[Topical anti-inflammatory activity of dexamethasone 17-valerate (author's transl)].

The anti-inflammatory activity of dexamethasone 17-valerate ointment (DV-17, 0.12%) was investigated by topical application in mice and rats, and the effects compared with those of dexamethasone (DX, 0.12%), betamethasone 17-valerate (BV-17, 0.12%), beclomethasone 17,21-dipropionate (BE, 0.025%) and hydrocortisone 17-butyrate (HC, 0.1%) which were prepared with the same ointment base. DV-17 inhibited markedly the superficial inflammation such as increased vascular permeability induced by intradermal injection of histamine or bradykinin in rats and edema induced by a drop of croton oil into the mouse ear. DV-17 also inhibited significantly rat paw edema induced by carrageenin, yeast, nystatin and mustard. The inhibitory activity of DV-17 on those acute inflammatory responses was similar to that of DX and BV-17. In the inhibitory activity on carrageenin induced paw edema, DV-17 was less potent than that of DX when given orally, however was similar to DX in topical application. DV-17 also inhibited granulation tissue proliferation by subcutaneous paper disk implantation and nontreated foot swelling in adjuvant arthritic rats, but the inhibitory activity of DV-17 on the inflammation of these distant areas was lower than that of DX. On the other hand, systemic effects such as decrease in weight of adrenal or thymus and body weight loss were most evident in the case of DX and lower with DV-17. DV-17 prolonged wound healing and inhibited the delayed-type hypersensitivity induced by picryl chloride. The activity was equivalent to that of DX and BV-17. From the above results, it may be considered that DV-17 possesses potent anti-inflammatory activity, whereas it has fewer side effects and is a useful glucocorticoid for external application.

The high prevalence of Wuchereria bancrofti infections in indigenous tribes in northern Mindanao, Philippines.

Human filariasis was detected in an anthropological and medical survey among the indigenous Mamanwa people of the northernmost region of Mindanao, Philippines in specimens of thin blood smears and cultures of leucocytes from blood samples taken in the day time in September, 1978. Microfilaria positives were 23.2% (22/95) at Bo. Pangaylan, Santiago, 26.7% (28/105) at Kitcharao, Agusan del Norte, 19.3% (16/83) at Urbistondo, Surigao del Norte, Mindanao and none (0/50) at Abucay, Bataan, Luzon. Morphological characteristics of microfilaria were those of Wuchereria bancrofti. The periodicity of microfilaria observed in 3 Mamanwa people was nocturnally periodic with the peak hours at 12 o'clock midnight and the periodicity index at 124.4 following the method of Aikat & Das, or 92.35 following the method of Sasa & Tanaka. Microfilaria positives in the culture of leucocytes were as high as 41.2% (28/68). Since blood samples were taken in the day time and the filaria in these areas was nocturnally periodic, the prevalence of infection among these people is assumed to be much higher than the observed prevalence.

Comparison of the blood film, Millipore filter and Nuclepore filter techniques for the detection of microfilaremia in a field survey in the Philippines.

The blood film, Millipore filter and Nuclepore filter techniques were applied in a filariasis field survey at a village in the Philippines. venous blood from each person was examined by the three techniques to compare their efficacy of microfilariae detection. Overall positive rate was 16.1% by the combined techniques. Microfilaria positive rate was highest in Nuclepore filter method (15.6%) followed by Millipore technique (13.6%) and blood film (11.5%), but their difference was not significant statistically. The ratio of the total microfilaria counts by the three techniques was 1:29.8:30.3, which seems to reflect the ratio of the blood volumes used. There was no significant difference between the microfilaria counts by the three techniques adjusted to the same volume of 30 microliter and examined by F-test. It is considered that the blood film is still of practical value in field surveys.

Human lung cancer cell line (KSNY) producing colony-stimulating activity which affects both human and mouse marrow cells.

A cell line (KSNY) in vitro, which produces colony-stimulating activity (CSA) for human and mouse marrow cells, has been established. A biopsy was performed on the tumor mass of a lung cancer patient who had developed extensive leucocytosis. A piece of the tumor was transplanted to a nude mouse. The seconarily transplanted mice, in turn, developed extensive leucocytosis. The mouse tumor was then removed and placed in culture bottles. To date, the KSNY cells have been maintained in vitro continuously for 15 months. By the use of a methylcellulose bone-marrow colony-formation technique, a high level of CSA in the supernatant of the tumor cell culture was recognized. Doubling time of the cell line is 46 hr. The modal chromosome number is 52, ranging from 45 to 106.

Evaluation of the intrathoracic injection method for screening of filaricides.

Intrathoracic injection technique was utilized to examine its value in screening of antifilaria drugs in Litomosoides carinii in the cotton rat, Sigmodon hispidus, using existing filaricides such as diethylcarbamazine, Mel W. metrifonate, suramin, arsenics and antimonials. Diethylcarbamazine at a dose of 100 mg/kg for 5 days given intrathoracically caused marked decrease of more than 95% of the microfilaria count in the blood 1 week after the initial injection which is the same effect as was observed by intraperitoneal injection. Whereas adult worms in the pleural cavity were not affected. Mel W at a dose of 10 mg/kg for 5 days killed all adults but had no effect on microfilaria density in the blood. When it was given intraperitoneally, even a larger dose of 50 mg/kg did not affect adults in the pleural cavity. In cotton rats tolerating suramin at 80 and 40 mg/kg for 5 days, adult worms were intact. However, a reduced dose of 20 mg/kg and 10 mg/kg given weekly for 6 weeks showed remarkable macrofilaricidal activity. Thus the slow action of suramin was reproduced by this method. There was no significant change in the microfilaria density in the blood. Aresenics such as Mapharsemin and Neo Neo Arsemin, and antimonials such as stibophen and tartar emetic were shown to have macrofilaricidal activity by intrathoracic injection with no effect on microfilaria density in the blood within a week. Effect of existing filaricides by this method showed remarkable coincidence with their action Wuchereria bancrofti.
