Relationships between milk levels of hormones and growth or puberty of offspring in mice.

Milk levels of prolactin, growth hormone (GH) or progesterone on days 12-13 of lactation and their relationships with the growth or the puberty of offspring were examined in primiparous SHN mice. Milk prolactin level had negative correlations with the body weights on days 12 and 20, the growth rate between days 0 and 20 or the age and the body weight at vaginal opening of offspring. There was a significant correlation between milk GH level and the growth rate between days 0 and 12 of offspring. A positive correlation was also found between milk progesterone level and the age at vaginal opening. These results suggest that hormones in milk would have effects on the growth or the puberty of offspring.

Synthesis and biological activity of a new cephalosporin, BMY-28232 and its prodrug-type esters for oral use.

The synthesis and structure-activity relationships of 7-[(Z)-2-(2-aminothiazol-4-yl)-2-hydroxyiminoacetamido]-3-[( Z)-1- propenyl]-3-cephem-4-carboxylic acid (BMY-28232), its 3-alkenyl analogs (6 and 7) and O-substituted derivatives of the oxyimino moiety (10) are described, as well as the oral pharmacokinetics and in vivo activities of the 1-acetoxyethyl ester of BMY-28232 (BMY-28271) and its analogous esters (11). The 3-alkenyl groups were introduced by the Wittig reaction of the ylide (2) prepared from the 3-chloromethyl cephem (1) to afford the Z (main) and E (minor) isomers regarding the 3-side chain. The O-substituted derivatives (10) were prepared by 7-N-acylation of the 7-amino cephem (4a) with the corresponding O-substituted side chain acids (8). The prodrug esters (11) were prepared by esterification of BMY-28232 with an appropriate halide. BMY-28232 was the most active among the 3-alkenyl analogs tested against Gram-negative organisms and much more active than the O-substituted derivatives against Gram-positive bacteria. BMY-28271 showed good oral bioavailability (66%) and good in vivo efficacy in mice against infections of Staphylococcus aureus Smith (PD50, 0.68 mg/kg) and Escherichia coli Juhl (0.54 mg/kg).

[Ectopic ureteral opening with common urogenital sinus. Review of the literature of 648 cases in Japan].

A 3-year-old girl was admitted with continuous enuresis. Urological examination and operation disclosed the left upper ureter ectopically opened into the vagina, and the vagina opened into the urethra (common urogenital sinus). Vesicoureteroneostomy was performed, and after the operation, incontinence disappeared. The 648 cases reported in Japan were discussed.

Mammary gland growth and serum levels of prolactin and growth hormone in four strains of male mice in comparison with females.

Male mammary gland growth in 4 strains of mice (SHN/Mei, SLN/Mei, GR/AMei and C3H/HeMei) was studied and compared to that in females of the same strains. The area of the mammary glands in female mice was greater in SHN and SLN, as compared to GR/A and C3H/He strains on both 20 and 60 days of age. A similar trend was seen in mammary gland growth in males on day 20. However, on day 60, there was a marked growth in SLN and GR/A strains, slight growth in SHN mice and no glands were found in C3H/He males. These patterns of mammary gland growth were not related to circulating levels of prolactin or growth hormone (GH) or to body growth in either sex. These findings provide the first description of these characteristic patterns of mammary gland growth in male mice.

Interleukin 4 (IgG1 induction factor): a multifunctional lymphokine acting also on T cells.

A cDNA encoding the murine interleukin 4 (IL4) (IgG1 induction factor/B cell-stimulating factor no. 1) was recently cloned (Noma et al., Nature 1986.319: 640; Lee et al., Proc. Natl. Acad. Sci. USA 1986. 83: 2061). In this report we tested recombinant IL 4 in various T cell assays. It was found that IL 4 activated the murine T cell line CTLL to increased DNA synthesis but not to growth. It also activated normal concanavalin A (Con A)-stimulated T cells both to increased DNA synthesis and to growth. These T cell growth factor-like activities were not inhibitable by anti-IL 2 receptor antibodies. Evidence is given that both Lyt-2+ and L3T4+ T cells responded to IL 4. Finally, IL 4 acted synergistically with phytohemagglutinin or Con A on normal T lymphocytes as well as on thymocytes. These data, as well as those of others, imply that lymphokines have a broader range of activity than previously anticipated.

Sequence analysis of a cDNA clone of a gene encoding a component of a putative phosphorylcholine-specific T suppressor factor and functional property of its gene product.

We have isolated a putative phosphorylcholine (PC)-T cell suppressor factor (TsF) cDNA clone, p6-5, from a cDNA library of a T hybridoma which constitutively secretes a PC-TsF in vitro [8]. In the present study, we determined the nucleotide sequence of the p6-5 gene and found that the p6-5 sequence is 86% homologous to rat preproelastase 1 gene, one of the serine protease genes. An oligopeptide (14 mer, TsF14) deduced from the p6-5 sequence was synthesized and antisera against TsF14 were prepared in rabbits. Anti-TsF14-conjugated Sepharose 4B specifically absorbed the PC-TsF activity from the culture supernatant of PC-TsF-secreting T hybridomas. In contrast, the binding molecule eluted from the anti-TsF14-conjugated Sepharose suppressed the antibody response PC specifically. These results indicated that the p6-5 polypeptide is a component of the PC-TsF molecule.

IgG1 induction factor: a single molecular entity with multiple biological functions.

A cDNA clone coding for the murine IgG1 induction factor has been isolated. The translation products directed by this clone were analyzed in different biological assays. The data obtained show that the IgG1 induction factor: Is involved in the regulation of IgG responses, by increasing IgG1 and decreasing IgG3 and IgG2b secretion; Induces hyper-Ia expression on resting B lymphocytes; Synergizes with anti-Ig in inducing DNA synthesis in resting B lymphocytes; Synergizes with DxS in inducing DNA synthesis by B lymphocytes; It induces DNA synthesis by either the T cell line CTL-L or Con-A blasts. Thus, this lymphokine in addition to IgG1 inducing activity has also BSF-1, BCGF-II and TCGF like activities. The fact that a single molecule can perform all the above listed functions has implications for our view of lymphocyte activation. It indicates that considering the B cell response as an ordered series of independently controlled events, is an oversimplified view of the dynamic process through which B cells are activated and also indicate the functional interconnection of the different elements of the immune system.

Purification and partial characterization of fimbriae of Vibrio cholerae O1.

Fimbriae of Vibrio cholerae O1 were purified from a strain of the classical biotype, Inaba serotype (Bgd 17), and a strain of the El Tor biotype, Ogawa serotype (K23), grown on TCG agar medium by the following procedure; homogenization of the cell suspension to detach fimbriae, ultracentrifugation to remove remaining cells and their debris, concentration of the supernatant containing fimbriae with ultrafiltration, and 20 to 40% sucrose linear gradient centrifugation of the concentrated material. The fimbriae in both preparations were flexible, long fibres readily distinguishable under the electron microscope from those of CFA/I, CFA/II seen in ETEC strains. Their structural subunit was a protein of 16 kdaltons. Fimbriae isolated from both serotypes and biotypes shared antigenic determinants.
