Cytotoxicity and antitumor activities of fungal bis(naphtho-gamma-pyrone) derivatives.

Cytotoxicities of twenty-seven fungal naphtho-gamma-pyrone derivatives to KB cells were examined. Chaetochromins A-D (1-4) and ustilaginoidins D (10) and E (11) exhibited strong effects and the structure-activity relationships are discussed. The antitumor effects in vivo of some of these compounds were examined but, due to their toxicity and to their marginal activity, development as antitumor agents was abandoned. Deoxyribonucleic acid, ribonucleic acid and protein synthesis in KB cells were equally inhibited by chaetochromin A (1), cephalochromin (5), and ustilaginoidin A (7) in parallel with the cytotoxicity of the drugs. Thus it was suggested that other still unknown mechanism(s) might induce cytotoxic lesion, resulting in the inhibition of the macromolecule syntheses.

[Evaluation of postoperative hepatobiliary function by deconvolution analysis of hepatobiliary image data by 99mTc-N-pyridoxyl-5-methyltryptophan].

Dynamic hepatobiliary image data were analyzed by the mathematical deconvolution to derive the transfer function (TF) which represents the impulse response function of the liver following direct bolus injection of a tracer into the liver. Biliary flow was evaluated by TF in patients with previous hepatobiliary surgery to detect abnormal flow causing problems such as cholangitis. Serial image data were collected for 60 min after intravenous injection of 37-75 MBq (1-2 mCi) of 99mTc-N-pyridoxyl-5-methyltryptophan (99mTc-PMT). TF was obtained by the matrix algorithm, using regional hepatogram as output and time-activity curve over the heart as input function. Minimum, mean and maximum transit time (Max. TT) were determined from TF. The functional image for each of the transit times was constructed by displaying the distribution of the values for matrix elements with a color scale. In this study, a critical Max. TT of 60 min was used to detect abnormal biliary flow. Of 30 cases, 9 positive cases were discriminated from 21 negative cases on the basis of the finding of presence or absence of areas with Max. TT of more than 60 min anywhere in the liver. Bowel activity overlapping the liver was excluded in the judgment. The validity of this method was evaluated in comparison with the clinical courses after surgery with (8 cases) or without (22 cases) problems. The results were sensitivity of 88%, specificity of 91%, accuracy of 90% and positive predictive value of 78%.(ABSTRACT TRUNCATED AT 250 WORDS)

[Continuous superselective intra-arterial infusion with BUdR and 5 FU in combination with radiotherapy in the treatment of head and neck tumors].

Since 1984, a new method for intra-arterial administration of anti-cancer drug and radiosensitizer, named 'superselective continuous intra-arterial infusion therapy' has been used in combination with simultaneous irradiation, for the treatment of 67 patients with the head and neck tumors. Under fluoroscopy, a preshaped catheter was introduced retrogradely from the superficial temporal artery and inserted superselectively into the main feeding artery of the tumor. Through this catheter, continuous intra-arterial infusion chemotherapy was done. The primary therapeutic effects were appreciated as complete or partial response in all cases. It was considered that this method was more effective and with less adverse reactions for the treatment of the head and neck tumors compared with the conventional method.

[A case of radiation induced aneurysm of extracranial carotid artery].

An unusual case of post-irradiation aneurysm of extracranial internal carotid artery is presented. A 70-year-old man, complaining of left cervical throbbing mass with focal pain, was admitted on February 8, 1985. It was noted, from his past history, that he had had surgery of the removal of cervical lymph-nodes and that unknown dosage of irradiation had been added to the cervical region 30 years before. Left carotid angiography (on admission) demonstrated a giant aneurysm in the cervical portion of internal carotid artery. Right carotid angiography with compression of left carotid artery revealed good cross filling through anterior communicating artery. Computed tomography with contrast media showed a ring like enhanced mass, which was thought to showed a ring like enhanced mass, which was thought to suggest that a large part of the aneurysm was filled with intraluminal thrombosis. During 30 days of evaluation, the aneurysm grew larger and his cervical pain became untolerable. Operation, the resection of the aneurysm and the reconstruction (of circulation) with vein graft, was challenged n March 12. It was so difficult with meticulous work that the ligation of left common carotid artery was performed after all. Seven days after the operation, he suffered from the gastrointestinal bleeding, which was enough to lead him to hypovolemic shock. Thereafter, right hemiparesis and aphasia were brought about. Somatotopic organization of facial nucleus of rabbit. With particular reference to intranuclear representation of perioral branches of the facial nerve.

Somatotopic organization of the facial nucleus was examined in the rabbit by the retrograde HRP (horseradish peroxidase) method; HRP was applied to the peripheral branches of the facial nerve. The facial nucleus is cytoarchitectonically divided into 5 division: the ventromedial, medial, dorsal, lateral and intermediate divisions. The ventromedial division contains neurons supplying the cervical branch. The medial division supplies the anterior auricular branch as well as the posterior auricular branch. The dorsal division is small and contains motoneurons innervating the periorbital regions through the zygomatico-orbital branchlets as well as the anterior auricular branch. Motoneurons innervating the perioral regions are numerous and distributed in the lateral and intermediate division; each of the lateral and intermediate divisions supplies both superior labial and inferior labial branches.

Immunohistochemical demonstration of coexistence of enkephalin-and substance P-like immunoreactivities in axonal components in the lumbar segments of cat spinal cord.

Coexistence of enkephalin-like and substance P-like immunoreactivities (ENK-LI and SP-LI) was shown in axonal components in the lumbar segments of cat spinal cord by using the double immunofluorescence method. Although axonal components labeled with both ENK-LI and SP-LI were distributed throughout the spinal gray, they were most dense in the ventral horn, and most sparse in the superficial part of the dorsal horn.

Representation of the main branches of the facial nerve within the facial nucleus of the Japanese monkey (Macaca fuscata).

The facial nucleus of the Japanese monkey was divided cytoarchitectonically into the ventral, medial, intermediate, dorsal and lateral divisions. When horseradish peroxidase (HRP) was applied to the inferior labial, cervical or posterior auricular branch of the facial nerve, HRP-labeled neurons were seen in the lateral, ventral or medial division of the facial nucleus, respectively. After applying HRP to the anterior auricular-zygomatico-orbital branch, labeled neurons were observed mainly in the intermediate and dorsal divisions. HRP applied to the superior labial branch labeled neurons within the dorsal and lateral divisions.

Opioid mechanisms involved in the slow potential change and neuronal refractoriness during cortical spreading depression.

The slow potential change (spc) accompanying spreading depression (SD) was studied in rats and in a seizure-sensitive strain of Mongolian gerbil under three different experimental paradigms, each involving the use of naloxone. Gerbils undergoing electroconvulsive shock treatment displayed SD during the post-ictal phase, which was blocked by the intraperitoneal (i.p.) administration of naloxone (20-50 mg kg-1). Topical application of naloxone to the exposed cortex of the anaesthetized gerbil and rat blocked the spc of SD evoked by KCl. Microiontophoretic ejection of naloxone during extracellular recordings reversed cell refractoriness following the spc, demonstrated by the observation of a maintained sensitivity to iontophoretic pulses of glutamate. The results suggest a possible involvement of naloxone-sensitive processes in the mechanism responsible for cortical SD.

Appearance of frontal mid-line theta rhythm during sleep and its relation to mental activity.

Fm theta occurred not only during mental tests in the waking state but also during nocturnal sleep in which it was most frequent during REM and secondarily most frequent during stage 1 of NREM sleep. As for the inner experience of the subjects, they frequently reported dream and distinct mentations on awakening from REM sleep whether or not it was accompanied by Fm theta. They reported dream and distinct mentations more frequently when awakened from stage 1 of NREM sleep with Fm theta than when awakened from stage 1 of NREM sleep without Fm theta. Relationship of Fm theta to mental activity during sleep was briefly discussed.

Synthesis and antitumor activity of tropolone derivatives. 4.

Modifications of monotropolone 2 having poor potency against P388 in mice were studied. The alpha-ethoxy group of 2, prepared from hinokitiol and benzaldehyde diethyl acetal, was replaced with a phenolic or heteroaromatic compound by heating 2 with the appropriate nucleophile. Structure-activity relationships indicated that an acidic hydroxyl and a proton-accepting group situated in the neighboring position, which permits the formation of a chelate with a metal ion, contributed to enhanced activity. Among the compounds studied, the 8-hydroxyquinoline analogue 10f was the most favorable compound.
