Staining of serum concanavalin A-binding proteins after micro two-dimensional electrophoresis and blotting.

Human serum proteins separated by micro two-dimensional electrophoresis in the absence of denaturants were electrophoretically transferred to nitrocellulose sheets (blots), and glycoproteins on the blots were stained by sequential incubation with the lectin concanavalin A, the glycoprotein horseradish peroxidase and the substrates of peroxidase diaminobenzidine-hydrogen peroxide. The stained serum concanavalin A-binding glycoproteins were identified by referring to the "identification map" of human plasma proteins, which we have prepared by immunochemical staining of the proteins on the blots. The concanavalin A-peroxidase method applied for the blots after two-dimensional electrophoresis provided information on the location of serum concanavalin A-binding glycoproteins, and showed greater sensitivity than Coomassie Brilliant blue R-250 staining for several serum proteins.

Immunohistochemical localization of the secretory products of rat Clara cells.

We used proteins in rat lung lavage fluid to successfully produce an antiserum against Clara cell secretory products. When used with the immunoperoxidase method, the antiserum specifically stained cells of the bronchiolar lining, which are morphologically consistent with Clara cells, as well as a few columnar cells in the bronchial and tracheal mucosa. B-5-fixed lung tissue furthermore demonstrated the immunoreactive layer over the bronchiolar epithelium. The alveolar and bronchial lining layers, on the other hand, were not immunoreactive, although a trace of granular immunoreactivity was seen in the latter. It was suggested that Clara cells produce and secrete some proteinaceous materials, which are mainly localized in the bronchiolar area after secretion and are seldom transferred into the alveolar lining layer. Our antibody cross-reacted with the Clara cells of mice, but not with those of hamsters, guinea pigs, rabbits, dogs, cats, monkeys, and man. The high degree of specificity of this antisera to Clara cells in formalin-fixed materials should make it a valuable tool for identifying Clara cell change in non-neoplastic lung pathology and in obtaining some insights into cell origin in neoplastic diseases.

Plasma levels of secretory IgA in patients with gastric cancer.

The levels of plasma secretory IgA were measured in patients with gastric cancer and found to be slightly higher (9.6 +/- 6.2 micrograms/ml) than those in healthy controls (7.0 +/- 2.6 micrograms/ml, 0.05 less than P less than 0.1). Secretory IgA levels in those with hepatic metastases (19.7 +/- 12.2 micrograms/ml) were significantly higher than those in patients without hepatic metastases (P less than 0.001). In the latter, there was no significant relationship between plasma secretory IgA levels and the deepest layer of cancerous invasion or lymph node metastases. The secretory IgA levels in cases of well differentiated tubular adenocarcinoma were significantly higher than those with poorly differentiated adenocarcinoma (P less than 0.05). Although there is small diagnostic value in the detection of gastric cancer by measuring the levels of secretory IgA, high levels of secretory IgA in gastric cancer patients may be indicative of the presence of hepatic metastases.

The role of plasma secretin during starvation in dogs.

Plasma secretin levels were measured during a 72-hour starvation period in dogs. Plasma secretin rose significantly and insulin levels fell significantly during starvation. Intravenous glucose loading at the end of the 72-hour starvation period suppressed plasma secretin and free fatty acid levels with a concomitant rise of glucose and insulin levels. These findings suggest that secretin might be regulated by plasma glucose and insulin levels and might play a role in lipolysis during starvation.

Ripple pigmentation of the neck in atopic dermatitis.

Ripple pigmentary lesions of the neck were observed in 12 patients with atopic dermatitis. Biopsy specimens from three cases revealed no amyloid substances. From the review of cases with either atopic dermatitis or ripple pigmentation seen in our hospital, it was concluded that ripple pigmentation of the neck was always associated with atopic dermatitis and appeared in 1.7% of all 751 patients with atopic dermatitis. It is suggested that ripple pigmentation is not necessarily specific or diagnostic of amyloidosis, as is generally believed. Possible relationships involving ripple pigmentation, atopic dermatitis, and macular amyloidosis are also discussed.

High levels of anti-mitochondrial antibodies in MRL mice. Influence of anti-DNA antibodies on anti-mitochondrial antibodies measured by an enzyme-linked immunosorbent assay.

We developed an easy enzyme-linked immunosorbent assay (ELISA) for anti-mitochondrial antibodies, and detected high levels of anti-mitochondrial antibodies in MRL/Mp-lpr/lpr (MRL1/l) mice. The influence of the presence of anti-DNA antibodies in the tested sera on this assay was also evaluated. The monoclonal anti-DNA antibodies, which were made from non-immunized MRL1/l mice, did not react with the mitochondrial antigens adhering to polystyrene plates. Absorption with DNA had little effect on the levels of mitochondrial antibodies in MRL1/l sera. Our assay for mitochondria-binding antibody was very easy and sensitive, although it could not differentiate among heterogeneous anti-mitochondrial antibodies.

Benign renal angiomyolipoma with regional lymph node involvement. Report of a case showing enlargement five years after nephrectomy.

A case of renal angiomyolipoma followed by massive enlargement of the angiomyolipoma in regional lymph nodes five years after nephrectomy is reported. On the basis of its histology and the absence of distant metastasis, we considered this to be a benign, hamartomatous, and multicentric lesion. Considering the rather rapid growth, however, this case raises some questions with regard to the treatment of this condition, i.e., whether removal of a renal angiomyolipoma should be accompanied by lymphadenectomy, and whether the lymph nodes should be carefully scrutinized by CT scanning or echography preoperatively.

Effects of ifenprodil glucuronide derivative on platelet aggregation and vasocontraction.

It has been supposed that ifenprodil glucuronide derivative, detectable in large amount in rabbit plasma, is related to the pharmacological actions by ifenprodil tartrate. However, a synthesized ifenprodil glucuronide derivative was found to have no effect on platelet aggregation and vasocontraction in vitro. These results indicate that ifenprodil itself rather than its glucuronide derivative manifested the pharmacological actions.

Plasma apolipoproteins in Tangier disease, as studied with two-dimensional electrophoresis.

Tangier disease is characterized by a deficiency of high-density lipoproteins and of their major protein constituent, apolipoprotein (apo) A-I. We used high-resolution two-dimensional electrophoresis to examine the principal plasma apolipoproteins (A-I, A-II, A-IV, E, C-II, and C-III) of three persons with Tangier disease, one homozygous patient and his two heterozygous children, comparing the patterns with those for healthy subjects. Characteristic abnormalities were found in the distribution of the isoproteins of apo A-I, there being a normal concentration of pro apo A-I but dramatically decreased concentrations of the other apo A-I isoproteins. We also found hitherto-undescribed polypeptide abnormalities in apo C-III: sialylated and nonsialylated forms of apo C-III appear as double spots having the same isoelectric points but different molecular masses. No other substantial difference was detected in the polypeptide distribution of the other plasma apolipoproteins.

Evaluation of lipoproteins and apolipoproteins in serum of a Tangier patient by micro-scale two-dimensional electrophoresis.

We examined lipoproteins and apolipoproteins in serum of a Tangier-disease patient. We used three different techniques of micro-scale two-dimensional electrophoresis: (a) no denaturants; (b) with sodium dodecyl sulfate (SDS) used only in the slab gel electrophoresis; (c) and with urea and a detergent used in isoelectric focusing and with SDS in slab gel electrophoresis. By technique a, an extremely low concentration of high-density lipoproteins (HDL) in the Tangier serum was seen, and lipoproteins that cannot form HDL complexes were detected as multiple spots in the acidic (pl 4 approximately 5) and relatively low apparent molecular mass (20,000 approximately 80,000) region. By technique b, Tangier low-molecular-mass lipoproteins were dissociated into their constituent apolipoproteins, and we observed a higher proportion of apoC-III, together with lower proportions of apoA-I and apoA-II, than in the normal HDL fraction. Technique c showed the total content of apolipoproteins in the whole Tangier serum, as several workers have reported. The presence of low-molecular-mass lipoproteins and a high concentration of apoC-III in this lipoprotein fraction characterized the Tangier serum.

[The availability of digital subtraction angiography using continuous intra-arterial infusion tubes founded various malignancy].

DSA was employed using continuous intraarterial infusion tubes for various malignancies (73 cases) which were examined a total of 135 times. In head and neck malignancy (50 cases), the general position of the infusion tube had been determined beforehand by dye infusion, but DSA from the tube showed that the tubes in 24 cases (48%) were located in the wrong position, especially in tongue cancer (21 cases) where many tubes were discovered to be in an erroneous position (71%) such as the common carotid artery. We were unable to determine the effect of chemotherapy and radiation using DSA only. In 9 cases of breast cancer for which fixation of the tube was not attempted under X-ray fluoroscopy, 7 (78%) showed an unusual tube position such as the intraaortic arch. In 5 cases of abdominal malignancy, only the tube position for sigmoid colon cancer was unusual. We were able to observe the effect of chemotherapy by DSA in 2 cases. For DSA in one out of 3 hepatomas using a Port-A-Cath, we observed that infusion of anticancer drug with degradable starch microspheres caused a reduction in tumor size. However, in the two remaining cases, we were unable to observe any effect of infusion of these drugs by DSA for various mechanical reasons. DSA from an infusion tube revealed not only the location of the tube accurately and promptly, but also the effect of chemotherapy.

[A case report of a solid and cystic tumor of the pancreas in a 33-year-old woman].

An unusual case of a solid and cystic tumor of the pancreas is presented. A 33-year-old woman with a 15-year history of an abdominal tumor underwent a pancreatectomy. The tumor was encapsulated by a thick connective tissue and exhibited two main patterns: solid sheets of relatively uniform small cells; and, a papillary proliferation with a central fibrovascular talk. The tumor cells had PAS and d-PAS reactive granules in the cytoplasm. Immunohistochemically, a positive reaction for alpha l-antitrypsin and a carcinoembryonic antigen, as well as a weekly positive reaction for a vasoactive intestinal polypeptide also were found in the cytoplasm. Our results lead us to suggest that an S-C tumor is likely to be a tumor of a acinar cell origin.
