Protein synthesis in heterotopically transplanted rat hearts.

This study examines protein synthesis in heterotopically transplanted rat hearts and several tissues of recipient rats. Donor hearts and recipient tissues synthesized many of the normally occurring proteins observed in tissues of unstressed rats. In addition, a stress-induced protein with a molecular mass of 71 kilodaltons was synthesized in donor heart, recipient heart and lung. Donor hearts incorporated more L-[35S]-methionine than did recipient hearts. Tissues of recipient rats also incorporated more label than the respective tissues of sham-recipient rats. These results suggest that ischemia, endured by the donor hearts during transplantation, induced these changes in protein synthesis.

Venous involvement in leprosy: a venographic and histopathological correlation.

Venous system involvement was studied by venography and vein histology in 30 leprosy patients irrespective of age, sex, duration of the disease, and treatment. Vascular abnormalities by venography were seen in 96.3% and by histopathological studies in 76.6% of patients. The percentage and severity of radiological and histological changes increased from tuberculoid to lepromatous in the disease spectrum. Perfect correlation of histology and venography was found in 40% of the paucibacillary patients and good correlation was found in 100%. In multibacillary patients perfect correlation was found in 29.4%, good correlation in 41.1%, and no correlation in 29.4% of the patients.

Microbial pathogens from goat mastitis and phage-typing of Staphylococcus aureus isolates.

Examination of milk from goats yielded 41 strains from 40 clinically affected halves; 15 were Staphylococcus aureus, 6 Staph. epidermis, 1 Streptococcus agalactiae, 2 Strept. dysgalactiae, 5 Strept. uberis, 2 Corynebacterium pyogenes, 3 Escherichia coli, 3 Pasteurella spp. and 4 Mycoplasma spp. One half had dual infection of Staph. aureus and Strept. dysgalactiae. Twenty two of the 297 milk samples from apparently normal halves also harboured pathogens comprising of 9 Staph. aureus, 1 Strept. agalactiae, 2 E. coli, 2 Pasteurella spp., 2 Candida albicans and 6 Mycoplasma spp. Most of the bacterial isolates were sensitive to many broad spectrum antibiotics. Twenty of the 24 Staph. aureus isolates were phase typable by a set of 23 human Staphylococcal International Phages suggesting the utility of these phages for the typing of goat strains. The isolates were grouped into 15 phage-types, many of which have been reported from human infections in Iraq. This indicates the possibility of association of human strains of Staph. aureus in caprine mastitis. No definite correlation could be noted between antibiogram and phage types of Staph. aureus strains.

Immunoprofile of reactions in leprosy.

The immunologic profile during erythema nodosum leprosum (ENL) revealed increased percentage and absolute count of B-lymphocytes, in addition to considerably lowered levels of complement component C3. Serum immunoglobulins--IgG, IgA, and IgM--were significantly raised after subsidence of ENL, whereas upgrading reaction showed an increase of absolute count of total T lymphocytes and percentage and count of B-lymphocytes. In addition, a raised serum IgM level was noted after regression of ENL. Downgrading reaction demonstrated a decreased percentage of early T-lymphocytes and raised the absolute count of B-lymphocytes. Serum IgA levels were found to be increased after amelioration of ENL.

Na+-Ca2+ exchange contributes to increase of cytosolic Ca2+ concentration during depolarization in heart muscle.

The possible role of Na+-Ca2+ exchange in contributing to depolarization-induced increase in cytosolic Ca2+ concentration ([Ca2+]i) of isolated rat ventricular myocytes was investigated. Measured with the Ca2+-sensitive indicator quin 2, [Ca2+]i increased from 177 +/- 12 (mean +/- SE, n = 11) to 468 +/- 41 nM when cells were depolarized with solutions containing 50 mM KCl [high extracellular K+ concentration ([K+]o)]. Approximately 73% of this high-[K+]o-induced increase in [Ca2+]i was abolished by the Ca2+ channel blocker verapamil (5 microM). For cells pretreated with 10 mM caffeine to deplete the Ca2+ stored in sarcoplasmic reticulum, 50 mM KCl still produced an increase in [Ca2+]i, even in the presence of 5 microM verapamil. However, if extracellular Na+ was replaced by Li+ or tris(hydroxymethyl)aminomethane, this increase was completely abolished. The results suggest that, in addition to voltage-sensitive Ca2+ channels, voltage-sensitive Na+-Ca2+ exchange can also contribute to the increase in [Ca2+]i on depolarization. Therefore both Ca2+ transport systems may play important roles in regulating cardiac excitation and contraction.

Pleuroscopy in the diagnosis of pleural effusion using a fiberoptic bronchoscope.

Forty patients with undiagnosed pleural effusions were investigated by pleuroscopy, using a fiberoptic bronchoscope. Pleural biopsies diagnostic of tuberculosis were obtained in 17 cases, and in 12 of these tubercle bacilli were cultured from the pleural fluid. Pleural biopsy at thoracoscopy is superior to the percutaneous intercostal route and for this purpose the fiberoptic bronchoscope is preferable to the rigid thoracoscope.
