Influence of hypothalamus on glucose utilization by adipose tissue of normal rats.

This study was designed to investigate whether Ventro-Medial and Ventro-Lateral Hypothalamus exert a direct effect upon the glucose uptake by rat adipose tissue. VMH, VLH and pieces of cortex as controls were removed from normal rats fasted for either 6 h (Exp. A), or 14 h (Exp. B), or made hyperglycemic by i.p. injection of glucose (Exp. C). The tissue pieces were then incubated for 30 minutes at 37 degrees C in Gey-Gey buffer. Aliquots of these incubates were then added to flasks containing epididimal fat pads of rats fasted 6 h, U-14C-glucose and unlabeled glucose to a final concentration of 1 mg/ml and incubated for additional 3 h at 37 degrees C. In exp. A the glucose utilization by adipose tissue was not affected by the addition of either VMH or VLH incubation media, when compared to controls. In exp. B a significant increase of glucose utilization was elicited by the addition of VLH incubates, while in exp. C the addition of VLH incubation media produced a significant decrease of glucose uptake by adipose tissue. The addition of VMH incubates had no significant effect in any of these experimental conditions. These results support the hypothesis that a hypothalamic factor may have a direct action upon glucose utilization by the adipose tissue, independently of the control exerted by hypothalamus on insulin and glucagon release.

[Vitamins in the prevention of hyperlipoproteinemia and atherosclerosis (author's transl)].

Primary and secondary vitamin deficiencies cause many metabolic diseases which possible are life-threatening. Nevertheless none of the hypovitaminosis is known to give rise to hyperlipoproteinemias and/or atherosclerosis. On the other hand, the hypolipemic effect of several vitamins, like retinol, ascorbate, tocopherol and nicotinate, has clearly been demonstrated. This effect takes place either on clinically non-demonstrable hypovitaminosis, or by a true pharmacological activity of those factors. In the last case several different mechanisms are probably involved, which are described in the present paper. The use of vitamins in metabolic disorders is justified both for the primary prevention, in order to correct the causes of hyperlipoproteinemias, and in the secondary one, to diminish the blood fat levels, which increase the risk of atherogenesis.

Gonadotropin-releasing hormone test for hypogonadic men.

Gonadotropin patterns before and after stimulation with gonadotropin-releasing hormone (GnRH) have been studied in 69 hypogonadic men of various types: patients with expansive hypothalamus--pituitary disorders before and after surgery, patients with hypogonadotropic hypogonadism, and patients with oligozoospermia or azoospermia who have primary partial or total testicular deficiency. Three characteristic gonadotropin patterns were found: (a) low basal values of LH and FSH with either absent or decreased and delayed responses; (b) normal basal values and pituitary responses above the normal range; or (c) high basal values and pituitary responses above the normal range. These gonadotropin patterns were correlated with disorders of the hypothalamus--pituitary--testis axis. The advantages and disadvantages of the GnRH test for the clinical evaluation of male hypogonadism are discussed.

[Effect of calusterone on the stem cell compartment after suppression with busulfan in mice].

These studies were undertaken to evaluate the effect of Calusterone (a weakly androgenic steroid) on hemopoiesis in mice. Cellular proliferation was suppressed by a single (IP) injection of busulfan (BU) (40 mg/Kg). Calusterone (CA) was administered s.c. SC daily (10 mg/Kg); controls received an equivalent injection of oil vehicle. Hemopoiesis was characterized by measurement of peripheral blood neutrophils, bone marrow cellularity, differentials and stem cell content. This included pluripotent (CFU-S), granulocytic (CFU-C) and erythroid (CFU-E) progenitor cells. Only a minimal decrease in narrow cellularity was observed after busulfan; similar values were obtained in calusterone recipients. Neutrophils fell by day 4, showed an abortive rise on day 8 and subsequently fell to 32% of control values. Calusterone recipients showed a 2 fold higher value (62%) on day 12. CFU-S, CFU-C, and CFU-E were depressed to 20-40% of control values by day 2 after busulfan. Although CFU-S and CFU-C remained depressed through the 14th day, CFU-E recovered by day 8 CA stimulated an overshoot in these cells to 288% of control values. These findings correlated with an increase in marrow erythroid cells to 182% on day 10. CFU-S remained low (20%) by day 14 and gradually increased to 50% of control by day 24. A delayed 10 day course of CA more than doubled the CFU-S recovery. These findings show that BU markedly suppress hemopoietic stem cells: a differential recovery is noted between CFU-E and the other progenitor cells. CA increase the recovery of all 3 hemopoietic stem cell compartments when given either immediately or in a delayed schedule. This suggests that this compound may be of use in the therapy of bone marrow hypoplasia.

[The problem of infections in acute leukemias: origin, therapy and prevention by protective isolation].

The most important factors of infection in leukemic patients and immunodepressive effect of chemotherapy are evaluated. The microorganisms causing infections most frequently are considered. The associations of antibiotics for therapy of severe infections are reported. The employement of granulocyte transfusion, in patients with severe neutropenia and infections antibiotico-resistents, is also considered. Dealing with patients liable of infections the possibility of infection prevention and therapy in protected environment is faced. 12 patients affected by acute nonlymphocitic leukemia were treated with cytostatic drugs to induce remission in laminar air flow room. The number of infections is significantly decreased in comparison with of patients treated in open ward. In protected environment it is possible to treat patients with the maximal dose of antiblastic drugs without interruption obtaining higher number of complete remission.

LH and FSH (basal values and pituitary responses to GnRH) in hyperthyroidism.

Gonadotropin behaviour (basal plasma values and pituitary responses to GnRH) in hyperthyroid patients was studied. LH values (both basal and pituitary responses) higher than the controls were documented. Thyroid hormones seem to modulate pituitary response to GnRH for LH only in hyperthyroid patients of both sexes. In order to explain the mechanism of the action of thyroid hormones, rather than the possibility of peripheral effect on binding capacity of the proteins for circulating steroids, a central interaction at medial eminence level is suggested.

Possible interaction of PGF2 alpha with hypothalamus-pituitary-thyroid axis in man.

The possible interactions of PGF2 alpha on the hypothalamus-pituitary-thyroid axis are the object of this study. Firstly a significant direct effect of PGF2 alpha infusion (mg2,5/270 min) on TSH,PRL,LH,FSH and GH pituitary secretion was excluded. Thereafter the possible PGF2 alpha on PRL and TSH pituitary response to TRH was considered: in only two cases PGF2 alpha was able to increase the TSH response. Finally the Authors studied T3 response to endogenous TSH rise induced by TRH: if they consider the mean peak responses of T3 the increase is significant only when PGF2 alpha infusion is performed.
