[Subrenal capsule assay--an experimental study and clinical application to chemosensitivity tests].

We have attempted to investigate the usefulness of the subrenal capsule assay (SRCA). Initially, we compared the results of antitumor activities of 27 anticancer drugs obtained using the SRCA in BDF1 mice and the subcutaneous transplantation assay in nude mice, and it became clear that these results correlated well. It was thus apparent that the antitumor activity of drugs could be evaluated faster, more cheaply and more easily using the SRCA. In the SRCA, the overall success rate for growing tumors (with a tumor growth rate of more than 1.0) from patients was 79% (106/134) and the success rates for each tumor type were as follows: malignant lymphoma 90%, ovarian cancer 87%, sarcoma 77%, breast cancer 76%, colorectal cancer 73% and renal cancer 58%. In a retrospective analysis of correlation between the antitumor activities assayed by the SRCA and the respective clinical responses, 11 out of 25 trials were true positive (79%) and 7 trials were true negative (64%). The sensitivity and specificity levels of this test were 73% and 70%, respectively. The tumor growth inhibition rates were correlated well with the clinical responses. The advantages of the SRCA are: it has a high success rate; it allows rapid evaluation; it can be applied for masked compounds; and it has a good clinical correlation. These advantages indicate that the SRCA is of potential value as a predictive test for selection of anticancer drugs on an individual patient basis.

[Results in oligodendroglioma: postoperative radiotherapy combined with chemotherapy].

The retrospective analysis of 39 cases of supratentorial oligodendroglioma included the years 1965 - 84. The 5-year and 10-year survival rates were 73.1% and 32.2%, respectively. Although oligodendrogliomas were benign tumor, the 10-year survival rate was not good. Recurrence of tumor appeared in 9 cases and some cases were noticed about 10 years after the treatment. So, long term follow-up is important for oligodendrogliomas. Our treatment for oligodendrogliomas was surgical removal and postoperative radiochemotherapy (radiation + ACNU + FT-207 + PSK) for several years. The previous cases who had only subtotal removal of tumor showed short survival time. On the other hand, the cases treated with postoperative radiotherapy or radiochemotherapy showed relatively good results.

A multivariate study on enzymatic changes in limb muscles and heart muscle of dystrophic mice.

The present study was undertaken to elucidate further the enzymatic changes in dystrophic muscle using multivariate analysis. The activities of 14 kinds of enzymes, including 6 exopeptidases, 4 endopeptidases, beta-N-acetyl-D-glucosaminidase, phosphatase, esterase, and ribonuclease, were examined in forelimb and hindlimb muscles as well as in cardiac muscle of dystrophic mice and their controls. Two principal components identified from the enzymatic spectrum proved to be related especially to aminopeptidases and to serine proteinases, respectively. The enzymatic changes in forelimb muscle were very similar to those in hindlimb muscle when both were compared to those in cardiac muscle. The changes in aminopeptidases were unique to the limb muscles, whereas those of serine proteinases were unique to cardiac muscle of dystrophic mice. In the future, more attention should be focused on the role of exopeptidases in pathogenetic mechanisms of muscular dystrophy, because of the possibility that they play a major role in the initial stage of muscular dystrophy.

[Regional cerebral blood flow in children--normal value and regional distribution of cerebral blood flow in childhood].

The changes in CBF and rCBF through the entire age range include the rapid period in childhood was reported. Sixteen children between the ages of 1 and 15 and 14 adults were studied. These 30 subjects were either volunteers of out-patients without CT and EEG abnormalities. rCBF was measured by the 133Xe intravenous injection method using Varmet rCBF analyzer. Ther relationship between age and CBF, the correlation coefficient was calculated based on the regression equations and the regression curve with the highest correlation was chosen. For the analysis of rCBF, he mean rCBF values (ISI) of 3 channels corresponding to the frontal, temporal, parietal and occipital lobes were expressed as a percentage of the hemispheric CBF. The hemispheric blood flow (ISI and CBF gray) of children less than 5 years of age was approximately twice that found in adults. This value decreased rapidly with age and in the 10-15 years the blood flow was approximately 1.3-fold that of adults. Thereafter, there was a slow decrease and a negative correlation with age was found. The decrease showed the correlation on the following equations; y = 146.5 - 58.5 log x. (r = -0.903) for ISI and log y = 2.26 - 0.29 log x. (r = -0.881) for CBF gray, which was statistically significant. In contrast, the CBF white showed a slightly higher value in the 1-2 years old children, but thereafter the CBF did not show a notable decreases with age. Through the entire age range, a best fit for the Fw values was found with : y = 18.3 + 37.5/x. (r = 0.798), which was also statistically significant.

Enhanced chemosensitivity of cells from malignant effusions under condition of exposure to high temperature.

Utilizing a clonogenic assay, the effects of hyperthermia and selected chemotherapeutic drugs on growth of cells from malignant effusions were studied. Fourteen of 25 samples obtained from 25 patients with various carcinomas formed at least 30 colonies per plate. Exposure of the cells to heat at 42 degrees C for 1 hr before the plating slightly inhibited the colony growth. The drugs, adriamycin (AM) and mitomycin C (MMC), were tested at 3 different concentrations. When the cells were treated with these two drugs for 1 hr at 42 degrees C, the percent of surviving colonies was significantly decreased, as compared to findings at 37 degrees C, in both groups, at 3 different concentrations. The combination of drugs and hyperthermia appeared to function synergistically in one-third of such cases. These results suggest that cells from malignant effusions in patients with various carcinomas were more sensitive to AM or MMC, under condition of a higher temperature (42 degrees C).

A simplified method for determination of tritiated thymidine incorporation into cells from tissue in soft agar culture.

We designed a simple method for in vitro chemosensitivity testing of tritiated thymidine incorporation by cells from tumor tissue plated in soft agar. Cells from tumor tissue were cultured in a plastic dish, of which a Millipore membrane had been sealed to the underside, containing two layers of agar. After labeling the cells with tritiated thymidine, the dishes were placed in a boiling water bath to solubilize the agar. The labeled cells could be readily collected on the membrane by evacuating the solubilized medium through membrane filters sealed underside of the dish. Using this system, 11 (55 per cent) of 20 tumor specimens from 20 patients with various carcinomas showed an incorporation of over 200 dpm above background and 80 per cent or greater inhibition of thymidine uptake by sodium azide treatment. This method offered several advantages over the thymidine incorporation assay, including technical simplicity and a shorter time course.

Enhancement of colonization of Ureaplasma urealyticum in the mouse genital tract by estrogen treatment.

Experimental colonization by Ureaplasma urealyticum in the vagina of Balb/c mice was greatly enhanced by administration of estrogen, whereas it was not affected by treatment with progesterone. Ureaplasma was recovered from almost half of the estrogen-treated animals 7 days after inoculation, and the organism persisted in all of these animals until 21 days after inoculation; on the other hand, the organism was isolated in less than 20% of the untreated, progesterone-treated, or danazol-treated mice 7 days after inoculation and was no longer isolated in any of these groups 21 days after inoculation. At least a hundredfold more organisms were recovered from the estrogen-treated mice as compared to the untreated mice 14 days after inoculation and thereafter. U. urealyticum was also isolated from the uterine cavity and salpinx in approximately half of the mice in which the organism was isolated from the vagina.

Early radiographic changes in radiation bone injury.

A chronologic series of periapical radiographs was evaluated for the purpose of detecting damage to bone and tooth-supporting tissues in a patient receiving radiation therapy for a basal cell carcinoma of the mandibular gingiva. Widening of the periodontal space was one of the early radiographic changes observed. It is suggested, from the sequence of radiographic changes, that radiation-induced changed in the circulatory system of the bone might be primarily responsible for the resulting changes.

Synthesis and antiviral activity of sulfonamidobenzophenone oximes and sulfonamidobenzamides.

To find antiviral agents, various sulfonamidobenzophenone oximes (II) were synthesized from the appropriate m-sulfonamidobenzophenones by hydroxylamine reaction. The reaction products were generally obtained as syn/anti mixtures which were separable by fractional crystallization. The anti isomer had more potent antipoliovirus activity than the syn isomer. Various sulfonamidobenzamides (III) which were structurally related to II were synthesized by the reactions of amino-substituted benzamides with sulfuryl chloride or amines with (aminosulfonyl)benzoyl chloride. Antiviral activity was examined by the plaque-inhibition test. Compounds 5, 36, and 69 exhibited strong antipicornavirus activity. The structure-activity relationships are discussed.

Prostatic cancer presenting as metastatic adenocarcinoma of sphenoid sinus.

Prostatic cancer is commonly manifested by obstructive uropathy, regional lymphatic metastases, and hematogenous metastases to the axial skeleton. It is relatively rare that initial signs begin with the involvement of other sites. Intracranial metastases especially are seldom found and may be unfamiliar to not only pathologists but also to physicians. In this article, we present a case where the metastasis was first manifest as a sphenoid sinus tumor prior to the demonstration of the primary site and the prostate was confirmed to be primary by biopsy specimen with immunoperoxidase method. In addition to discussing the route of the tumor spread, we deal with a prostatic specific antigen efficient for identifying the primary site.

Nuclear bodies in human prostate with special reference to appearance rate.

The Nuclear Body Appearance Rate in seventeen human prostatic cases was statistically analyzed according to three lesion areas--the hyperplastic nodule, non-nodule and atrophic--in the secretory epithelium and basal cells. It was meaningfully high in the secretory epithelium of the hyperplastic nodule, but not in the other two lesion areas. There was no meaningful result in basal cells. Though there is a wide variety of reported data on the human prostate, particular care must be taken in analysis at the electron microscopic level, keeping in mind that even in a single specimen this organ has a notable variety of tissue changes. The Nuclear Body Appearance Rate reflects cellular hyperactivity although it does not have a specific known function at present.
