Macromolecular collagen components in normal liver and at the different stages of human liver cirrhosis were studied under various extraction conditions. The collagen content at the typical stage of liver cirrhosis was more than five-fold higher than that of the normal state. Pepsin-solubilized collagens extracted successively accounted for 90% of the total collagen and were subjected to determination of the collagen types by salt differentiated fractionation and SDS-polyacrylamide gel electrophoresis. Both type I and III collagens, especially the former, increased, reflecting enhanced total collagen with the progression of liver cirrhosis. The ratio of type I to type III was 1.02 - 1.22 in normal liver and at the early stage of liver cirrhosis, but increased to 1.58 and 1.60 at the typical and advanced stages of liver cirrhosis, respectively. At the early stage, the remarkable increase in type V collagen started much earlier than at the typical stage when the ratio of type I to type III changed. The enhancement of type V collagen may result from a cell proliferative phenomenon at the earlier stage of liver cirrhosis.
A case of simultaneous squamous cell carcinoma and papillary adenocarcinoma in the thyroid gland is reported. A pure squamous cell carcinoma arose in the right lobe, and a pure papillary adenocarcinoma without squamous metaplasia arose in the left lobe. Both cancers came into collison at the isthmus. The case is unique in that the tumor was a so-called collison tumor but not a mixed tumor.
Using seventeen human tumor cell lines derived from a variety of tissues, specific binding sites for epidermal growth factor (EGF), a mouse submandibular gland-derived growth factor, has been characterized. A significant amount of membrane-bound EGF receptors, although considerably varied, was demonstrated in all the tumor cell lines studied. Epidermoid carcinoma appeared to have more EGF receptors than adenocarcinoma. One small cell carcinoma of the lung, one choriocarcinoma of the stomach and three bone tumors also possessed EGF receptors comparable to those of epidermoid carcinoma, while one adenoacanthoma of the stomach had less EGF receptors comparable to adenocarcinoma. Among a variety of phorbol esters tested, tetradecanoyl phorbol acetate, a potent tumor promotor, was shown to be the most effective compound in inhibiting 125I-labeled EGF binding to its receptors. Our results indicate that human tumor cells contain varying amounts of membrane-bound receptors for EGF and that phorbol esters interact with these EGF receptor sites. However, the relationship between EGF receptor sites on tumor cells and cellular proliferation and/or differentiation awaits further study.
A quantitative study of carcinoembryonic antigen (CEA) was made in nine human gastric cancer cell lines. Six of them were found to produce CEA in vitro. The production of CEA in the three cell lines derived from well differentiated tubular adenocarcinomas began at the mid-exponential phase of cell growth and reached its peak at the late stationary phase, the amount of CEA per 10(5) cells and the frequency of CEA-positive cells on immunostaining increased with culture time. In contrast, CEA in the three cell lines derived from poorly differentiated adenocarcinomas including a signet-ring cell carcinoma was produced immediately after plating and the amount of the antigen per 10(5) cells and the frequency of CEA-positive cells were almost constant throughout the cell growth phases. Serum CEA content in nude mice was low or not detectable in the case of subcutaneous heterotransplantation of gastric cancer cells, irrespective of CEA productivity of the cell lines in vitro. Intraperitoneal inoculation, however, led to high CEA levels in sera of nude mice bearing human gastric cancers. No significant difference was found between the two kinds of inoculation in terms of the total tumor weight and the frequency of CEA-positive cells in tumor tissues. One reason for the above findings may be that the transport of CEA in the subcutaneous tumors to the systemic blood flow is hindered.
Using human trophoblastic (SCH) and nontrophoblastic (HeLa S3) tumour cell lines, specific binding sites for epidermal growth factor (EGF), a potent stimulator of growth in many tissues, and its effect on secretion of human chorionic gonadotrophin (hCG) and/or its subunits were compared between these two tumour cells. Both SCH and HeLa S3 cells possessed two populations of specific binding sites for 125I-labelled EGF: the high affinity (Kd approximately 10(-10) M) and the low affinity (Kd approximately 7 x 10(-10) M) system. Tetradecanoyl phorbol acetate (TPA), a tumour promotor, showed a potent competitor of labelled tracer binding to its receptor sites in both cell lines. EGF stimulated both hCG-alpha and hCG and/or hCG-beta secretion in a dose-responsive manner from SCH cells, whereas it had no effect on hCG-alpha secretion from HeLa S3 cells. In contrast, dibutyryl cyclic AMP plus theophylline, a phosphodiesterase inhibitor, enhanced hCG-alpha secretion from both cells, while TPA had no effect in either cells. These data suggest that EGF may play a physiological role in hCG secretion from trophoblastic tissues and that the mechanism by which hCG and/or its subunits are secreted may differ between trophoblastic and non-trophoblastic tumour cells.
In an attempt to elucidate the role played by neoplastic epithelial cells in the formation of stromal collagen, the synthesis of collagen by two cloned human gastric carcinoma cell lines was studied. The presence of antigenicity of procollagen alpha 1(I) chain in the cytoplasms of both carcinoma cell lines growing in culture was demonstrated by an immunocytochemical technique using specific antibodies. After denaturation of the radiolabeled collagenous proteins extracted from the combined cells and culture media, two components comigrating with authentic alpha 1(I) and alpha 2 chains on sodium dodecyl sulfate:polyacrylamide gel electrophoresis were found. Electrophoretogram analysis revealed further a dense band slightly slower than the alpha 1(I) chain, most likely representing alpha 1(I) trimer. These radioactive components disappeared after exposure of the samples to bacterial collagenase. The relative activity of collagen synthesis determined by using purified collagenase was slightly higher than that of fibroblasts derived from human synovial membrane in culture. The same antibodies to procollagen alpha 1(I) chain also labeled the cytoplasms of carcinoma cells, and extracellular matrix of the tumors developed after transplantation of one of the cell lines into the nude mice. Our data indicate that both human gastric carcinoma cell lines synthesize type 1 collagen in vivo as well as in vitro and suggest that carcinoma cells may play an active role in the formation of stromal collagen in most human carcinomas.
Two cell lines, Kuramochi and CKS, established from human ovarian cancer, were analyzed in vitro drug sensitivity to Mitomycin C and Carboquone. These values were compared to those of eight human gastric cancer cell lines composed of various histological types. The CKS cell line, derived from serious cystadenocarcinoma, was less sensitive to both test-drugs than the groups of gastric cancer cell lines. Therapeutic indices of the two drugs were very low. So it was considered that the two drugs were unsuitable to chemotherapy to serous cystadenocarcinoma. The Kuramochi cell line, derived from undifferentiated carcinoma, was less sensitive to Mitomycin C than the groups of gastric cancer cell line. However, it was as sensitive to Carboquone as high-sensitive gastric cancer cell lines. Therapeutic index of the Carboquone in the undifferentiated carcinoma was high. So it was considered that Carboquone was suitable to chemotherapy to undifferentiated adenocarcinoma. The cytocydal actions of the two drugs were dependent upon dose and time. Time-dependency was, however, smaller in Carboquone than in Mitomycin C.
The present study first dealt with characterization of two cell lines, Kuramochi and CKS, established from human ovarian cancer. Secondary, the quantitative analyses of sensitivity of the cell lines to Mitomycin C was reported. The two cell lines showed typically epithelial features in vitro. Kuramochi cells, derived from undifferentiated carcinoma, had microvilli on the cell surface, cell continuity with the relative developed junctional complex, but no secretary granules and vesicles. The chromosomal analysis revealed the hyperdiploid modal number of 50. The population doubling time was about 26 hours. CKS cells, derived from serous cystadenocarcinoma, had secretary vesicles, microvilli, but no developed junctional complex. The chromosomal analysis reveals the hypodiploid modal number of 37. The population doubling time was about 34 hours. Biochemical markers of alpha-fetoprotein, human chorionic gonadotropin and carcinoembryonic antigen were not detected in both cell lines. 90% lethal dose of 2-hr-treatment with Mitomycin C was 0.42 microgram/ml in Kuramochi cell line and 1.13 microgram/ml in CKS cell line. These values were higher than the one of the control cell line, KATO-III, derived from signet-ring cell carcinoma of stomach. Ovarian cancers were suggested resistant to Mitomycin C.
Several experimental methods inducing murine amyloidosis were tested using six strains of mice including congential asplenic mice and athymic nude mice. Traditional induction method of amyloidosis by casein injections failed to cause amyloidosis in C3H/He mice. Single injection with complete Freund's adjuvant reinforced with Mycobacterium butyricum was also unable to induce amyloidosis in C3H/He, CBA, and BALB/c mice. Six or four injections with complete Freund's adjuvant at an interval of once a week successfully induced amyloidosis in CBA mice with high incidence but not in C3H/He, ICR/SLC, and BALB/c mice. Athymic nude mice with genetic background of BALB/c and congenitial asplenic mice, cross-bred with C57BL/6 X C3H, were free from amyloidosis after six or fourteen injections with the adjuvant. Experimental amyloidosis in mice, therefore, might mainly depend on the strain of mice used and the induction method chosen.
Leupeptins, protease inhibitors, suppress the appearance of experimental amyloidosis in CBA mice induced by the injections with complete Freund's adjuvant. This substance, however, should be administered continuously from 1 week prior to amyloid induction to the end of the experiment. On the other hand, Trypan blue, inhibitors of lysosomal enzymes, accelerates experimental amyloidosis in the mouse model above-mentioned. Trypan blue is effective when given either prior to or at the same time of the initiation of amyloid induction. Organic Germanium has not been confirmed to be a potent suppressor of experimental murine amyloidosis but the experimental group administered this substance continuously from 1 week prior to the induction shows a rather low incidence of amyloidosis, and the average number of amyloidotic organs per affected mouse is about half of that of the control group. The suppression and acceleration of experimental murine amyloidosis presented here are a useful tool for investigating the pathogenesis of amyloidosis.
Spontaneously hypertensive rats develop periarteritis nodosa spontaneously with high incidence. Immunohistology of periarteritis nodosa reveals that fibrinogen constantly deposits in the wall of the diseased artery in exceptional combination with IgG but never with complement (C3). This fact suggests that periarteritis nodosa in spontaneously hypertensive rats may not be arteritis of immune complex type. Permeability test using mainly peroxidase as a tracer demonstrates that increased arterial permeability in spontaneously hypertensive rats is recognizable since 4 to 6 months of age without sexual difference in its intensity. In male spontaneously hypertensive rats periarteritis nodosa is observed in 6 of 11 and the arterial permeability increases more pronouncedly in the positive group, whereas 5 female spontaneously hypertensive rats of various ages are free from periarteritis nodosa irrespective of high arterial permeability. Therefore, high arterial permeability does not always lead to regular development of periarteritis nodosa especially in female spontaneously hypertensive rats.
A continuous cultured cell line, CKS, was established from the ascites of a serous cystadenocarcinoma of the ovary and has been maintained in vitro for more than 3 years. The cells grew in a medium containing human umbilical cord serum and form a monolayer in a mosaic fashion with polyhedral or angular cells of varying size together with occasional giant cells. Hemicyst formation was evident in confluent culture. The ultrastructural features noted, were well developed microvilli and poorly developed junctional complex on the cell surface associated with abundant secretory vesicles in the cytoplasm. Population doubling time obtained from the growth curve was about 79 hr. The chromosomal modal number was near-diploid with minimal aberrations. Heterotransplantation of the cells was successful in ATS-treated hamsters and athymic nude mice. Histological feature of the heterograft much resembled those of the original tumor.

Steroid receptors in endometriosis and adenomyosis were investigated to clarify their clinical significance. The receptor levels were determined by Scatchard plot analysis (4 degrees C, by dextran-coated charcoal). In the cytosols of both tissues, the 17 beta-estradiol-estrogen receptor (ER) complex demonstrated a dissociation constant (Kd) of 4.5 x 10(-10) M; the Kd of the progesterone-progesterone receptor (PR) complex was 1.5 x 10(-9) M; and the Kd of the dihydrotestosterone-androgen receptor (AR) complex was 4.0 x 10(-10) M. Seven cases of ovarian endometriosis were studied. The ER and PR levels in endometriosis seemed to be lower than those in the corresponding normal endometrium. AR was also present. There was a suggestion that most endometriosis is least responsive to progestogens. Ten cases of adenomyosis were studied. Histologic dating revealed a delay in the most aberrant endometrial tissue in adenomyosis, as compared with dating of corresponding normal endometrial tissue. ER and AR were detected in all cases. PR was not detected in some cases and, when detected, the content seemed to be lower, possibly suggesting the delayed dating.
