Loop-like structure of dGTTG in solution.

A strong NOESY cross peak was observed between G1H8 and G4H8 for a single-stranded tetradeoxyoligonucleotide dGTTG in aqueous solution. If dGTTG is in an expanded form, NOE should not be observed because the distance between G1H8 and G4H8 is estimated to be about 13 A. This NOESY cross peak shows that the guanine residues of the both ends of dGTTG exist with short distance (< 5 A), and that dGTTG has a distorted loop-like structure in aqueous solution. This is the shortest single-stranded oligodeoxynucleotide that has a rigid structure in solution.

Mechanistic aspects of the interaction of antisense DNA.

To investigate the mechanism of antisense effects, diastereoisomerism of oligodeoxynucleotides with modified internucleotide phosphate linkages such as oligodeoxynucleoside phosphoramidates (OPA) and oligodeoxynucleoside phosphorothioates (OPT). Reversed-phase HPLC was used to separate the diastereoisomers of OPA (up to tetramer) and OPT (up to pentamer), and it was found that the complete purification of all diastereoisomers could be achieved by the two-step chromatographic approach. From the circular dichroism measurements, it was found that some diastereoisomers formed certain hybrids with their complementary oligonucleotides and some did not.

[Osteolytic and osteosclerotic lesion of the femur associated with pustulosis palmaris plantaris].

The cases with pustulosis palmaris et plantaris associated with lesions in sterno-cost clavicular region, spine and ilio sacral joint have been in the previous literatures. We encountered four rare cases of this pustulosis in association with femoral lesion. We mainly reported their radiological and pathologic findings. Three cases had lesions in the diaphysis of the femur, while one case had lesion in the lesser trochanter. The average age was 55.5 years (range 53-60) All patients complained of thigh pain. Three cases suffered sterno cost clavicular pain also. Laboratory data showed the elevation of white blood cell number, ESR and tested positive for C-reactive protein. The radiological findings showed osteolytic lesions of the femoral cortex at the onset of the disease, followed by thickening and sclerosis in the cortex. A biopsy was performed in three cases and the pathological results showed no malignancy. The specimens showed only chronic inflammation. Pathological results and radiological abnormalities suggested that femoral lesion of pustulosis palmaris et plantaris originated from the periostenum or the cortex of bone.

[Expression of activation antigen CD69 on eosinophils].

The antigen, CD69, has been shown to be expressed on activated lymphocytes. CD69 has also been shown to be associated with signal transduction process. Recently, we demonstrated the expression of CD69 on eosinophils at inflammatory sites. Whatever the disease, lung eosinophils obtained from the bronchoalveolar lavage fluid or pleural effusion expressed significant levels of CD69, whereas most peripheral blood (PB) eosinophils did not express CD69. Further, in vitro stimulation of PB eosinophils by IL-3, IL-5, GM-CSF and PMA induced CD69 expression on PB eosinophils. These findings suggest that CD69 expression on eosinophils is restricted to local inflammatory sites and CD69 may be a useful marker for activated eosinophils at inflammatory sites.

In vivo expression of CD69 on lung eosinophils in eosinophilic pneumonia: CD69 as a possible activation marker for eosinophils.

The antigen, CD69, has been demonstrated to be expressed on activated T cells and natural killer cells. There have been no studies concerning the expression of CD69 on eosinophils. In this article, we demonstrate that lung eosinophils obtained from the bronchoalveolar lavage fluid of patients with eosinophilic pneumonia expressed significant levels of CD69, whereas peripheral blood (PB) eosinophils did not express CD69. We also activated PB eosinophils in vitro using phorbol myristate acetate and cytokines to determine whether CD69 was expressed. PB eosinophils expressed CD69 after short-term culture with phorbol myristate acetate and eosinophil hemopoietic cytokines (interleukin-3, granulocyte-macrophage--colony-stimulating factor, and interleukin-5). These findings suggest that CD69 may be a useful marker for activated eosinophils at inflammatory sites.

Syntheses and properties of fluorescent labeled oligonucleotides containing deoxyethenoadenosine at 5' end.

The fluorescent labeled oligodeoxyribonucleotides which contain deoxyethenoadenosione (d epsilon A) at their 5' end were prepared by treating CPG bound oligonucleotides with 5'-DMTr-deoxyethenoadenosine-3'-H-phosphonate. The hybrid formation of d epsilon A-oligonucleotide with its complementary DNA was studied by fluorescence spectroscopy. The fluorescence of d epsilon A in a single strand was largely quenched by stacking interaction with the base at 3' position. When d epsilon A-oligonucleotides hybridized with their complementary strands, relative fluorescence quantum yields (Qrel) against d epsilon A changed in specific manners. These results suggest that d epsilon A-oligonucleotides are applicable to study the local structure of DNA in solution.

Separation and characterization of diastereoisomeric oligonucleotide.

For developing the antisense method, two types of oligonucleotides suitable for antisense molecules were explored for their essential properties. One is oligonucleoside phosphorothioate, and all their possible R/S diastereoisomers were separated and purified by reversed phase liquid chromatography. Isolated diastereoisomers have been investigated for the correlation between their configuration and hybridization manner. Separation, R/S characterization and spectroscopical properties of these oligonucleotides will be discussed.

Effects of base damages on DNA replication--mechanism of preferential purine nucleotide insertion opposite abasic site in template DNA.

DNA polymerase preferentially inserts purine nucleotides opposite non-instructive lesions such as abasic sites during DNA replication. In order to elucidate the mechanism of the preferential insertion, a DNA template containing a model abasic site and primers containing 4 different nucleotides (A,G,C,T) at primer terminus were synthesized. The stability of the primer terminus nucleotide placed opposite the abasic site was evaluated on the basis of its sensitivity to 3'-5' exonuclease associated with DNA polymerase.

NMR studies on solution structure of single-stranded oligonucleotides causing line broadening.

Unusual line broadening of 1H-NMR lines attributable to the proton (8H) of guanine residues was observed for all tetradeoxyoligonucleotides tested here which have a specific base sequence of dGXXG (X = A or T). For the same samples, line broadening was also obtained in the 31P-NMR spectra. These broadened signals did not become sharp up to 60 degrees C. This unusual spectral phenomenon has been attributed by 2D-NMR and differential NOE to the compact solution structure of the oligonucleotides.

Sequence-specific DNA recognition by basic peptides.

A chiral template with C2 symmetry has been used for modeling a dimeric interface of DNA binding protein. An oligopeptide derived from the basic region of MyoD, a recently described "helix-loop-helix" class of DNA binding protein, has been tethered to the template. Among the four models which differ in chirality and polarity with respect to the arrangement of two subunits, only one dimer model with right-handed and C-terminus to C-terminus arrangement of the peptide subunits binds DNA containing native MyoD binding sequence.

[Evaluation of tissue eosinophils in eosinophilic pneumonia].

Transbronchial biopsy specimens from ten patients with eosinophilic pneumonia were analysed to see whether the tissue contained activated eosinophils or secreted eosinophil cationic protein. The specimens showed many infiltrated tissue eosinophils in the intraalveolar spaces, and thickened alveolar septae and perivascular areas. Most of these eosinophils stained clearly with degranulation by immunohistochemical studies with monoclonal antibody EG2 (for the secreted form of eosinophil cationic protein), indicating that they were activated eosinophils. On the other hand, although pleural tissue obtained at bullectomy performed after pneumothorax showed eosinophilic infiltration, EG2-positive cells were few. Activated eosinophils may play an important role in the development of pulmonary lesions of eosinophilic pneumonia.

Expression of activation antigens CD69, HLA-DR, interleukin-2 receptor-alpha (IL-2R alpha) and IL-2R beta on T cells of human decidua at an early stage of pregnancy.

T cells of human decidua at an early stage of pregnancy were examined by flow cytometry for the expression of the T-cell-activation antigens CD69, HLA-DR, interleukin-2 receptor-alpha (IL-2R alpha) and IL-2R beta. The decidua contained a small number of T cells and both CD4+ and CD8+ subsets expressed CD69, HLA-DR, IL-2R alpha and IL-2R beta antigens significantly whereas, in peripheral blood, only a small number of T cells expressed these activation antigens. These findings indicate that T cells in the decidua in the first trimester of pregnancy are regionally activated.

Structural polymorphism in the major groove of a 5S RNA gene complements the zinc finger domains of transcription factor IIIA.

Metal complexes that bind to DNA on the basis of shape-selection have been used to map the conformational features of the DNA binding site for transcription factor IIIA. Conformationally distinct segments are detected on the 5S rRNA gene that correspond closely to the binding sites identified for the individual zinc finger domains of the protein. The local conformations are characterized by a major groove opened because of a change in base pair inclination and/or displacement at a central 5'-pyrimidine-purine-3' step, flanked by a widened minor groove, as would arise at the junctions between alternating B- and A-like DNA segments. Docking experiments with a consensus structure of a zinc finger reveal that the mixed A-B binding site accommodates the peptide domain better than either canonical B- or A-DNA helices. The close structural matching of the conformational variations in the 5S rDNA both to the proposed sites of zinc finger binding and to the shape of an individual zinc finger domain points to DNA structural polymorphism as providing an important determinant in recognition. In particular, shape selection in the 5' half of the internal control region may orient the multiple finger domains.

Flow cytometric analysis of anti-platelet antibodies in patients with chronic idiopathic thrombocytopenic purpura (ITP) using acid-treated, formalin-fixed platelets.

Acid treatment of normal washed platelets prepared by albumin density gradient centrifugation followed by formalin fixation effectively eliminates HLA class I antigens from the platelet surface, without altering the immunoreactivity of platelet membrane glycoproteins Ib and IIb/IIIa complex. Using these platelets, flow cytometric analysis was performed on a total of 20 sera from different patients with chronic ITP, and revealed an inverse linear correlation between the platelet count and the mean fluorescence intensity which represents a relative amount of platelet-bound IgG. These results suggest that acid-treated, formalin-fixed platelets can be used as specimens to detect anti-platelet antibodies to differentiate from those to HLA-class I antigens.

Accumulation of CD16-CD56+ natural killer cells with high affinity interleukin 2 receptors in human early pregnancy decidua.

Most human peripheral blood natural killer (NK) cells express the phenotype CD16+CD56+. However, a very minor subset of NK cells express CD16-CD56+, and these NK cells bear both interleukin 2 receptor (IL-2R)alpha (p55) and IL-2R beta (p75) (high affinity IL-2 receptors). In this report, we demonstrate that in human early pregnancy decidua--an interface between maternal immunocompetent cells and fetus (placenta)--abundant (approximately 83%) CD16-CD56+ NK cells with high affinity IL-2 receptors were present, and these cells responded to low amounts of IL-2 (4.5 pM). These CD16-CD56+ NK cells significantly expressed an early activation antigen, CD69, in vivo, whereas peripheral CD16-CD56+ NK cells did not express CD69. These findings suggest that CD16-CD56+ NK cells in early pregnancy decidua may be activated in vivo, and may play an important role in immunoregulation during early pregnancy. Also, decidual lymphocytes may be useful materials to study the mechanism of MHC-unrestricted cytotoxicity of this type of NK cells.

Unicompartmental replacement with the Marmor Modular knee: operative procedure and results.

Unicompartmental arthroplasty using the Marmor Modular knee was performed in 27 knees of 24 patients--three men and 21 women--aged 51-82 years (average age 68.9 years). The disease was osteoarthritis in 19 knees, steroid arthropathy in two knees, Charcot joint in three knees, and rheumatoid arthritis in three knees. Medial replacement was done in 24 knees and lateral replacement in three knees. Follow-up was from 2-7 years (average 4 years 9 months). Post-operatively, 14 knees were reported to be free of pain and 11 only occasionally mildly painful. Walking distance improved to more than 1 km in 17 patients. The knee score improved from 40.4 to 72.2 points (p less than 0.01). Complications included two knees with loosening due to postoperative limb malalignment; there were no instances of infection or fracture.

DNA recognition by synthetic peptides as a model for the dimeric proteins.

A Chiral template possessing a C2 axis has been utilized to study an effect in the dimer formation of oligopeptides. Oligopeptide rich in basic amino acids are used as subunits that would interact with DNA, and the subunits are aligned according to the C2 axis of the template. Synthesis, characterization and DNA binding study of these dimeric peptides will be discussed.
