Extranodal formation of lymphoid follicles was morphologically studied in experimental bronchopneumonia. Control gnotobiotic mice had no peribronchial lymphoid follicles and only lymphatic vessels were traced from the terminal bronchiolar region toward larger bronchi. During the week after intranasal inoculation of mycoplasma pulmonis, lymphoid follicles developed in the terminal portion of the lymphatics by the accumulation of small lymphocytes. A loose network of mesenchymal cells and early infiltration of macrophages, following stromal edema, seemed to play an important role in the early accumulation of lymphocytes. Blastic transformation was seen frequently in the center of the accumulated lymphocytes. Two weeks after inoculation plasma cells emerged conspicuously in the periphery of the lymphoid follicles, and the acute phase of bronchopneumonia began to subside. Typical germinal centers with tingible body macrophages and dendritic reticulum cells developed when the bronchitis persisted in a chronical manner.
Fifty-two cases of non-Hodgkin's lymphomas were studied with enzyme histochemical methods. Forty-four cases of these were also investigated for surface markers with immunological techniques, and results of histochemical, routine histological and immunological observations were correlated. Twenty-one of 27 B-cell lymphomas showed prominent ATPase activity, while all 13 T-cell lymphomas, except one case, did not show such activity. Nodular lymphomas, though of B-cell nature, were often negative for ATPase and it remained negative after diffuse evolution in some. Four of 7 A1Pase positive lymphomas were of B-cell origin. Dot-like localized AcPase and beta-glucuronidase activity characterized T-cell lymphomas while 5 T-cell PDL, including lymphoblastic type with double markers, showed localized esterase activity. Enzyme histochemical characteristics of lymphomas were fairly honest reflection of those of various functional units in the normal lymph nodes. Enzyme histochemical methods appeared to be a useful tool for the study of lymphomas.
Twenty-nine cases of malignant lymphoma were studied by the enzyme histochemical method and membrane surface markers. Strong adenosine triphosphatase (ATPase) activity was demonstrated in 15 cases of 20 B lymphomas and in one case of null-cell lymphoma (1/1) by a light- and electron-microscopic enzyme histochemical method according to Wachstein et al. Neoplastic cells in nodules of 2 nodular lymphomas (0/2) and in 8 cases of T lymphomas (0/8) did not show any ATPase activity enzyme-histochemically. The ATPase activity of B lymphomas was electron-microscopically demonstrated on the plasma membrane of lymphoma cells but not in the cytoplasmic organelles. The specificity of ATPase activity in B lymphomas was also examined enzyme-histochemically with reference to normal lymphoid tissues. Such specificity was similar to that of non-neoplastic B lymphocytes of primary follicles and the mantle zone of secondary follicles in lymphoid tissues. Therefore, ATPase is one of the useful enzyme histochemical markers in differentiating T and B subclasses of malignant lymphomas.
Recently, urinary pyridinoline and deoxypyridinoline have been commonly employed as bone resorption markers. We studied these markers in 17 patients with hyperthyroidism, 15 undergoing long-term anticonvulsant drug therapy, and 28 with postmenopausal osteoporosis. Both markers had significantly higher levels than those in age-matched control groups. Values of urinary pyridinoline and deoxypyridinoline correlated well with urinary hydroxyproline levels in patients with hyperthyroidism (r = 0.856, p < 0.001 for pyridinoline and hydroxyproline; r = 0.919, p < 0.001 for deoxypyridinoline and hydroxyproline); however, poor correlations were observed, especially between urinary deoxypyridinoline and urinary hydroxyproline (r = 0.357, NS) in patients with postmenopausal osteoporosis. To compare the discriminatory ability of urinary pyridinoline and deoxypyridinoline, receiver operating characteristic (ROC) curves were generated for each of these patient groups using data from age-matched healthy females as the control group. The areas under the curves for both markers were 100.0% in hyperthyroidism. The areas under the curves for pyridinoline in patients undergoing long-term anticonvulsant drug therapy (mean +/- SE; 98.1 +/- 2.8%) and postmenopausal osteoporosis (77.9 +/- 5.7%) were significantly higher than those for deoxypyridinoline in anticonvulsant drug therapy (92.4 +/- 3.3%) and in osteoporosis (64.9 +/- 4.3%). Using data from premenopausal healthy females as the control group, areas under ROC curves for urinary pyridinoline (100.0%) and deoxypyridinoline (94.8 +/- 5.9%) were significantly higher than those for urinary hydroxyproline (73.8 +/- 9.4%) in patients undergoing long-term anticonvulsant drug therapy. In patients with postmenopausal osteoporosis, those for urinary pyridinoline (97.0 +/- 2.8%) were also significantly higher than those for urinary hydroxyproline (74.0 +/- 6.4%).(ABSTRACT TRUNCATED AT 250 WORDS)
BACKGROUND/AIMS: Immunological status has been considered to correlate to the response to interferon therapy for chronic hepatitis C. The aim of this study was to evaluate the correlation between humoral immunity and long-term response to interferon treatment for chronic hepatitis C. METHODOLOGY: Seventy-one patients with chronic hepatitis C received 10 million units of interferon-alpha 2b three times a week for 24 weeks. Peripheral blood mononuclear cells were obtained before interferon-alpha 2b was administered and were cultured for 7 days. Immunoglobulin concentration in the culture supernatants was measured by enzyme-linked immunosorbent assay and correlation with the response to the therapy was evaluated. RESULTS: Serum ALT levels normalized in 51.4% and hepatitis C virus RNA disappeared in 35.7% six months after the end of therapy. Immunoglobulin production was significantly lower in the patients in whom serum ALT levels normalized than those in whom serum ALT levels remained elevated. The similar result was obtained when efficacy was evaluated on the basis of hepatitis C virus RNA disappearance. CONCLUSIONS: These results suggest that the less humoral immunity, the better response to interferon will be obtained in patients with chronic hepatitis C, meaning that the balance in T-helper function is one of key factors in the response to interferon treatment.
