Heparin inhibits the progression phase of subcultured endothelial cell proliferation in rat aorta.

The anti-proliferative effects of heparin on subcultured endothelial cells (EC) of rat aorta were investigated to determine whether heparin inhibits the competence phase or the progression phase in the cell cycle using the starting time and the rate of proliferation. Fetal bovine serum (FBS)-stimulated EC proliferation increased to 1.5-times in cell number, but decreased to 0.5-times in 3H-thymidine incorporation, compared with the proliferation of rat primary cultured smooth muscle cells (SMC). The FBS-effects on EC proliferation were attributed to the progression phase rather than the competence phase. Heparin (1, 10 or 100 micrograms/ml) significantly inhibited the proliferation of FBS (5%)-stimulated EC, and the extent of inhibition was the same in both cell number and 3H-thymidine incorporation. In the 3H-thymidine incorporation every 3 hr, heparin reduced the rate of incorporation into G0-arrested EC, but did not affect the starting time of DNA synthesis. When the index of competence phase, starting time, was plotted against that of progression phase, rate of proliferation, the inhibition of heparin was attributed to the progression phase. These results demonstrate that heparin selectively inhibits the progression phase in subcultured EC of rat aorta.

[A case of small liver cancer presenting as a huge mediastinal mass].

A 61-year-old male was admitted because of hemoptysis. He had a 9 year history of liver cirrhosis associated with HB viral chronic hepatitis. Physical examination revealed no abnormalities. Laboratory investigations revealed positive HBs antigen with normal alpha-fetoprotein. Chest X-ray film showed large mediastinal lymph nodes and an endobronchial polypoid mass in the distal end of the right main bronchus. The right main PA was narrowed due to compression by the mediastinal mass. Bronchoscopic examination revealed a polypoid mass in the right main bronchus. The biopsy specimen was histologically diagnosed as undifferentiated large cell carcinoma. The patient developed respiratory failure, and died 3 weeks after admission. Autopsy revealed a small liver cancer of 1.3 cm diameter within the cirrhotic liver, associated with a small abdominal lymph node metastasis and large mediastinal lymph node swellings. Thromboembolism in the bilateral main pulmonary arteries was concluded to be the cause of death. The mediastinal mass which directly invaded into the right main bronchus had a close histological similarity with the liver cancer, showing undifferentiated carcinoma cells with bizarre nuclei and abundant cytoplasm. An immunohistological study revealed cells positive for alpha-fetoprotein in the mediastinal lymph nodes. The patient was diagnosed as having small liver cancer with mediastinal lymph node metastases. A survey of the literature revealed only a few cases of advanced hepatoma associated with prominent mediastinal metastases. This is the first reported case of small liver cancer presenting with large mediastinal lymph node metastases.

The background adaptation of the flatfish, Paralichthys olivaceus.

The background adaptation of the flatfish, Paralichthys olivaceus, was studied by sliding beneath the fish a long strip of plastic sheet with various patterns to serve as a background. The body shade and pattern differed depending upon the pattern used. In general, the fish showed maximal pallor on a white background and darkened when lying on a black background. With a pattern of large checks, several black patches became conspicuous. The fish showed the same tendency in terms of response even when the left eye was covered. However, the fish was prevented from manifesting these changes when the right eye was covered. The results suggest that the left eye of the fish does not see the immediate background. This conclusion is supported by an anatomical study.

The weak calcemic vitamin D3 analogue 22-oxacalcitriol suppresses the production of tumor necrosis factor-alpha by peripheral mononuclear cells.

The effect of 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) and its analogue 22-oxacalcitriol (OCT), which was reported to have very weak bone resorbing activity, on the production of tumor necrosis factor (TNF)-alpha was investigated. Mononuclear cells (MNC; 10(6)/ml) were incubated in 5% FCS/RPMI-1640 medium containing 1 microgram/ml lipopolysaccharide (LPS) in the presence or absence of 10(-8) M 1,25(OH)2D3 or 10(-8) M OCT for up to 96 h. Both 1,25(OH)2D3 and OCT suppressed TNF-alpha release by LPS-stimulated mononuclear cells, from the early to late stage of the incubation period, while neither 1,25(OH)2D3 nor OCT shifted the peak time point of TNF-alpha release clearly. MNC (10(6)/ml) were incubated with 1 microgram of LPS in the presence of various concentrations of 1,25(OH)2D3 or 10(-8) M OCT for 48 h. 1,25(OH)2D3 reduced TNF-alpha levels of LPS-stimulated MNC culture supernatant at 48 h in a dose-dependent manner. The half-maximal dose (ED50) for this suppressive effect was 3.7 x 10(-9) M. OCT decreased TNF-alpha levels of culture supernatant at 48 h with a half-maximal dose of 7.8 x 10(-11) M, which indicates that it is approximately 50 times more potent than that of 1,25(OH)2D3. These results indicate that OCT may be applicable as an immunosuppressive agent with limited calcium metabolic activity.

Interspecific variation in the emetic response of anurans.

1. The emetic response of seven species (four genera) of frogs to apomorphine hydrochloride, copper sulfate, antimonyl potassium tartrate and mechanical stimulation at the esophageal orifice was surveyed. Xenopus laevis and Rhacophorus schlegelii were more sensitive to systemically administered apomorphine than were the other species tested. 2. The sensitivity of Rana rugosa to apomorphine varied with the season. 3. All of the species showed vigorous vomiting behavior after the oral administration of either copper sulfate or antimonyl potassium tartrate. 4. Mechanical stimulation also induced vomiting in all species. Although the species differed in sensitivity to the different emetic stimulants, the adaptive significance of this interspecific variation is not known. 5. From the stand point of the mechanics for ejecting gastric contents, there is little difference between frogs and mammals. 6. Frogs, particularly Xenopus laevis, may be a useful non-mammalian model for studying emesis.

Synthesis and biological evaluation of substituted benzenesulfonamides as novel potent membrane-bound phospholipase A2 inhibitors.

A novel series of 4-[N-methyl-N-[(E)-3-[4-(methylsulfonyl)phenyl]-2- propenoyl]amino]benzenesulfonamides has been prepared and evaluated as membrane-bound phospholipase A2 inhibitors. A structure-activity relationship study indicated that the optimum potency was realized with the N-(phenylalkyl)piperidine derivatives 3 and 4. These compounds inhibited the liberation of arachidonic acid from the rabbit heart membrane fraction with IC30 values of 0.028 and 0.009 microM, respectively. Several compounds (3, 4, and 28), which proved to be potent inhibitors in vitro, significantly reduced the size of myocardial infarction in coronary occluded rats by iv administrations prior to the ligation. N-(1-Benzyl-4-piperidinyl)-4-[N-methyl-N-[(E)-3-[ 4-(methylsulfonyl)phenyl]-2-propenoyl]amino]-benzenesulfonamide (3, ER-3826), which showed the protective in vivo effects at doses higher than 0.3 mg/kg iv, was finally chosen as a leading candidate.

Intracellular Ca2+ and Mg2+ regulation for insulin-stimulated glucose uptake into mouse diaphragm muscles.

We investigated whether the changed levels of intracellular Ca2+ and Mg2+ in mouse skeletal muscles affected insulin-stimulated glucose uptake. Insulin alone had no effect on 45Ca2+ efflux and uptake. A23187 at 20 microM increased 45Ca2+ influx and inhibited insulin-stimulated 14C-glucose uptake with normal external concentrations of Ca2+ and Mg2+ or in the absence of both cations. These results suggest that the increase in Ca2+ influx into, and in Mg2+ efflux from, skeletal muscles may inhibit insulin-stimulated glucose uptake.

Isolation of two endo-beta-N-acetylglucosaminidases with different specificities from Pseudomonas sp.

Two endo-beta-N-acetylglucosaminidases (PI and PII) have been isolated from the culture fluid of Pseudomonas sp. The substrate specificity of the PI enzyme was very similar to that of Endo-H from Streptomyces plicatus. On the contrary, the PII enzyme had a novel substrate specificity that degraded both high-mannose type and hybrid type oligosaccharides derived from ovalbumin, and the core structure of complex type oligosaccharides derived from human transferrin and porcine pancreatic lipase.

An endogenous Ca2+ channel agonist, endothelin-1, does not directly activate partially purified dihydropyridine-sensitive Ca2+ channel from cardiac muscle in a reconstituted system.

To elucidate the action of tentative endogenous Ca2+ channel activator, endothelin (ET)-1, on a voltage-dependent Ca2+ channel in the heart, a dihydropyridine (DHP)-binding protein was solubilized from porcine ventricular muscle, partially purified by wheat germ agglutinin-affinity chromatography and reconstituted into proteoliposomes. Ca2+ flux into the proteoliposomes was determined using a fluorescent probe, Quin-2. The initial Ca2+ entry rate was dose-dependently activated by either a K(+)-depolarization or a synthetic Ca2+ channel agonist, Bay K8644, and inhibited by several Ca2+ entry blockers or cadmium ions. Using the same reconstituted system, it was demonstrated that sufficient dose of ET-1 yielded no effect on the Ca2+ channel function, indicating that the ET-1 action was not directly mediated by the voltage-dependent, DHP-sensitive Ca2+ channel.

Movements of the large intestine in the anuran larvae, Xenopus laevis.

1. The contractile behavior of the large intestine of Xenopus laevis tadpoles was studied. 2. The large intestine is divided into a colon and rectum, and shows three types of movements: rhythmic ascending (antiperistaltic) waves of contraction originating at the anal end of the large bowel, rhythmic longitudinal contractions in the rectum and colon, and irregular contractions. The first two patterns occur in the large bowel in situ and thus appear mature. The last one occurred only in older preparations, and thus appeared pathological. 3. Antiperistaltic waves of contractions and longitudinal contractions are generated independent of each other, suggesting that circular muscles and longitudinal muscles contract separately. 4. Acetylcholine, adrenaline and noradrenaline augment motility. 5. The premetamorphic motility of the large bowel is similar to that seen in adult frogs. Comparable motility was not observed elsewhere in the larval alimentary tract. The large intestine appears to be the first portion of the anuran alimentary tract to acquire the adult physiological and morphological profile.

Response of intracellular Ca2+ transients in cultured vascular smooth muscle cells to angiotensin II, vasopressin, acetylcholine and atrionatriuretic peptide.

Modulation of intracellular Ca2+ concentration [( Ca2+]i) is a signal for the contraction of vascular smooth muscle cells responding to vasoreactive substances. We prepared confluently cultured smooth muscle cells from rat aorta, loaded them with Ca2+ sensitive fluorescent dye, fura-2, and measured the [Ca2+]i transient by microscopic spectrofluorometry. The [Ca2+]i was distributed heterogeneously in cytosol. Angiotensin II (10 nM) transiently doubled the [Ca2+]i. It was also increased by arginine-vasopressin (10 nM), even after stimulation by angiotensin II was saturated. In contrast, acetylcholine (10 microM) or rat atrionatriuretic peptide (10 nM) did not change the [Ca2+]i in the same detecting field of the same cell, contradicting previous reports.

[Experimental study of periodontal tissue regeneration using biodegradable membranes].

This study was designed to investigate the effect on new attachment formation using a biodegradable membrane as a barrier to the regeneration of periodontal tissue. One-wall wide periodontal osseous defects with exposed root surfaces were prepared in three adult mongrel dogs. After surgical debridement of the periodontal defects, 3 types of biodegradable membranes, [Poly(L-lactic acid); (P-L-LA)] membrane, [Poly(lactic acid-co-glycolic acid); (PLGA)] (81: 19 mole%) membrane and PLGA (50: 50 mole%) membrane, were arranged to cover the denuded root surfaces. Specimen blocks were removed 8 weeks postoperatively for histological evaluation of their effect on the regeneration of periodontal tissue. The results were as follows: 1. Gingival regions containing P-L-LA or PLGA (81: 19 mole%) membranes showed delayed wound healing macroscopically. 2. Almost all P-L-LA membranes were present in the experimental sites failing to be resorbed during the 8-week experimental period. PLGA (81: 19 mole%) membranes were also present but showed with some degree of resorption and fragility, while PLGA (50: 50 mole%) membranes were completely resorbed. 3. This showed that the membranes used in this experiment are possible barriers to the generation of new attachment. It is important to make sure keeping the membrane. 4. There was no relationship between osteogenesis and cementogenesis. It seems that osteogenesis was depressed by membrane shrinkage and the gingival pressure which limited "the regenerative space of bone." 5. Moderate resorption was observed on the root surface, and new cementum was formed at the resorbed surface. 6. Ankylosis was observed between the new cementum and bone at the experimental sites. Newly formed cementum was seen on coronal sections of this area.

[A comparative study on the responses of blood vessels of oral mucosa to thermal stimulation in dentulous and edentulous subjects].

The purpose of this experiment was to clarify the characteristic changes of vasculature in edentulous alveolar mucosa and the effect of wearing a complete denture during sleep. Thermal stimulations were applied to the oral mucosa of five normal subjects and five completely edentulous patients and the responses of blood vessels were observed using both Impedance Plethysmography (IMP) and Reflection Photoelectric Plethysmography (RPP). Quantitative comparisons were made between edentulous patients and dentulous normal subjects, and between patients wearing complete dentures during sleep and those who do not. The following results were obtained. 1) The responses of blood vessels were divided into four groups. Type 1 showed dilatation in IMP and RPP. Type 2 showed dilatation but had late initial rising in IMP. Type 3 showed initial contraction in either or both IMP and RPP, and followed by dilatation few seconds later. Type 4 showed no changes in RPP but dilatation in IMP. 2) There was definite difference between edentulous and dentulous subjects in the frequency of exhibition of each type. Especially for the edentulous patients type 4 was observed in all stimulation temperature. 3) For the dentulous subjects there were more cases showing responses of RPP in digital apex than those do not. This was most conspicuous at the 50 degrees C stimulation. However for the edentulous patients there were about equal number of each cases. 4) The responses of blood vessels for both dentulous and edentulous subjects became greater and blood volume increased both with time at each temperature and as the stimulation temperature were raised. However the increase in the response was greater for the edentulous patients when the stimulation temperature was raised from 0 degrees C to 50 degrees C. 5) The responses of blood vessels became greater after two weeks when complete dentures were worn during sleep at night. And when dentures were not worn during sleep the responses decreased to a level lower than what it used to be after three weeks. 6) From the above results it was suggested that oral mucosa of the edentulous patients were losing their sensitivity due to wearing complete dentures and were not receiving as much effect of the contractile control from body as the dentulous subjects. It was also suggested that wearing denture during sleep at night cause inflammation of oral mucosa and increase the metabolism of tissue in edentulous patients.

Endogenous digoxin-like activity of mammalian-lignans and their derivatives.

A comparative study was made on the endogenous digoxin-like activity of sixteen mammalian-type lignan derivatives including enterolactone and enterodiol. Cross-reactivity to antidigoxin antibody, inhibition of dog kidney Na+,K+-ATPase, and ouabain displacing activity against [3H]ouabain binding to human erythrocytes on the part of these derivatives were examined and compared in all cases with that of ouabain. meso-2,3-Dibenzylbutane-1,4-diol (meso-HA-1) was found to possess the most potent cross-reactivity to antidigoxin antibody. Several lignans, particularly HA-1 (either meso or dl), dl-2,3-bis(3-methoxybenzyl)butane-1,4-diol(HA-4), dl-2,3-bis(3,4-dimethoxybenzyl)butane-1,4-diol(HA-10), dl-2,3-bis(3,4-dimethoxybenzyl)-1,4-dimethoxybutane(HA-11), and trans-2,3-bis(3,4-dimethoxybenzyl)-gamma-butyrolactone(HA-14) were capable of inhibiting Na+,K+-ATPase activity with IC50 at a concentration less than 5 x 10(-4) M. Meso-HA-1, HA-11 and HA-14 also showed [3H]ouabain displacement activity with IC50 at a concentration of 10(-4)-10(-3) M. These determinations of activity were made at doses 100-1000 times those of ouabain. The synthetic lignans are considered to derive in vivo from plant material usually present in fibrous food. Based on the data obtained, some lignans may possibly be endogenous digitalis-like substances.
