Groups of three or four male and female beagle dogs were maintained for 6 months on diets containing BHA at concentrations of 0 (control group), 0.25, 0.5 and 1.0% (w/w). The average daily intake of BHA was about 60 mg/kg body weight in the 0.25% group, 110 mg/kg in the 0.5% group and 220 mg/kg in the 1.0% group. All animals survived the entire experimental period without showing signs of toxicity other than a dose-related retardation of growth. Serum biochemical examinations conducted at 1, 3 and 6 months revealed a slight decrease in albumin concentration and an elevation of alkaline phosphatase and leucine aminopeptidase activity in the higher dose groups. Histopathological and histometrical examination showed no evidence of mucosal alteration in the stomach, oesophagus or duodenum attributable to the administration of BHA. The mitotic index in the squamous epithelium of the distal oesophagus was comparable in the control and BHA-treated groups. Liver weights were increased in BHA-treated groups, but there were no related histopathological changes. From these data it is concluded that feeding of BHA-supplemented diets at palatable concentrations for 6 months has no pathological effects on the stomach, oesophagus, duodenum or liver of beagle dogs.

The ciliated protozoan Paramecium contains hemoglobin in heterogeneous monomeric forms. In particular, Paramecium caudatum is characterized by the presence of a major component called Hb10 and a basic component named bHb. We found that in P. caudatum both of these hemoglobin components show some variation according to stock. The types and distributions of these hemoglobin components were examined on 16 stocks in five different syngens and one stock in an unidentified syngen using high performance liquid chromatography. The results indicate that in a variety of stocks the major component, Hb10, was divided into three types, A, B or A + B, and that the basic hemoglobin component was composed of a combination of two or three variants out of four possible, i.e. bHb 1, bHb 2, bHb 3 and bHb 4. Neither the Hb10 types nor the bHb variants, however, could be used to distinguish syngen in P. caudatum, since all of the Hb10 types and bHb variants were widely distributed over syngens and identical profiles appeared to some stocks in different syngens.
The presence of circulating anti-p53 antibodies (p53Ab) was investigated in 81 gastric cancer patients by ELISA. Preoperatively, anti-p53Ab were revealed in 13 cases (16%), including 3 early gastric cancers. No significant correlation of p53Ab with patient age, sex, pathological parameters and tumor markers, such as CEA and CA19-9 was observed, although p53Ab positivity tended to be associated with a higher rate of lymph node metastases (p = 0.073). We have found no significant correlation between p53Ab and poor survival rate (p = 0.325). In conclusion, we have observed that p53Ab are detectable in gastric cancer patients, even at early stages of disease, however, p53Ab have not served as a predictor of poor prognosis in gastric cancer in this series.
We have investigated the antitumor effect of murine colon carcinoma cells developed to produce cytokines. Retrovirally transduced cells were inoculated into syngeneic mice, and their survivals were examined. Inoculation of interleukin-4 (IL-4)- or granulocyte macrophage-colony stimulating factor (GM-CSF)-producing cells alone did not affect the survival of the mice. In the tumors derived from an admixture of IL-4 and GM-CSF producers, however, regression developed spontaneously, and prolonged survival of the challenged mice was consequently observed. Histological examination revealed predominant infiltration of neutrophils around the tumor of a mixed population of IL-4 and GM-CSF producers.
We aimed to evaluate the subcutaneous tissue reaction to a new calcium hydroxide paste for root canal fillings developed by Neo Dental Chemical Products Co., Tokyo, Japan. We injected calcium hydroxide paste and a water-based control subcutaneously into the dorsal area of 6-week-old anaesthetized male ddY mice. The tissue surrounding the injection sites was removed from mice in each group, 2 days, 1 week, 3 weeks and 12 weeks after the injection, and histopathologically examined. We found necrotic and degenerative changes, as well as foreign body reaction or proliferation of granulation tissue in specimens from both groups until 3 weeks after the injection. Some lymphocyte infiltration was observed in the 12-week specimens of the control group only. We conclude that calcium hydroxide paste is safe to use as a root canal filling material.
The significance of serum p53-Abs in patients with esophageal squamous cell carcinoma was determined. Examination of clinicopathological features and assessment of tumor marker sensitivities of carcinoembryonic antigen (CEA), squamous cell carcinoma antigen (SCC-Ag) and CYFRA21-1 were performed. Thirty-three (58%) of 57 patients were positive for serum p53-Abs, however, no relation with cancer progression existed. Fourteen of the 33 sero-positive patients revealed normal levels of all tumor markers tested. Thus, serum p53-Abs appears to be a useful marker for the detection of esophageal squamous cell carcinoma.
