[Body acceleration measurement and oxygen uptake during jumping].

The integral of the positive acceleration per unit time was defined as physical activity. The device that measured physical activity during jumping was developed and the relationship between physical activity by acceleration measurement and energy expenditure was evaluated. Acceleration wave forms of back were measured with strain gauge type accelerometer. After amplification and rectification, the positive acceleration wave forms integrated. When the output of the integrator reached the threshold of comparator, the output of the comparator contained trigger signal. The physical activity was determined by the total counts of the trigger signal. 10 males jumped at four jumping cycles (20, 40, 60 min-1 and random) and 24 children jumped at random jumping. Oxygen uptake, physical activity and heart rate were simultaneously measured. The results showed the oxygen uptake, physical activity and heart rate of males were linearly related to jumping cycle. However, oxygen uptake of random jumping was less than one of 60 min-1 jumping in spite of the greater physical activity. This result indicated that the random jumping was operated efficiency due to the effect of elastic bounce. The oxygen uptake/body weight of males had good correlation to physical activity (r = 0.922), and heart rate (r = 0.902). Also the oxygen uptake/body weight of males and children had good correlation to physical activity (r = 0.953). The difference of ages and weights was not significant during jumping. These results mentioned to be useful in providing an objective evaluation of the physical activity of human subjects during jumping.

Folate turnover in chronically alcoholic monkeys.

To study the pathogenesis of folate deficiency in chronic alcoholism, monkeys were fed nutritionally complete liquid diets containing 50% of energy as ethanol for 4 years. The initial excretion and subsequent turnover of a tracer dose of intramuscular 3H-folic acid was measured during a 25-day period prior to autopsy. Within the first 3 days, the excretion of tritium in the urine and feces was significantly greater in the ethanol-fed monkeys than in their pair-fed controls, and most of the folate recovered in the urine of the third day was unchanged 3H-folic acid. Analysis of total body tritium retention revealed a rapid phase of elimination (0 to 3 days), in which the half-time of elimination was shorter in the ethanol-fed group, and a slow phase (4 to 25 days) with similar half-times of elimination. At autopsy, the levels and total amounts of endogenous folates in the liver were decreased in the ethanol-fed monkeys, as was the recovery of tritium. These studies suggest that folate deficiency in chronic alcoholism is caused in part by increased urinary and fecal losses and resultant decreased hepatic incorporation of exogenous folate whereas long-term folate turnover is unchanged.

Separation of mono-, di-, and trihydroxy stereoisomers of bile acids by capillary gas-liquid chromatography.

Capillary gas-liquid chromatographic separation was studied for the complete set of the 26 theoretically possible isomers of mono-, di-, and trihydroxylated 5 beta-cholanic acids, which differ from one another in the number, position, and configuration of hydroxyl groups at C-3, C-7, and/or C-12 in the nucleus, as well as for some of their related acids. The bile acid samples were chromatographed as their methyl ester-trimethylsilyl (TMSi) ether derivatives and analyzed on three capillary columns coated with nonpolar OV-1, slightly polar OV-17, and polar SP-2340 as liquid phases. The retention times on capillary gas-liquid chromatography (GLC) responded dramatically to the minor structural differences, and almost complete separation of the positional and stereochemical isomers was achieved by the combined use of SP-2340 and OV-17 (or OV-1) capillary columns.

Long terminal repeat (LTR)-derived recombination of retroviral DNA: sequence analyses of an aberrant clone of baboon endogenous virus DNA which carries an inversion from the LTR to the gag region.

Nucleotide sequences of a cloned proviral DNA of baboon endogenous virus M7 were analyzed, which carried an internal inversion. The inversion of 2.2 kilobase pairs was occurred between the junction of two tandem LTRs and a site locating in the p30 region of the gag gene. The ATAA sequence was a target for recombination generating the inversion, which was duplicated at both ends of the inverted segment. AAA and CA were lost at the 5'- and 3'-ends of the LTRs by the inversion, respectively. On both sides of the target sequence, long AG-rich stretches were detected, which may specify the site of recombination together with the target sequence. The characteristic base changes in the inversion are concluded to result from an illegitimate recombination associated with LTRs, as well as in case of provirus integration into the host cell DNA. We propose and discuss models to explain the processes of recombination to generate both inversion and integration.

[The ovulation-inducing effect of CB-154 on normoprolactinemic anovulatory subjects (author's transl)].

Several investigators have reported that CB-154 induces ovulation in patients with normoprolactinemic anovulation as well as those with hyperprolactinemic amenorrhea. In the present research, the ovulation-inducing effects of CB-154 were studied in normoprolactinemic subjects with special reference to the feedback effect of estradiol on LH release. Thirty female subjects aged 20 approximately 32 years with ovulatory disturbances were studied. Basal serum PRL, LH and FSH were determined by radioimmunoassay, and both hyperprolactinemic and hypergonadotropic anovulatory patients were excluded. A 2mg dose of estradiol benzoate was administered intramuscularly to each subject and 8ml samples of venous blood were taken at 0, 6, 24, 30, 48, 54, 72, 78 hr. The subjects under study were divided into two groups, A and B, according to the effect the estradiol benzoate had an LH release. Group A subjects (nine in all) failed to show any positive feedback release of LH in response to the estradiol benzoate. Group B subjects (twenty-one in all) showed a more than twofold increase in circulating LH as compared with the initial serum LH value, and this was taken as an indication of positive feedback release. All the subjects in group B were given clomiphene (50 approximately 100mg daily for five days). The clomiphene therapy was effective in eleven subjects, and four became pregnant (three in the first or second cycle of treatment and one in the third). The therapy was ineffective in the remaining six subjects, four of whom were diagnosed as suffering from polycystic ovary syndrome. Clomiphene was judged to be effective when the subjects undergoing therapy with this drug ovulated during three successive treatment cycles, and ineffective when the subjects did not fulfill this criterion (criterion for effectiveness of clomiphene). With the exception of four cases of polycystic ovary syndrome and three pregnancies which occurred in the first or second cycle, the rate of effectiveness of clomiphene in group B was 12 out of 14. It was concluded from these results that clomiphene was effective in group B subjects except in cases of polycystic ovary syndrome. Treatment with clomiphene alone was effective in none of the seven subjects in group A. However, administration of CB-154 for several weeks prior to and during the clomiphene treatment cycle (combined therapy of CB-154 and clomiphene) led to remarkably improved ovulation rates in five subjects in group A. Four patients in group A were selected, and given estradiol benzoate prior to (control) and during (study) CB-154 administration. In each case administration of CB-154 elicited marked positive feedback release of LH as compared with the control period, that is to say, CB-154 transformed group A patients into group B patients. This effect of CB-154 may explain why therapy combining CB-154 and clomiphene improved ovulation rates in group A...

[Calf blood flow at rest evaluated by thermal measurement with tissue temperature and heat flow and 133Xe clearance (author's transl)].

The regional blood flow in the calf was determined simultaneously by thermal measurement and by 133Xe clearance technique. Calf blood flow (Ft) by thermal measurement was accounted for by the equation of the form Ft = (CdTd + Ho - Mb)/rho bCD (Ta--Td), where Cd is thermal capacitance of the calf compartment, Td is the change of calf tissue temperature, Ta is arterial blood temperature, Td is calf tissue temperature, Ho is the heat dissipation from the compartment to the environment, Mb is estimated metabolism of the calf tissue and rho bCb is the product of density and specific heat of blood. The healthy men were chosen for the experiments. Total calf blood flow was 2.53 +/- 1.31 ml/(min-100 ml calf), and muscle blood flow was 2.63 +/- 1.69 ml/(min-100 ml muscle) and skin blood flow 7.19 +/- 3.83 ml/(min-100 ml skin) measured by 133Xe clearance. On the basis of the results, an estimate has been made of the proportions of the calf volume which can be ascribed to skin and muscle respectively. Estimated muscle and skin blood flow were correlated with total calf blood flow (r = 0.98).

Monoclonal antibody against the carboxy terminal peptide of pp60src of Rous sarcoma virus reacts with native pp60src.

A monoclonal mouse antibody has been prepared against a synthetic peptide corresponding to the six carboxy-terminal amino acids (C' peptide) of the src gene product pp60v -src of Rous sarcoma virus (RSV). The antibody was able to precipitate pp60v -src and to bind pp60v -src kinase activity in a competition test, indicating that this peptide can serve as an antibody-binding site (epitope). Furthermore, the finding that three out of 28 pp60src-specific tumor-bearing rabbit (TBR) sera contained antibody against the C' peptide argues for an in vivo role for the carboxy terminus of pp60src. C' peptide-specific IgG was purified from one TBR serum using affinity chromatography, and was shown to precipitate significant amounts of pp60src, and bind most of the pp60src kinase activity from SRA, PrA, and B77-C strains of avian sarcoma virus (ASV), but not endogenous pp60c -src, a cellular homologue to the viral pp60v -src. Similar results were obtained with IgG isolated from a C' peptide immune rabbit serum. None of the three C' peptide-specific IgGs could serve as a phosphate acceptor in an immune complex protein kinase reaction.

Overlapping genes in the heat-labile enterotoxin operon originating from Escherichia coli human strain.

We have determined the nucleotide sequence at the distal end of the heat-labile enterotoxin subunit A (LT-A) gene (toxA) originating from human enterotoxigenic Escherichia coli. The sequenced region covers the entire LT-A2 region and a part of the LT-A1 region. In confirming our previous prediction based on product analysis of clones toxA regions, the data suggest the overlapping of the distal end (5'-TTA TGA) of toxA with the proximal end (5'-ATG AAT) of the LT subunit B gene (toxB), in the sequences 5'-TTATGAAT. Some additional characteristics of the LT operon as well as of the products are discussed.

Amoxicillin concentrations in serum, jaw cyst, and jawbone following a single oral administration.

Forty-four patient who underwent enucleation of jaw cysts were administered amoxicillin preoperatively. Specimens of venous blood (44), walls and fluids from periodontal (31) and dentigerous (13) cysts, and jawbone (26) were obtained during the operation and assayed for amoxicillin content. Measurable concentrations were found in all specimens. The levels were higher in periodontal cysts than in dentigerous cysts, and higher in maxillary bone than in mandibular bone. Since amoxicillin can easily and rapidly pass through the epithelial lining according to the change in concentration, the penetration by amoxicillin of blood/cyst wall/cyst fluid probably depends on simple diffusion.
