Entamoeba histolytica: inhibition in vitro by bithionol of respiratory activity and growth.

Endogenous and 2-propanol-supported respiration of intact trophozoites of of Entamoeba histolytica (stain HM-1:IMSS) were inhibited by bithionol, an effective chemotherapeutic agent for some trematode and cestode infections in humans. Dichlorophene and hexachlorophene also inhibited 2-propanol-supported respiration of the parasite. In contrast, ethanol formation by E. histolytica extract in the presence of N2 was scarcely inhibited by bithionol. The compound also inhibited in vitro growth of axenic (HM-1 strain) and polyxenic (strain HJ-1:KEIO) amoebae in culture. It took less than 24 hr to kill and disrupt virtually all amoebae of either strain with 0.28 mM bithionol. Omission of bovine serum from BI-S-33 medium resulted in considerably less disruption of HM-1 strain amoebae by the compound. However, organisms that looked undisrupted were strained with trypan blue. Moreover, the number of amoebae incubated for 10 min in the serum-free BI-S-33 medium containing 0.14 mM bithionol did not increase, even after incubation for 24 hr following replacement of the experimental culture fluid with fresh complete BI-S-33 medium free of the compound. These findings suggest that, although serum appears to diminish the antiamoebic action, some halogenated bisphenols (in particular bithionol) may be useful for treatment of amoebiasis.

The efficient use of microcolumns in chromatographic systems.

Microcolumn techniques involving pre-column concentration and post-column derivatization make good use of micro-scale high-performance liquid chromatography. The efficient use of microcolumns is illustrated by some applications to the analysis of amino acids, bile acids and catecholamines. The potential of new detector techniques based on the multichannel photodiode array detector, and on immobilized enzyme post-column reaction systems with fluorimetric detection, is also discussed.

Development of human fetal muscles: a comparative histochemical analysis of the psoas and the quadriceps muscles.

The development of human muscle can be divided into three distinct phases on the basis of the enzyme histochemistry (Dubowitz 1965). On the other hand, muscles can be classified into trunk muscles innervated by the motoneurons in the anteromedial nucleus of the anterior horn and limb muscles innervated by those of the lateral nucleus. We compared histochemically the development of the psoas muscle as a trunk muscle with the quadriceps. Four legally aborted fetuses and sixteen autopsied preterm or full-term infants with conceptional ages ranging from sixteen to fourty-two weeks, served as subjects. Serial crossly cut frozen sections of the psoas and the quadriceps were stained with HE, modified Gomori-trichrome, NADH-TR, myofibrillar ATPase (routine, preincubation at pH 4.6, 4.3), and analyzed quantitatively. In the psoas and the quadriceps, all muscles consisted of undifferentiated type IIC fibers before the eighteenth week of gestation. Type I fiber began to appear around the eighteenth week in both muscles. Before the thirtieth week, both muscles comprised a great number of smaller type IIC fibers and scattered large type I fibers. The psoas had already a little more type I fibers than the quadriceps in this phase. After the thirtieth week, the percentage of type I fibers increased greatly to about 50% at term in the psoas, gradually to 20-23% at term in the quadriceps. Type IIA and IIB fibers appeared suddenly around the thirty-seventh week. Total percentage of these two type fibers increased at term to about 50% and 70% in the psoas and the quadriceps, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Spontaneous resolution of chronic epidural hematomas: report of three cases.

We report three cases of the spontaneous resolution of a traumatic chronic epidural hematoma. The patients were young adults with only mild headache after trauma. All patients were treated conservatively because of the very mild symptoms and the time lapse of 1 to 2 weeks between trauma and our unexpected detection of the frontal epidural hematomas. Slow, spontaneous resolution of the hematoma occurred in all three cases.

Study on serum antistreptococcal esterase among school children.

Antistreptococcal esterase (an antibody to extracellular esterase produced by Group A streptococci: ASE) was evaluated according to Hayano's method by using an assay kit on sera of children from two schools, one (F-school) in a non-epidemic state and the other (I-school) in an epidemic state of Group A streptococcal infection. Out of 104 children of F-school, 51 with negative throat cultures for Group A streptococci showed a mean serum ASE titer of 191 +/- 144 unit, and 53 with positive throat cultures for Group A streptococci showed a mean serum ASE titer of 360 +/- 147 unit. Follow-up determinations on serum antibodies to four Group A streptococcal antigens were performed on the children from I-school during a period of 13 months after an epidemic infection of Group A streptococci. The initially elevated serum ASE titer showed a decreasing trend on subsequent bleedings during the study period as well as antistreptolysin O (ASO) and antideoxyribonuclease B (ADN-B) titers. However, the changing pattern of the antistreptococcal polysaccharide (ASP) titer was different from that of the other three antibodies. A positive correlation was observed statistically between ASE and ASO titers, and also between ASE and ADN-B titers. The ASE tests appear to be useful for serological evidence of a streptococcal infection.

No evidence of direct cardiac action of leucine-enkephalin in canine heart-lung preparation.

Intravenous administration of leucine-enkephalin (30 micrograms/kg) caused significant decreases in mean blood pressure, cardiac output, and left ventricular dP/dt in intact dogs. However, cardiovascular effects of leucine-enkephalin (30-120 micrograms/kg) did not occur in the heart-lung preparation uninfluenced by extracardiopulmonary factors. These results provide no evidence of a direct cardiac action of leucine-enkephalin.

Blood mobilization by regional vascular beds during cerebral ischemic pressor response in rabbits.

The mobilization of blood volume from regional vascular beds during cerebral ischemic pressor response was studied in anesthetized rabbits. The visceral beds of the kidney, jejunum, and liver served as volume reservoirs from which blood was mobilized during cerebral ischemia. The renal bed gave the largest volume decrease, from 24 to 13%. The magnitude of weight change in the renal and intestinal regions varied in parallel to the volemic state of the animal. Superimposition of pressure oscillation on the systemic pressor response was created by progressive restriction of the blood supply to the brain. Regional tissue weights also oscillated with a period equal to that seen in arterial pressure but almost 180 degrees out of phase. Such variations gave evidence of active venoconstriction in the reservoir response of all regional beds except the hind paw. The responses noted in the hind paw were passive except during complete restriction of the blood flow to the brain. Autoregulation of skeletal muscle and especially liver volume was present with moderate elevations in systemic arterial pressure.

Pharmacological evidence that central alpha 2-adrenoceptors are involved in cardiovascular responses to intravenously injected enkephalins in anesthetized dogs.

In anesthetized dogs, intravenous infections of 30 micrograms/kg of leucine- and methionine-enkephalin (L- and M-enk respectively) produced a significant reduction of mean blood pressure (MPB) and heart rate (HR). The peak decreases in MBP and HR occurred within 1 min after injection and recoveries to pre-injection levels within 3 min. The depressor responses to both L-enk and M-enk were abolished by intracisternal pretreatment with yohimbine (0.5 mg/kg) but not with prazosin (0.1 mg/kg). Pretreatment with either drug did not alter the bradycardia. All cardiovascular effects of both L-enk and M-enk were prevented by intravenous pretreatment with naloxone (2 mg/kg). These results suggest that central alpha 2-adrenoceptors may participate in the central actions of enkephalins on blood pressure regulation. HR responses to enkephalins may be elicited by a different mechanism.

Cellular uptake and efflux and cytostatic activity of 4'-O-tetrahydropyranyladriamycin in adriamycin-sensitive and resistant tumor cell lines.

Cellular uptake and cytostatic activity of 4'-O-tetrahydropyranyladriamycin (THP) in various sublines resistant to anthracycline antibiotics of mouse lymphoblastoma L5178Y, Chinese hamster ovary (CHO) and mouse leukemia P388 cells were studied. All the sublines resistant to adriamycin (ADR) showed slightly decreased uptake of THP as compared with each sensitive lines, but THP was still taken up much more quickly than ADR by each of the ADR-resistant cell lines. Efflux of both anthracycline glycosides from the ADR-resistant P388 cells was faster than that from the ADR-sensitive P388 cells. The percentage of THP retained at equilibrium was higher than that of ADR in both ADR-resistant and -sensitive P388 cells. Cytotoxicity of THP to ADR-resistant cell lines was considerably lower compared with that for each of the sensitive lines but THP inhibited growth of ADR-resistant tumor cells at a concentration about 10 times lower than that for ADR. Thus THP was taken up more quickly, effluxed more slowly than ADR from the ADR-resistant cells, and showed stronger cytostatic activity than ADR on the cells.
