For the purpose of verifying the effectiveness of peplomycin, one of the derivatives of bleomycin, against carcinoma of the esophagus and the safety of it, the analysis of the data for total 113 cases collected from 25 institutions in Japan was made. The results are as follows. It was effective in 19 out of 74 evaluable cases of carcinoma of the esophagus (25.7%). In case of treatment with peplomycin alone, it was effective in 6 out of 39 cases (15.4%). In case of the combination treatment with peplomycin and some other therapy, it was effective in 13 out of 35 cases (37.1%). As for the side effects, the incidence of fever was the highest in both the cases of peplomycin alone and the combination treatment such as 39.6% and 37.0%, respectively. Anorexia, nausea, vomiting, respiratory symptoms and tiredness were found in relatively many cases. In the clinical laboratory tests, the vital capacity after the treatment tended to be lower than that before the treatment, but there was little change in the hematological tests, pulmonary function test and renal function test.

We reported two cases of multicentric glioma. The first case was 60-year-old female. In CT scan findings, separate mass lesions were observed in the right parieto-occipital area, left parietal lobe and left thalamic region. The second case was 53-year-old male. In CT scan findings, separate mass lesions were observed in the right fronto-parieto-temporal area and left frontal lobe. These two cases were considered to be metastatic brain tumor and radiation therapy and chemoimmunotherapy were performed. But in the postmortem examinations, both cases were evident to be not metastatic brain tumor but glioblastoma multiforme. And tumor cells were not found in commissural or other nerve pathways and cerebrospinal fluid system histologically. So our cases were considered as multicentric glioma. The pathogenesis of multicentric glioma are not understood till now. Therefore, it is interesting that the different histological findings were observed in the each tumor lesion of the first case.

Pharmacokinetics of 3-sulfamoylmethyl-1,2-benzisoxazole (AD-810), a new possible antiepileptic compound, were evaluated in Phase I trials in nine healthy male volunteers. The subjects, divided into three groups, were given single oral dose of 200 mg (Group A), and oral doses of 200 mg/d (B) and 400 mg/d (C) for two d, respectively. AD-810 was well tolerated by all subjects. No neurologic side effects were observed. Evaluation of the laboratory screening profiles (hematological and biochemical examination) showed no clinically significant abnormalities. Mean peak plasma concentrations of AD-810 at 5.3-6.0 h after intake were 2.9, 5.1 and 13.0 micrograms/ml in Group A, B and C, respectively, and mean elimination half-life was about 60 h. Mean concentrations of AD-810 in erythrocytes were higher than those in plasma. The unchanged AD-810 and a major metabolite, the glucuronide of the N-O cleft compound, were excreted at 48.7% and 12.4% of the given dose in the urine for 2 weeks in Group C, respectively. The results suggest that AD-810 would induce no severe adverse effects in man at plasma concentrations over 10 micrograms/ml, the therapeutic concentrations predicted from animal experiments.
From the echocardiographic files of 1,231 patients, eight revealed mitral annular calcification (MAC), in whom four had hypertrophic obstructive cardiomyopathy (HOCM). In the remaining four patients without HOCM, whose ages ranged from 69 to 80 years with a mean of 73 years, an additional calcification was observed in the either aorta or aortic valve. Therefore, MAC was thought to be a part of the aging process. Of the four patients with HOCM two were females (71 and 78 years, respectively) and two were males (50 and 51 years, respectively). The latter two younger male patients had MAC without another calcific lesion, and the cause of MAC was suspected as the pressure and/or mechanical strain on the mitral orifice due to excessive movements of either the chordae tendineae or papillary muscle.
