The BTB domain of bric à brac mediates dimerization in vitro.

The gene bric à brac (bab) is required for the proper development of the limbs and ovary in Drosophila melanogaster. bab encodes a BTB domain (also called a POZ domain), an approximately 115-amino-acid conserved motif found primarily in the N termini of zinc finger proteins. In this paper, we show that the BTB domain of bab can mediate protein dimerization in vitro. In addition, we demonstrate that the first 51 amino acids of the bab BTB domain are sufficient for dimerization, and we identify amino acids within this region that are required for binding.

Quantitation of hepatic glucose fluxes and pathways of hepatic glycogen synthesis in conscious mice.

Mice were studied with the euglycemic hyperinsulinemic and the hyperglycemic clamp techniques after a 6-h fast: 1) euglycemic (6.7 +/- 0.2 mM) hyperinsulinemia (approximately 800 microU/ml); 2) hyperglycemic (15.3 +/- 0.4 mM) hyperinsulinemia (approximately 800 microU/ml). All mice received an infusion of [3-3H]glucose and [U-14C]lactate. Basal hepatic glucose production (HGP) averaged approximately 170 mumol.kg-1.min-1 in both groups. During euglycemic and hyperglycemic hyperinsulinemia, HGP decreased by 53% (to 76.7 +/- 11.1 mumol.kg-1.min-1; P < 0.01) and 74% (to 43.3 +/- 7.2 mumol.kg-1.min-1; P < 0.01), respectively. Hyperglycemia increased glucose cycling (by 2.1-fold; P < 0.01) and the contribution of gluconeogenesis to HGP (88 vs. 43%; P < 0.01) while decreasing that of glycogenolysis (12 vs. 57%; P < 0.01). The percentage of neosynthetized hepatic glycogen formed via the direct pathway was markedly increased during hyperglycemia (53 +/- 2% vs. 23 +/- 3%; P < 0.01): These data indicate that the assessment of hepatic glucose fluxes can be accomplished in conscious unrestrained mice and that, in the presence of hyperinsulinemia, hyperglycemia causes 1) a further inhibition of HGP mainly via inhibition of glycogenolysis and increase in hepatic glucose cycling; and 2) about a fivefold stimulation in the direct pathway of hepatic glycogen formation.

L-glutamine and L-asparagine stimulate ODC activity and proliferation in a porcine jejunal enterocyte line.

We studied the effect of L-glutamine (Gln), the principal intestinal fuel, on proliferation of a porcine jejunal cell line, IPEC-J2. In cells synchronized by serum deprivation for 4 h, Gln stimulated ornithine decarboxylase (ODC; EC 4.1.1.17) in a dose- and time-dependent manner, with maximal effects at 10 mM in 3 h (P < 0.01). Similar effects were seen for the structurally related amino acid L-asparagine and serum. The Gln effect on ODC was specific, as isosmolar mannitol, glucose, methyl-beta-D-glucoside, L-phenylalanine, ammonia, and aminoisobutyric acid were ineffective. The alanine aminotransferase inhibitor aminooxyacetate (AO) inhibited the ODC stimulation by Gln in a dose-dependent manner (half-maximal inhibitory concentration = 0.5 mM). AO was not toxic to cells, as determined by propidium iodide uptake into nuclei. In addition, Gln stimulated a twofold increase of cellular 24-h [3H]thymidine incorporation above rates of control cells bathed in standard media (P < 0.01); this effect was also blocked by AO. Gln and phorbol 12-myristate 13-acetate stimulated ODC in a synergistic manner. The Na+/H+ exchange inhibitor methylisobutyl amiloride blocked the enhancement of ODC by Gln. Gln also induced the mRNA of the immediate-early gene c-jun. Gln stimulates proliferation in a porcine jejunal cell line through a mechanism requiring transamination and intact Na+/H+ exchange. This stimulation of enterocyte proliferation by Gln suggests that therapeutic Gln administration could facilitate epithelial recovery in the injured small intestine.

A redox-based mechanism for cardioprotection induced by ischemic preconditioning in perfused rat heart.

Recent studies have suggested that mild redox alterations can regulate cell function. Therefore, we tested the hypothesis that alteration in the thiol redox state might be responsible for the cardioprotective effects conferred by ischemic preconditioning in the perfused rat heart. We find that preconditioning with four 5-minute periods of ischemia, each separated by 5 minutes of reflow, is associated with a significant loss of glutathione (3.98 +/- 0.32 mumol/g dry wt, n = 8) compared with no preconditioning (6.38 +/- 0.24 mumol/g dry wt, n = 14). We further find that the addition of N-acetylcysteine (NAC, a glutathione precursor and antioxidant) during the preconditioning protocol not only blocks the loss of glutathione (5.60 +/- 0.31 mumol/g dry wt, n = 9) but also blocks the protective effects of preconditioning. It is observed that after 20 minutes of ischemia followed by 20 minutes of reflow, untreated hearts recover 38 +/- 7% (n = 5) of their initial preischemic contractile function, whereas preconditioned hearts recover 91 +/- 11% (n = 7). Hearts preconditioned in the presence of NAC recover 24 +/- 3% (n = 7) of their preischemic function. Similarly, the addition of NAC reverses the protective effect of preconditioning on creatine kinase release. On reflow after 60 minutes of ischemia, creatine kinase release from control hearts was 271 +/- 20 IU.20 min-1.g dry wt-1 (n = 5), whereas preconditioned hearts release only 170 +/- 26 IU.20 min-1.g dry wt-1 (n = 6), and hearts preconditioned in the presence of NAC release 361 +/- 30 IU.20 min-1.g dry wt-1 (n = 5). We also find that hearts preconditioned in the presence of NAC have less attenuation of the decline in pHi than hearts preconditioned in the absence of drug. Thus, a redox-sensitive mechanism may be involved in the protection afforded by ischemic preconditioning.

In vivo glucosamine infusion induces insulin resistance in normoglycemic but not in hyperglycemic conscious rats.

To test the hypothesis that increased flux through the hexosamine biosynthetic pathway can induce insulin resistance in skeletal muscle in vivo, we monitored glucose uptake, glycolysis, and glycogen synthesis during insulin clamp studies in 6-h fasted conscious rats in the presence of a sustained (7-h) increase in glucosamine (GlcN) availability. Euglycemic (approximately 7 mM) insulin (approximately 2,500 pM) clamps with saline or GlcN infusions were performed in control (CON; plasma glucose [PG] = 7.4 +/- 0.2 mM), diabetic (D; PG = 19.7 +/- 1.1), and phlorizin-treated (3-wk) diabetic rats (D + PHL; PG = 7.6 +/- 0.9). 7-h euglycemic hyperinsulinemia with saline did not significantly decrease Rd (360-420 min = 39.2 +/- 3.6 vs. 60-120 min = 42.2 +/- 3.7 mg/kg.min; P = NS). GlcN infusion raised plasma GlcN concentrations to approximately 1.2 mM and increased muscle and liver UDP-GlcNAc levels by 4-5-fold in all groups. GlcN markedly decreased Rd in CON (360-420 min = 30.4 +/- 1.3 vs. 60-120 min = 44.1 +/- 3.5 mg/kg.min; P < 0.01) and D + PHL (360-420 min = 29.4 +/- 2.5 vs. 60-120 min = 43.8 +/- 2.9 mg/kg.min; P < 0.01), but not in D (5-7 h = 21.5 +/- 0.8 vs. 0-2 h = 24.3 +/- 1.1 mg/kg.min; P = NS). Thus, increased GlcN availability induces severe skeletal muscle insulin resistance in normoglycemic but not in chronically hyperglycemic rats. The lack of additive effects of GlcN and chronic hyperglycemia (experimental diabetes) provides support for the hypothesis that increased flux through the GlcN pathway in skeletal muscle may play an important role in glucose-induced insulin resistance in vivo.

Oral transforming growth factor-alpha enhances jejunal mucosal recovery and electrical resistance in piglet rotavirus enteritis.

A randomized, investigator-masked trial determined the effects of oral recombinant human transforming growth factor-alpha (TGF alpha) on jejunal mucosal recovery in 75 piglets with rotavirus diarrhea. Rotavirus inoculation of artificially reared piglets induced subtotal (approximately 50%) villus atrophy and watery diarrhea. Dietary TGF alpha was associated with significant restoration of villus surface area by 4 d postinoculation (p.i.) and complete restoration by 8 d p.i., whereas saline-treated animals required 12 d for recovery. Jejunal segments from clinically recovered TGF alpha-treated piglets showed an increase in electrical resistance across the epithelial barrier in vitro which was proportional to villus height. TGF alpha treatment for 12 d also produced a 30-50% increase in jejunal mucosal mass (protein content and wet weight), compared with the corresponding values in saline-treated piglets and in uninfected controls. However, oral TGF alpha did not hasten the resolution of diarrhea, enhance the specific activities of jejunal mucosal digestive enzymes, or increase jejunal glucose-stimulated Na+ absorption in vitro. We conclude that dietary TGF alpha stimulates jejunal mucosal hypertrophy, improves barrier function, and enhances regrowth of villi in rotavirus enteritis; however, it does not facilitate the restoration of functional activity or mucosal digestive enzymes. Oral TGF alpha can facilitate intestinal epithelial recovery in diseases associated with mucosal damage.

Studies of cuticle drugs from natural sources. III. Inhibitory effect of Myrica rubra on melanin biosynthesis.

The inhibitory effect of a 50% ethanolic extract obtained from the dried leaves and the bark of Myrica rubra, was investigated in vitro on melanin biosynthesis which is closely related to hyperpigmentation. These extracts inhibited tyrosinase activity which converts dopa to dopachrome in the biosynthetic process. Furthermore, the extracts inhibited the production of melanin from dopachrome by autoxidation and also showed superoxide dismutase (SOD)-like activity. After bioassay-guided fractionation, quercetin, myricetin and myricetin 3-O-rhamnoside were isolated from the leaves. As they showed the inhibitory effect on tyrosinase activity, the activity is partially attributable to them in the extract of M. rubra. These results suggested that the leaves or the bark of M. rubra might be used as a whitening agent for the skin.

Alternative pork carcass evaluation techniques: I. Differences in predictions of value.

Dissected and predicted wholesale and lean boneless values for 154 pork carcasses representing seven genotypes with substantial variation in carcass composition and percentage of lean were determined. Dissected carcass value was determined using a component pricing model, and four alternative models were specified to predict that value. The models included measurements from a ruler (RULER) and two carcass evaluation technologies, Hennessy probe (PROBE) and electromagnetic scanner (EMS1). A combination of the PROBE and EMS1 models (EMS2) was also used. For wholesale value, R2 were .40, .70, .59, and .74, and the RSD were 8.18, 5.77, 6.76, and 5.38 ($/100 kg of carcass value) for RULER, PROBE, EMS1, and EMS2, respectively. For lean boneless value, the R2 were .41, .73, .59, and .74, and the RSD were 8.34, 5.67, 6.99, and 5.51 ($/100 kg of carcass value) for RULER, PROBE, EMS1, and EMS2, respectively. The results indicate that a combination of probe and electromagnetic scanner measurements provided the best fit to dissected value.

Alternative pork carcass evaluation techniques: II. Statistical analysis of error attributable to sex, genotype, and weight.

Carcasses of 154 hogs representing seven genotypes with substantial variation in carcass composition and percentage of lean were completely dissected and analyzed. Measurements from a ruler, Hennessy probe, and electromagnetic scanner were each used to predict wholesale and lean boneless carcass value. Error, defined as dissected value minus predicted value, due to the omission of sex, genotype, weight, and their interactions was estimated for each model. The errors were significantly different from zero for the models using ruler and electromagnetic scanning measurements separately (P < .01). Errors due to sex, genotype, weight, and their interactions were greatest for the less lean barrows. A combination of probe and electromagnetic scanner measurements resulted in the least error. The value of barrows with low percentage of lean was consistently overpredicted, whereas the value of leaner gilts was underpredicted for the models using ruler and electromagnetic scanning separately (P < .001).

Interleukin-6 in synovial fluid and HLA-DR expression in synovium from patients with temporomandibular disorders.

Interleukin-6-dependent mouse hybridoma cell line KD83 was used to test the biologic activity of interleukin-6 in synovial fluid from 37 patients with temporomandibular disorders. The results showed that the interleukin-6 level was greater than 100 U/mL in 13 of 18 patients with degenerative joint disease and in five of 12 patients with temporomandibular disc displacement. However, the interleukin-6 level was less than 100 U/mL (range, 20 to 75 U/mL) in all patients with masticatory muscle disorder. It has been found that degenerative joint disease tends to have acute and chronic stages, and interleukin-6 activity was probably related to the acute stage in the patients. Histologic studies of the synovium from seven patients with degenerative joint disease showed a variable degree of hyperplasia of the synovial lining cells and chronic inflammation in five of eight specimens. Immunostaining studies clearly showed the presence of significantly more HLA-DR-expressing cells (human leukocyte antigen-D-related) in synovium. Although it is unlikely that immune responses play an important primary role in initiating synovial inflammation and cartilage destruction, immune reactions may be one important factor in the maintenance and severity of some patients with temporomandibular disorders.

[Measurement of the reserve function of inspiratory muscle and its clinical significance].

The principles of measuring inspiratory muscle tension-time index (TTim) and the ratio of the works of inspiration over the maximal works of inspiration (Wi/Wi(max)) were investigated and their formulae were deduced, i.e. TTim = (Pi x Ti)/(MIP x Ttot) and Wi/Wi(max) = (Pi x VT)/(MIP x IC). The importance of the inspiratory pressure and the maximal inspiratory pressure (MIP) measured at function residual capacity (FRC) level was emphasized. Both TTim and Wi/Wi(max) were measured in 35 normal subjects and 89 patients with chronic obstructive pulmonary disease (COPD). The results showed that normal value of TTim was 0.0253 +/- 0.0055 which corresponds to the normal value of the diaphragm tension-time index (TTdi = 0.02-0.03) reported by Bellemare. Patients with COPD had a mean TTim much higher than that of normals (P < 0.01). The works of inspiration (Wi) in patients with COPD increased, while the maximal works of inspiration (Wi(max)) declined, so Wi/Wi(max) became significantly greater than that of normals (P < 0.01). The results also showed that there was a linear relationship between Pi/Pimax and TTim or Wi/Wimax (r = 0.7891, 0.9738, 0.6459, 0.9327, P < 0.01). Therefore, we suggest that both TTim and Wi/Wimax can be used as clinical indices to reflect the reserve function of inspiratory muscles.

Asparagine stimulates piglet intestinal Cl- secretion by a mechanism requiring a submucosal glutamate receptor and nitric oxide.

Amino acids are potential components of oral rehydration solutions for infants, which could combine with glucose to further stimulate intestinal Na+ and water absorption. L-Glutamine, the principal fuel of the intestine, stimulates neutral NaCl absorption and enhances enterocyte DNA synthesis, but is unstable in solution. L-Asparagine (ASN), a more stable amino acid with similar structure to L-glutamine, also stimulates enterocyte proliferation. We determined the effects of ASN on electrolyte transport across piglet jejunum in Ussing chambers. Mucosal but not serosal ASN produced electrogenic Cl- secretion (delta JClnet = -1.8 +/- 0.3 microEq/cm2.hr-1). ASN, when added at 0.1 to 30 mM, increased short-circuit current in a dose-dependent manner with a K1/2 of approximately 5 mM and maximal effect at approximately 10 mM. The stimulation of Cl- secretion by ASN was blocked by pretreatment with serosal tetrodotoxin and bumetanide and was inhibited by preincubation with capsaicin (8-methyl-N-vanillyl-6-nonenamide) or substance P. Inhibition of nitric oxide synthesis with the structural analog of L-arginine, NG-monomethyl-L-arginine, reduced ASN-stimulated secretion by > 70%. Additionally, serosal 6-cyanonitro-quinoxaline 2-3-dione, which is a nonspecific blocker of neural non-N-methyl D-aspartate (NMDA) glutamate receptors, fully inhibited the ASN response (IC50 = 10(-6) M). Inhibition was specific for neurally mediated secretion. We found no inhibition of ASN-stimulated secretion by atropine, ketanserin, indomethacin or L-2-amino-5-phosphonovalerate (specific for NMDA receptors). When compared to ASN, L-glutamate was a weaker stimulator of jejunal Cl- secretion.(ABSTRACT TRUNCATED AT 250 WORDS)

Studies on kinetics of substrate utilization of hydrogen production from wastewater with immobilized cells of photosynthetic bacteria.

The kinetic characteristics of substrate utilization by immobilized cells of Rhodopseudomonas capsulata 386 and Rhodopseudomonas sp. D for H2 production was investigated. The results showed that substrate utilization did not proceed simultaneously with H2 evolution, H2 producing capacity of immobilized cells with agar was higher than that of alginate immobilized cells, but the appearance of maximum H2 producing activity was later than the last one. The kinetics of substrate utilization (glucose) by immobilized cells of strain D followed the first-order reaction, rate constant k value was 1.2 x 10(-2) L/h, analysis of macrokinetics indicated that substrate utilization by immobilized cells in H2 evolution process was governed by biochemical reaction other than diffusion transfer limitation, because of the Thiele modulus were 0.125 and 0.154 for immobilized cells with D = 3.6 mm and 4.4 mm particles, the corresponding effective factor were determined as 0.998 and 0.988, respectively. An immobilized cell bioreactor, fed by glucose and lactate, was employed for continuous H2 evolving system. It was found that hydrogen production were 0.659 L/d and 0.477 L/d in immobilized cells system of strain 386 and D when lactate used as H2 evolving substrate, while the volumetric H2 rate were all up to 1.0 L/L.d.

[Observations on serum and amniotic fluid hormone levels and ultrastructure of uterine myometrium before and during term labor].

OBJECTIVES: To evaluate the relationship between serum and amniotic fluid (AF) estradiol (E2), estriol (E3), progesterone (Po), cortisol (Co), human placental lactogen (hPL) concentrations and uterine contractions. To observe the ultrastructural changes of uterine myometrium before and during labor. METHODS: Sixty-eight term-pregnant women (before labor 18, in normal labor 16, uterine inertia 34) were enrolled for study. Serum and AF E2, E3, Po, Co, hPL concentrations were measured by radioimmunoassay in 56 out of the 68 subjects. Myometrium gap junction (GJ), dense body (DB), sarcoplasmic reticulum (SR) were observed under electron microscope in the remaining 12 subjects (before labor 6, Uterine inertia 6). RESULTS: (1) E2, E3 levels and E2/Po, E3/Po ratio in AF, but not in serum, increased successively in the groups before labor, in normal labor and uterine inertia with significant differences (P < 0.01). (2) Both serum and AF Co, hPL concentrations were significantly different before and during labor (P < 0.01, P < 0.05). (3) Serum E2, E3, Co and hPL levels elevated significantly in cases with uterine inertia after successful treatment with estradiol benzoate or oxytocin infusion. (4) Compared with the group before labor, the amount of myometrium GJ, DB, SR increased significantly after the onset of labor. CONCLUSIONS: The elevation of serum and AF Co, hPL concentrations may be involved in the initiation of labor. This may be one of the major factors affecting the uterine contractions. Appropriate serum and AF estrogen (E), progestin (P), E/P ratio may be related to normality of uterine contractions during labor. The increase of GJ, DB, SR amount in uterine myometrium may be the histologic basis for the onset of labor.

[Acute effect of milrinone on pulmonary hemodynamics in patients with hypoxic pulmonary hypertension].

Milrinone is a phosphodiesterase inhibitor that has vasorelaxant activity. The authors studied its effect on pulmonary haemodynamics in 12 patients with hypoxic pulmonary hypertension. After treatment with milrinone, mean pulmonary arterial pressure (from 4.61 +/- 1.41 kPa to 3.36 +/- 1.20 kPa, 1kPa = 7.5mmHg) and pulmonary vascular resistance (from 557.56 +/- 167.70 dyne.sec.cm-5 to 392.60 +/- 133. 81 dyne.sec.cm-5) decreased (P < 0.05), while cardiac output increased (from 5.11 +/- 1.62 L/min to 5.62 +/- 1.66 L/min) (P > 0.05). The systemic blood pressure (from 11.50 +/- 1.63 kPa to 10.71 +/- 1.83 kPa) and systemic vascular resistance (from 1476.64 +/- 531.52 dyne.sec.cm-5 to 1,236.49 +/- 410.18 dyne.sec.cm-5) also decreased (P > 0.05). but there was no change in arterial oxygen saturation (SaO2). The incidence of side effects were low, 4 patients developed tackycardia, and 2 patients developed ventricular premature beats. It is therefore, shown that milrinone is a potent pulmonary vasodilator.

[Clinical study of improving effect of shuxinyin on abnormal left ventricular diastolic function in coronary heart disease].

There was evidence that abnormal left ventricular diastolic function was common in patients with coronary heart disease which led to symptoms such as dyspnea on exertion, distress in chest etc, even overt congestive heart failure. The changes of left ventricular diastolic function were examined with pulsed doppler echocardiography as well as of clinical symptoms, signs, electrocardiographic findings and plasma lipoprotein after Shuxinyin (SXY) therapy in 31 patients with coronary heart disease and abnormal left ventricular diastolic function. The SXY mainly consists of medicinal herbs capable of tonifying Qi and Yin. After treatment for one month the symptoms, signs and ECG were improved, the symptomatic effective rate was 87.1%. Plasma lipoprotein was reduced (P < 0.05). The main changes in doppler echocardiography were as follows. The peak of E wave, the area of E wave and Dec rate increased significantly (P < 0.01). The ratio of A wave peak to E wave peak was decreased. The changes in peak of A wave, the area of A wave and time index were insignificant (P > 0.05). It was concluded that SXY mainly improved the early left ventricular diastolic functions, increased the peak velocity and volume of rapid left ventricular filling blood flow during the early diastole, and thus alleviated symptoms as well as ECG findings.
