The inhibitory effect of iodide on growth of rat thyroid (FRTL-5) cells.

The effect of iodide on growth of rat thyroid cells (FRTL-5) was studied. TSH-stimulated cell growth was inhibited by iodide in a concentration-dependent manner, and an effect of iodide was detected at 10(-6) mol/l. KClO4 or 1-methylimidazole-2-thiol blocked the effect of iodide, suggesting that iodide uptake and its organification are required to produce the inhibitory effect of iodide on cell growth. Iodide not only decreased TSH-stimulated cAMP production in FRTL-5 cells but also cell growth induced by cAMP. These observations suggest that iodide inhibits TSH-stimulated growth of the cells by attenuating cAMP production and also by acting on the step(s) distal to cAMP generation. The inhibitory effect of iodide was also seen in growth stimulated by insulin, insulin-like growth factor-I or 12-O-tetradecanoyl phorbol 13-acetate, suggesting multiple sites of action of iodide in the process of growth of FRTL-5 cells.

Phorbol ester pretreatment attenuates the growth hormone (GH) response to GH-releasing factor in cultured rat pituitary cells.

The effect of phorbol ester pretreatment on rat (r) GH release induced by GH-releasing factor (GRF) or 8-bromo-cyclic (c)AMP was investigated using rat pituitary cells cultured in monolayers. Pretreatment with 12-O-tetradecanoylphorbol-13-acetate (TPA) for 3 h significantly suppressed the rGH release induced by GRF, but not that by 8-bromo-cAMP 20 h later; this suppressive effect of TPA was concentration-dependent from 8 to 160 nmol/l, and complete suppression was observed after pretreatment with 80-160 nmol TPA/l. Production of cAMP by pituitary cells stimulated with GRF was similarly attenuated in TPA-pretreated cells. The rGH responsiveness to GRF of these cells was fully recovered on prolonged culture (40 h), suggesting that the inhibitory effect of TPA is reversible. In contrast, pretreatment with GRF (5 nmol/l) resulted in suppression of the rGH response to subsequent exposure to GRF (5 nmol/l) or 8-bromo-cAMP (10 mmol/l), but not to TPA. These observations suggest that pretreatment with TPA modifies the rGH response to GRF at steps before the formation of cAMP.

Effects of nifedipine on intracranial pressure in neurosurgical patients with arterial hypertension.

The effects of nifedipine, 20 mg administered via a nasogastric tube, on intracranial pressure (ICP) and cerebral perfusion pressure (CPP) were examined. Nifedipine was administered to treat arterial hypertension (greater than 180 mm Hg, systolic). Ten measurements were made in eight patients with cerebrovascular disease or head trauma. The mean arterial blood pressure (MABP) and ICP were measured before and for 30 minutes after the administration of nifedipine. The MABP gradually decreased and reached its lowest value at approximately 10 minutes after initiation of nifedipine administration, and thereafter remained unchanged. The MABP decreased significantly from 128 +/- 8 (mean +/- standard deviation) to 109 +/- 7 mm Hg, and the CPP decreased from 105 +/- 11 to 84 +/- 10 mm Hg. The ICP increased by 1 to 10 mm Hg in eight of 10 measurements, and the mean change of ICP from 19 +/- 7 to 22 +/- 6 mm Hg was statistically significant. These changes were not accompanied by alterations in neurological signs. The results suggest that enteral nifedipine produces a small but statistically significant increase in ICP. Accordingly, neurological signs must be closely observed to detect deterioration, which can be caused by an increase in ICP and/or a decrease in CPP.

[Testing performance of the improved pinhole collimator and its application to small animals].

Testing performance of the improved pinhole collimator of 1.5 mm aperture attached to the gamma camera was carried out with three kinds of radionuclides, 99mTc, 67Ga and 131I, using a hand-made phantom. The results obtained in this experiment were compared to those obtained with a human pinhole collimator of 4 mm aperture. Those were nearly understood by taking account of the effective aperture of a pinhole collimator for each effective photon energy. And all scintigrams obtained from rats with various scintigraphy showed high resolution images.

[Effects of pre-operative intrahepatic-arterial infusion in liver cancer with special reference to hepatic resection].

The following conclusions were obtained in terms of the histological response of preoperative TAI and TAE in 27 hepatectomized patients with 15 primary and 12 metastatic liver cancers. 1) TAE was superior to TAI in tumor 100% necrosis rate. 2) TAE showed higher necrosis rate than TAI in tumors with capsule formation smaller than 2 cm. 3) Even TAE reveals no histological response in tumors with intracapsular infiltration.

Effect of lipophilic extract of Chlorella vulgaris on alimentary hyperlipidemia in cholesterol-fed rats.

The effect of glycolipid (GL) and phospholipid (PL) fractions obtained from Chlorella on serum lipid level and fecal excretion of steroids were examined in cholesterol-fed rats. The increase of the level of serum lipids were inhibited by the feeding of GL, PL and Chlorella powder almost the same degree. Fecal excretion of steroids (mostly of cholesterol, deoxycholic and lithocholic acid) were increased by feeding of GL and PL fractions. It is concluded that the feeding of each fraction inhibits the absorption of exogenous steroids and promotes turnover of bile acids in liver to suppress the increase of serum cholesterol level caused by administration of high cholesterol diet.

[Metastatic tumor of the spermatic cord from gastric cancer].

A 70-year-old male who had undergone gastrectomy due to gastric cancer 6 years previously visited our department complaining of perineal and bilateral scrotal mass. Biopsy specimen of the perineal mass showed well-differentiated adenocarcinoma. The spermatic cords were resected en bloc with the entire scrotal content, penis and perineal skin. Histological examination of the spermatic cord, epididymis and perineal subcutaneous tissue revealed well differentiated adenocarcinoma similar to that of the lesion resected from the stomach 6 years earlier. The patient was followed for 2 years with no evidence of recurrence. Metastatic tumors of the spermatic cord and epididymis resulting from gastric cancer are rare and 31 cases previously reported in the Japanese literature are reviewed.

A rare case of double aortic arch: right thoracotomy and residual retroesophageal aortic arch.

The case of a 2-year-old boy with tetralogy of Fallot and a complete double aortic arch (both arches patent) is reported. The left dominant aortic arch ran retroesophageally to the right and joined with the right smaller arch to form the descending thoracic aorta on the right side. We employed a right thoracotomy and performed a division of the right nondominant arch at the connection with the descending aorta. The surgical implications of an unusual type of double aortic arch are discussed.

Modified Fontan operation for single ventricle with common atrium and abnormal systemic venous drainage: usefulness of an additional superior vena cava to pulmonary artery anastomosis.

In two patients (aged 15 and nine years old) with single ventricle, common atrioventricular valve, common atrium, and bilateral abnormally draining superior vena cava (SVC), a modified Fontan procedure was performed together with a SVC to pulmonary artery (PA) anastomosis. This avoided complex intraatrial septation. A bilateral anastomosis was carried out in one patient and a unilateral one in the other, the PA not being divided in either patient. This procedure provided simple intraatrial septation while avoiding possible obstruction of either the pulmonary or systemic blood pathways if complete intraatrial septation had been carried out.

Granular cell tumor of the ascending colon: a case report.

A granular cell tumor (granular cell myoblastoma) of the ascending colon in a 48-year old male is reported. The tumor was detected by barium enema study as a sessile polyp, and colonofiberscopy revealed submucosal tumor. It was removed by endoscopic polypectomy. Macroscopic examination showed the characteristic features of granular cell tumor. The avidin-biotin-peroxidase complex (ABC) method for detection of S-100 protein demonstrated that the cytoplasm of tumor cells and the pleomorphic nuclei were strongly stained with anti-S-100 protein serum, which supports the concept of the Schwann cell origin of granular cell tumor.

Stromal reactions to squamous cell carcinoma of the cervix.

The characteristics of the stromal reaction to invasion by cancer were studied in 23 cases of cervical squamous cell carcinoma (stage Ib to IIb) and 86 control cases of benign disease of the uterus. Tissues taken from sites adjacent to tumor or from comparable sites in the nonmalignant uteri were analyzed for number of round cells, density of collagen and reticular fibers, and collagenase activity. The results were correlated with the depth of invasion and the histologic appearances of the tissues adjoining the cancer. Plasma cells and reticular fibers were most numerous in patients with slight invasion and no direct contact between the malignant and benign tissues (C type in CPL classification of Imai). Glycosaminoglycans (mainly hyaluronic acid and chondroitin sulfate) were present in the greatest amounts in C type stroma. The prognosis showed no correlation with either collagen content or collagenase activity but it was a better prognosis correlated with the collagen/collagenase ratio. It is concluded that increased numbers of plasma cells and reticular fibers and a high collagen/collagenase ratio are among the mechanisms protective against the invasiveness of cervical carcinoma.
