[Induced rat ovarian carcinoma showing mainly squamous components--an experimental approach to common epithelial tumors].

Female Wistar strain rats were treated with a chemical carcinogen DMBA (7,12-Dimethylbenz (a) anthracene) implanted directly into their ovary. In this method of tumor induction, adenocarcinoma commonly developed in the ovary by the direct carcinogenic effect of DMBA during a 30 weeks observation period. However, two cases of primary ovarian carcinoma showing mainly squamous components have been found in a recent experiments. The histological characteristics of these indicated that they were adenoacanthoma and adenosquamous cell carcinoma and both were derived ultimately from the ovarian surface epithelium with interposition of endometrioid type tumor. These two cases of experimental rat ovarian cancer consist of mainly squamous element are studied morphologically and their histogenesis is discussed.

Light and electron microscopic observations of the spleen in the musk shrew, Suncus murinus.

In the spleen of the musk shrew, a marginal zone was clearly demonstrable between the white pulp and the red pulp. The white pulp was separated from the marginal zone by a septum, a circumferential layer of reticular cells and fibres, and between the marginal zone and red pulp there was a population difference of blood cells. The white pulp consisted of periarterial lymphatic sheaths and lymphoid follicles including germinal centres. The marginal zone was filled by numerous erythrocytes, some lymphocytes, granulocytes and a few macrophages, while the red pulp had a large number of neutrophils, eosinophils, megakaryocytes and their precursors. In the red pulp erythroblasts tended to form islets, and there were a few basophils, plasma cells, plasmablasts and macrophages. Haematopoiesis was also active in the bone marrow.

Fibronectin in corneal wound healing: appearance in cultured rabbit cornea.

Fibronectin (FN) has been detected at the site of corneal wounds. To investigate whether FN is supplied by the cornea itself or by extracorneal sources, we cultured small blocks of rabbit cornea in vitro with and without autologous serum (5%). The sliding rate of epithelial cells was determined and localization of FN in the corneal block examined by indirect immunofluorescent microscopy. Epithelial cells slid more rapidly in the presence of serum and covered most of the stromal surface in about 24 hours. FN was detected at the surface edge of the cut stroma shortly after culturing began. During epithelial cell sliding, FN-specific fluorescence was noted beneath the migrating cells. The location of FN was essentially the same in cultures with and without autologous serum. These results suggest that FN might be supplied by the cornea itself and that it may play a role in epithelial cell sliding.

Rapid preparation of purified autologous fibronectin eyedrops from patient's plasma.

Fibronectin (FN), a glycoprotein present in plasma and the extracellular matrix, has been reported to be effective on corneal trophic ulcer. This study reported a rapid method for preparing purified FN eyedrops from a patient's own plasma. Plasma FN concentration in healthy Japanese was found to be 235 +/- 62 micrograms per ml(age 21-31 years old). Men had a higher plasma FN level than women (264 +/- 62 vs. 206 +/- 44 micrograms/ml). The FN eyedrops were prepared by gelatin-coupled Sepharose 4B affinity chromatography and gel filtration by Sephadex G-25 (PD-10). Biochemical and immunochemical analyses revealed that the prepared FN eyedrops contained electrophoretically pure FN and no other plasma proteins were detected. Average FN concentration in the eyedrops was 425 micrograms per ml and recovery rate was 54%. The FN eyedrops were found to be stable at room temperature up to 10 days in biological activity as far as we examined. The total time required to prepare the FN eyedrops was within 2 hours. Thus it is possible to apply them to outpatients as well as inpatients.

Synthetic study on human C-peptide and its related peptides.

Syntheses of eighteen peptides related to human C-peptide and the connecting peptide are described. The syntheses were performed exclusively by the azide fragment condensation method. The immunochemical results justified the utilization of these peptides for development of human C-peptide radioimmunoassay system. The synthetic peptide fragments may be extremely important substrates for assessment of specificity of human C-peptide antisera and thus usefulness of the antisera in practical measurement of blood levels of human C-peptide immunoreactivity.

Complement-dependent cytotoxicity in rats bearing human adenovirus type 12-induced primary retinoblastoma-like tumor in the eye.

Using an animal model of retinoblastoma in inbred rats and cultured human adenovirus type 12-induced retinoblastoma-like tumor cells (RAO 188), complement-dependent cytotoxicity was determined by measuring release of 3H-uridine labelled RNA. Sera from rats in which tumors did not grow after adenovirus type 12 inoculation had higher cytotoxicity against RAO 188 cells than sera from rats bearing primary adenovirus type 12-induced retinoblastoma-like tumor. These results showed that the rat which could raise antibodies against adenovirus type 12-induced retinoblastoma-like tumor cells did not allow the tumor growth in the eye after virus inoculation.

Development of ovarian tumors experimentally studied in mice by 60Co-ray irradiation.

Female C3H strain mice aged 5 to 6 weeks were exposed to a single dose of 400 Rad irradiation to their lower abdomens and the morphological changes of their ovaries were observed until 72 weeks after irradiation. The incidence of tumor development increased gradually from 34 weeks after irradiation till 72 weeks, when ovarian tumors were noted in more than 90% of the animals. Three types of ovarian tumors developed after a prolonged period, postirradiation: tubular adenoma, granulosa cell tumor and luteinizing tumor. Mixed tumor types account for 68% of the tissue examined. The tubular adenoma is believed to arise from the germinal epithelial inclusion cyst, the granulosa cell tumor and luteinizing cell tumor arise from the interstitial cells. In the case of the granulosa cell tumor, it is believed that tumor cells one capable of secreting estrogen, because vaginal smears demonstrated estrus and histological sections of endometrium demonstrated hypertrophy.

Ovarian tumors in children and adolescents less than 20 years age.

Reports are made of the analytical results of 76 cases with ovarian neoplasma under 20 years of age who had been treated at the Kurume University Hospital from January 1952. The incidence of ovarian tumors was 5.5% in all age groups, and the youngest patient was 5 years of age, the mean age being 15 years. Histologically, these tumors comprised 48 cases (63%) of germ cell origin, showing characteristically of ovarian tumors occurring in childhood and adolescence. The total 76 cases included 40 of malignant tumors, which were subdivided into 13 of embryonal carcinoma, 12 of solid teratoma, 9 of dysgerminoma, 5 of simple carcinoma, and 1 case of granulosa cell tumor. In all the cases the clinical stage was important factor for prognosis, and the prognosis of embryonal carcinoma was poorest, with only 4 cases of survival out of 13 cases. But the prognosis of dysgerminoma was comparatively good, with 8 cases of survival out of 9 cases. It is worthy of note that this tumor was highly radiosensitive, since 3 out of 4 cases at stage III and stage IV had been surviving for over 5 years.

Chemotherapy of malignant ovarian tumors; therapeutic results of ifosfamide.

Here are reported the therapeutic results of ifosfamide on primary ovarian malignancies in 30 cases which were treated at Kurume University Hospital from September 1978 to December 1980. These cases were advanced or recurrent ones, and histologically, they consisted of serous cystadenocarcinoma in 22 cases, mucinous cystadenocarcinoma in 2, unclassified adenocarcinoma in 1, and malignant teratoma in 5. In the ifosfamide therapy, 2 grams or 40 mg/kg per day were given intravenously for 5 consecutive days, and as observing the blood state and the general condition, it was repeated every 3 weeks. The drug was given from 1 to 7 courses, and the average was 3.3 courses. The effect was evaluated by Koyama-Saito's criteria and Karnofsky's criteria; the response rate was 48% in the adenocarcinoma group and 20% in the malignant teratoma by the former criteria, while the rate was 64% and 20%, respectively, by the latter one. As main side effects, there were seen hemorrhagic cystitis, nausea and vomiting, and the myelotoxicity was low.

Malignant degeneration of benign cystic teratomas of the ovary.

In 10 years from January 1971 to January 1981, 145 cases with cystic teratoma were treated at Kurume University Hospital, and malignant degeneration was seen in 7 of them (4.8%). Their age distribution was from 43 to 78 years, and the mean age was 59 years; the distribution shifted to the older side. The malignant degeneration was squamous cell carcinoma in all of the cases, and in 1 case carcinoma in situ leads to invasive carcinoma was observed continuously to a normal epithelium; it was interesting histogenetically. In their prognoses 3 out of 7 cases died (1 case dying from decrepitude) but the rest 4 have been surviving for 4 to 10 years.
