Comparative study on the structure of the light chains of human immunoglobulins. II. Assignment of a new subgroup.

The primary structure of the variable region of the human lambda type Bence Jones protein NIG-48 was determined by analysis of the N-terminal sequence of the completely reduced and aminoethylated protein, as well as of five cyanogen bromide fragments. The variable region of NIG-48 contains 112 amino acid residues. The protein NIG-48, having a unique sequence of the variable region, a low degree of homology (about 50%) with lambda chains of the five other subgroups and the addition of two residues around 65, may represent a new subgroup, namely V lambda VI.

Pitfalls in the use of chromosome variants for paternity dispute cases.

A huge 'satellite' (22s+) was observed in a mentally retarded and malformed girl. It consisted of constitutive heterochromatin based on the C-band technique (CBG). In neither of the parents was the variant observed. Genetic marker studies revealed that the father was, indeed, the father of the proposita (probability, 99.8%). It is emphasized that caution must be taken in the use of chromosome variants for paternity dispute cases.

Dopamine-beta-hydroxylase activity in serum and cerebrospinal fluid in neuropsychiatric diseases.

A 24-hour rhythm in serum dopamine-beta-hydroxylase (DBH) activity in chronic schizophrenic patients showed a similar change as that in normal subjects: the highest activity during day time with a peak at 12.00 h and the lowest activity at night with a bottom at 24.00 hr, 4 h after sleep onset. Between a group of schizophrenic patients who have sleep disturbance and another group of patients who have normal sleep patterns, no significant difference was observed in the 24-hour rhythm in serum DBH activity. Very low but significant DBH activity was found in cerebrospinal fluid of neuropsychiatric patients, and the values were approximately 1/1,000 of the serum activity. A case after cerebral injury showed a high DBH activity in cerebrospinal fluid.

Comparative structural studies on the light chains of human immunoglobulins. I. Protein Ka with the Inv(3) allotypic marker.

The primary structure of the variable region of the human type K Bence-Jones Protein Ka with Inv(3) genetic marker was determined by sequence analysis of 25 major tryptic peptides covering 214 residues isolated from completely reduced and aminoethylated protein. The complete sequences of 18 of these peptides were determined. These comprised the entire variable NH2-terminal half and 7 peptides from the COOH-terminal half of the protein. For the remaining peptides covering the COOH-terminus, only partial sequences or the amino acid compositions were determined. On the basis of these results all the tryptic peptides could be arranged in order. The sequence of the variable region differed from other previosly reported in 21 to 51 residues, but no variation was found in the sequence of the last 107 residues. On the basis of sequence homology the protein was classified in the kI subgroup. A brief discussion of the possible genetic mechanism of sequence variability in relation to subgroup specificity is presented.
