[Dependence of lethality and incidence of chromosome aberrations induced by treatment of synchronized human diploid fibroblasts with sodium fluoride on different periods of the cell cycle].

The cytotoxic and clastogenic effects of sodium fluoride during various phases of cell cycle of human cultured diploid fibroblasts were examined. The cells in confluence were synchronized at G1/G0 phase by a period of growth in medium containing 1% serum (low serum medium). To obtain the cells in S phase and G2 phase, exponentially growing cells were cultured in low serum medium with a subsequent blockage of the cells at the G1/S boundary by hydroxyurea. Synchronized cells were treated with sodium fluoride for 3 hr during the G1 phase or G2 phase, and for each of three 3-hr periods during the S phase which lasted 9 hr. The cytotoxicity, as determined by a decrease in colony-forming ability, was dependent upon the phase of cell cycle during which treatment was administered. The highest lethality was observed for treatment in early to middle S phase, whereas scare lethality was observed in G1 phase. Inducibility of chromosome aberrations of the cells following treatment with sodium fluoride was also dependent upon the phase of cell cycle. Significant increase in the incidence of chromosome aberrations was observed only in cultures treated during early and/or middle S phases of the cell cycle. These results indicate that cytotoxicity and clastogenicity of sodium fluoride to cultured human diploid fibroblasts are cell phase dependent, and that the cells in early and middle S phases are more sensitive to these effects.

[Correlation of the ability of Di(2-ethylhexyl)phthalate to induce cell transformation, chromosome aberrations, and peroxisome proliferation in cultured Syrian hamster embryo cells].

Di(2-ethylhexyl)phthalate (DEHP), a commonly used plasticizer, induces peroxisome proliferation in liver cells and hepatocellular carcinomas in rodents. To study possible mechanisms for DEHP-associated cancer, we have measured induction of morphological transformation, chromosome aberrations, and peroxisome proliferation of cultured Syrian hamster embryo (SHE) cells. Molphological transformation was weakly induced by treatment with DEHP. The transformation frequency of DEHP was enhanced in the presence of rat liver postmitochondrial supernatant. DEHP induced chromosome aberrations in the cells only in the presence of exogenous metabolic activation. Clofibrate, a widely used hypolipidemic drug, failed to induce morphological transformation or chromosome aberrations. Treatment with [4-chloro-6-(2, 3-xylidino)-2-pyrimidinylthio]acetic acid (Wy-14, 643), which is a more potent carcinogen than DEHP or clofibrate, elicited a lower frequency of morphological transformation than DEHP in the presence of exogenous metabolic activation. Similar levels of peroxisome proliferation, as determined by an intensity of diaminobenzidine (DAB) staining, were observed in cultures treated for 2 hr with DEHP, clofibrate or Wy-14, 643. The results suggest a possible involvement of genetic damage by DEHP metabolites in induction of transformation of SHE cells. No clear relationship between inductions of peroxisome proliferation and cell transformation was observed.

Immunodeficiency associated with selective loss of helper/inducer T cells and hypogammaglobulinaemia in a child with intestinal lymphangiectasia.

A patient with intestinal lymphangiectasia (IL) was studied for his immunological abnormalities. The patient had hypoproteinaemia with severe hypogammaglobulinaemia. The results of lymphocyte subpopulation studies revealed a decrease of CD4+ cells and a decrease of surface immunoglobulin (sIg)-positive B lymphocytes. T cell functions determined by the proliferative responses against Concanavalin A (Con A) or phytohaemagglutinin (PHA) and by delayed type cutaneous hypersensitivity (DTH) response to purified protein derivatives (PPD) and PHA were normal. No immunoglobulin (Ig)-secreting cells were induced when his peripheral blood lymphocytes (PBL) were cultured with pokeweed mitogen (PWM). His T cells showed suppressor function to the PWM-induced differentiation of normal B cells. This suppressor activity was sensitive to 3000 rads irradiation. His B cells also failed to differentiate into Ig-producing cells with the help of normal T cells and PWM. Thus, in this patient, the decrease of Ig-synthesis in vitro could be attributed to suppressor T cells, lack of T helper cells and an intrinsic B cell defect. Therefore, this patient appears to have immunological abnormalities which differ from previously reported IL patients.

[A clinical study on clarithromycin especially on its granular form for children in pediatrics].

Clarithromycin (TE-031, A-56268), a new macrolide antibiotic, was administered to a total of 25 child patients (age range: 4 months-12 years) to treat infections; the patients consisted of 23 children who received the drug in the form of granules for children and 2 patients who were given tablets. Daily dosages were 14.3 mg/kg (in 2 divided doses) in 1 patient, and 18.3-30.3 mg/kg (in 3 divided doses) in the other 24 patients. Lengths of administration ranged from 4 to 13 days with 6-8 days for 18 of the patients. The results obtained are summarized as follows. 1. Clinical efficacy evaluations for various infections were as follows: 1 excellent, 4 good and 2 fair cases in 7 cases of pertussis; 3 excellent, 2 good and 1 fair cases in 6 cases of tonsillitis; 1 good case in tonsillitis with bronchitis; 1 excellent and 3 good cases in 4 cases of bronchitis; 1 excellent and 1 good cases in 2 cases of pneumonia; and 4 excellent and 1 good cases in 5 cases of enteritis. Thus, evaluations of a total of 25 patients showed 10 excellent, 12 good and 3 fair cases, with no poor cases. The overall clinical efficacy rate was, therefore, 88.0%. These results were attributed to good antimicrobial activity and absorption of TE-031. 2. Causative bacteria were isolated from 12 of the patients. Bacteriological efficacies of TE-031 on those strains were investigated with the following results. Of 5 Haemophilus influenzae strains, 3 were eliminated, 1 was reduced, and no change was observed in the other.(ABSTRACT TRUNCATED AT 250 WORDS)

Gamete physiology and timing of ovulation and fertilization in dogs.

Ovulation in most bitches occurs between 48-60 and 72 h from the onset of oestrus. The eggs are ovulated as primary oocytes and are not capable of being fertilized until about 60 h after ovulation when they undergo the first meiotic division to become secondary oocytes. By this time the ova have descended through two-thirds of the oviduct. Ova are fertilizable from the time of undergoing the first meiotic division at 60 h after ovulation until about 108 h after ovulation, i.e. for a period of about 48 h. The period during which the spermatozoa remain capable of fertilization in the female reproductive tract is thought to be about 5 days. The mating period during which conception is possible is thus about 7 days from the onset of oestrus.

[Impedance analysis using the random noise oscillation method and a body box].

The frequency characteristics (at 10-40 Hz) of airway (Za) and tissue (Zt) impedances in patients with chronic bronchitis (CB), chronic pulmonary emphysema (CPE) and interstitial pneumonitis (IP) were examined using the random noise oscillation method and a body box. Compared with normal subjects, Zar (r:real part) was elevated in cases of CB, but not in those with CPE. Zai (i:imaginary part) was decreased in cases of CB and CPE. Applying an electromechanical analogy to these results, it was inferred that CB increases both central and peripheral airway resistance, while CPE only increases peripheral resistance. In CPE, but not in CB, Ztr fell; only Zti decreased in IP, which suggests a decrease in mass per unit of lung tissue in CPE and a loss of pulmonary compliance in IP. We also measured the impedance of a dog with acute pulmonary edema induced by oleic acid. Zar an Ztr were increased but Zti was decreased, which corresponded with the increase of airway resistance due to hypersecretion and the decrease of lung compliance. We conclude that this method is useful in differentiating interstitial disorders from COLD, and also CB from CPE, because it can detect lung stiffness and can identify sites of airway narrowing.

[A clinical study on cefteram pivoxil granule in the field of pediatrics].

The clinical effectiveness of cefteram pivoxil (CFTM-PI) granule, a new oral cephalosporin, was studied in pediatric patients. The results are summarized as follows. 1. CFTM-PI was given orally to 17 children in daily doses of 9.5 to 31.8 mg/kg in 3 to 4 divided portions for 2 to 10 days. Clinical evaluations were made on 14 patients. Clinical effects of CFTM-PI were excellent in 4, good in 5 of 9 patients with tonsillitis or pharyngitis, excellent in all cases of 2 patients with pneumonia, 1 patient with scarlet fever and 1 patient with pyelonephritis, and fair in 1 patient with purulent cervical lymphadenitis. Overall clinical effects were excellent in 8, good in 5, and fair in 1 with an efficacy rate of 92.9%. 2. No side effects were observed in any of the 17 patients. Hematological tests showed a slight elevation of blood platelet counts in 1 patient. 3. The taste and odor of CFTM-PI granule were well accepted by the children. 4. CFTM-PI is a useful oral antibiotic for the treatment of bacterial infections in pediatrics.
