[Effect of E. coli endotoxin on the amount of 2,3-diphosphoglycerate in rabbits].

Fifteen rabbits were injected i/p with an endotoxin at the rate of 6 mg each. Studied were the amounts of 2,3-diphosphoglycerate (DPG), hemoglobin, and erythrocytes in the blood 1, 2, 2.5, and 3 hours following treatment. A clearly manifested lowering trend was observed with the level of hemoglobin up to the 3rd hr. The changes concerning DPG and the erythrocytes also consisted in lowering their values, which was more strongly expressed with DPG than with Hb, reaching its lowest values 2.5 hours following treatment. From that moment on a rise was noted with the values of DPG and erythrocytes. The changes that set in with the latter had no effect on the DPG/Hb coefficient, their being a lowering of its values up to the 2nd hour, whereas from the 2nd to the 3rd hour there followed a rise. The rise of the DPG and the DPG/Hb values after the 2.5 and the two hours respectively, was referred to as a compensatory mechanism through which the body lowered the level of hypoxia, developing under the effect of the E. coli endotoxin.

A study on the mutagenic effect of some Bulgarian drugs using the Salmonella/microsome test.

It has been shown that mixtures of phenobarbital and 3- methylcholantrene may be used in the Salmonella/microsome test as inductors of the S9 liver fraction, instead of "Aroclor 1254". The Salmonella/microsome method has been used to study the mutagenic action of Flatogel (2.5% polymethylsyloxanum activatum and 2% ethylium -p- aminobenzoicum , suspended in sorbitol-balanced gel of aluminum hydroxidatum and magnesium hydroxidatum ), Nitrolong (2,2-bis/hydroxymethyl/-1,3- propandioltetranitrate ) and Cymethidine (N-cyano-N'-methyl-N"/2-/5-methyl-4-imidazolyl-methylthio/ethyl/guanidin e). These substances are produced as drugs by the Bulgarian chemical and pharmaceutical industry ( PHARMACHIM ). The results obtained show that the drugs studied have no mutagenic action, both without and with activation with S9 liver fraction isolated from rats treated with "Aroclor 1254" or combinations of phenobarbital and 3- methylcholantrene .

[Nomograms developed for determining the acid-base and oxygen status of cattle].

Investigations were carried out on a total of 66 lactating cows in order to work out an Octane nomogram for the direct recording of the PO2/HbO2 interrelationship and determine the normal alkali-acidic range in cattle. It is stated that these studies have made it possible to speculate on the clinical veterinary normal values of each particular index of the alkali-acidic balance and its prompt calculation.

[Determination of total lipids, esterified and nonesterified fatty acids in the blood plasma of guinea pigs treated with CC14].

Studied was the effect of CCI4 intoxication of guinea pigs on the dynamics of changes taking place in the level of the total lipids and the esterificated and unesterificated fatty acids. It was found that following treatment with CCI4 the amount of the total lipids in the blood rose by the eighth hour and reached its peak value at the 24th hour. The unesterificated fatty acids rose considerably, while the esterificated ones showed but a slight rise. It is concluded on the basis of correlation coefficients that the changes in the level of the unesterificated fatty acids have the highest diagnostic value in the tetrachlormethane intoxication of the liver.

[Mechanism of glycogen depletion in the liver in endotoxin-treated rats].

Studied were the rate and mechanism of glycogen breakdown in rats treated with Salmonella abortus ovis endotoxin. It was found that tissue homogenates from treated animals showed higher glycogenolytic activity in vitro as compared with homogenates from untreated animals, which demonstrated the direct mechanism of endotoxin action. The higher lytic activity with treated animals seemed to be associated with the higher activity of glycogen-phosphorylase, which showed peak values at the third hour following treatment. The higher activity of glycogen-phosphorylase was, in turn, the result of the enzyme reactivation under the effect of the endotoxin, and was not linked with the synthesis or new enzyme molecules.

[Changes in the amount of free amino acids in infections due to Gram-negative agents].

Changes of free amino acid content in blood plasma from sheep and hens infected with S. abortus ovis and P. multocida or treated with purified lipopolysaccharides (endotoxins) of the respective agent, were studied. It was established that the infection reduced total free amino acid content by about 46% in sheep and by 36% in hens. The reduction in total free amino acid content did not affect to the same extent each individual amino acid. Treatment of sheep and hens only with purified lipopolysaccharides of respectively S. abortus ovis and P. multocida resulted also in reduced total free amino acid content equal to 33% in sheep and about 59% in hens. The reduced content of total free amino acids in this case also did not concern equally the individual amino acids. However, in all cases observed, when a given amino acid was reduced during the infection period the same reduction was evident following endotoxin treatment only. The conclusion is drawn that disturbances in cell metabolism arising under the influence of endotoxins let free by the respective agent are the main cause for the reduction both of total free amino acid content and of the individual free amino acid content.

[Biochemical and histological studies of cows with ketosis].

Biochemical and histological investigations were carried out with cows affected with ketosis in a subclinical and clinical form. Parellel studies were performed with normal (control) cows originating from the herds with the ill animals and manifesting the same physiologic status. It was found that the blood level of glucose in the diseased cows was lowered more than 2.6 times, and the content of ketone bodies was higher-- it was increased 4.6 times, that of total lipids was 2.6 times higher, and that of free fatty acids was 3.5 times higher as compared to the same indices in the controls. The blood content of pyruvate in the affected cows was also increased. The liver glycogen in these animals was lowered from 2.1 to 4.1 times. The activity of glucose-6-phosphatase was lower, and that of glucose-6-phosphatedehydrogenase was higher as against the same indices shown by normal cows.

[Changes in carbohydrate metabolism in hens experimentally infected with Pasteurella multocida kokoshki].

Studied were the concentration of blood sugar, the activity of glucose-6-phosphatase and glucose-6-phosphatedehydrogenase, and the level of glycogen in the liver of P. multocida infected chickens. An abrupt decrease of the liver glycogen was found as well as a negligible rise of the blood sugar. The activity of glucose-6-phosphatase remained unchanged, and that of glucose-6-dehydrogenase was twice as high. It is concluded that the intense exhaustion of liver glycogen is in connection with the activation of the pentosephosphate pathway.

[Comparative study of the optimum pH value of serum alkaline phosphatase in various species of farm animals].

Investigations were carried out on the alkaline phosphatase in the sera of cattle, horses, pigs, sheep, goats, and chickens, the pH value of the buffer used being 9.0-9.8-10.0-10.2-10.6 and 11.0, and the method applied--that of Richterich. The pH value at which the serum alkaline phosphatase in the various farm animals and birds was most active was found to vary to a large extent. Optimal values for the enzyme's activity usually range as follows: cattle, 10.2; pigs and goats, 10.0; sheep,--10.2; horses,--9.8; chickens,--10.6.
