After phenol-acetic-acid extraction the following Haemophilus strains resp. their proteins were subjected the polyacrylamid-gel-electrophoresis in presence of 8 M urea: Strains of the serovar A of H. paragallinarum: 0083, 1516, 1598, 2213, 1645, 1646, Löhren, 2671, 1385, 758, 17756; strains of serovar B of H. paragallinarum: 0222, 2600, 733, 2028, 1596, 2026, 1676, 245, the S and R-form of 2403 as well as the strains 782 and 1655, which were not serotyped; strains of H. paravium sp. nova (HINZ: Inst. J. Syst. Bacteriol. in press): 1762, 62 (Serovar 1), 2654, 2659 (Serovar 2), 780 (Serovar 3), 94 (Serovar 4) and 1254, 0002, 0003, which were not serotyped. H. parainfluenzae (NCTC 4101) and H. parasuis were examined in the same way. The Coomassie Blue-stained protein patterns show that each of the strains tested developed its characteristic protein pattern, with exception of the S- and R-form of the strain 2403, which developed identical pattern. Interrelations between electrophoretic pattern and biological properties such as biochemical activities or pathogenicity could not be proved. However, the procedure described seems to be suitable for strain- or clon-identification on the subspecies level. The electrophoresis apparatus, which was made according to our instructions was less expensive than corresponding available equipments and proved to be usable for the polyacrylamid-gel-electrophoresis.

A method for selective detection of terminally alpha 2-3 and alpha 2-6 sialylated neolacto-series gangliosides has been developed. The procedure involves separation of gangliosides on high performance thin layer chromatography plates, fixation of the silica gel, treatment with Vibrio cholerae neuraminidase and incubation of the plates with nLcOse4Cer-specific antibodies. Alkaline phosphatase-conjugated second antibodies were used to visualize bound first antibodies by generating a blue dye from 5-bromo-4-chloro-3-indolyl phosphate. Neolacto-series gangliosides from human granulocytes with terminally alpha 2-3 and alpha 2-6 linked N-acetylneuraminic acid served as examples. Neuraminidase treatment of gangliosides with alpha 2-3 substituted sialic acid is necessary prior to immunostaining whereas alpha 2-6 sialylated gangliosides can be detected without enzyme treatment. Steric hindrance of sialic acid bound in position 3 to terminal galactose prevented binding of the antibody to the Gal beta 1-4GlcNAc sequence whereas sialylation in position 6 to terminal galactose does not hinder recognition. The procedure is viable for the detection of amounts down to 10 ng of gangliosides. This method should be useful to screen gangliosides from different tissues or cell lines for the presence of such components, especially if only small quantities of material are available.

QUESTION: Disorders of coordination, strength and proprioception are frequently described after surgery of the knee joint. These muscular coordination disorders have not yet been studied sufficiently. The purpose of this study was the objective-quantitative characterization of muscular coordination disorders of the M. quadriceps femoris by means of EMG mapping in the arthrotic knee joint and their progress after the implantation of a total knee arthroplasty and after in-patient rehabilitation. METHODS: The extent and pattern of activation (EMG mapping) of the M. quadriceps femoris were investigated by means of a 16-channel EMG technique in 38 patients before and after the implantation of a total knee arthroplasty (type LCS). RESULTS: EMG mapping permitted an objective-quantitative characterization of the extent and pattern of activation in the M. quadriceps femoris before and after the implantation of a total knee arthroplasty and therefore the objectivation of muscular coordination disorders. Muscular coordination disorders were present already before surgery and were also caused by the implantation of a total knee arthroplasty. These coordination disorders can be improved by an inpatient rehabilitation. CONCLUSIONS: The development of exercise programs adequate for everyday life is necessary to remove quickly and actually the demonstrated coordination disorders.
AIM: Based on our own previous surface EMG investigations (EMG mapping), it has been shown that disturbances in coordination, proprioception and strength measured before and after implantation of a total knee arthroplasty (TKA) can be treated successfully with standard physical therapy during in-patient rehabilitation. The objective of the present study was to evaluate the therapeutic effect of a special type of hydrotherapy. METHOD: 25 patients with TKA were treated for 3 weeks with a standardized hydrotherapy training during in-patient rehabilitation. Several diagnostic methods (EMG mapping, Isokinetics, Ultrasound) were applied to objectify and quantify changes in muscular coordination and strength at 4, 7, and 26 weeks postoperative. These results were compared with the results of 38 TKA patients treated with a "standard rehabilitation program" and with the results of 20 healthy persons. RESULTS: EMG mapping as well as isokinetics and ultrasound permitted an objective characterization of the results. Patients treated with the special hydrotherapy showed greater improvements in the investigated items of coordination and strength than patients treated with the "standard rehabilitation program". For example, in the hydrotherapy group a significantly better contraction capacity of the M. vastus intermedius was measured. The intensive hydrotherapy was tolerated by all patients and reached good acceptance. CONCLUSION: The investigated hydrotherapy technique leads to better muscular coordination and strength, which can provoke a better stabilization of the knee joint. Therefore, the early and intensive application of hydrotherapy for improving coordination and strength in the rehabilitation of patients with total knee arthroplasty is advisable.
AIM: The objective of the present study was to evaluate the long-term functional results after implantation of a total knee arthroplasty (TKA) objectively and quantitatively. METHOD: Functional parameters (muscular coordination, strength and range of motion) and the subjective satisfaction of the patients were measured by means of EMG mapping, isokinetics, ultrasound and a questionnaire in 21 patients with TKA 5.1 years after implantation of the TKA. RESULTS: 81 % of the investigated patients were subjectively satisfied. The isokinetic results showed a significant improvement of the extensor strength and of the relation between flexors and extensors. Patients who had been more physically active over these 5 years reached significantly better results. The results of EMG mapping showed good overall retention of the satisfactory coordination levels resulting from the initial in-patient rehabilitation. But the coordination patterns demonstrated clear changes showing highly activated areas in the region of the M. vastus medialis. The range of motion and the contraction capacity showed no further improvement compared with the results 26 weeks after implantation of the TKA. CONCLUSION: Long-term results after implantation of a TKA showed a good muscular coordination, strength and range of motion. A higher physical activity level led to better functional results and greater overall satisfaction of TKA patients 5 years after implantation.
A statistical approach was used to establish a new classification system of Marek's disease virus (MDV) on the basis of neurologic responses. To develop the system, neurologic response data from 15x7 chickens inoculated with 30 strains of serotype 1 MDV were statistically analyzed by a cluster analysis. The goal was to identify a statistical system that would verify if three neurovirulence groups correlated with the three pathotypes previously described. The system was also validated in two additional strains of specific-pathogen-free (SPF) chickens, SPAFAS and line SC (Hy-Vac). The proposed system is based on analysis of three variables: 1) frequency of birds showing transient paralysis between 9 and 11 days postinoculation (dpi), (2) mortality before 15 dpi, and (3) frequency of birds showing persistent neurologic disease between 21 and 23 dpi. By use of this system, a MDV may be classified in one of three groups, designated neuropathotypes A, B, and C, which roughly correspond to the virulent, very virulent, and very virulent plus pathotypes, respectively. However, correlation between neuropathotype and pathotype was not absolute, and neuropathotyping is more a complement to the current pathotyping system than a replacement for it. Our results showed that neuropathotyping studies can be conducted in two types of commercial SPF chickens by the use of the same variables, although the system would first have to be standardized by the use of prototype viruses. Neuropathotypes can also be estimated with our statistical analysis with reasonable accuracy. By use of this analysis, we established that MDV strains within the very virulent pathotype may be subdivided into neuropathotypes B and C, thus establishing a previously unrecognized pathotypic classification. This finding illustrates how neuropathotyping may extend important information not identified by conventional pathotyping.
