Synthesis and properties of 5-membered heterocyclic disulfides: application for enzyme modification.

Synthesis of the disulfides 2,2'-dithiodithiazole, 2,2'-dithiobis(4-methylthiazole), and 5,5'-dithiobis(3-methylisothiazole), which are thiol-specific reagents, is described and the uv-absorption spectra of the disulfides and corresponding thiols and/or thiones are presented. The utility of these and several other reagents for the modification of thiol groups in enzymes has been demonstrated with lactate dehydrogenase and malate dehydrogenase and compared with the frequently applied modification reagents 5,5'-dithiobis(2-nitrobenzoic acid) and 2,2'-dithiodipyridine. The 5-membered heterocyclic disulfides permit the spectrophotometric determination of all sulfhydryl groups of malate dehydrogenase and lactate dehydrogenase without requiring their prior denaturation. Application of these reagents makes possible the determination of SH groups in the pH range less than or equal to 7.

Action of FPL 52694 on gastric acid secretion in the healthy human stomach.

In a double blind crossover experiment in 16 healthy male volunteers, the effects of oral FPL 52694 and a matching placebo upon pentagastrin stimulated gastric acid secretion, gastric mucosal histamine content, and gastric mucosal mast cell count were compared. There was a significant increase in acid secretion, and a fall in tissue histamine and the mast cell count after treatment with the placebo. All these changes were inhibited by FPL 52694, and there was a significant difference between the effects of this compound and the placebo in all three parameters. It is concluded that FPL 52694 caused significant inhibition of acid secretion, and that the likely mechanism of action is by stabilisation of mast cells and inhibition of histamine release.

[Cell electrophoresis studies of peripheral blood lymphocytes in patients following kidney transplantation].

In 111 patients (control group, dialysis patients, kidney recipients) the behaviour of T lymphocytes was observed using an automatic measuring apparatus ( Parmoquant cytopherometer ) and the spontaneous rosette test. The Parmoquant method gives no information on the process of rejection, but it helps to identify risk situations due to bacteria of viruses. In cases of rejection after the second post-operative week, the results of the two methods are reversed.

[Chlamydia infections in the gynecology outpatient clinic].

Chlamydia trachomatis is a common agens of non-specific genitourinary inflammatory conditions. We examined 269 female outpatients of the clinic. For 16 per cent of the patients Chlamydia trachomatis could be isolated. In these positive cases we found the following distribution: 32.5 per cent in the cervix and urethra, 60.5 per cent only in the cervix and 7 per cent only in the urethra. The clinical symptoms and importance of the disease are discussed.

[Iatrogenic skin-tissue embolism in the lung].

A rare autopsy finding of an embolic fragment of skin in the lung after intravenous injection and central venous catheterization is described. Frequency, origin and fate of these microembolisms are discussed.

[Leptospiroses with acute renal failure--report on 6 dialyzed patients].

From 1968 to 1981 6 patients with severe leptospiroses were treated: one with leptospirosis icterohaemorrhagiae (Weil's disease), two with leptospirosis copenhageni and three with leptospirosis grippotyphosa. The diagnosis must at first clinically be made from exposition anamnesis and symptoms. In three patients the agglutination lysis reaction had a negative result still between the 10th and 19th day after the beginning of the disease. In all 6 cases a hypercatabolic acute renal failure with azothaemia of high degree was present. The polyuric phase developed 10 to 18 days after the beginning of the disease, the serum creatinine values normalized 20 to 35 days after the beginning of the disease. The parenteral nutrition and the dialysis therapy were particular points in the complex therapy of the patients. 4 patients were haemodialysed - 3 of them after primary peritoneal dialysis - 2 patients were only peritoneally dialysed. On account of the hypercatabolism and the intestinal paralysis the haemodialysis is to be preferred, as a rule, to the peritoneal dialysis. Also in the haemodialysis of patients with leptospirosis modern developments should be taken into consideration: bicarbonate-containing dialytic solution and in haemorrhagic diathesis the heparin-free dialysis. All 6 own patients survived. When connecting these data with other reports from Central and South Europe the lethality of dialysed cases with leptospirosis is about 42% (25 of the 59 patients died). In 4 of 5 of our patients with leptospirosis a clear hyperamylasaemia was present, apparently the expression of the involvement of the pancreas.

[Topical treatment of acne vulgaris with erythromycin and benzoylperoxide (author's transl)].

16 healthy persons were treated for 7 weeks with a 1% erythromycin containing ointment. Another 25 persons (among them 10 acne patients) were treated during the same period of time with a combination therapy consisting of erythromycin (1% in an unspecific ointment base) and benzoylperoxide (5% in a gel base). The combined therapy reduces the micrococcaceae in the pilosebaceous ducts significantly. On the other side the therapy consisting only of erythromycin causes only initially a decrease and then an increase in the number of micrococcaceae. Induction of resistance to erythromycin is in both cases considerable. In both cases a reduction of propionibacteria which are most important for the pathogenesis of acne must be assumed. Because of the probable pathogenetic role of staphylococci in acne vulgaris an additional improvement of the therapeutical results can be expected in the case of combination therapy. In vitro experiments demonstrate further that an erythromycin treatment leads to considerable resistance induction in S. aureus, regardless of whether it is applied alone or in combination with benzoyl peroxide.

[Long-term study in congenital refractory anemia and secondary siderosis].

It is reported on the course of the disease in a female patient with congenital therapy-refractory anaemia, in whom a transfusion siderosis of high degree with functional disturbances of the myocardium and the liver, with endocrine disturbances (primary hypothyroidism and normogonadotropic hypogonadism) and with small growth became clinically manifest. A reduction of the iron overload by a continuous desferrioxamine therapy in form of long-term subcutaneous infusions over two years could be proved. Possibilities and problems of treatment and conclusions relevant to practice are described.

Effect of N-methyl-formimino-methylester on the vomeronasal neuroepithelium of mice.

N-methyl-formimino-methylester (MFM), a highly volatile chemical substance, causes massive, transient sensory-cell degeneration in the main, but not in the vomeronasal olfactory sensory epithelium of mice. After MFM-treatment it appears possible to study the accessory olfactory system after chemical "deafferentation" of the main system.
