Alterations in serum pancreatic elastase 1 content in acute and chronic pancreatitis: comparison with alpha-amylase activity.

Serum pancreatic elastase 1 content in 45.9% of 203 samples from 32 patients with acute pancreatitis was found to be greatly elevated to greater than 1000 ng/dl, much higher than the control values (90 to 270 ng/dl), whereas serum alpha-amylase activity, in 62.3% of the same samples, was within the control range (100 to 460 IU/L). The increased pancreatic elastase 1 content persisted for several days after serum alpha-amylase activity returned to normal, suggesting that, compared with serum alpha-amylase activity, pancreatic elastase 1 content truly reflects the clinical course of acute pancreatitis. The measurement of pancreatic elastase 1 content appears to be more valuable in the diagnosis and prognosis of acute pancreatitis than that of serum alpha-amylase activity. In 41 patients with chronic pancreatitis, on the other hand, pancreatic elastase 1 content and serum alpha-amylase activity were within the normal range in 38.3% and 56.2% respectively, of 336 samples.

Effects of adrenalectomy on gonadal hormone-induced changes in hepatic microsomal carboxylesterase activities in rats.

Hepatic microsomal carboxylesterase (E.C. 3.1.1.1) showed a different capacity for the hydrolysis of various substrates in rats. Adrenalectomy in castrated male rats slightly decreased the activities towards p-nitrophenyl-acetate and malathion. The magnitude of testosterone-induced changes in the hydrolase activities by liver microsomes in adrenalectomized-castrated male rats was markedly decreased castrated male rats. However, when 5 alpha-dihydrotestosterone was administered to adrenalectomized-castrated male rats, malathion and p-nitrophenylacetate hydrolase activities were significantly increased. But in all cases, isocarboxazid and acetanilide hydrolase activities were not affected by physiological states. These results suggest that hepatic microsomal carboxylesterases may be, at least in part, regulated by adrenal gland and gonadal hormones which exert different effects on the several isozymes of carboxylesterases.

Photoactivation of hyaluronidase by chlorpromazine.

The effects of UVA and chlorpromazine on the activity of hyaluronidase were investigated. Hyaluronidase was slightly activated by UVA alone. It was also activated by chlorpromazine, but the activation was enhanced by concomitant UVA irradiation. By pre-irradiation of chlorpromazine, it was found that a stable photoproduct of chlorpromazine was formed that activated hyaluronidase. As activation of hyaluronidase has been described in relation to tissue inflammation, the onset of chlorpromazine-induced photosensitivity may, in part, be related to the hyaluronidase activating effect of chlorpromazine plus UVA.

Increase in anti-carboxylesterase action of organophosphorothioates by nicotinamide adenine dinucleotide (NAD) in vitro.

The purpose of the present study was to investigate the relationship between chemical structure of organophosphorus insecticides and potentiation of anti-carboxylesterase (CEase) action of these insecticides by NAD in vitro (NAD-effect). Experiments using with three organophosphorothioates having ethoxy group except for diazinon exhibited greater NAD-effect than those having methoxy ones such as methylparathion and fenitrothion. In contrast, none of five organophosphates tested showed NAD-effect. And, 3-acetylpyridine adenine dinucleotide (Ac-py AD) among four derivertives of NAD was also found to have NAD-effect. These results suggested that P = S group in the molecule of organophosphorus insecticides was needed to occur NAD-effect. In addition, the extent of NAD-effect using acetylcholinesterase (AChE) was lesser than that of CEase, therefore, a higher susceptibility of liver microsomal CEase to organophosphorus insecticides may be explained, at least inpart, by NAD-effect.

The ovarian enzyme catalyzing conversion of 13,14-dihydro-15-ketoprostaglandin F2 alpha to 13,14-dihydroprostaglandin F2 alpha in rats.

The preliminary experiments concerning enzymes involved in the production system of 13,14-dihydroprostaglandin F2 alpha (13,14H2-PGF2 alpha) in rat ovary were carried out. The enzyme which converted 13,14-dihydro-15-keto-prostaglandin F2 alpha (15KD-PGF2 alpha) to 13,14H2-PGF2 alpha required NADPH as a cofactor, and the enzyme activity was inhibited by sulfhydryl reagents as well as quercitrin. Thus, the enzyme studied here seems to be a carbonyl reductase possessing SH group(s) in the molecule.

Early changes in the levels of serum triiodothyronine (T3), thyroxine (T4), T3/T4 ratio and microsomal carboxylesterase activity in rats following treatment with CCL4.

To study the relationship between the activity of nonspecific carboxylesterase located in the smooth endoplasmic reticulum (SER) of hepatocytes, and serum thyroid hormones including triiodothyronine (T3), thyroxine (T4), and T3/T4 ratio, we determined these hormones, liver carboxylesterase activity, and serum alanine amino-transferase (SGPT) before, and 5 to 720 min after treatment with a single oral dose of CC14 (1 ml/kg) in rats. The serum T3/T4 ratio decreased 5 to 10 min after administration. Each of 16 male and 16 female rats showed a significant correlation between carboxylesterase activity and T3/T4 ratio (male, r = 0.79, p less than 0.01; female, r = 0.84, p less than 0.01), but revealed no correlation between carboxylesterase and T3 or T4, alone. SGPT contents were elevated 6 to 12 h after, whereas T3/T4 ratio decreased gradually during the course. The SER increased mildly 5 min after, but was irregularly distended 6 h after. Changes in the microvilli were not found 5 min after, but the disruption was seen 6 h after. These results suggest that serum T3/T4 ratio is closely correlated with microsomal carboxylesterase activity in CC14-treated rats and represents a microsomal function.

[A study on the levels of cefoperazone in urological organs].

The concentration of Cefoperazone (CPZ) in urological organs and serum was measured after the intravenous administration of 2 g of CPZ. Serum levels on healthy adults attained a maximal value of 122.1 +/- 4.2 micrograms/ml at 30 minutes. Serum levels of patients operated under general anesthesia attained a maximal value of 159.0 +/- 13.9 micrograms/ml at 30 minutes. Serum concentration on operated cases continued to be 1.3-2.2 fold higher than that of the healthy adults. Prostate tissue levels attained a maximal value of 80.9 +/- 3.6 micrograms/g at 30 minutes, vesical tissue levels were 52.1 +/- 1.3 microgram/g at 60 minutes and renal tissue were 94.0 +/- 14.5 micrograms/g at 90 minutes. Judging from the minimal inhibitory concentration of CPZ, CPZ seemed to be clinically useful in the treatment of urological infections.

[A case of malignant lymphoma in the skull after head injury associated with multiple bone tumors].

A case of malignant lymphoma in the skull after head injury associated with whole bone metastasis is reported. The patient was a 66-year-old man who was admitted to Almeida Memorial Hospital because of headache and general fatigue 2 months after head injury. After admission tumors appear in the frontal and occipital region and grew rapidly. Plain craniogram revealed large map-like bone destructions and multiple punched out lesions. Bone scintigram with 99mTc-MDP revealed multiple accumulations of RI in the skull, vertebrae, ribs and pelvis. CT scan revealed destructive, markedly enhancing bone tumor which was compressing the brain as an extradural mass in the left frontal and occipital regions. Pathological examination of the tumor revealed malignant lymphoma of non-Hodgkin type and diffuse pleomorphic type. Though combination chemotherapy with ACNU, FT 207, PSK, CHOP (Cyclophosphamide, Adriamycin, Vincristine and Predonisone) and Acracinomycin A was performed after operation, and brought forth regression of tumor size and improvement of clinical symptoms transiently, he died 6 months after the onset because of recurrence in many bones with pathological fracture and complications such as pneumonia, DIC and acute renal failure. At autopsy the tumors were found to be localized only in the bones, but in none of lymphnode or visceral organs. Malignant lymphoma appearing initially as a skull tumor is rare, and its diagnosis and treatment were discussed.

Metastatic patterns of prostatic cancer. Correlation between sites and number of organs involved.

Prostatic cancer in 1885 autopsy cases was classified according to the number of organs involved in metastasis, and a comparison was made concerning the frequency of metastasis to the various organs. The frequencies of metastasis to the lungs and para-aortic lymph nodes were low in cases with single-organ involvement (4.6% and 2.3%, respectively), but increased rectilinearly in accordance with the number of organs involved and became high in cases with metastasis to three or more organs (49.1% and 21.8% in total, respectively). On the other hand, the frequencies of local extension to the bladder and invasion of the pelvic lymph nodes were high even in cases with single-organ involvement (34.5% and 9.2%, respectively) and were not significantly changed regardless of the number of organs involved. No significant correlation was seen between pelvic and para-aortic lymph node involvement. In cases with single-organ involvement, metastasis to the lumbar spine occurred frequently, but those to the ribs, sternum, and ilium occurred less frequently. There may be multiple metastases in cases with metastases to the para-aortic lymph nodes, sternum, and ilium. The number of metastatically involved organs is useful in analyzing the mode of metastasis.

Dose-dependent pharmacokinetics of aprindine in healthy volunteers.

The disposition of aprindine following a single oral dose can best be described by a two-compartment open model. The mean plasma half-life (t 1/2 beta) increased from 8.0 +/- 2.1 h (SD) after a 25 mg dose of 9.4 +/- 2.9 h after 50 mg and to 15.8 +/- 2.6 h after 100 mg, with a decrease in total plasma clearance (Cl/F) and volume of distribution at steady state (V dss/F) and during beta-phase (V d beta/F). The area under plasma concentration-time curve (AUC), maximum plasma concentration (C max) and the amount of unchanged aprindine excreted in the urine increased in a non-linear fashion with the increase in dose. The t 1/2 beta after multiple oral doses showed a 3-fold increase over the single dose value. These results indicate that aprindine shows dose-dependent non-linear kinetics.

Brain extract induces the tetrodotoxin-sensitive action potentials in a rat skeletal muscle cell line (L6).

Tetrodotoxin (TTX) usually blocks the action potential in adult vertebrate innervated skeletal muscle, i.e. the action potential is operated by a TTX-sensitive sodium channel. The L6 cultured myotube, however, has TTX-resistant action potential that is usually found in skeletal muscle following denervation or during fetal and neonatal stages in rat. The TTX-resistivity of the L6 myotubes might be due to a lack of a neurotrophic substance. The present study revealed that brain extract had a trophic substance that is capable of inducing the TTX-sensitive sodium channels in uninnervated cultured L6 myotubes. Seventeen to 18 days old cultures were used and the maximum rate of rise of action potential was measured as an index of the sodium channel density. In control cultures, the maximum rate of rise was about 120 V/s and did not change by TTX application. However, in cultures with the brain extract the value was 142 V/s and reduced to 86 V/s by TTX. The reduced maximum rate of rise by TTX in the latter cultures means that exposure of L6 myotubes to brain extract resulted in an increase in TTX-sensitive sodium channels and a decrease in TTX-resistant sodium channels. Therefore, exposure to a factor in brain extract can elicit development of TTX-sensitive sodium channels in isolated L6 muscle cells that have been grown as a cell line in the absence of neurons for over 10 years.

Redox properties of membrane-bound b-type cytochromes and a soluble c-type cytochrome of nitrate reductase in a photodenitrifier, Rhodopseudomonas sphaeroides forma sp. denitrificans.

In order to identify the b-type cytochrome involved in the nitrate reduction in a photodenitrifier, Rhodopseudomonas sphaeroides forma sp. denitrificans, the b-type cytochromes in the spheroplast membranes were characterized. Difference spectra at 77K of spheroplast membranes indicated the presence of two b-type cytochromes with a bands at 556.5 and 562 nm. Three components considered to be of the b-type cytochrome were resolved by anaerobic potentiometric titration at 560-572 nm. Their midpoint potentials at pH 7, Em,7, were - 135 mV, +40 mV and +175 nm and their approximate reduced minus oxidized maxima were determined to be at 565 nm (562 nm at 77K), 560 nm (556.5 nm) and 560 nm (556.5 nm), respectively. These values are almost the same as those reported for R. sphaeroides. The Em,7 value of the cytochrome c involved in the nitrate reductase of this denitrifier was determined to be 250 mV. A b-type cytochrome reduced with NADH and FMN was oxidized by nitrate in chromatophore membranes. The possibility that cytochrome b (Em,7 = 175 mV) is involved in the nitrate reduction is discussed.

Potentiation of EPN-induced inhibition of liver microsomal carboxylesterase by addition of liver cytosol from 6-aminonicotinamide-treated, starved rats.

Addition of liver cytosol from 16 hr-starved rats treated with 6-aminonicotinamide to rat liver microsomes caused potentiation of the anti-carboxylesterase action of ethyl-p-nitrophenyl phenylphosphonothioate (EPN). This was not found when liver cytosol from non-pretreated rats after 16 hr-starvation was used. This potentiation of EPN-induced inhibition of carboxylesterase may be, at least in part, explained by the fact that treatment of rats with 6-aminonicotinamide resulted in a significant increase in NADPH level in liver cytosol which, in turn, stimulated formation of an EPN oxygen analog, a potent inhibitor of carboxylesterase, through cytochrome P-450-coupled monooxygenase.
