Inhibition of tumor promotion in mouse skin by 1 alpha,25-dihydroxyvitamin D3.

1 alpha,25-Dihydroxyvitamin D3 [1 alpha,25(OH)2D3], a hormonally active form of vitamin D3, was found to inhibit the promotional phase of 7,12-dimethylbenz[a]anthracene-induced skin carcinogenesis in female Sencar mice. Topical application of 1 alpha,25-(OH)2D3 once a week at a dose of 1 micrograms or less, a tolerable dose from hypercalcemia, dose dependently inhibited tumor promotion by 12-O-tetradecanoylphorbol-13-acetate (TPA). When 1 micrograms of 1 alpha,25(OH)2D3 was applied 30 min before 5 micrograms of TPA, the times required for 50 and 100% tumor incidence were delayed about 2.5 and 7 weeks, respectively, and the number of tumors per mouse was decreased by 25-30%. This inhibitory effect was more pronounced when examined by a two-stage promotion protocol, in which a single application of 5 micrograms of TPA (Stage I) was followed by repeated applications of 5 micrograms of mezerein once a week for 19 weeks (Stage II). When 1 alpha,25(OH)2D3 at 1 micrograms was applied at Stage I + II or Stage II, tumor formation was markedly suppressed, resulting in decrease of about 70-80% in the incidence and 87-90% in the number of tumors per mouse. Application of 1 alpha,25(OH)2D3 at Stage I only did not inhibit tumor formation, indicating that 1 alpha,25(OH)2D3 specifically inhibited Stage II promotion. These results are in good agreement with the previous and present findings that 1 alpha,25(OH)2D3 inhibited induction of epidermal ornithine decarboxylase by TPA and mezerein. The possibility that 1 alpha,25(OH)2D3 suppressed tumor promotion by killing initiated cells rather than inhibiting promotion was ruled out by an experiment in which TPA was applied to the 1 alpha,25(OH)2D3 alone-treated animals.

Presence of immunoreactive dynorphin in human cerebrospinal fluid.

Immunoreactive dynorphin (I-dynorphin) was measured by radioimmunoassay in human cerebrospinal fluid (CSF). I-dynorphin concentration in CSF was 30 +/- 2 pg/ml. Sephadex G-25 column chromatography showed the main peak eluted at the position of dynorphin-(1-17). HPLC elution profile of this major peak from gel filtration showed a large peak corresponding to the position of dynorphin-(1-17) and small peaks corresponding to the positions of dynorphin-(1-13), dynorphin-(1-12) and other unknown peptides.

Serial alterations in endothelial cell shape and pattern after intraocular surgery.

We serially examined the endothelial cellular pattern and thickness of the central cornea after intracapsular cataract extraction and penetrating keratoplasty. The endothelial cellular pattern was analyzed with a computer-assisted digitizer. We used the frequency of hexagonal cells as the index to describe the degree of hexagonality in the cellular pattern. In the intracapsular cataract extraction group (20 eyes), the frequency of hexagonal cells was 68% preoperatively. During the first four weeks this frequency decreased rapidly to a significantly lower level (52%). The corneal thickness was significantly increased within the first week (0.57 mm) and steadily decreased to preoperative values at four weeks (0.53 mm). During the period from four to 24 weeks, the frequency of hexagonal cells increased gradually to 64%. In the penetrating keratoplasty group (ten eyes), the frequency of hexagonal cells and the thickness at one month were 62% and 0.64 mm respectively. During the next six months the frequency of hexagonal cells decreased significantly to 53%, and the thickness decreased to 0.53 mm. There was a gradual increase in the frequency of hexagonal cells to 62% during the two years after penetrating keratoplasty.

1 alpha, 25-Dihydroxyvitamin D3 directly induces fusion of alveolar macrophages by a mechanism involving RNA and protein synthesis, but not DNA synthesis.

The results of our present study indicate that 1 alpha, 25-dihydroxyvitamin D3[1 alpha, 25(OH)2D3] directly induces fusion of mouse alveolar macrophages without any participation of T-lymphocytes by a mechanism involving RNA and protein synthesis but not DNA synthesis. We have reported that 1 alpha, 25(OH)2D3 induces fusion of alveolar macrophages by a direct mechanism and by a spleen cell-mediated indirect mechanism [(1983) Proc. Natl. Acad. Sci. USA 80, 5583-5587]. Alveolar macrophages pretreated with or without anti-Thy 1.2 antibody and complement fused similarly when they were incubated with 1 alpha, 25(OH)2D3. The vitamin suppressed DNA synthesis, but it significantly enhanced RNA and protein synthesis. The 1 alpha, 25(OH)2D3-induced fusion was blocked by adding actinomycin D or cycloheximide, but not by hydroxyurea.

Identification of 24,25,26,27-tetranor-23-hydroxyvitamin D3 as a product of the renal metabolism of 24,25-dihydroxyvitamin D3.

By cochromatography, mass spectrometry, and chemical derivatization, we have shown that a metabolite isolated from the perfused rat kidney incubated with 24-(R),25-dihydroxyvitamin D3 is indistinguishable from chemically synthesized 24,25,26,27-tetranor-23-hydroxyvitamin D3. The new metabolite is also produced from 24-oxo-25-hydroxyvitamin D3 but not from 23(S),25-dihydroxyvitamin D3. Enzymes required for the synthesis of the new metabolite are absent in the vitamin D deplete animal but are induced along with the 25-hydroxyvitamin-D3 24-hydroxylase by vitamin D repletion. The pathway of 24,25-dihydroxyvitamin D3 metabolism in the perfused kidney is stimulated by pre-treatment of the rat with large doses of vitamin D3, suggesting that the pathway is a degradative one.

Quantitative analysis of endothelial mosaic pattern changes in anterior keratoconus.

The corneal endothelia of 21 eyes with anterior keratoconus and 15 eyes of age-matched controls were investigated with a specular microscope. Of the 21 eyes, 15 had definite keratoconus while six apparently normal fellow eyes were designated as latent. The corneal endothelial photographs were subjected to a computerized digitizer analysis of the area and shape of individual cells. The endothelium in keratoconus showed a significant increase in the extent of polymegethism (the coefficient of variation in cell area; 0.36 +/- 0.07) as compared with controls (0.24 +/- 0.03), with the mean cell area staying within normal limits. The relative frequency of hexagonal cells in keratoconus (50.5 +/- 5.7%) was significantly lower than that of controls (70.6 +/- 5.5%). Additionally, an increase of various cell shapes was noted, indicating that there was also a significant increase in cellular pleomorphism. In the latent group, alterations in cellular structure could not be detected by analysis of cell area when compared to controls. However, when these eyes were subjected to cell shape analysis and compared to fellow keratoconic eyes, both eyes showed similar pleomorphic characteristics. These results suggest that in the fellow eye in unilateral keratoconus, even when there are no ocular signs and symptoms, diagnosis can be made early with specular microscopy and computerized cell shape analysis.

A rapid and sensitive in vitro assay of 25-hydroxyvitamin D3-1 alpha-hydroxylase and 24-hydroxylase using rat kidney homogenates.

A sensitive and rapid in vitro assay of 25-hydroxyvitamin D3 [25-(OH)D3]-1 alpha- and 24-hydroxylase activities was developed using rat kidney homogenates. A potent inhibitor of the enzymes in rat plasma was removed by thoroughly perfusing rats with saline. Kidney homogenates prepared from vitamin D-deficient rats preferentially produced tritiated 1 alpha,25-dihydroxyvitamin D3 [1 alpha,25(OH)2D3] from 25(OH) [3H]D3. Addition of 10 microliter or more of rat plasma to 3 ml of 10% kidney homogenates suppressed 1 alpha-hydroxylase activity dose-dependently. Thyroparathyroidectomy (TPTX) of vitamin D-deficient rats greatly abolished 1 alpha-hydroxylase activity. Administration of parathyroid hormone to the TPTX rats increased 1 alpha-hydroxylase activity and that of 1 alpha,25(OH)2D3 enhanced 24-hydroxylase markedly. Since this assay is technically simple, rapid and sensitive, it will be useful in studying the regulatory mechanism in the renal metabolism of 25(OH)D3 in mammals.

Inhibition by 1 alpha,25-dihydroxyvitamin D3 of dimethyl sulfoxide-induced differentiation of Friend erythroleukemia cells.

Regulation of erythroid differentiation by vitamin D3 derivatives was examined in Friend erythroleukemia cells. After Friend cells were cultured for 5 days with 1.5% dimethyl sulfoxide (DMSO), as much as 70% of the cells became benzidine-positive and the hemoglobin content increased in parallel with the increase of benzidine-positive cells. The DMSO-induced erythroid differentiation was markedly inhibited by concurrent addition of the active form of vitamin D3, 1 alpha,25-dihydroxyvitamin D3 [1 alpha,25(OH)2D3]. Of the vitamin D3 derivatives tested, 1 alpha,25(OH)2D3 was the most potent in inhibiting DMSO-induced erythroid differentiation. 1 alpha,25(OH)2D3 alone was totally ineffective in both cell growth and erythroid differentiation. These results together with our previous reports indicate that 1 alpha,25(OH)2D3 is somehow involved not only in myeloid differentiation, but also in erythroid differentiation.

Isolation, identification, and metabolism of (23S,25R)-25-hydroxyvitamin D3 26,23-lactol. A biosynthetic precursor of (23S,25R)-25-hydroxyvitamin D3 26,23-lactone.

23S,25R,26-Trihydroxyvitamin D3 and (23S,25R)-25-hydroxyvitamin D3 26,23-lactol were chemically synthesized, and the metabolism of the two compounds to (23S,25R)-25-hydroxyvitamin D3 26,23-lactone in chick kidney homogenates was studied. 23S,25R,26-trihydroxyvitamin D3 was efficiently metabolized to the lactone in kidney homogenates from vitamin D-supplemented chicks, but not from vitamin D-deficient chicks. In contrast, the (23S,25R)-25-hydroxyvitamin D3 26,23-lactol was converted to the lactone in kidney homogenates regardless of the vitamin D status of the animals used. A new metabolite was isolated in pure form from the incubation mixture of 23S,25R,26-trihydroxyvitamin D3 with kidney homogenates prepared from vitamin D-supplemented chicks. The metabolite was identified as (23S,25R)-25-hydroxyvitamin D3 26,23-lactol by its ultraviolet and mass spectra and by derivatization. The structure was confirmed by direct comparison with an authentic sample on high pressure liquid chromatography. The evidence suggests that the stereochemistries of the isolated lactol at the 23- and 25-positions are S and R, respectively.

Analysis of pure and mixed murine mast cell colonies.

Mast cells have been proposed to originate from diverse sources, including connective tissues, macrophages, T lymphocytes, and hemopoietic cells. Evidence for a hemopoietic origin of mast cells includes the presence of mast cell precursors in spleen colonies and the presence of mast cells in hemopoietic colonies in culture. Here we report a detailed analysis of mouse spleen mixed hemopoietic colonies containing mast cells. All of the colonies in cultures plated at low cell densities were individually removed for analysis by May-Grunwald-Giemsa staining on day 15 of culture. Examination of five dishes which contained a total of 82 colonies showed 16 pure mast cell colonies and 36 mixed mast cell colonies. Sixteen different combinations of cell types were seen and were not distinguishable from each other in situ. The most diverse type of mixed colony contained macrophages (m), neutrophils (n), eosinophils (e), mast cells (Mast), megakaryocytes (M), erythroid cells (E), and blast cells. The clonal origin of mixed mast cell colonies was established by the replating of single cells obtained from blast cell colonies. Individual cells were removed with a micromanipulator, replated, and allowed to grow for 15 days. Cytospin preparations of 10 such colonies showed diverse combinations of cell lineages which were seen in the different types of mixed mast cell colonies described above. Replating studies of mixed mast cell colonies were carried out and a high incidence of replating was seen. Approximately one half of these colonies formed only mast cell colonies upon replating. Further studies showed that pure mast cell colonies could be serially replated four to five times. The replating efficiency of cells in the primary mast cell colonies varied over a wide range (2.5-44%) with an average replating efficiency of 13%. The data also revealed that cells containing metachromatic granules possess significant proliferative capacity. From these studies of pure and mixed mast cell colonies, we concluded that mast cells are in wide variety of types of mixed colonies and that the in situ identification of mixed colonies is unreliable, that mast cells are derived from pluripotent hemopoietic stem cells, and that mast cells with metachromatic granules can have a high proliferating ability.
