A subpopulation of large ganglion neurons express IsK protein mRNA: an in situ hybridization analysis in the rat eye.

Rat IsK protein is a 130-amino acid membrane protein and subserves generation of K+ outward current. The expression of this putative K+ ion channel mRNA was explored in the eye with in situ hybridization. The ganglion cell layer of the retina expressed IsK mRNA in a subpopulation of ganglion cells composed of large cell bodies. The hybridization-positive cells were scattered throughout the retina. The hybridization signal was also localized to the epithelial cells throughout the cornea. The results provided evidence for IsK message in neuronal cells. Expression of IsK message in the eye was thus shown to be restricted to particular cell types.

Altered production of nerve growth factor in cultured vascular smooth muscle cells from genetically hypertensive rats.

1. Nerve growth factor (NGF) is a neurotrophic peptide for peripheral sympathetic nerves. Hypotrophy of sympathetic nerve ganglia and reduced content of neuropeptide Y were reported in genetically hypertensive (GH) rats. These facts led us to consider that production of NGF might be altered in GH rat cells. 2. Vascular smooth muscle cells (VSMC) were prepared from the aortic media of 12 week old GH rats and age-matched normotensive (NT) rats by enzyme digestion method. Growth rates and the maximum cell densities were fairly equivalent between GH cells and NT cells. 3. The NGF content in the medium was measured using an enzyme immunoassay against mouse beta NGF. The amount of NGF secreted from VSMC of GH were 1.2 pg/10(4) cells per 48 h in the progressive phase and 0.7 pg/10(4) cells per 48 h in the quiescent phase, while those of NT were 8 pg/10(4) cells per 48 h and 1 pg/10(4) cells per 48 h, respectively. The levels of NGF secretion per cells were greater in NT cells. In summary, NGF secretion from VSMC were reduced in GH. 4. These results suggested that a reduced production of NGF from VSMC might be involved in the reported hypotrophy of sympathetic nerve cells in GH.

High incidence of Haemophilus influenzae in nasopharyngeal secretions and middle ear effusions as detected by PCR.

PCR was used to detect Haemophilus influenzae in samples of nasopharyngeal secretion and middle ear effusion (MEE). Nasopharyngeal secretions were collected from 102 patients with otitis media with effusion and from 111 healthy subjects. Eighty samples of MEE were collected from patients with otitis media with effusion. A pair of primers was designed to amplify a DNA segment of the gene encoding P6 outer membrane protein of H. influenzae. The amplified PCR product was detected with an internal probe that hybridized specifically to the P6 DNA of H. influenzae. Samples of MEE and nasopharyngeal secretion were also examined by a conventional culture method. The incidence of P6 gene DNA in nasopharyngeal secretions detected by PCR was about two times higher than that of H. influenzae detected by the conventional culture. Culture-positive samples were all positive in the PCR test. In MEEs, the rate of detection of the P6 gene DNA target was about five times higher than that of H. influenzae detected by the culture method. All patients who had P6 gene DNA in MEEs were found to have the DNA in nasopharyngeal secretions. These findings suggest that the presence of H. influenzae in MEEs and in nasopharyngeal secretions is more common than previously reported.

[Vein patch angioplasty for coronary revascularization to ischemic heart disease: report of two cases].

Two cases of vein patch angioplasty for ischemic heart disease were presented. The first case (59-year-old female) admitted to our hospital because of posterolateral infarction. Preoperative coronary angiogram (CAG) showed stenosis of 4-PD of RCA, proximal LAD and distal LCX. The second case (62-year-old male) admitted for operation of abdominal aortic aneurysm. Preoperative CAG showed stenosis of proximal RCA and mid LAD. Stenosis of 4-PD and proximal RCA was relieved by patch angioplasty using saphenous vein graft and coronary artery bypass to LAD was performed by left internal thoracic artery graft. Both cases showed successful enlargement of RCA stenosis and good graft patency at the postoperative CAG. Vein patch angioplasty is thought to be an alternative method for coronary revascularization to ischemic heart disease.

Immunohistochemical proof of intraneural localization of herpes simplex virus in experimental retinitis.

A unilateral intravitreal inoculation of herpes simplex virus type 1 (HSV-1) induced bilateral retinitis and encephalitis in the rat. Immunohistochemistry was employed to clarify the pathway of transmission of HSV-1 from the inoculated eye to the contralateral eye, and to identify localization of the viral antigen in the retina, optic nerve and brain. At an early stage of the post-inoculation period, HSV-1 immunoreactivity was first seen in the cytoplasm of Müller cells and then in various cells of the inner nuclear layer in the inoculated eye. At a later stage, HSV-1 immunoreactivity was seen in some retinal ganglion cells and optic nerve fibers in the inoculated eye. In the brain, HSV-1 was found in the optic chiasm, bilateral primary visual centers (bilateral suprachiasmatic nuclei, bilateral lateral geniculate nuclei, bilateral superior colliculus and bilateral pretectum) and bilateral visual cortex. HSV-1 was also demonstrated in the retinal ganglion cells, neural cells in the inner nuclear layer, and Müller cells in the contralateral eye. We found that HSV-1 antigen in the retina of the contralateral eye was transmitted through the optic nerve, visual pathway and nuclei, but not through the blood stream.

Mutation of low affinity nerve growth factor receptor gene in spontaneously hypertensive and stroke-prone spontaneously hypertensive rats: one of the promising candidate genes for hypertension.

There is evidence that abnormal formation of the sympathetic nervous system might be important for pathogenesis of hypertension. In the present study we analyzed nucleotide sequences of low affinity nerve growth factor receptor (LNGFR) genes in the spontaneously hypertensive rat and its stroke-prone substrain. There was a point mutation generating an amino acid substitution in signal peptide of these LNGFRs. This result suggested that the mutated LNGFR gene is one of the promising candidate genes for hypertension.

Epstein-Barr virus in gastric carcinoma with lymphoid stroma. Special reference to its detection by the polymerase chain reaction and in situ hybridization in 99 tumors, including a morphologic analysis.

BACKGROUND: Gastric carcinoma with lymphoid stroma (GCLS) has been reported to have a more favorable prognosis than ordinary gastric carcinoma, however, the precise mechanism of the pathogenesis for GCLS remains unclear. METHODS: The authors analyzed 99 GCLS in 94 patients for Epstein-Barr virus (EBV) sequences using polymerase chain reaction (PCR) and in situ hybridization (ISH); these were compared with 42 ordinary gastric carcinomas. RESULTS: Two series of PCR showed 81 (82%) and 46 (47%) of the 99 GCLS to have EBV sequences, which were significantly higher compared with ordinary gastric carcinoma (50% and 9.5%, respectively). With ISH using thymine-thymine dimerized oligonucleotide probes corresponding to EBV-encoded small RNA 1 (EBER1), 82 (83%) of 99 GCLS showed clear, intense hybridization signals localized over the nuclei of the tumor cells, in contrast to only 4 (9.5%) of 42 ordinary carcinomas (P < 0.001). A comparative morphologic analysis of EBER1-positive and negative GCLS revealed that typical features of GCLS, such as mild cellular pleomorphism, rare mitoses, a marked degree of lymphoid stroma, and mild fibrosis, together with a lymphoid infiltration within the cancer cell nests were significantly more frequent in EBER1-positive GCLS. CONCLUSIONS: More than 80% of GCLS were associated with EBV. The presence of EBV association in GCLS was characterized by the above morphologic features.

Distribution of primary afferent fibers projecting from hindlimb cutaneous nerves to the medulla oblongata in the cat and rat.

The dorsal column nuclear complex, one of the most important relays for tactile perception, has well been known to be somatotopically organized. Topographical arrangements of terminal sites of individual cutaneous nerves within the dorsal column nuclei, however, have not been examined systematically, although many studies have been done upon primary afferents to the medulla oblongata, including the dorsal column nuclear complex. Thus, in the present study, distribution of primary afferent fibers projecting from the hindlimb cutaneous nerves to the medulla oblongata was examined in the cat and rat by means of the transganglionic transport method with horseradish peroxidase. Cutaneous primary afferent fibers projecting from the hindlimb to the medulla oblongata were distributed mainly in the ipsilateral gracile nucleus. Terminal labeling in the gracile nucleus was seen at all rostrocaudal levels of the nucleus, occasionally including the nuclear part straddling the midline (the median or accessory nucleus). The labeled axon terminals in the gracile nucleus were more densely distributed in the middle and caudal parts of the nucleus than in the rostral part. Although the fields of termination of the hindlimb cutaneous nerves overlapped in the gracile nucleus, the foci of the terminal labeling of the nerves innervating the distal parts of the hindlimb were located more medially or dorsomedially than those of the nerves innervating the proximal parts. Terminal labeling was further found in a small zone immediately medial to the rostromedial border of the external cuneate nucleus. This hitherto undescribed zone (U zone) contained a small cluster of medium-sized neurons in the cat. Although no particular cell cluster was found in the U zone of the rat, convergence of the primary afferent fibers of the cutaneous nerve from the hindlimb appeared to occur as in the U zone of the cat.

A subpopulation of olivocerebellar projection neurons express neuropeptide Y.

By neuropeptide Y (NPY) immunohistochemistry and in situ hybridization histochemistry with digoxigenin-labeled oligonucleotide probes for this peptide, a subpopulation of neurons in the caudal portions of the rat dorsal and medial accessory olives were found to express NPY immunoreactivity and mRNA. In the cerebellum, NPY-immunolabeled climbing fibers were distributed to the flocculus and parts of vermal cortex. The results suggest particular association of NPY with the climbing fiber system presumably mediating spinal, visual and vestibular inputs.

Mu opioid receptor: expression and vagotomy-induced depletion of the mRNA in medullary preganglionic neurons.

By in situ hybridization with digoxigenin-labeled RNA probes, distinct expression of mu opioid receptor mRNA was found in rat medullary preganglionic neurons for vagal parasympathetic outflow. Two days or more following unilateral vagotomy, the expression was almost completely abolished on the operated side. The results, taken together with previous findings indicating proximodistal axonal transport of radioligand-labeled opioid receptors, suggest that the medullary preganglionic neurons thus identified are the source for mu opioid receptor acting in cardiorespiratory and gastrointestinal tissues.

Colonic submucosal tumors: comparison of endoscopic US and target air-enema CT with barium enema study and colonoscopy.

PURPOSE: To compare the imaging characteristics of colonic submucosal tumors at endoscopic ultrasound (US) and target air-enema computed tomography (TACT) with those at conventional double-contrast barium enema study and colonoscopy. MATERIALS AND METHODS: Twenty consecutive patients with suspected colonic submucosal tumors at barium enema study and colonoscopy underwent endoscopic US, TACT, or both. Morphologic features and posture-related change in shape of tumor were evaluated with barium enema study, color and consistency of tumor with colonoscopy, internal echogenicity of tumor and layer of origin in normal colonic wall with endoscopic US, and CT attenuation number with TACT. RESULTS: Eight lipomas, seven carcinoids, three leiomyomas, four lymphangiomas, and one hemangioma were found at histologic examination. Lipomas and lymphangiomas had characteristic findings at endoscopic US and TACT. The differential diagnosis of the other submucosal tumors was facilitated by using endoscopic US. CONCLUSION: Endoscopic US and TACT may play a valuable role in the evaluation of colonic submucosal tumors.

A case of orthostatic and postexertional hypotension with ischemic electrocardiographic changes.

A case of orthostatic and postexertional hypotension with ischemic electrocardiographic changes is reported. The etiology was considered to be the partial dysfunction of efferent sympathetic nerve endings. Peripheral adrenergic receptors (alpha and beta) were up-regulated, which might have caused pseudoischemic electrocardiographic changes and abnormal vasodilation after exercise.

The effects on blood flows of coronary artery by-pass grafts during intra-aortic balloon pumping.

The internal thoracic artery (ITA), as well as aorto-coronary by-pass grafts, has been used for widely coronary artery by-pass grafting. Intra-aortic balloon pumping (IABP) is the first choice for left ventricular support when low output syndrome occurs during coronary artery by-pass surgery. However, the effect of diastolic augmentation by IABP may vary to the type of grafts. Graft flow with and without IABP support were measured in six patients undergoing elective coronary artery bypass surgery requiring IABP at Kanazawa National Hospital. The patients ranged in age from 59 to 67 years, with a mean age of 63 years, and included one woman and five men. In all cases, the left ITA was dissected from the thoracic wall as pedicle, and anastomosed in situ to the left anterior descending artery. Saphenous vein grafts (SVGs) were used for aortocoronary by-pass to the obtuse marginal branches, the first diagonal branches, the left circumflex branches, and/or the right coronary artery. Blood flow in 6 ITAs, 11 SVGs to the left coronary artery systems, and three SVGs to the right coronary artery was measured by ultrasound transit-time flowmeter simultaneously with the electrocardiogram. Blood flows in ITA grafts and SVGs were measured during IABP assist and unassisting under hemodynamically stable conditions after discontinuing cardiopulmonary by-pass. The systolic and diastolic flows of each graft were measured using the peak of the R wave and the end of T wave on the electrocardiogram as the references for systole. Systolic flow during IABP were similar to unassisted flow in both ITA and SVGs.(ABSTRACT TRUNCATED AT 250 WORDS)

[The effects of sex steroid hormones, gonadotropins, and gonadotropin releasing hormone agonist on the proliferation of ovarian cancer cell line (KOC-2S)].

The present study was designed to determine the effects of sex steroid hormones (estradiol, progesterone), gonadotropins (FSH, hCG), and gonadotropin releasing hormone agonist (GnRHa: Buserelin) on the proliferation of the ovarian serous adenocarcinoma cell line KOC-2S in vitro. The results showed: 1. The colony formation rate and fraction growth in the colony stem cell assay were suppressed by estradiol (5,000 pg/ml) and progesterone (50 ng/ml). The colony formation rates were 2.96% (control); 2.49%, 0.99% (estradiol: 500 pg/ml, 5,000 pg/ml); 2.58%, 0.53% (progesterone: 5.0 ng/ml, 50.0 ng/ml); 4.80%, 3.34% (hCG: 10 mIU/ml, 100 mIU/ml); and 1.79%, 2.96% (GnRHa: 1.0 ng/ml, 10 ng/ml). 2. The doubling time (DT) in the growth inhibition test was shortened by FSH and the saturation density (SD) became greater. The SD was suppressed by hCG. 3. The cytogenic features of the cells treated with FSH, estradiol and GnRHa did not show obvious morphologic change. The spontaneous floating cells were observed following treatment with progesterone. Cytoplasmic enlargement was observed following treatment with hCG. 4. Neither erb B nor erb B-2 was expressed in KOC-2S cells, and neither was induced by sex steroid hormones, gonadotropins or GnRHa.

[Immunohistochemical localization of herpes simplex virus type 1 in experimental retinitis--2. The brain, contralateral optic nerve and retina].

Unilateral intravitreal inoculation of herpes simplex virus type 1 (HSV) induced bilateral retinitis and encephalitis in rats. In a previous paper, we demonstrated immunohistochemical localization of HSV in the ipsilateral retina. In this paper, we report immunohistochemical localization of antigen to HSV in the brain and opposite eye. HSV was found in the optic chiasm, and bilaterally in the suprachiasmatic nuclei, lateral geniculate nuclei, superior colliculus, pretectum, and visual cortex. HSV was also found in the retinal ganglion cells, neural cells in the inner nuclear layer, and Müller cells in the opposite eye. We demonstrated HSV infection in the retina of the opposite eye via the optic nerve, visual pathway, and nuclei, but not through the blood stream.

Time trends of early gastric carcinoma. A clinicopathologic analysis of 2846 cases.

BACKGROUND: Despite recent improvements in the diagnostic accuracy and techniques for identifying early gastric carcinoma, gastric carcinoma remains a major cause of death in the world. METHODS: The clinicopathologic features of 2846 surgically resected early gastric carcinomas from 1962-1987 were reviewed, and the time trends of a variety of clinicopathologic factors were studied using both an univariate and a multivariate analysis. RESULTS: The differentiated type of carcinoma increased in frequency from 34-50%. The percentage of patients older than 60 years of age increased from 32-56%. The number of lesions measuring less than 3 cm increased from 28-56%. The frequency of shallow-type-IIc carcinomas measuring less than 3 cm, which had been previously difficult to detect and were mainly composed of differentiated carcinomas, increased from 46-69%. The relative proportion of the differentiated type of carcinoma was significantly higher in patients older than 60 years of age than in those younger than 60. The relative proportion of differentiated-type carcinomas measuring less than 3 cm, which had been easy to detect as opposed to undifferentiated-type carcinomas, significantly increased from 56-76%. CONCLUSIONS: The above time trends may indeed reflect genuine change. However, the recent increase in the rate of differentiated-type, small-sized carcinomas also suggests the possibility that such results may, in fact, be due mainly to improvements in both diagnostic accuracy and medical technology. Thus, a large number of undifferentiated-type, small-sized carcinomas might have merely failed to be found in routine gastric examination.

Hepatoid adenocarcinoma of the stomach. A clinicopathologic and immunohistochemical analysis.

BACKGROUND: The clinicopathologic and immunohistochemical features of the primary gastric hepatoid adenocarcinomas still remain unclear. METHODS: The authors evaluated 28 hepatoid adenocarcinomas (HC) of the stomach on the basis of characteristic histologic features resembling hepatocellular carcinoma, which were selected from the 7200 cases of primary gastric carcinoma in their files. Additionally, 22 alpha-fetoprotein (AFP)-positive adenocarcinomas without hepatoid features (APC) were also selected. RESULTS: The HC cases fell into the following two groups: HC with AFP-positive tumor cells (Group 1; 15 cases) and HC without AFP-positive cells (Group 2; 13 cases). Histologically, the glycogen granules and hyaline globules were common features in HC. The incidence of a venous invasion of HC (Groups 1 and 2) was higher than that of APC (Group 3). There were no significant differences among the three groups regarding clinical features, macroscopic features, and the incidence of lymphatic permeation. An immunohistochemical study showed that HC had differentiation into various directions. As for the advanced carcinomas, the 5-year survival rates of patients with HC (Groups 1 and 2) and those with APC (Group 3) were 11.9% (21.4%, 8.3%) and 38.2%, respectively. The prognosis of Group 1 was similar to that of Group 2, but was poorer than that of Group 3. CONCLUSIONS: The findings suggested that HC, as shown by its characteristic histologic features, had a poor prognosis whether producing AFP or not, and that HC should therefore be distinguished from AFP-positive carcinoma without hepatoid features.

On the expression of Fos-like protein in the subthalamic nucleus and basal ganglia output systems following kainic acid injections into the rodent striatum.

By immunohistochemistry, Fos-like protein induced in response to intrastriatal kainic acid injections was explored in the basal ganglia and the ventrolateral thalamic nucleus of the rat. In the globus pallidus, entopeduncular nucleus, substantia nigra pars reticulata, subthalamic nucleus and ventrolateral thalamic nucleus, Fos was expressed in neuronal cell nuclei showing the maximal number at 4 h and disappeared within 48 h. The substantia nigra pars compacta neurons lacked prominent Fos induction, suggesting that a different mechanism may operate for changes of dopamine systems. Concurrent Fos expression noted in selected regions suggests the link of relay and output stations of the basal ganglia with the ventrolateral thalamus at the early stage of striatal cell degeneration.
