Collision of malignant lymphoma and multiple early adenocarcinomas of the stomach.

An unusual case of coexisting malignant lymphoma and adenocarcinoma of the stomach is described. A 61-year-old man with weight loss, malaise, and tarry stool demonstrated diffuse lymphoma, large-cell type, and two early gastric carcinomas. Each of the two early carcinomas collided with malignant lymphoma, respectively. Two histologically different tumors were precisely diagnosed preoperatively in this case. Frequent endoscopy with biopsies from various sites of the lesion is important in achieving a correct diagnosis.

Identification of essential arginine residue(s) for Mg-ATP binding of human argininosuccinate synthetase.

Human argininosuccinate synthetase (ASS) activity was found to be inactivated by alpha-dicarbonyls such as 1,2-cyclohexanedione and phenylglyoxal in accordance with pseudo first-order kinetics. The enzyme was almost completely protected from this inactivation by Mg-ATP and partially by its analogues. The strongest protective effect against inactivation was found with Mg-ATP, followed by Mg-ADP, AMP, adenosine and Mg-inorganic pyrophosphate. These results suggest the importance of arginine residue(s) for Mg-ATP binding. We determined the amino acid sequence of the peptide with the highest specific radioactivity derived from ASS which had been labeled with [14C]phenylglyoxal and then cleaved by cyanogen bromide treatment. The sequence obtained, PEFYNRFKGRNDLM, corresponds to residues 148-161 of the amino acid sequence deduced from the cDNA nucleotide sequence determined by Bock et al. [Nucleic Acids Res 11:6505-6512, 1983], and has a high homology with the sequences of ATP-binding sites proposed for several ATP-requiring enzymes.

Isolation of a new type of human papillomavirus (HPV52b) with a transforming activity from cervical cancer tissue.

Substantial evidence has implicated human papillomaviruses (HPVs) as etiological agents of human cervical cancer. We detected and cloned an unidentified HPV genome in cellular DNA extracted from a surgical specimen of a Japanese patient with cervical cancer. Hybridization studies suggested that this HPV was the most homologous to HPV33 among more than 51 types of HPV ever identified. A recently identified new HPV, isolated by W. Lancaster and designated HPV52, is also most homologous to HPV33 (W. Lancaster et al., unpublished data). Our HPV was found to be homologous but not identical to HPV52 and hence was designated HPV52b. By introducing it into NIH3T3 cells, we found that HPV52b DNA had growth-stimulating activity as HPV16 DNA. This newly identified HPV52b DNA was present as episomes in cervical cancer cells of two out of two specimens so far examined and the integrated copies were not detected. HPV52b was present in three out of 15 (20%) specimens in Japan. In addition, the presence of three additional unidentified HPV sequences homologous to HPV52b was detected in three other specimens. The results suggest that this group of HPV may be prevalent and involved in carcinogenesis of cervical cancer in Japan.

Interaction of human parathyroid hormone-related peptide with parathyroid hormone receptors in clonal rat osteosarcoma cells.

Synthetic peptides corresponding to the amino-terminal region of the human parathyroid hormone-related peptide (hPTHrp) were used to characterize the interaction of hPTHrp with parathyroid hormone (PTH) receptors in clonal rat osteosarcoma cells (ROS 17/2.8). Both hPTHrp-(1-34) and [Tyr40]hPTHrp-(1-40) showed full agonist activity in stimulating cyclic AMP accumulation in ROS cells; human PTHrp-(1-34) was approximately 2.5-fold as potent as hPTH-(1-34). Both [Tyr-40]hPTHrp-(3-40) and hPTH-(3-34) inhibited the cyclic AMP increase induced by either hPTHrp or PTH with parallel dose-inhibition curves. Binding to intact ROS cells of a 125I-labeled [Tyr40]hPTHrp-(1-40) (125I-[Tyr40]hPTHrp-(1-40)) which retains full biological activity was time- and temperature-dependent and reversible. Binding of 125I-[Tyr40]hPTHrp-(1-40) and 125I-labeled [Nle8, Nle18, Tyr34]bovine PTH-(1-34)NH2 to ROS cells was competed for, to the same extent and with the comparable potency, by either unlabeled hPTHrp or PTH peptides. The binding capacity and affinity of receptors in ROS cells were strikingly similar for hPTHrp and PTH. Affinity cross-linking with either radioligand resulted in high affinity, specific labeling of an apparently identical macromolecule centering at Mr = 80,000, which was detected in sodium dodecyl sulfate-polyacrylamide gel electrophoresis in both reducing and nonreducing conditions. The data indicate that hPTHrp and PTH, their amino-terminal fragments at least, interact with the identical receptors with regard to affinity, capacity, specificity, and physicochemical characteristics in osteoblastic ROS 17/2.8 cells.

Human parathyroid hormone related protein fragment-(1-34) had glucose-6-phosphate dehydrogenase activity on distal convoluted tubules in cytochemical bioassay.

In the present study, the action of parathyroid hormone related protein (PTHrP) on glucose-6-phosphate dehydrogenase (G6PD) activity of the distal convoluted tubules was examined utilizing cytochemical bioassay (CBA). Recently full amino acid residues of human PTHrP (hPTHrP), one of the causative agents of HHM, was identified based on the cDNA clone using BEN cells. We synthesized hPTHrP-(1-34) and examined the effect of this protein on G6PD activities on the distal convoluted tubules, and compared its bioactivity to that of human parathyroid hormone (hPTH)-(1-84). hPTHrP-(1-34) stimulated G6PD activity in a log linear fashion with equivalent activity to that of hPTH-(1-84) on a molar basis. Conclusively, we found that PTHrP act on distal convoluted tubules similar to hPTH.

Receptors for calcitonin gene-related peptide on the rat liver plasma membranes.

Specific binding sites for calcitonin gene-related peptide (CGRP) were identified in the rat liver plasma membrane. The binding of 125I-[TyrO]rat CGRP to rat liver plasma membrane was time dependent, saturable and reversible. Scatchard analysis of the data revealed a single class of binding sites with apparent dissociation constant of 260.8 pM and a maximal binding capacity of 26.6 fmol/mg of protein. Rat, chick, and human CGRP and their synthetic analogues inhibited label binding in a dose-dependent manner with relative potencies as follows; chick greater than rat greater than human greater than [TyrO]rat CGRP. Salmon, human and [Asu1'7]eel calcitonin also inhibited label binding but only at higher concentrations. These results clearly indicate the presence of specific binding sites for CGRP in rat liver plasma membrane and suggest that CGRP has possible biological actions on the rat liver.

Sensory input to cerebellocerebral relay neurons in the cat thalamus.

Intracellular recording techniques were used to study the synaptic responses of feline ventrolateral neurons to dorsal column, spinothalamic and sensory cortex stimulation. More than 75% of the cells responded with short- and long-latency excitatory or inhibitory postsynaptic potentials to the one or/and other stimulation sites. These findings indicate that a considerable amount of somatosensory integration in the cerebellocerebral circuit occurs in the thalamic relay neurons.

Simultaneous determination of noradrenaline and 3-methoxy-4-hydroxyphenylglycol in plasma with high-performance liquid chromatography using electrochemical detection.

We herein report the simultaneous determination of the levels of noradrenaline (NA), and 3-methoxy-4-hydroxyphenylglycol (MHPG), a major metabolite of NA. The sample was subjected to a Sep Pak C18 cartridge prior to the NA and MHPG assay by high-performance liquid chromatography with an electrochemical detector. The results correlated well with the established methods. The average percentage of recovery was 91.2 and 98.7% for NA and MHPG, respectively. The intraassay coefficients of variation were 3.7 and 4.6% for NA and MHPG. The interassay coefficients of variation were 3.5 and 7.5% for NA and MHPG, respectively.

Hepatotoxicity of gardenia yellow color in rats.

Acute toxicity by gardenia yellow color was studied in rats. Oral administration of the colorant at doses of 800 mg/kg up to 5000 mg/kg caused diarrhea and increases in serum alanine aminotransferase and aspartate aminotransferase activities in a dose-dependent manner. After 24 h of oral treatment with 2000 mg/kg of the colorant, liver showed partially hemorrhagic changes and the intestinal tract, especially the duodenum, appeared blue. The toxicity induced by the colorant was stronger by oral administration than by intraperitoneal administration. The content of geniposide, an iridoid compound, was estimated to be 28% of the colorant, and this iridoid accounted for almost all the hepatotoxic activity of the colorant.

The heterogeneous distribution of argininosuccinate synthetase in the liver of type II citrullinemic patients. Its specificity and possible clinical implications.

The authors analyzed the heterogeneous distribution of hepatic argininosuccinate synthetase of type II citrullinemia in reference to its specificity and clinical implications. The low content of the enzyme in the liver of type II citrullinemic patients is associated with two kinds of the enzyme distribution that can be visualized by means of an immunohistochemical method (Saheki and colleagues. Biomed Res 1983;4:235-238). Among the 25 cases of type II citrullinemia examined, 11 exhibited homogeneous distribution of the enzyme, as in the control livers. On the other hand, 14 presented the clustered distribution, in which the hepatocytes stained positively with antisera to argininosuccinate synthetase formed a cluster among the poorly stained cells. No clustered distribution of the enzyme was present in the liver of control patients either with or without liver diseases. No clustered distribution of arginase and aldolase B was observed even in the liver of type II citrillinemic patients. These results suggest that clustered distribution is specific to argininosuccinate synthetase in the liver of type II citrullinemic patients. From considerations concerning the heterogeneous distribution of the enzyme and certain clinical parameters as well, the authors suggest that the clustered type in type II citrullinemia has a less favorable prognosis with regard to fatality.

Progression of the phenotype of transformed cells after growth stimulation of cells by a human papillomavirus type 16 gene function.

Alteration of the growth properties of the established murine fibroblast cell lines NIH 3T3 and 3Y1 was studied in monolayer cultures and in cells suspended in semisolid medium after introduction of a cloned human papillomavirus type 16 (HPV16) DNA. HPV 16 DNA stimulated both cell lines to grow beyond their saturation densities in monolayer cultures without any apparent morphological changes or tendency to pile up. These cells were also stimulated to grow in soft agar. Since essentially all the cells that received the viral gene were stimulated to grow, the growth-stimulatory activity of HPV16 appeared to be due to the direct effect of a viral gene function. The NIH 3T3 cells showed an additional change in growth properties upon prolonged incubation of dense monolayers of cells containing the HPV16 DNA; morphologically recognizable dense foci appeared at a frequency of about 10(-3). These cells, when cloned from the foci, grew more rapidly in soft agar than the parental cells and were morphologically transformed. In other words, there were two sequential steps in cell transformation induced by HPV16. Practically all the viral DNAs were present in the cells as large rearranged multimers and were integrated into host chromosomal DNA. There was no obvious difference in the state of viral DNA in the cells of the original clone or the three subclones derived from it as dense foci. There was no difference in the amount or the number of viral RNA species expressed in the cells at these two stages. The secondary changes in the growth properties of NIH 3T3 cells appear to be due to some cellular alterations.

[Pharmacokinetics of N4-behenoyl-1-beta-D-arabinofuranosylcytosine after intraperitoneal administration].

The patients with cartinomatous peritonitis were treated with the intraperitoneal administration (ip) of N4-behenoyl-1-beta-D-arabinofuranosylcytosineine (BH-AC: analogue of Ara-C), and the pharmacokinetics of BH-AC ip was studied. The following results were obtained (1) Immediately after ip administration, the concentration of BH-AC in ascites became as high as 10(6) ng/ml. At 24 hours following BH-AC ip, 10(4) ng/ml of BH-AC was detected in ascites. (2) Immediately after ip administration the concentration of Ara-C derived from BH-AC in ascites became as high as 10(3) ng/ml. At 24 hours, more than 10 ng/ml of Ara-C was detected in the ascites. (3) Ara-U in ascites was detected also soon after BH-AC ip was performed. Accordingly, it is expected that deaminase may be present in ascites. (4) As compared with in ascites, Ara-C in plasma showed very low level (less than 1 ng/ml). These findings indicate that BH-AC is suitable drug for intraperitoneal administration, because BH-AC revealed low peritoneal and high plasma clearances.

Effect of human parathyroid hormone (PTH(1-34)) on experimental osteopenia of rats induced by ovariectomy.

The effect of human PTH(1-34) on the development of osteopenia induced by ovariectomy was investigated in immature female Sprague-Dawley rats. After the surgery, human PTH(1-34) was injected subcutaneously three times a week for 25 weeks. A reduction of serum calcium level and a tendency to increased serum alkaline phosphatase activity were seen in ovariectomized rats. The ovariectomized rat bones were characterized by reduction of dry weight, ash weight, calcium content and phosphorus content, but there was no change in length, volume and ash content in these bones. Human PTH(1-34) prevented the reduction of dry weight, ash weight, calcium content and phosphorus content dose dependently (1.5-6.0 micrograms/kg) in ovariectomized rats. It was concluded that pulsatile administration of human PTH(1-34) prevented the development of osteopenia induced by ovariectomy.

[A case report of advanced ovarian cancer with chronic renal dysfunction treated by CDDP i.p].

Reported in this paper is a case of a 56-year-old female patient who was diagnosed as having an ovarian cancer (stage T 4) with a renal dysfunction (CCr 30.5 ml/min). We instilled CDDP intra-peritoneally (CDDP-ip), taking into consideration the peritoneal clearance (CLp.). The administration of sodium thiosulphate also was combined. The low CLp. of this case (299.2 ml/h) made it possible to instill a high dose in the peritoneal cavity and a low dose in the plasma. This method provided enough of anti-tumor effect and kept the renal function safe. We believe that the CLp. is the most important factor in determining the effect and side effects of CDDp-ip.
