[Double tracer autoradiography of bone tumor using 3H-thymidine and radionuclides for diagnostic purpose].

Whole body distribution and accumulation of radionuclide for diagnostic purpose such as 99mTc-MDP (methylene diphosphonate), 67Ga-citrate (67Ga) and 201Tl-chloride (201Tl) in implanted osteosarcoma of human origin were evaluated by macroautoradiography of Balb/c nude mouse. 3H-thymidine of which uptake has a close relation to growth pattern of tumors was used for a partner of target nuclide in double tracer technique to examine the distribution characteristics. Separated distribution pattern of coupled nuclides was imaged by a difference both of half life and energy of radiation between radionuclides. The distribution pattern of each target nuclide and 3H-thymidine were compared to clarify distribution characteristics respectively in specimen. 67Ga and 201Tl were taken around the tumor and the mode of their distribution was identical to that of 3H-thymidine. Uptake of 67Ga in tumor was much higher. 99mTc-MDP accumulated in the central zone of the tumor and its pattern was quite different from that of 3H-thymidine. From above result it was demonstrated that the accumulation mechanism of 67Ga, 201Tl and 99mTc-MDP are quite different in the tumor each other. So we concluded that our method of double tracer technique was useful for evaluation of distribution pattern of radionuclides for diagnostic purpose.

[Clinical evaluation of transaxial bone imaging with 99mTc-phosphate compounds].

Single photon emission computed tomography using newly designed whole body imaging system which was composed of opposing dual gamma cameras, a rotating gantry and a sliding table was clinically evaluated for bone imaging. Two hundred and seventeen portions of various bone diseases were performed single photon emission computed tomography following conventional bone imaging with 99mTc-MDP (methylen diphosphonate) or -HMDP (hydroxymethylene diphosphonate) and the results were discussed. Transaxial images were sometimes superior than conventional images in the diagnosis of the tumor localization and extension of the facial bone. Combination of emission computed tomography to conventional methods also presented more exact informations for the diagnosis of the vertebral change. Separation of the overlapped radioactivities of the bone and the soft tissue is another advantage of the tomographic image, and cross-sectional images with enoughly high qualities were available in early time after administration of 99mTc-phosphate compounds. Useful diagnostic informations could be obtained by tomographic bone imaging combined with conventional imagings.

[Clinical effect of long-term administration of PSK in a combination in patients with malignant urologic cancer].

Long-term administration of PSK was made in 40 patients with urological cancer. Mean duration of the administration was 201, 358 and 243 days for the patients with bladder cancer, renal cancer and other urological cancers, respectively. The clinical evaluations were performed by the standard of Japan Society for Cancer Therapy and historical control study. In some cases, it seemed that the immunological therapy by PSK was a benefit to some degree. However, more accurate evaluation should be made by the matched control study.

Use of saliva for monitoring unbound free cortisol levels in serum.

In order to verify the clinical usefulness of saliva in assessing the adrenocortical function, we measured saliva cortisol levels by a radioimmunoassay after extraction with dichloromethane, and compared the results with the levels of serum unbound cortisol determined by the method of equilibrium dialysis. Paired samples of saliva and serum were obtained from 10 healthy male volunteers. Morning levels of saliva cortisol and serum unbound cortisol were 0.99 +/- 0.42 and 1.56 +/- 0.54 microgram/100 ml, respectively, where serum total cortisol was 19.28 +/- 3.56 microgram/100 ml. A well-defined diurnal rhythm and a greater response to ACTH stimulation were observed in saliva cortisol than in serum total cortisol. Kinetic study of injected cortisol revealed almost identical values for the compartments of saliva cortisol and serum unbound cortisol. The correlation coefficient (r) between the levels of saliva cortisol and serum unbound cortisol was 0.893 (p less than 0.001, n = 150). From these results we concluded that the measurement of saliva cortisol can be used sufficiently to monitor unbound free concentrations in serum.

[Scinti-angiographic investigation of aortic dissection (author's transl)].

We perform scinti-angiography regularly in suspected cases of dissecting aneurysms. In our experience, the dissection is well demonstrated by this method. In correctly selected cases, scinti-angiography is a method which is well tolerated by the patient and which carries very little risk, since the complications due to contrast media are absent.

Inhibition by sugars of opsonization of Staphylococcus aureus in fetal calf serum.

Phagocytosis of Staphylococcus aureus by human peripheral polymorphonuclear leucocytes in vitro in the presence of fetal calf serum was inhibited by D-galactose, N-acetyl-D-glucosamine and asialofetuin but not by fetuin. Absorption of opsonin in the serum with the bacteria, inhibition of the absorption by sugars and the effect of glycosidase treatment of the bacteria on phagocytosis showed that terminal structures of polysaccharides on the surface of bacteria play an important role in binding of opsonin to the target.

[Two-dimensional echocardiographic observation of ruptured chordae tendineae of the mitral valve: Its localization and uneven development of papillary muscles (author's transl)].

Twenty-three cases with ruptured chordae tendineae (RCT) of the mitral valve and 9 normal control cases were studied by two-dimensional echocardiography. Mitral valve replacement (MVR) was performed in 13 of the 23 cases. The preoperative diagnosis of RCT was proven accurately at operation in all except 2 cases with bacterial endocarditis. Whether RCT occurred in the antero-lateral or in the postero-medial commissural side could be determined perfectly. RCT was observed in the postero-medial side more often than in the anterolateral side, i.e., postero-medial side in 16 (9 operated) and antero-lateral side in 5 (2 operated). This tendency has also been noted in other Japanese reports regardless of the cause of RCT. In the short axis view at the level of the papillary muscles, the area of the posterior papillary muscles was smaller than the area of the anterior papillary muscles in the RCT cases. It is speculated that the smaller size of the posterior papillary muscles has a certain relation with the more frequent RCT in the postero-medial commissural side.

Restoration of E-rosette formation by bestatin in patients with bladder cancer.

Incubation of peripheral blood lymphocytes from patients with bladder cancer in bestatin (specific inhibitor of aminopeptidase B and leucine aminopeptidase) solution resulted in a significant increase in the proportion of E-rosette-forming lymphocytes. The increase in the proportion of E-rosette-forming lymphocytes was abolished after reincubation with 100% autochthonous serum. The fraction of E-rosette-forming lymphocytes from patients with bladder cancer which had restored receptors for sheep erythrocytes (SRBC) by incubation with bestatin also expressed receptors for the Fc portion of IgG and IgM. The proportion of EAC-forming lymphocytes remained unchanged after the incubation with bestatin. When 600 mg of bestatin was administered for 1 day to patients with bladder cancer, restorative effect on the proportion of E-rosette forming lymphocytes was also demonstrated. The proportion of EAC-forming lymphocytes, however, did not change significantly.

Surface markers of lymphocytes infiltrating seminoma tissue.

Infiltrating lymphocytes were separated from human seminoma tissue obtained surgically. The relative distributions of B and T lymphocytes, and a subset of T lymphocytes with IgG-Fc receptors were measured by the rosette-forming method in peripheral blood and in seminoma tissue from 7 patients with pure seminoma of the testis. The proportions of T and TG cells were increased in seminoma tissue compared to those of peripheral blood from the same patients, whereas the proportion of B cells did not change. This result might correlate with the favorable prognosis of seminoma usually accompanied with lymphocytic infiltration.
