Cloning and sequencing of an MPB70 homologue corresponding to MPB83 from Mycobacterium bovis BCG.

MPB70 is secreted in high concentrations by Mycobacterium bovis BCG substrain Tokyo (BCG Tokyo), but little by substrains Pasteur (BCG Pasteur) and M. tuberculosis. The gene encoding a MPB70 homologue secreted by BCG Tokyo was found at the upstream region of the gene encoding MPB70, with approximately 2.3 kilobase pairs (kbp) spacing: the same gene was also found in BCG Pasteur. This gene was cloned and sequenced from BCG Tokyo. The DNA sequence which contained a 663 base pair (bp) open reading frame beginning at position 1 and ending with a TAA codon at position 661 was found. Its theoretical molecular mass was calculated to be 22.068 kDa. This gene was highly homologous to the coding region of mpb70 and the deduced amino acid sequence was very similar to MPB83 reported by Harboe et al. It was speculated that the gene the authors characterized probably corresponded to the mpb83 gene.

The crystal structure of lipase II from Rhizopus niveus at 2.2 A resolution.

The crystal and molecular structure of Lipase II from Rhizopus niveus was analyzed using X-ray single crystal diffraction data at a resolution of 2.2 A. The structure was refined to an R-factor of 0.19 for all available data. This lipase was purified and crystallized as Lipase I, which contains two polypeptide chains combined through non-covalent interaction. However, during crystal growth, Lipase I was converted to Lipase II, which consists of a single polypeptide chain of 269 amino acid residues, by limited proteolysis. The structure of Lipase II shows a typical alpha/beta hydrolase fold containing the so-called nucleophilic elbow. The catalytic center of this enzyme is analogous to those of other neutral lipases and serine proteases. This catalytic center is sheltered by an alpha-helix lid, which appears in neutral lipases, opening the active site at the oil-water interface.

Malignant eccrine poroma with multiple visceral metastases: report of a case with autopsy findings.

An autopsy case of malignant eccrine poroma (MEP) with multiple visceral metastases is reported. A flat, dark tumor of 1 cm in diameter developed on a pre-existing pigmented spot at the left side of the waist of a 58-year-old male. The histopathology suggested the tumor to be malignant melanoma. Nine years later, a painless swelling occurred in the left lower leg, resulting from the obstruction of lymphatics at the left inguinal region. The swelling continued and spread with pain, reaching the inguinal region. Two years later, several papules appeared around the left knee, from which an extensive lymphorrhea occurred. The histopathology of the resected papules suggested epidermoid carcinoma or trichilemmal carcinoma, mainly localized within the lymphatics of the upper dermis. Re-examination of the first skin tumor and electron microscopy of the tumor obtained at autopsy revealed that both tumors were MEP. Although the metastases to the local cutaneous regions and lymph nodes via the lymphatics occur in 20% of MEP, cases with multiple visceral metastases are very few.

Intracellular calcium response to hydraulic pressure in human trabecular cells.

AIMS: To understand the mechanism for regulation of intraocular pressure, human trabecular cells were examined to determine whether they could respond to the change in hydraulic pressure. METHODS: Human trabecular cells were cultured from trabeculum tissue fragments excised during trabeculectomy in four eyes of three patients with primary open angle glaucoma and exposed to the change of hydraulic pressure in a tissue culture flask connected to a glass syringe. The pressure was exerted by automatic infusion of the piston of the syringe and monitored by a pressure gauge. The intracellular calcium concentration was measured in real time with a calcium binding fluorescent dye, fluo-3. RESULTS: A small number (about 10%) of cells appearing morphologically to be trabecular cells showed transient elevations or oscillations of the intracellular calcium concentration in response to the elevation of hydraulic pressure to 20-30 mm Hg, indicating that a part of the human trabecular cells could sense the change in hydraulic pressure. CONCLUSION: Some cells in the human trabecular tissue seem to sense the change in intraocular pressure and might play a role in its regulation.

Characteristics of the insulin resistance syndrome in a Japanese population. The Jichi Medical School Cohort Study.

We investigated the relationships between hyperinsulinemia (a major indicator of the insulin resistance syndrome), blood pressure, dyslipidemia, and coagulation factors in 2606 community-dwelling Japanese individuals as part of the Jichi Medical School Cohort Study. An age-related decrease of the fasting insulin level was found in men but not in women. Body mass index, systolic and diastolic blood pressure, triglyceride and fasting glucose levels, and factor VII activity all increased in both sexes as the insulin level became higher, while the HDL cholesterol level decreased. In addition, total cholesterol and LDL cholesterol levels increased as the insulin level became higher and lipoprotein(a) levels decreased in the men. Fibrinogen levels were not related to the insulin level in either sex. Multiple logistic regression analysis revealed that fasting insulin levels were positively correlated with body mass index and fasting glucose and factor VII activity levels, whereas they were negatively correlated with HDL cholesterol in both sexes. In addition, fasting insulin levels were positively correlated with LDL cholesterol levels in men and with triglyceride levels in women. Our results indicate that hyperinsulinemia is associated with high factor VII activity in a general Japanese population as well as with high blood pressure and dyslipidemia. The accumulation of these cardiovascular risk factors in hyperinsulinemic subjects appears to contribute to cardiovascular events in the Japanese as well as in westerners.

Factor VII hyperactivity and endothelial cell damage are found in elderly hypertensives only when concomitant with microalbuminuria.

We studied the relationship among albuminuria, factor VII (FVII) hyperactivity, and endothelial cell damage in 6 elderly hypertensive subjects. The plasma levels of activated FVII (FVIIa), FVII coagulant activity, FVII antigen (FVIIag), von Willebrand factor (vWF), and thrombomodulin were measured to assess FVII hyperactivity and endothelial cell damage, and urinary albumin excretion rate (UAE) was calculated using 12-hour nighttime (7 pm to 7 am) urine collection (mean for 2 consecutive nights). We performed 24-hour ambulatory blood pressure monitoring in all 61 hypertensive patients and classified them into a white-coat hypertension group (n=12) and a sustained hypertension group (n=49). For the levels of FVII, vWF, and thrombomodulin, there were no differences between the white-coat hypertension group and normotensive control subjects (n=25). In the sustained hypertensive group, only the microalbuminuric subgroup (UAE, 15 to 300 microgram/min: n=30) showed significant elevation compared with the normotensive group for the level of FVIIa (mean [95% confidence interval]: 4.0 [3.6 to 4.4] versus 3.0 [2.6 to 3.3] ng/mL, P<.001), the FVIIa/FVIIag ratio (an indicator of activation of FVII zymogen to FVIIa) (1.33 [1.19 to 1.50] versus 1.04 [0.92 to 1.19], P<.01), the level of vWF (188 [165 to 214] % versus 144 [129 to 160] %, P<.01), and thrombomodulin (11.7 [10.3 to 13.3] versus 9.3 [8.5 to 10.3] ng/mL, P<.01). In contrast, none of these levels in the normoalbuminuric hypertensive group (UAE <15 microgram/min, n=19) differed from that in the normotensive control group. These results suggest that among elderly hypertensives, only those with microalbuminuria show enhancement of FVII activation and endothelial cell damage, while patients with white-coat hypertension and normoalbuminuric hypertensives do not show these accompanying abnormalities. Thus, increased levels of FVII activity and markers of endothelial cell damage might account for the higher risk of cardiovascular events in essential hypertension with microalbuminuria.

'Silent' cerebral infarction is associated with hypercoagulability, endothelial cell damage, and high Lp(a) levels in elderly Japanese.

"Silent" lacunar stroke, often found in the elderly, has been proposed as a predisposing condition for clinically overt stroke. However, the risk factors related to this condition have not been studied thoroughly. We conducted brain magnetic resonance imaging and measured the levels of fibrinogen, molecular markers of coagulation activation [prothrombin fragment 1 + 2 (F1 + 2)] and endothelial cell damage [von Willebrand factor (vWF) and thrombomodulin], and lipid profiles including lipoprotein (a) [Lp(a)] in 178 asymptomatic, high-risk, Japanese subjects aged 44 to 93 years. We also studied 32 symptomatic patients with lacunar stroke (symptomatic lacunar group). The prevalence of silent lacunar stroke increased with age up to 85 years but decreased with age in those 85 years old and older. Of the 160 elderly subjects ( > or = 60 years) 84 (53%) had > or = 1 lacunar infarcts (silent lacunar group) and the remaining 76 were considered as the nonlacunar group. Fibrinogen and F1 + 2 levels in the silent lacunar group were significantly higher than those in the nonlacunar group (P < .01). Mean Lp(a) levels and the prevalence of subjects with an Lp(a) level > 30 mg/dL were significantly higher in the symptomatic lacunar group than the nonlacunar group (P < .05), whereas these levels in the silent lacunar group were intermediate to those of the other two groups. When we further classified the silent lacunar group into three subgroups based on the number of lacunes (few lacunes, 1 or 2; moderate number of lacunes, 3 or 4; and numerous lacunes, > or = 5), levels of Lp(a), F1 + 2, vWF, and thrombomodulin were significantly higher and Lp(a) levels > 30 mg/dL more common in the numerous-lacune than in the few-lacune subgroup. We conclude that silent lacunar stroke is often found in asymptomatic, high-risk, elderly Japanese patients and that silent multiple lacunar stroke is associated with hypercoagulability, endothelial cell damage, and high Lp(a) levels.

Nocturnal fall of blood pressure and silent cerebrovascular damage in elderly hypertensive patients. Advanced silent cerebrovascular damage in extreme dippers.

To study the relation between diurnal blood pressure variations and silent cerebrovascular damage, we performed both 24-hour ambulatory blood pressure monitoring and brain magnetic resonance imaging in 131 elderly asymptomatic hypertensive patients. Silent cerebrovascular damage was identified by the magnetic resonance imaging findings of lacunae (low intensity in T1-weighted images and high intensity in T2-weighted images) and advanced periventricular hyperintense lesions (on T2-weighted images). The frequency of silent cerebrovascular damage in the 100 patients with sustained hypertension was greater than that in the 31 patients with white coat hypertension. We further classified the former group into nondippers (nocturnal reduction of systolic pressure by < 10% of awake systolic pressure; n = 46), dippers (reduction by > or = 10% to < 20%; n = 38), and extreme dippers (reduction by > or = 20%; n = 16). The extent of silent cerebrovascular damage was least severe in the dipper group (P < .05). This J-shaped relation was not found either with the cardiac hypertrophy detected by electrocardiography or with the renal damage assessed by urinary albumin excretion. More than half of the extreme dippers were patients with isolated systolic hypertension, and this prevalence was significantly greater than that in dippers or in nondippers (21% and 30%, respectively). Extreme dippers also had greater variability of pressure (standard deviation of awake systolic pressure) than dippers. Our results indicate that in addition to nondipping, extreme dipping (marked nocturnal fall of blood pressure) should be considered a type of abnormal diurnal blood pressure variation in elderly patients with hypertension who are likely to have advanced silent cerebrovascular damage.

S-methyl methanethiosulfonate, bio-antimutagen in homogenates of Cruciferae and Liliaceae vegetables.

The isolation of a new type of bio-antimutagen, S-methyl methanethiosulfonate (MMTS), from cauliflower (Brassica oleracea var. botrytis) and the distribution and formation of MMTS in Cruciferae and Liliaceae vegetables are described. For the separation and purification, cauliflower curds were homogenized, extracted with acetone, and then purified by organic solvent extraction and by various processes of chromatographic separation. The chemical structure of an active principle was identified as MMTS by GC-MS and 1H-NMR analyses. MMTS was widely found in vegetable homogenates of the Cruciferae and Liliaceae species, and is found from its precursor S-methyl-L-cysteinesulfoxide (SMCS), by wounding the vegetable tissues. Wounding may induce C-S lyase, which converts SMCS to MMTS. The amount of MMTS formed was affected by the pH value for C- lyase, but not by the SMCS content in a tissue homogenate.

Autoantibodies and immunoglobulins in atomic bomb survivors with human T-lymphotropic virus type I.

The association of human T-lymphotropic virus type I (HTLV-I) with autoimmune disorders was investigated on the basis of prevalence of antinuclear antibody (ANA), rheumatoid factor and anti-thyroglobulin antibody as well as immunoglobulin (Ig) serum level (IgG, IgA, and IgM). The subjects, all atomic bomb survivors, were 59 HTLV-I-seropositive people without HTLV-I-associated myelopathy or adult T-cell leukemia and 149 HTLV-I-seronegative persons. The mean serum level of IgM was higher in HTLV-I-seropositive subjects than in HTLV-I-seronegative subjects, and a significant association with HTLV-I and sex was indicated in the IgM serum level. No association with HTLV-I was indicated in the prevalence of the autoantibodies except for ANA. These results suggest some clear humoral immunity differences between HTLV-I-infected and noninfected subjects, but whether HTLV-I infection can lead to autoimmune disorders remains uncertain.

Effect of beef tallow diet on alpha 2-adrenergic receptor binding in rat brain regions.

We examined the effects of dietary fats consisting of different fatty acids on alpha 2-adrenergic receptor binding in the hypothalamus and cerebral cortex. Sprague-Dawley male rats were meal-fed isoenergetic diets based on safflower oil or beef tallow for 8 weeks. alpha 2-Adrenergic receptor bindings were determined with [3H]para-aminoclonidine. Binding affinities of alpha 2-adrenergic receptor in the hypothalamus and cortex were significantly higher in the beef tallow diet group. The values of the polyunsaturated/saturated fatty acid ratio and the fluidity of the plasma membrane in the hypothalamus and cortex were lower in the beef tallow diet group than in the safflower oil diet group. These results suggest that the beef tallow diet decreases membrane fluidity by altering the fatty acid composition in rat brain regions. Consequently, alpha 2-adrenergic receptor binding affinities in the brain regions are higher in rats fed the beef tallow diet than in rats fed the safflower oil diet.

Effect of dietary fat type on beta-oxidation of brown adipose tissue and Na+ channel density of brain nerve membrane in rats.

Effect of dietary fat type on beta-oxidation of brown adipose tissue and Na+ channel density of brain nerve membrane was studied in rats. Rats were fed an experimental diet containing lard, high oleic safflower oil, safflower oil or linseed oil for 12 weeks. The activities of carnitine palmitoyltransferase and cytochrome oxidase in brown adipose tissue were significantly lower in rats fed the lard diet than in those fed the high-oleic safflower oil diet, safflower oil diet or linseed oil diet. However, the peroxisomal beta-oxidation activity in brown adipose tissue was significantly higher in rats fed the lard diet than in those fed the other diet. The Na+ channel density in brain nerve membrane was not significantly different among the diet groups. These results suggest that intake of the lard diet rich in saturated fatty acids, compared with the vegetable oil diets rich in unsaturated fatty acids decrease mitochondrial beta-oxidation in brown adipose tissue, and that the dietary fat type has a differential effect on mitochondrial and peroxisomal beta-oxidation in brown adipose tissue.

Capsaicin in diet does not affect glycogen contents in the liver and skeletal muscle of rats before and after exercise.

Effects of capsaicin, a pungent principle of hot red pepper, on glycogen contents in the liver and skeletal muscle at rest and during exercise were studied in rats. The contents of glycogen in the liver and soleus muscle, and the concentrations of serum glucose, lactate, free fatty acid and glycerol were examined. Capsaicin was supplemented at 0.014% of the experimental diet. Each group of rats was fed the capsaicin-diet ad libitum for 7 days, and then both groups of rats were fed isoenergetic diets with or without capsaicin for 7 days. Rats were trained running with a treadmill for 14 days. At the final stage of the experiments, 1 h running was loaded after the capsaicin diet or the control diet. The glycogen contents in the liver and soleus muscle were not significantly different between the capsaicin-diet group and the control-diet group after the meal and during exercise. The concentrations of serum glucose, lactate, free fatty acid and glycerol were not significantly different between the two dietary groups after the meal and during exercise. The results in this study suggest that the intake of capsaicin have little effect on glycogen contents in the liver and soleus muscle at rest and during exercise in rats previously fed a capsaicin-diet ad libitum for 1 week.

Beef tallow diet decreases uncoupling protein content in the brown adipose tissue of rats.

The effects of dietary fats consisting of different fatty acids on the content of mitochondrial uncoupling protein in the interscapular brown adipose tissue were studied in rats. Sprague-Dawley male rats were meal-fed an isoenergetic diet based on either beef tallow or safflower oil for nine weeks. The gain in body weight during the experimental period did not differ between the two dietary groups. The weight of the brown adipose tissue was similar in the two dietary groups, whereas the weight of the abdominal white adipose tissue was larger in rats fed the beef tallow diet. The content of mitochondrial uncoupling protein in brown adipose tissue was lower in the beef tallow diet group than in the safflower oil diet group without differing mitochondrial mass between the two dietary groups. These results suggest that, in rats, a beef tallow diet reduces the content of uncoupling protein in brown adipose tissue, resulting in lower diet-induced thermogenesis as compared to a safflower oil diet.
