[Effects of hyper- and dyslipidemias on activity of angiotensin-converting enzyme (biochemical disturbances in metabolic syndrome)].

AIM: To study influence of lipoprotein complexes on activity of angiotensin-converting enzyme (ACE). Material and methods. Lipoproteins fraction was isolated from the serum of 53 healthy males and females and was studied for lipoproteins action on ACE in vitro. In addition, the examination covered 783 individuals. The data were analysed mathematically for relationships between ACE activity and parameters of lipid metabolism. RESULTS: It was found that blood serum lipoproteins inhibit activity of soluble ACE. Serum ACE activity depends on concentration of total cholesterol (TC) and triglycerides. Maximal activity of ACE is observed in TC 9.41 mM/l and TG concentration 3.05 mM/l. Albumin-related free fatty acids affect ACE activity neither in aqueous solution nor in the presence of lipoprotein complexes. CONCLUSION: . Development of most common atherogenic hyperlipidemias raises 2-fold plasma ACE activity. Elevated ACE activity may be an integral component of biochemical disturbances in metabolic syndrome.

[Acoustic (ultrasound) determination of concentration of whole protein and protein fractions in blood serum].

A new method is suggested for determining the concentration of whole protein and the distribution of protein fractions in blood serum by means of an ultrasound device. The acoustic method is based on measuring the proliferation rate and absorption of the ultrasound signal in the examined serum. The findings of the method were compared with those obtained by routine tools in examining both blood sera of patients and controls. A high correlation factor was registered for the biuret method in the determination of the concentration of whole protein (R = 0.95; p < 0.05). It was demonstrated as possible to use the discussed method as a screening tool in evaluating the distribution of protein fractions. The examination findings are generated by the instrument as in the electrophoretic examination. The method is simple in use. No reagents are needed.

[Current technologies for enhancing efficiency of laboratory techniques].

The current approaches to promoting the efficiency of applying the laboratory tools were comprehensively analyzed. It was for the first time that the above issue was considered from the standpoint of a scientifically substantiated management scheme applicable to the laboratory examination volume and within the framework of the modern conclusive medicine. The problem can be solved through starting, in each clinic, an independent research project and through patient management (critical course--technological card) with respect to available technological and personnel resources. Finally, the basic principles are outlined that are needed for designing the technological cards; the related role of experts in clinical laboratory diagnostics is demonstrated.

[Lipoproteins of blood serum and the activity of angiotensin-converting enzyme].

We studied the influence of native lipoproteins on the activity of the angiotensin-converting enzyme (ACE). Lipoproteins of blood serum were inhibiting the activity of soluble ACE. The ACE activity in blood serum was found to depend on a concentration of total cholesterol and of triglycerides. The maximal ACE activity was observed at a concentration of total cholesterol of 9.41 mM/l and at a concentration of triglycerides of 3.05 mM/l. The albumin-linked non-etherified fatty acids affected the ACE activity neither in water solutions nor in presence of lipoprotein complexes. According to the study, the progression of most frequent atherogenic hyperlipidemia results in a 2-fold increase of the ACE activity. It is suggested that an intensified ACE activity in blood plasma can be regarded as an integral component of biochemical disorders in metabolic syndrome.

[The content of double bonds and lipids in blood serum of healthy people and patients with hyperlipidemia].

The number of dual bonds (DB) was determined by the ozonization method in the lipid fraction of blood serm (BS) in healthy subjects and in patients with hyperlipidemia (HLP). Single-direction changes were detected in the DB quantity and in the lipid level in case of HLP; besides, a highly reliable positive correlation was established between the DB concentration and the basic BS lipid classes. The results suggest that an increasing DB quantity in HLP is preconditioned by a growing lipid contents and show their tense relation with components of the lipid transport system (LTS). The determination of the DB concentration, as of an index of the total unsaturation of the BS lipid fraction, provides for obtaining extra data useful in describing the LTS in different-type impairments of lipid metabolism.

[Involvement of kappa-opioid receptors in mechanisms of aggressive and submissive types of behavior in male mice].

Effects of the kappa-opioid receptor agonist U-50.488H (0.0, 0.6, 1.25, 2.5 mg/kg. s.c., 30 min) on behavior of the winner with repeated experience of victories and the losers with repeated experience of social defeat in 20 daily agonistic confrontations as well as the control mice were investigated in the tests estimating exploratory activity (open-field) and communication (partition test). Different effects of drug on behaviors of animals with different social story were shown in both tests. In the losers, all doses of U-50.488H had anxiolytic effect, increasing the communication in the partition test. In the winners, the drug induced an increase of aggressive motivation. The control mice were less sensitive to the treatment. In the open-field test, U-50.488H increased the locomotor and exploratory activity in high anxious losers. Winners significantly differed in their reaction to drug treatment in most behavioral forms in comparison with the controls and losers. It was concluded that kappa-opioid receptors are specifically involved into mechanisms of formation of aggressive or submissive types of behaviors under positive or negative social experience.

Increase in expression of brain serotonin transporter and monoamine oxidase a genes induced by repeated experience of social defeats in male mice.

Serotonin transporter and monoamine oxidase (MAO) A are involved in the inactivation of serotonin. The former is responsible for serotonin re-uptake from the synapse, whereas the latter catalyzes serotonin deamination in presynaptic terminals. Expression of serotonin transporter and MAO A genes was investigated in raphe nuclei of midbrain of CBA/Lac male mice with repeated experience of social victories or defeats in 10 daily aggressive confrontations. The amount of cDNA of these genes was evaluated using multiplex RT-PCR. Two independent experiments revealed that the defeated mice were characterized by significantly higher levels of serotonin transporter and MAO A mRNAs than the control and aggressive animals. Increased expression of MAO A and serotonin transporter genes is suggested to reflect the accelerated serotonin degradation in response to activation of the serotonergic system functioning induced by social stress. Significant positive correlation between MAO A and serotonin transporter mRNA levels suggests common pathways of regulation of transcriptional activity of these genes.

[The change in the activity of angiotensin converting enzyme in patients with pneumonia and chronic obstructive pulmonary diseases].

The activity of angiotensin-converting enzyme (ACE) was measured in the serum and bronchial contents of 69 patients with pneumonia and 77 with chronic obstructive pulmonary diseases (COPD). ACE activity was decreased both in the blood and bronchial contents during the acute phase of pneumonia. With resolution of the inflammatory process, ACE activity normalized. In patients with COPD, the activity of ACE is decreased during remission in comparison with the mean values in the population. During COPD exacerbation the activity of ACE increases both in the blood and bronchial contents. Changes in ACE activity in pneumonia and COPD are more pronounced in the bronchial contents than in the blood. Presumably alteration of the enzyme concentration is the only cause of alteration of its activity in patients with COPD and pneumonia.

[Modifying effect of the repeated experience of agonistic confrontations on effect of naltrexone in male mice].

In mice with different experience of agonistic confrontations: victories or social defeats during 3 and 10 days (T3 and T10 winners and T3 and T10 losers, resp.), T10 winners displayed a lesser aggression and a more hostile behaviour than T3 winners. Naltrexone dose-dependently decreased attacks in the T3 winners and did not affect aggressive grooming, diggings, autogrooming, and exploratory activity. Naltrexone was ineffective in T10 winners. The naltrexone effects were similar in T3 and T10 losers and its high and low doses contrarily affected different parameters of submissive behaviour. The repeated experience of agonistic confrontations seems to modify the naltrexone effects depending on a neurochemical background, differing in winners and losers.

[Effect of long-term restraint stress on behavior of female C57BL/6J and CBA/Lac inbred mice. Dispersion and cluster analysis].

An augmented exploratory behaviour and motor activity and diminished anxiety after a restraint stress were found in CBA/Lac female mice [corrected] but not in C57BL/6J ones. In the Porsolt test the result was exactly opposite. A possibility of inherent anxiety-depressive pathological condition in the C57BL/6J mice [corrected] developing under the effect of repeated psychological stress, is assumed.

[Acarbose (glucobai) effects on lipid metabolism in patients with non-insulin dependent diabetes mellitus].

AIM: To investigate the effect of acarbose, intestinal alpha-glucosidase inhibitor, on lipid metabolism in patients with non-insulin-dependent diabetes mellitus (NIDDM). MATERIAL AND METHODS: Glucobai was given for 6 months in a dose 300 mg/day to 63 NIDDM patients aged 40-69 years. The examination covered indices of glycemia, glycosylated hemoglobin, pancreatic insulin secretion, total cholesterol (TC), triglycerides (TG), HDLP and LDLP cholesterol, atherogenic index. RESULTS: Glucobai has improved glycemic profile, reduced insulinemia and body mass index, TC, TG, atherogenic index. Blood levels of HDLP cholesterol elevated by 62%. CONCLUSION: Long-term intake of acarbose improves carbohydrate metabolism and produces a hypolipidemic effect.
