Characterization of phage-Xp10-coded RNA polymerase.

A bacteriophage-coded RNA polymerase was isolated from bacteriophage-Xp10-infected Xanthomonas campestris pv. oryzae. The enzyme was purified to homogeneity through precipitation by poly(ethylene glycol) and chromatography on DEAE-cellulose, heparin--Sepharose 4B and blue-dextran--Sepharose 4B. It is composed of a single polypeptide of Mr96,000. The enzyme preferred denatured Xp10 DNA, calf thymus DNA, host bacterium DNA and poly[d(A-T)] as templates. The optimal concentration of MgCl2 is 16 mM. The optimal temperature and pH are 37 degrees C and 8.0, respectively. The Km of ATP is 26 microM. DNA, MgCl2 and four ribonucleotides were required for enzyme activity. If ATP alone was present, half of the Xp10 RNA polymerase activity was retained. The enzyme activity was inhibited by KCl, spermidine, actinomycin D, heparin, blue dextran and ethidium bromide; it was resistant to rifampicin and streptovaricin. N-Ethylmaleimide did not affect the enzyme activity. The transcription site and product of Xp10 RNA polymerase upon Xp10 DNA were analyzed by DNA/RNA hybridization and polyacrylamide-agarose composite gel electrophoresis. The enzyme could specifically transcribe the late region of Xp10 genome and produce two RNA bands.

Tinea infection histologically simulating eosinophilic pustular folliculitis.

A biopsy taken from an annular eruption on the thigh of an infant boy showed features of Ofuji's eosinophilic pustular folliculitis, yet fungal hyphae were demonstrated in the lesion. This case is reported to illustrate that tinea infection can stimulate eosinophilic pustular folliculitis histologically.

Charcot-Leyden crystals in pemphigus vegetans.

In the skin lesion of a patient with pemphigus vegetans, Charcot-Leyden crystals were observed in the eosinophilic abscess cavities of the epidermis. The finding is documented for the first time. In addition, some of the intraepidermal eosinophilic abscess cavities have been shown to be due to accumulation of eosinophils in dilated acrosyringia, but their formation from acantholysis and spongiosis can not be entirely excluded.

Adult T-cell leukemia/lymphoma in Taiwan. A clinicopathologic observation.

The retrovirus-associated adult T-cell leukemia/lymphoma (ATL) has not been previously documented in Taiwan. Five cases identified recently by the authors are reported. Three of the patients were women, and their ages ranged from 36 to 60 years. The most important diagnostic clue was the observation of polylobated lymphoid cells in the peripheral blood. Other variably observed significant features included hypercalcemia, cutaneous eruptions, osteolytic bone lesion, hepatomegaly, and lymphadenopathy. Surface marker studies revealed that the leukemic or lymphoma cells were T-helper cells. Histopathologic examination revealed one case of pleomorphic type and three cases of medium-sized cell type. No tissue was available for study in one case. The diagnosis of ATL was confirmed by the indirect immunofluorescence test on MT-1 cell for antibodies to adult T-cell leukemia virus-associated antigen (ATLA). Three patients were dead within 6 months, and two patients had been in clinical remission for 7 and 10 months, respectively. These two latter cases were similar to the so-called smoldering type of ATL. Two descendents among nine relatives of the patients were also positive for anti-ATLA (22%). Two husbands were negative. Four of the five patients lived in the same county in northeastern coastal Taiwan, which suggested a possible clustering of ATL in that region.

Serological survey of antibodies to the adult T-cell leukemia virus-associated antigen (HTLV-A) in Taiwan.

A serological survey of antibodies to the adult T-cell leukemia virus-associated antigen (HTLV-A) conducted in Taiwan revealed II seropositive individuals among 2,565 serum donors tested, which included 1,187 hospital or free clinic patients, 1,065 blood donors and 313 aborigines. The overall prevalence rate was 0.43% and was 0.90% (9/995) for adults aged 40 years or over, which was similar to the rates observed in the non-endemic areas in Japan. No highly prevalent area was demonstrated. The II seropositive individuals identified were 8 hospital or clinic patients, 1 blood donor and 2 aborigines. Among 108 various leukemia/lymphoma patients also studied, 7 were positive. These 7 anti-HTLV-A-positive patients also had clinicopathologic features of adult T-cell leukemia/lymphoma (ATL). Among 21 relatives of 3 seropositive individuals in the epidemiologic study, 2 were positive; and among 25 relatives of 5 ATL patients, 6 were positive. Our data support the contention that horizontal familial transmission of human T-cell leukemia virus (HTLV-I) is an important route and HTLV-I infection can occur on a different genetic background. Furthermore, the antibody titer did not correlate with the manifestation or the state of the disease.

Histiocytoid hemangioma of the heart with peripheral eosinophilia.

A histiocytoid hemangioma of the heart is reported, which was found incidentally in a man with unusually high eosinophilia. The eosinophilia subsided dramatically following removal of the tumor. The "histiocytoid" or the "epithelioid" appearance of the tumor cells and the presence of vacuolated cells were the characteristic microscopic features. The endothelial origin of this tumor was verified by positive immunostaining for factor VIII-related antigen and ultrastructural demonstration of intracytoplasmic lumen formation, abundant cytoplasmic filaments, pinocytotic vesicles, and prominent basal lamina. The presence of mitotic activity, cellular pleomorphism, and tumor necrosis raised the possibility of its malignant potential. The occurrence of this tumor in the heart may be mistaken for a myxoma clinically and a metastatic carcinoma pathologically.

Cutaneous manifestations of adult T cell leukemia/lymphoma. Report of three different forms.

The clinical and pathologic features of cutaneous lesions observed in three adult T cell leukemia/lymphoma (ATL) patients identified in Taiwan are described. They represent one classical case of ATL and two "smoldering" variants. The classical ATL patient when first seen had numerous erythematous or purpuric papules, nodules, and plaques with or without ulceration. The two "smoldering" cases developed encrusted purpuric plaques with subcutaneous erythematous nodules in one patient and pompholyx-like vesicular eruptions with tumor masses in another patient. The pompholyx-like eruptions have not been described before. Pruritus was the major complaint in two patients. Histopathologic studies revealed pleomorphic infiltration in the classical case and monomorphic infiltration with medium-sized cells in the two "smoldering" variants. Therefore, the cutaneous lesions of ATL are diverse and not pathognomonic. The diagnosis requires a high index of suspicion, detection of circulating characteristic multilobated lymphoid cells with T helper/inducer cell marker, and demonstration of serum antibody against the adult T cell leukemia virus-associated antigen.

Hepatocyte hepatitis B surface antigen. Diagnostic evaluation of patients with clinically acute hepatitis B surface antigen-positive hepatitis.

Patients who were diagnosed as having acute hepatitis B by conventional criteria were reevaluated by a correlative study of tissue hepatitis B surface antigen (HBsAg) and serologic tests for anti-hepatitis A virus, IgM, and/or anti-hepatitis B core antigen (anti-HBc) IgM. The results demonstrated that the diagnosis of acute hepatitis B in an endemic area was complicated by superinfection or reactivation of unrecognized hepatitis B carriers or chronic hepatitis B patients simulating acute hepatitis clinically. Anti-HBc IgM failed to discriminate newly infected acute hepatitis B from reactivation of chronic hepatitis B liver disease, especially when the enzyme-linked immunoassay index value was low. Our data suggest that a combination of different tissue HBsAg expression patterns usually indicate the presence of underlying chronic liver disease, while the tissue HBsAg was practically all negative in newly infected cases. Therefore, the study for tissue HBsAg expression by the immunoperoxidase method is an invaluable aid in identifying true acute hepatitis B in patients in an endemic area.

Solid and papillary neoplasm of the pancreas. Report of three cases from Taiwan.

During a 2-year period, three young Chinese women in Taiwan were found to have a rare pancreatic tumor known as solid and papillary neoplasm of the pancreas. They had a large abdominal mass, found either incidentally or because of abdominal discomfort. Endoscopic retrograde cholangiopancreatography was useful in localizing the tumor. Grossly, the tumors all appeared well circumscribed with focal or extensive cystic change and hemorrhage. Microscopically, they were composed of small uniform eosinophilic cells forming papillary, microcystic, and solid structures with extensive hemorrhage resulting in necrosis and formation of cholesterol granulomas. No carcinoembryonic antigen was demonstrated by the immunoperoxidase method. None of these three patients experienced recurrence more than 2 years after surgery. Even the one who received only drainage and cyst-jejunostomy has been doing well. Long survival or even cure can be expected, but the accumulated experiences indicate that this neoplasm should be regarded as a low-grade, potentially malignant tumor. One of the patients developed diabetes mellitus postoperatively.

Lymphadenopathic type of Kaposi's sarcoma presenting with generalized petechial hemorrhages. A case report from Taiwan.

An 8-year-old Taiwanese girl presenting with generalized petechial hemorrhages, severe thrombocytopenia and lymphadenopathy was found to have primary Kaposi's sarcoma of lymph node without cutaneous involvement. This patient is a first-documented case of lymphadenopathic type of Kaposi's sarcoma in Taiwan. The associated severe thrombocytopenia resulting in generalized petechial hemorrhages as the initial manifestation is also unique in this case.

Mucicarminophilic histiocytosis. A polyvinylpyrrolidone (PVP) storage disease simulating signet-ring cell carcinoma.

In Taiwan, a peculiar type of cell with mucicarmine-positive bubbly or vacuolated cytoplasm was sporadically observed in a variety of surgical specimens removed for neoplastic or non-neoplastic conditions. They closely mimicked signet-ring cell carcinoma. Study of 14 such cases and review of the related literature led to the conclusion that the peculiar cells were storage histiocytes containing polyvinylpyrrolidone (PVP). Because they were shown to be mucicarmine-positive, we designated them " mucicarminophilic histiocytes." Particulate bodies sometimes observed in association with mucicarminophilic histiocytes were believed to be the extracellular counterpart of the cytoplasmic vacuoles. Our survey also revealed that PVP-containing intravenous injection fluids had been used in Taiwan as recently as 5 years ago. For surgical pathologists, PVP-containing mucicarminophilic histiocytes should not be mistaken for signet-ring cell carcinoma or signet-ring cell lymphoma. Mucicarminophilic histiocytes can be identified by the positive mucicarmine, colloidal iron, Gomori methenamine silver, Congo red, Sudan black B, Fontana-Masson argentaffin, and Victoria blue; but negative periodic acid-Schiff, alcian blue, and Giemsa stains.

A physical map of the filamentous bacteriophage Cf genome.

Twenty-three restriction endonucleases were used to cleave the genome of bacteriophage Cf. BamHI, EcoRI, HaeII, KpnI, PstI, SalI, SmaI and SstII each cut Cf DNA at one site, HindIII, PvuI, PvuII and XhoI cleaved at two sites, Bg/II cleaved at three sites, HincII cleaved at four sites, HinfI, BstNI, AluI, HpaII and HaeIII cleaved at 14, 19, 20, 24 and 26 sites, respectively, whereas HpaI and XbaI have no cleavage site on the Cf genome. HhaI and TaqI cleaved at more than 20 and more than 8 sites respectively. Based on the sizes of the fragments produced by digestion with these enzymes, Cf DNA was estimated to have 7.62 +/- 0.1 kilobase pairs. Using the EcoRI cleavage site as a reference point, nine restriction enzymes, BamHI, Bg/II, EcoRI, HindIII, KpnI, PstI, PvuII, SstII and XhoI were selected and a detailed physical map of the Cf genome was constructed.

Mesonephroid blastoma. A previously undescribed renal tumor of childhood.

We describe a renal tumor arising in a 4 1/2-year-old boy and characterized by the formation of epithelial tubules in a primitive mesenchymal stroma. We call attention to certain unique structural features of this tumor, which set it apart from Wilms' tumor. The tissue components regularly observed in Wilms' tumor were absent from the primary neoplasm despite exhaustive search, suggesting that this case should be considered distinct from nephroblastoma. However, a metastasis appeared that was formed entirely by undifferentiated nephrogenic cells. Based on structural appearance, histogenesis from mesonephros is tentatively proposed. No precedent was found in the literature of a tumor of this description.

[Transfer of plasmid RP4 into some phytopathogenic bacteria and its relation to their virulence].

Plasmid RP4 were transferred from Escherichia coli into Pseudomonas solanacerum, Xanthomonas campestris pv. oryzae, X. campestris pv. campestris, and X. campestris pv. citri at the frequencies of 1.8 X 10(-6), 2.8 X 10(-6), 1.4 X 10(-2) and 2.0 X 10(-3), respectively. The frequencies of transfer depended on bacterial species and conjugation conditions. Treatment of bromide at the concentration of 2 micrograms/ml and acridine orange at 100 micrograms/ml for 32 hrs the plasmid RP4 could be cured from the transconjugant of P. solanacerum at the frequencies of 75% and 95%, respectively. When transconjugant of P. solanacearum was inoculated into tomato plants, the decrease in virulence was observed, furthermore, no plasmids were detected both in the wild-type strain and the cured transconjugant. It thus appears that the virulence may not be related to plasmids.
