The double contrast hysterography, which was performed by the continuous CO2 insufflation with the ordinary HSG, was applied to 162 infertile patients. Forty-nine of 162 patients showed the abnormal findings by this method. Hysteroscopic or histological examinations revealed that 26 of these patients had hyperplasia, 15 had polyp, 2 had myoma and 1 had intrauterine foreign body. When these results were compared with those obtained by ordinary HSG, it was found that the double contrast hysterography would be more useful to investigate the detailed intrauterine changes such as hyperplasia or polyp than the ordinary HSG, and, therefore, would be a useful aid in the clinical care.

The synthesis of hCG and its subunits in temperature-sensitive, simian virus 40 tsA mutant-transformed human first trimester and term placental cells and in choriocarcinoma cells was studied in the presence and absence of glucocorticoid hormones. The tsA-transformed placental cells were studied at 33 C (the temperature at which the cells exhibit the transformed phenotype) and at 40 C (the temperature at which the cells regain their differentiated nontransformed phenotype). The glucocorticoids cortisol and dexamethasone did not affect the synthesis of hCG or hCG alpha in either transformed term placental cells or choriocarcinoma cells. In the tsA-transformed first trimester placental cells, however, glucocorticoids greatly inhibited hCG synthesis but induced hCG alpha synthesis. The inhibition on hCG synthesis was immediate, stronger at 40 C than at 33 C, and independent of the growth stage of the cells. The glucocorticoid effects were specific; neither estradiol nor progesterone affected the synthesis of hCG or hCG alpha. Furthermore, progesterone did not block the action of the glucocorticoid. These selective effects in cultured first trimester placental cells suggest that glucocorticoids may play an important role in the regulation of hCG production during early pregnancy.
The synthesis of pregnancy-specific beta 1-glycoprotein (PS beta G) was studied in simian virus 40 temperature-sensitive A mutant-transformed human first trimester and term placental cells. At the permissive temperature (33 C, transformed phenotype), they produced low levels of PS beta G. At the restrictive temperature (40 C), the transformed phenotype was lost, and the production of PS beta G was greatly enhanced. The PS beta G produced by these transformed placental cells resembled the purified placental PS beta G by several criteria. Both cell and placental PS beta G bound to Concanavalin A-Sepharose and were, therefore, glycoproteins. The cell PS beta G cochromatographed with placental PS beta G on a Bio-Gel A-0.5m column. Furthermore, the slopes of the dose-response curves for the cell PS beta G were indistinguishable from that for placental PS beta G. The synthesis of PS beta G at both 33 and 40 C in these placental cells was greatly induced by sodium butyrate and 5-bromo-2'-deoxyuridine. Sodium butyrate was a more effective inducer at 33 C, whereas BrdUrd appeared to be a better inducer at 40 C.
Effects of a new muscle relaxant E.M.P.P.(4'-ethyl-2-methyl-3-piperidino-propiophenone . hydrochloride) on afferent discharges of muscle spindle were studied in 10 healthy volunteer subjects. Single unit afferent discharges of muscle spindle were recorded microneurographically from the median or tibial nerve using a tungsten microelectrode with a tip diameter of about 1 micrometer (impedence: about 10 M omega). Effects of an oral administration of 150--300 mg of E.M.P.P. on spontaneous afferent discharges as well as dynamic and static responses of muscle spindle were analysed in the present study. 1. E.M.P.P. suppressed remarkably the frequency of spontaneous afferent discharges of muscle spindle. Meanwhile, mental activations such as speech of mental arithmetic recovered temporarily the frequency of afferent discharges to that before E.M.P.P. administration. 2. E.M.P.P. suppressed the dynamic and static responses of muscle spindle to stretch. It also suppressed the dynamic responses of muscle spindle in the decontraction phase of electrically-induced twitch contractions of the receptor-bearing muscle. Based on these results, it is concluded that E.M.P.P. suppresses the static and dynamic activities of muscle spindle in man. These effects seem to be due to modifications of descending influences from central structures on the static and dynamic gamma motoneurons which innervate the muscle spindle.

Alkaline phosphatase activity in human placental cells transformed by a tsA mutant of simian virus 40 (SV40) can be greatly induced by growing these cells at 40 degrees C, the temperature at which the tsA transformants regain their nontransformed phenotype. The induction of alkaline phosphatase in these cells requires the synthesis of both RNA and protein. The induced alkaline phosphatase from a SV40 tsA30 mutant-transformed term placental cell line (TPA30-1) was purified, characterized, and compared with alkaline phosphatase from term placenta and first trimester placenta. The form of alkaline phosphatase found in TPA30-1 cells differs from the phosphatase of term placenta in physiochemical and immunological properties. The TPA30-1 phosphatase is, however, indistinguishable from the alkaline phosphatase of human first trimester placenta by several criteria, including electrophoretic mobility, apparent molecular weight (Mr = 165,000), size of monomeric subunit (Mr = 77,000), heat lability, and sensitivity to inhibition by amino acids and EDTA. In addition, alkaline phosphatase from both TPA30-1 cells and first trimester placenta can be inactivated by antiserum to liver alkaline phosphatase but not by antiserum to term placental alkaline phosphatase. The induction of first trimester phosphatase in cells derived from term placenta provides a system for the study of alkaline phosphatase gene regulation in human placenta.
The plasma 1,25-(OH)2VD3 level in Fanconi syndrome was low but rose rapidly following the administration of 1 alpha-OHVD3. 1 alpha-OHVD3 was administered to 6 patients with different types of Fanconi syndrome. The effect of 1 alpha-OHVD3 proved to be 200-250 times as great as that of vitamin D2. From our results it was suggested that Ca malabsorption due to failure of 1 alpha-hydroxylation of vitamin D in the kidney, together with renal tubular acidosis and hypophosphatemia, is involved in the pathogenesis of the rickets of Fanconi syndrome.
