Inhibition of active oxygen generation in guinea-pig neutrophils by biscoclaurine alkaloids.

Effect of biscoclaurine alkaloids, such as cepharanthine, on active oxygen production of neutrophils was investigated. Cepharanthine inhibited both superoxide generation and luminol-dependent chemiluminescence (CL) induced by either formylmethionyl-leucyl-phenylalanine, opsonized zymosan, arachidonic acid or by phorbol myristate acetate. Ca2(+)- and phospholipid-dependent protein kinase (PKC) activity and the phosphorylation of cytoplasmic protein including 47 kDa proteins of neutrophils were also inhibited by cepharanthine; dose dependent inhibition of CL was quite similar to that of PKC. Among various biscoclaurines tested, the inhibitory effect of cepharanthine, tetrandrine and isotetrandrine was strong, but that of berbamine and cycreanine was weak; the inhibitory action of the former on lipid peroxidation and platelet aggregation were also stronger than those of the latter. These and other observations indicated that these alkaloids inhibited the active oxygen generation by way of stabilizing plasma membrane and inhibiting PKC and NADPH oxidase activation.

Benign fibrous histiocytoma involving the ends of long bone.

Five bone tumors, previously labelled as benign fibrous histicytoma in our Bone Tumor registry were reviewed. All the cases were reexamined clinically, radiologically, and histologically. Ages of the patients ranged from 17 to 68 years and all tumors involved the epiphysis or epiphysiometaphysis of long tubular bones. A diagnosis of giant cell tumor was suggested from initial radiographs in all cases. Three cases proved to be giant cell tumors of bone after thorough examination of previous histological specimens. In the diagnosis of benign fibrous histocytoma of epiphyseal origin in adults, the possibility of giant cell tumor of bone should always be kept in mind, and painstaking histological examination is required.

A case of alveolar rhabdomyosarcoma with a chromosomal translocation, t(2;13)(q37;q14).

We report on an alveolar rhabdomyosarcoma occurring in a 17-year-old girl. Histologically, the tumour exhibited a proliferation of small, round cells and formed alveolar structures. Immunohistochemical studies of the tumour cells showed desmin and creatine kinase MM positivity. In electron-microscopic studies, the tumour cells showed an abundance of glycogen granules, and myofilaments were recognized in the cytoplasm. Chromosome analysis revealed a translocation, t(2;13)(q37;q14), which is though to be common in this subtype of rhabdomyosarcoma. Conventional ultrastructural and immunohistochemical investigations and chromosome analysis thus appear to be a highly promising combination of methods for improved pathological diagnosis of alveolar rhabdomyosarcoma.

Protective effect of recombinant human granulocyte colony-stimulating factor (rG-CSF) against various microbial infections in neutropenic mice.

Protective effect of recombinant human granulocyte colony-stimulating factor (rG-CSF) on microbial infections was studied in cyclophosphamide (CPA)-induced neutropenic mice. The neutropenic mice showed severely decreased resistance against systemic infections of Pseudomonas aeruginosa, Escherichia coli, Serratia marcescens, Staphylococcus aureus, and Candida albicans. When such mice were injected subcutaneously with rG-CSF on four consecutive days beginning the day after CPA injection, the decreased anti-microbial resistance of the mice was restored to the level of that in normal mice. The anti-infective effect of rG-CSF was dose-dependent and the 50% effective doses (ED50) in various microbial infections tested were 1-10 micrograms/kg/day. The results suggest that rG-CSF is useful for protection of neutropenic patients from microbial infections.

Antimicrobial activities of thiolactomycin against gram-negative anaerobes associated with periodontal disease. f1.

Thiolactomycin (TLM), (4R)-(2E,5E)-2,4,6-trimethyl-3-hydroxy-2,5, 7-octatriene-4-thiolide, purified from a culture filtrate of a strain of the Nocardia species, was examined for antimicrobial activities against more than 100 strains of oral and periodontally associated bacteria. Nine other commonly used antibiotics were also included for the test. We found that TLM exhibited strong and selective antimicrobial activities against Bacteroides gingivalis and other oral black-pigmented Bacteroides species that may be etiologically associated with adult periodontitis. TLM also inhibited the growth of Actinobacillus actinomycetemcomitans, but did not affect the growth of oral streptococcal species and Eubacterium species. Strains of Eikenella corrodens were moderately susceptible to TLM, while Actinomyces viscosus strains were only slightly susceptible to it. Other antibiotics used for comparison showed a broad spectrum of antimicrobial activities in general. In conclusion, TLM exhibited highly selective antimicrobial activities to black-pigmented Bacteroides species and A. actinomycetemcomitans, both of which are implicated in the pathogenesis of human periodontal disease.

Morphological and cytogenetic studies of a human synovial sarcoma xenotransplanted into nude mice.

Specimens of a synovial sarcoma from the left thigh of a 60-year-old woman and cells from the tumor transplanted into nude mice were examined morphologically and immunohistochemically, and the karyotype was analyzed. Immunohistochemically, cells of both the parent tumor and the transplanted tumor were positive for vimentin and cytokeratin. The multicystic features of the parent tumor were not reproduced in the mice. On ultrastructural examination, the parent tumor cells were found to have dilated channels of endoplasmic reticulum as well as a junctional apparatus, and they formed extracellular spaces. The transplanted tumor cells, in contrast, had a poorly developed endoplasmic reticulum and were devoid of extracellular spaces. Chromosome analysis of the tumor cells revealed a translocation, t (X; 18) (p11; q11), as reported previously for synovial sarcoma, thus suggesting the diagnostic utility of the chromosome pattern for identification of synovial sarcoma.

Rotation of the tectal primordium reveals plasticity of target recognition in retinotectal projection.

Retinotectal projection is precisely organized in a retinotopic manner. In normal projection, temporal retinal axons project to the rostral part of the tectum, and nasal axons to the caudal part of the tectum. The two-dimensional relationship between the retina and the tectum offers a useful experimental system for analysis of neuronal target recognition. We carried out rotation of the tectal primordium in birds at an early stage of development, around the 10-somite stage, to achieve a better understanding of the characteristics of target recognition, especially the rostrocaudal specificity of the tectum. Our results showed that temporal retinal axons projected to the rostral part of the rotated tectum, which was originally caudal, and that nasal axons projected to the caudal part of the rotated tectum, which was originally rostral. Therefore, the tectum that had been rotated at the 10-somite stage received normal topographic projection from the retinal ganglion cells. Rostrocaudal specificity of the tectum for target recognition is not determined by the 10-somite stage and is acquired through interactions between the tectal primordium and its surrounding structures.

[Cerebellar infarction; the findings of CT, MRI and cerebral angiography].

The findings of CT, MRI and cerebral angiography were analysed in 18 cases with cerebellar infarction, of which 10 were men and 8 women. The areas of infarction were as follows; PICA in 9 cases, SCA in 8 cases, AICA and diffuse type in one case each. One case had two lesions. CT showed low density areas in 15 cases after 24 hours of the onset, but an abnormality was not demonstrated in one case which had an infarction of PICA area. MRI showed a high-intensity lesion on T2 weighted image during the first and tenth day after the onset, while a low-intensity lesion on T1 weighted image and high-intensity lesion on T2 weighted image were demonstrated after seventh day. MRI revealed the area of infarction better than CT. Vertebral angiography and DSA showed narrowing or poor visualization of the cerebellar arteries in 5 cases but this finding was not shown in 3 cases which were examined by IVDSA.

[Antitumor activity of a new platinum complex, (R)-(-)-2-aminomethylpyrrolidine (1, 1-cyclobutanedicarboxylato) platinum (II), against cisdiamminedichloroplatinum (II)-resistant murine leukemia cell line].

Antitumor activity of a new platinum complex, (R)-(-)-2-aminomethylpyrrolidine (1, 1-cyclobutanedicarboxylato) platinum (II) (DWA 2114 R) against cisdiamminedichloroplatinum (II) (CDDP)-resistant tumor was examined in in vitro and in vivo experiments. CDDP-resistant line was established from L 1210 mouse leukemia cells by continuous exposure to CDDP in dose-escalation manner. Six clones were isolated from parental resistant line and one of these clones, clone f, which was found to be highly resistant (30-40 fold) to CDDP, was used in the following experiments. Clone f showed 4-7 fold cross-resistance to DWA 2114 R and 11-19 fold to cisdiammine-1, 1-cyclobutanedicarboxylatoplatinum (II) (CBDCA) in in vitro growth inhibition assay. DWA 2114 R showed the most effective antitumor activity against mice transplanted with the resistant cells in the increase of life span (ILS%). About 100% of ILS and cured mice were observed in the treatment with DWA 2114 R. On the other hand, CDDP or CBDCA showed a little increase in the survival time (less than 40% of ILS) and all mice died. These results suggest that DWA 2114 R seemed to be more effective against CDDP-resistant tumors clinically than CDDP and CBDCA.

A long-term follow-up study of total hip replacement with bone graft. Correlations between roentgenographic measurement and hip mobility.

Fifty total hip replacement treating the acetabular deficiency with autologous bone graft from the femoral head were performed on 44 patients with congenital dislocation of the hip. The average age was 55 years. The average follow-up period was 5 years and 2 months, ranging from 3 years, 8 months to 6 years, 11 months. Solid union was demonstrated in all acetabular grafting. No loosening of either acetabular or femoral components occurred. Roentgenological measurements used as a parameter of good results showed that better results were obtained using the true acetabulum (ideal position) with appropriate bony coverage.

Oxidation of cytosolic NADH by the malate-aspartate shuttle in MC29 hepatoma cells.

The malate-aspartate shuttle activity for the reoxidation of cytosolic NADH was studied in MC29 avian hepatoma cells whose mitochondria preferentially utilized glutamine and produced aspartate for ATP formation. The tumour cells showed reoxidation of NADH, as evidenced by the accumulation of pyruvate, when incubated aerobically with L-lactate. The involvement of the respiratory chain and transaminase in the process was demonstrated by the addition of specific inhibitors. When the tumour cells were cultured in Eagle's medium with aminooxyacetate or in the absence of glutamine, a marked reduction in the cellular NAD/NADH ratio was observed. These results indicate that the malate-aspartate shuttle was actively functioning in the tumour cells and that this hepatoma may provide a suitable system for the investigation of the bioenergetics of malignant cells.

End products of glutamine oxidation in MC-29 virus-induced chicken hepatoma mitochondria.

Products of glutamine metabolism were examined in the MC-29 virus-induced chicken hepatoma mitochondria incubated in vitro. Glutamine oxidation proceeded in the tumor mitochondria exclusively via a pathway involving glutamic-oxalacetic transaminase. Malate stimulated aspartate production from glutamine, while pyruvate exerted suppressive effect on aspartate production with little alanine formation. The mitochondria of this hepatoma are unique in that the metabolic pattern and response to malate and pyruvate are essentially inconsistent with those reported in normal cells as well as those proposed by Moreadith and Lehninger in various tumor cells.

Syndrome of generalized (peripheral tissue and pituitary) resistance to thyroid hormone.

Generalized resistance to thyroid hormone (GRTH), or Refetoff syndrome, is a disease in which peripheral tissues show resistance to thyroid hormone. Three patients with this disease were investigated. Cases 1 and 2 involved identical 7-year-old female twins and case 3, a 5-year-old girl. All three patients had goiters, and cases 1 and 2 had sensorineural deafness. In all three, the blood levels of T4, free T4, and T3 were high, while the blood levels of TSH were normal or slightly elevated. The responses shown by blood levels of the thyroid hormone and TSH to administration of propylthiouracil and T3 suggest that the regulating mechanism in the hypothalamic-pituitary-thyroid system was functional. Upon administration of T3, no sign of hyperthyroidism was observed.

Synthesis and antitumor properties of N1-acyloxymethyl derivatives of bis(2,6-dioxopiperazines).

Many N1-acyloxymethyl derivatives VI of bis(2,6-dioxopiperazine) I, ICRF-154, were prepared and tested for antitumor activity. The treatment of I with formaldehyde gave a crystalline bis(N1-hydroxymethyl) derivative VII, which was acylated under various conditions to give bis(N1-acyloxymethyl) derivatives VI. Antitumor activity of VI against P388 leukemia in mice was studied. Several bis(N1-acyloxymethyl) compounds such as phenylacetyloxymethyl VI-6, methoxycarbonyloxymethyl VI-41, isobutoxycarbonyloxymethyl VI-44, and furancarboxymethyl VI-38 compounds were found to have potent antitumor activities. On the other hand, water-soluble esters having an amine or a carboxylic acid function in their acyl groups showed rather reduced activity. These bis(N1-acyloxymethyl) derivatives VI were presumably hydrolyzed into the parent bis(2,6-dioxopiperazine) I by nonspecific esterase in the body to exhibit their antitumor activity.

Glutamine synthetase and glutaminase activities in various hepatoma cells.

Glutamine synthetase and glutaminase activities in a series of hepatoma cells of human and rat origins were determined for comparison with normal liver tissues. Marked decrease in glutamine synthetase activity was observed in the tumor cells. Phosphate-dependent and phosphate-independent glutaminase activities were increased compared with those from normal liver tissues. Well coupled mitochondria were isolated from HuH 13 line of human hepatoma cells and human liver. Oxypolarographic tests showed that glutamine oxidation was prominent in the tumor mitochondria, while mitochondria from the liver showed a feeble glutamine oxidation. Glutamine oxidation was inhibited by prior incubation of the mitochondria with DON (6-diazo-5-oxo-L-norleucine), which inhibited mitochondrial glutaminase. These results indicate that the product of glutamine hydrolysis, glutamate, is catabolized in the tumor mitochondria to supply ATP.

[Effect of azelastine on bronchial hyperresponsiveness].

We have investigated the effect of azelastine on bronchial responsiveness in 25 asthmatics, using methacholine inhalation challenge, (Astograph). Three parameters, initial respiratory resistance (RrsC), bronchial sensitivity (Dmin), and bronchial reactivity (SGrs/GrsC) were studied. After 8 weeks' treatment with 2 mg azelastine b.i.d., Dmin was increased significantly and after 4 weeks an insignificant increase was observed. RrsC and SGrs/GrsC did not change significantly during the 8 week treatment. Recent advances have revealed that various chemical mediators, especially leukotrienes, are closely related to bronchial hyperresponsiveness. Accordingly, the improvement of Dmin observed during azelastine treatment might be ascribed to its inhibitory action on the synthesis and release of leukotrienes.
