Dispersion of liver tissue from rainbow trout at 10 and 25 degrees C resultet in cells which were cultivated in vitro at 20 degrees C regularly. Using this method a cell line (R1) has been established useful for testing the toxicity of chemical substances and waste water. The toxicity of samples influenced the morphology, growth and the living duration of the cells. These parameters gave a clear answer about the toxicity of samples testet in 1-12 h. The tests on the vitality of cells (using trypan blue) and the release of LDH did not show an agreement with the cytotoxicity in all cases. From 106 samples of waste water were 53.8% cytotoxic but only 35% showed a toxicity for fish. 93.44% of 61 toxic samples of waste water showed cytotoxicity, only 60.65% of the samples were fish-toxic. According to the results obtained it is concluded that fish cell tissue cultures are useful tools for determination of the toxicity of chemical substances and waste water. Compared with the fish test the cytotoxicity test ist more sensitive and it reduces the material, time and money needed.

A method for preparing enzymatically dispersed pituitary cell cultures of carp (Cyprinus carpio) is described. The cultures have been used to assay a synthetic analog of gonadotropin releasing hormone (GnRH) and to determine the specificity of steroids able to affect gonadotropin (GtH) release in vitro. Time course secretion studies indicated that by 48 h incubation, in the presence of 500 pM GnRH, cumulative secretion of gonadotropin (719 ng +/- 90/2.5 X 10(5) cells) had exceeded that of controls (446 ng +/- 106/2.5 X 10(5) cells). Estradiol-17 beta, progesterone, testosterone, and 11-ketotestosterone showed different inhibitory effects on pituitary basal GtH release. Based on the results, it was concluded that carp pituitary cell cultures can be applied to investigations of several aspects of the hypothalamo-hypophysial-gonadal system.

A new continuous cell strain (CaPi) was established from normal pituitary tissue of carp, Cyprinus carpio L. Cells of a fractionated dispersion (0.25% trypsin-PBS, 10 degrees C) of pituitaries formed monolayer (fibroblast- and epithelial-like-cells) at 25 degrees C using Eagle's minimal essential medium (MEM) supplemented with 10% fetal calf serum and non essential amino acids. The continuous cell strain, designated CaPi, was obtained by an enzymatical selection of epitheloid cells from the monolayer of the primary pituitary cell culture. This culture has been subcultured 70 times over a period of 24 months. CaPi-cells multiply over a temperature range of 15-35 degrees C with an optimum growth temperature of 30 degrees C at a seeding density of 1.7 X 10(6) cells/ml. Chromosome analysis indicates the cells are heteroploid and show modal numbers of 47 chromosomes. The CaPi cell line is susceptible to fishviruses (VHSV, PFR, SVCV) producing cytopathic effects (CPE). However, IPNV replicates without visible cell alterations.

CHSE-214-cells and IPN-virus persistently infected CHSE-cells showed no differences in morphology and in growth rates. Egtvedvirus was replicated in IPN-virus persistently infected cells but virus multiplication was about 90% suppressed compared with the virus yield of standard CHSE-cells.
A nonenveloped icosahedral virus measuring 60 (+/-5)nm in diameter has been isolated from pike fry (Esox lucius). The isolate is stable to heat, lipid solvent and acid. Iododeoxyuridine has no effect on its replication. Serologically, the virus related to the major European strains of infectious pancreatic necrosis virus (IPN) Sp and Ab. The isolate is neutralized by normal trout serum and is not pathogenic for trout fry or for young pike. A line of CHSE-214 cells persistently infected with the isolate was established and has been passed 30 times.
