Supersensitivity to prostaglandin in chronic neurogenic bladders.

The bladder response to prostaglandin (PG test) was investigated in chronic neurogenic bladders by isometric cystometry and compared with the results obtained from normal subjects. The urinary bladder was filled with 100 ml of saline and 100 micrograms of prostaglandin 995 were injected intravenously. The changes in bladder and intra-abdominal pressures were observed for up to 20 min. A rise in detrusor pressure of more than 20 cm of water was judged a positive response. Out of 21 neurogenic bladders 12 had a positive response. The result of the PG test demonstrated a good correlation with the supersensitivity test to bethanechol chloride which was performed on these patients on a different day. All of the normal subjects had negative responses. It was concluded that the chronically denervated bladder is supersensitive to prostaglandin 995.

Weight estimation of benign prostatic adenoma with urethral pressure profile.

An estimation of the weight of the benign prostatic adenoma by quantitative analysis of the urethral pressure profile has been correlated with the surgically removed specimen in 23 patients. It has been found that the length of the continence zone cane be used as an index of prostatic weight. After operation the length of the continence zone fell into the normal range of the age matched controls.

Neo-enactin, a new antifungal antibiotic potentiating polyene antifungal antibiotics. I. Fermentation, extraction, purification and physico-chemical and biological properties.

A new antifungal antibiotic, named neo-enactin, was isolated from the cultured mycelia of Strain H 829-MY 10, identified as a new subspecies of Streptoverticillium and named Streptoverticillium olivoreticuli subsp. neoenacticus. The antibiotic was produced with a tetraene antifungal antibiotic, found mainly in the cultured mycelia of Strain H 829-MY 10 and it was extracted with methanol. The antibiotic is of a basic nature and it can be extracted with ethyl acetate at alkaline pH. Purification of neo-enactin was carried out by partition chromatography on cellulose and elution with ethyl acetate bufferized with phosphate buffer (pH 8.0). Neo-enactin shows strong antifungal activity and potentiates the antifungal activity of polyene antifungal antibiotics.

Neo-enactin, a new antifungal antibiotic potentiating polyene antifungal antibiotics. II. Taxonomic studies of the producing microorganism and simultaneous production of bleomycin group and streptothricin-like antibiotics.

A new antifungal antibiotic, named neo-enactin, was produced mainly in the mycelia of strain H 829-MY 10. Strain H 829-MY 10 was identified as a Streptoverticillium, determined to be nonchromogenic, and fits in the white color-series. Although Streptoverticillium olivoreticuli is known to be chromogenic, strain H 829-MY 10 is most related to that species. Thus, strain H 829-MY 10 is named as Streptoverticillium olivoreticuli subsp. neoenacticus. Besides neo-enactin, two bleomycin-group antibiotics and two streptothricin-like antibiotics were simultaneously produced by strain H 829-MY 10.

Ileumycin, a new antibiotic against Glomerella Cingulata.

A new antifungal antibiotic, named ileumycin, was isolated from culture broth of streptomyces H 698-SY2, which was identified as S. lavendulae. The antibiotic was recovered from the culture filtrate by adsorption on Amberlite XAD-II and elution with aqueous methanol and was further purified by ion-exchange column chromatography on SE-cellulose and followed by partition chromatography on silica gel. The antibiotic was named ileumycin, because isoleucine was detected in the acid hydrolyzate of the antibiotic. Ileumycin exhibited antimicrobial activity against only a few species of fungi.

[Effect of furosemide on ouabain toxicity in guinea pigs (author's transl)].

An attempt was made to study the effect of furosemide on the toxic and lethal dose (LD) of ouabain. Ouabain was infused continuously in guinea pigs anesthetized with urethane. The toxic dose of ouabain was determined by the minimal dose which produced ventricular arrhythmias. The LD of ouabain was 300.8 +/- 13.8 microgram/kg (mean +/- SE) in control guinea pigs. When furosemide was administered in a dose of 2 mg/kg i.v. prior to ouabain, this was significantly decreased to 225.2 +/- 14.1 microgram/kg (P less than 0.01). Furosemide-treated guinea pigs also showed a marked decrease in toxic dose of ouabain. In another series of experiment, the ouabain uptake by the heart and its subcellular fraction were determined by the infusion of 3H-ouabain at a constant rate under the same experimental conditions aforementioned. Approximately 30% decrease in the LD of 3H-ouabain was also observed in the furosemide pretreated guinea pigs. 3H-ouabain concentration in blood, cardiac muscles and microsomal fraction were lower than control group. These findings suggest that potentiation of ouabain toxicity produced by furosemide is not associated with increased cardiac ouabain uptake, but with increased sensitivity at the site of action.

[Effect of chronic oral administration of clonidine on the endogenous catecholamine metabolism in spontaneously hypertensive rats (author's transl)].

The present experiments were undertaken to elucidate the effect of chronic oral administration of clonidine on the endogenous catecholamine metabolism in spontaneously hypertensive rats (SHR). A three month administration of clonidine (20 microgram/kg/day p.o.) produced a slight decrease in blood pressure and heart rate. No significant change was observed in the body weight. A one month administration of clonidine (300 microgram/kg/day p.o.) produced hypotension and weight loss. No significant effect was observed in heart rate. After a two month administration of clonidine (300 microgram/kg/day p.o.), the catecholamine content in the brainstem showed a significant decrease as compared with that in control. 3H-norepinephrine (NE) turnover rate after intraventr culari injection of 3H-NE tended to decrease in the brainstem of SHR while no significant difference was recognized from a view point of uptake, biological half life, elimination rate constant of 3H-NE between control and clonidine administered rats. Under the same experimental conditions, a marked rebound hypertension occurred when medication is discontinued in some cases of this group of SHR.
