progression.

This study provides information on the lipid content and composition of rat thymocytes during cell cycle progression. Cultured rat thymocytes were stimulated by concanavalin A and interleukin-2 to enter the cell cycle and to proliferate. At indicated intervals during a 96 h culture, cells were analyzed for lipid content and lipid synthesis. A growth-related 3.8-fold increase in the cellular cholesterol and a 2.8-fold increase in the phospholipid content was observed after 48 h of culture. Cholesterol synthesis, measured by [14C]acetate incorporation, increased 34-fold, whereas the synthesis of the other lipids was enhanced only 15-fold. Concerning the percentage distribution of the total fatty acids, arachidonic acid showed a significant decline between 24 and 72 h of culture, being mainly caused by the release of arachidonic acid from phosphatidylcholine. Addition of actinomycin D, an inhibitor of DNA replication, prevented the decline of cellular arachidonic acid content in a dose-dependent manner. These data suggest that both the pronounced rise of the cholesterol content as well as the decline of arachidonic acid content are closely linked to cell proliferation.
