From the screening of a number of new pyrazole derivatives, the title compound, PZ-177, was selected as the most significant derivative for analgestic and anti-edematous actions, in this paper, we report the anti-edematous activity of PZ-177 as assessed by detailed analysis. PZ-177 showed a markedly inhibitory effect against rat paw edema induced by various phlogists (carragenin, dextran, egg albumin, serotonin, formalin and bradykinin). It also inhibited edema induced by anti-rat rabbit serum. The activity of PZ-177 was more potent than that of mepirizole and the same as that of phenylbutazone. Though this agent has a central depressive effect, it is considered that the action has actually little influence on anti-edematous effect, as carrageenin-induced edema was inhibited in spinal rats. On the other hand, the anti-edematous effect of PZ-177 was reduced significantly in adrenalectomized rats. It is therefore suggested that the potent anti-edematous action of PZ-177 is exerted partially by a direct action at the inflamed site and mediated partially by stimulation of the hypophysis-adrenal system.
The effects of maprotiline, mainly on the CNS, were examined and compared with those of imipramine, methamphetamine and chlorpromazine. In the repeated administration and the stroboscope studies, maprotiline exhibited actions to increase behavioral activities of animals, just like amphetamine and imipramine. This appears to attest to pharmacological effects of maprotiline as an anti-depressant. It is noted, however, that whereas maprotiline shows a slight anti-apomorphine action in dogs and an inhibition of the spinal reflex action potential in cats, such actions are not recognized in other anti-depressants of the tricyclic groups including imipramine, amitriptyline, etc. These differences suggest the tranquilizing action of maprotiline at work, probably forming a specific pharmacological feature of maprotiline. Also, maprotiline increases body weight of rats strikingly, which seems to offer a wide range of clinical applications of this drug.

This experiment was designed to investigate and evaluate the characteristic behavior of mandibular free-end saddle removable partial denture using various retainers. The abutment tooth displacement and saddle movement were measured while a known vertical force was applied on the saddle. Fifteen different types of free-end saddle dentures were tested, using four various types of prefabricated attachments. Strain-measuring devices were used to measure the extent of abutment tooth displacement and saddle movement. The findings were as follows: 1) The force transmitted to the abutment tooth and the residual ridge were affected by (a) the selection of direct retainers, (b) splinting of abutment teeth, and use of indirect retainers, and (c) the variation in size and shape of the occlusal table on the saddle. 2) The influences by direct retainers a) With use of a resilient joint, the stress was heavily loaded on the residual ridge directly under the direction of force. b) With the joint incorporating the vertical rock-preventing device, a heavy proportion of lateral force was transmitted to the abutment tooth. 3) Both the lateral stress on the abutment tooth and movement of the saddle were reduced more effectively by use of indirect retainers than by splinting of abutment teeth. 4) By reduction of the occlusal area in bucco-lingual dimension, the denture saddle was better stabilized and the lateral stress on the abutment tooth was reduced effectively.
