Sodium dodecyl sulfate-gel electrophoresis of proteins employing short capillaries.

The performance of capillary gel-sodium dodecyl sulfate (SDS) electrophoresis for molecular-mass-dependent separation of proteins has been examined employing cross-linked polyacrylamide gels and untreated fused-silica capillaries of short length. An apparatus for capillary electrophoresis has been constructed, combining a UV detector equipped with a capillary holder, a power supply, and a micromanipulator for sample loading. The apparatus is capable of accommodating gel capillaries of different lengths and can be operated with two methods of sample loading: manual injection on top of the gel or electrokinetic injection. Standard proteins have been separated according to their molecular mass (ranging from 17000 to 116000) within 30 min, employing gel capillaries of effective lengths shorter than 10 cm and polyacrylamide gels ranging from 2.4 to 4.8%T (5%C). The results confirmed that gel capillaries of effective length less than 10 cm can be used for protein size-separation by SDS electrophoresis, with much higher performance than is achieved in rod gel-SDS electrophoresis.

Studies on the procedure for the construction of cellular protein databases employing micro two-dimensional electrophoresis: an HL-60 protein database.

The procedure to construct a cellular protein database employing a micro two-dimensional electrophoresis (2-DE) technique was studied. Protein information on human cultured leukemic cell HL-60 was collected as an HL-60 protein database, which consists of a 2-DE polypeptide map of 670 recognized spots with spot numbering and information on isoelectric point (pI), molecular mass, and percent integrated density. The procedure employed in this report will be applicable for the comparative studies of cellular proteins and will help to construct comprehensive cellular protein databases.

Morphological changes and proliferative activity of alveolar epithelium in mouse lungs treated with urethan.

We have observed sequential cellular changes in type II pneumocytes in mouse lungs following oral administration of 0.1% urethan solution. Histologically, scatterings of swollen alveolar cells were first noticed 25 days after administration. These cells continued to swell further, after which they aggregated more compactly to form a papillary structure by day 75. After day 100, distinct tumour nodules were found with atypical large cells in some areas. Necrotic foci appeared in large tumours of more than 4 mm diameter. At day 250, nucleoli became distinct and large bizarre and multinucleated cells were intermingled with some mitotic figures. These morphologies correspond to conventional descriptions; that is, hyperplasia (day 25-75), benign neoplastic changes (day 75-100) and malignant neoplastic changes (day 100-250). The proliferating cell nuclear antigen (PCNA) and nucleolar organizer regions (AgNOR) scores in these lesions increased with time, and proliferative activities during tumour progression seemed continuous. However, proliferative activity of the cells on specific days did not differ statistically from the values on neighboring days. We speculate that the hyperplasia-like lesions seen in our mice are neoplastic in nature from their outset.

Ultrastructural changes in the lung in Niemann-Pick type C mouse.

The biochemical and morphological aspects of BALB/c mice with many features of the Niemann-Pick disease type C in man (NP-C mouse) have been studied extensively. However, the pulmonary pathology has not been studied extensively and we describe here some unique ultrastructural features of the lung in the NP-C mouse. Ultrastructurally, macrophages in younger mice contained osmiophilic dense granules and annulolamellar structures, but larger multilamellar concentric structures increased in the macrophages of older mice. In contrast, endothelial cells and type I pneumocytes showed membrane-bound bodies with dense granules and vesicular or vesiculogranular structures as well as amorphous materials. Type II pneumocytes were unremarkable throughout. Our study suggests that endothelial cells and type I pneumocytes are the major site of metabolic derangement resulting in pronounced morphological changes with granular and round membranous structures in the lungs of NP-C mouse. Alveolar macrophages with multilamellar concentric structures may be a result of disturbed disposal of surfactant material from type II pneumocytes rather than that from storage material of type I pneumocyte.

Perforated acute appendicitis in a patient with AIDS/HIV infection: report of a case.

We report herein the case of a 40-year-old man with AIDS who was admitted to hospital with severe abdominal pain, fever, and chills. He underwent an emergency laparotomy which revealed a perforated appendix with suppurative peritonitis. An appendectomy with peritoneal drainage was carried out, but the postoperative course was complicated by fever without leukocytosis; however, he gradually improved following treatment with intravenous antibiotics, granulocyte colony-stimulating factor (G-CSF) and immunoglobulins, and made a complete recovery. His postoperative course demonstrates the effectiveness of this treatment regimen for patients with AIDS complicated by infection without an increase in the white blood cell count (WBC).

Formation of a fatal arterioportal fistula following needle liver biopsy in a child with a living-related liver transplant: report of a case.

A 2 1/2-year-old girl underwent a living-related liver transplantation for biliary atresia, following which four percutaneous liver biopsies were performed uneventfully during her hospital stay for the evaluation of recurrent rejection. Her condition improved without any complications, and she was discharged from hospital in a stable condition on postoperative day (POD) 51. However, 3 months later she was readmitted with marked ascites and a moderate elevation of her liver function tests. A color Doppler ultrasound (US) confirmed hepatofugal portal blood flow, and selective celiac arteriography demonstrated an intrahepatic arterioportal (A-P) fistula. An attempt to embolize the fistula failed and a surgical approach was attempted without success. The patient died from irreversible liver failure on POD 152. A review of the previous histologic analyses revealed that arterial components were present in the fourth biopsy specimen; for this reason it was hypothesized that this procedure may have led to the development of the A-P fistula.

Immunohistochemical staining for type IV collagen and laminin in the stroma of human pancreatic cancer.

Fourteen human pancreatic cancer, six normal pancreatic, and four alcoholic chronic pancreatitis tissue samples were examined by immunohistochemistry with antitype IV collagen and antilaminin monoclonal antibodies (MAb). The basement membranes of acinar, ductal, and endothelial cells from all of the normal pancreatic and alcoholic chronic pancreatitis tissue samples were continuously immunostained, but the stroma was negatively immunostained by both antibodies. On the other hand, all 14 pancreatic cancer tissues showed irregular immunostaining on the basement membranes surrounding cancer cells, and positive fibrillar immunostaining for type IV collagen in the stroma of the carcinomatous parenchyma. Six of the 14 pancreatic cancer tissue samples also showed positive immunostaining for laminin in the stroma. The stromal type IV collagen with or without laminin in human pancreatic cancer tissues may have originated from basement membranes degraded through cancer invasion and proliferation.

Case report: intraglomerular metastasis with neoplastic cell interposition.

A case is described of an 88-year-old man with lung cancer, nephrotic syndrome, and renal dysfunction who died suddenly of an acute myocardial infarction and whose autopsy revealed many adenocarcinoma cells stacked within glomerular capillary lumina of his kidney, entering into basement membrane zones (ie, neoplastic cell interposition). In addition, glomeruli showed a lobular transformation, doubling of glomerular basement membrane, and electron dense deposits along the glomerular basement membrane. These changes were similar to those of membraneoproliferative glomerulonephritis. The association of intraglomerular metastasis and membranoproliferative glomerulonephritis-like lesions led the authors to speculate that the latter glomerular change might have provided an attractive opportunity for circulating tumor cells to be trapped and grow within the glomerular lumina. This mode of metastasis has not been well-recognized. The authors describe the experience, review the literature, and discuss its possible pathogenesis.

Quantitative analysis of collagen and collagen subtypes I, III, and V in human pancreatic cancer, tumor-associated chronic pancreatitis, and alcoholic chronic pancreatitis.

The collagen content in human pancreatic cancer tissue, tissue of tumor-associated chronic pancreatitis (TACP), and normal pancreatic tissue was determined in 14 patients with pancreatic cancer by measuring the amount of 4-hydroxyproline. Four patients with alcoholic chronic pancreatitis (AlCP) were also analyzed. The mean collagen content in both pancreatic cancer tissue and TACP tissue was approximately threefold higher than in normal pancreatic tissue. Cyanogen bromide peptides of type I, III, and V collagens from invasive ductal carcinomatous tissue of the pancreas and from TACP tissue of eight patients were analyzed sequentially using high-performance liquid chromatography with ion-exchange and gel-permeation columns. No difference in the proportion of type I, III, and V collagens was detected between pancreatic cancer tissue and TACP tissue. The mean collagen content in AlCP tissue was significantly lower than that in TACP tissue, but no difference in the proportion of type I, III, and V collagens was detected between these two tissues. These results indicate a similar quantity and distribution pattern of fibrillar collagen in human pancreatic cancer and TACP.

Hyalinizing trabecular adenoma of the thyroid: its unusual cytoplasmic immunopositivity for MIB1.

The monoclonal antibody Ki-67 reacts with a human nuclear cell proliferation-associated antigen that is expressed in all active parts of the cell cycle and is well established as a marker of cell proliferation. However, the Ki-67 method requires fresh frozen material. Recently, MIB1 has been reported to give an immunohistochemical staining pattern identical to Ki-67 on paraffin-embedded tissue sections, frozen sections and cytological samples. This proliferation-associated antigen is apparently localized in the nucleus. Recently, we performed immunohistochemical staining using the monoclonal MIB1 antibody upon a variety of tumors and non-neoplastic conditions of the thyroid. Tumor cells of hyalinizing trabecular adenoma revealed an intense cytoplasmic immunopositivity for MIB1. In contrast, cytoplasmic immunostaining for MIB1 was negative in all other thyroid tumors and non-neoplastic lesions. Because of this unusual staining pattern, we repeated the staining of all cases and found the results to be reproducible. Therefore, we believe that positive cytoplasmic immunostaining for MIB1 is a characteristic finding of hyalinizing trabecular adenoma and is useful in differentiating it from other thyroid tumors.

Intracellular Ca2+ dynamics and in vitro secretory response in acute pancreatitis induced by a choline-deficient, ethionine-supplemented diet in mice.

In order to approach impaired stimulus-secretion coupling in acute pancreatitis induced by a choline-deficient, ethionine-supplemented (CDE) diet in mice, the agonist-evoked intracellular Ca2+ dynamics of dispersed pancreatic acini were evaluated by microspectrofluorometry. Mice were fed a CDE diet for 24 or 48 h, and the pancreas was dispersed to the acini. The in vitro amylase secretion analysis of the CDE groups demonstrated a poor dose-response curve which was significantly lower (p < 0.01) when 100 pM cholecystokinin (CCK) was administered. Both in CDE and control groups, the application of a physiological dose of CCK-8 (10 pM) evoked intracellular Ca2+ oscillations. Periodicity and amplitude of the oscillations in the CDE groups were not significantly different. The administration of a higher dose of CCK-8 (100 pM) evoked a large, sharp, and transient rise in intracellular Ca2+, followed by a small, continuous increase above basal levels for the duration of stimulation both in CDE and control groups. The peak Ca2+ level was lower in the CDE groups, but this was not statistically significant. In conclusion, during the early phase (from 24 to 48 h) of CDE pancreatitis, the pattern of agonist-evoked intracellular Ca2+ release is less affected. Other mechanisms subsequent to the onset of intracellular Ca2+ release are likely to be involved in the inhibition of enzyme secretion.

An intraperitoneal tumorous mass caused by granulomas of microfibrillar collagen hemostat (Avitene).

Microfibrillar collagen hemostat (Avitene) is an absorbable topical hemostatic agent prepared from purified bovine corium collagen. Herein, we report the case of a 56-year-old woman who had undergone an ovariohysterectomy for an ovarian mucinous cystadenocarcinoma 3 months previously and then required a second operation for an intraperitoneal mass associated with microfibrillar collagen hemostat. Microscopically, the resected mass was composed mostly of foreign-body granulomas with microfibrillar collagen hemostat materials. Microfibrillar collagen hemostat was used to control bleeding during the first operation. In a very small portion, however, a recurrent mucinous cystadenocarcinoma was also present. This case is worthy of note in that microfibrillar collagen hemostat may produce huge granulomatous masses that clinically mimic a neoplasm, inducing unnecessary and extensive treatment.

[Five cases of ASD-PDA complex: a review of 38 cases reported in Japan].

Five cases of ASD-PDA complex were experienced. The incidence of ASD-PDA complex was 1.2% of CHD in our institution. The age at operation was ranged from 10 days after birth to 2 years of age. Four patient were male and 1 was female. Also, 4 patients had noncardiac anomalies, including Down syndrome, microcephaly, cleft palate, micrognathia, and cranioschisis. All patient had moderate or severe pulmonary hypertension. Three patients received ASD and DA closure under cardiopulmonary bypass, however, 2 patients were succeeded in decreasing CTR and pulmonary vascular shadow by ligation of DA. Operative and hospital mortality was none. We also studied concerning clinical features and surgical treatment by the review of 38 cases reported in Japan and our 5 cases.

[A case of advanced gastric cancer treated effectively with neo-adjuvant chemotherapy using l-leucovorin and 5-FU].

A 59-year old man was diagnosed as having advanced gastric cancer, liver metastasis and lymph node metastases after a closed medical examination. Because Schnitzler metastasis was also suspected, chemotherapy was started pre-operatively. The schedule of administration was as follows. One course was l-LV 250 mg/m2 and 5-FU 600 mg/m2 injected intravenously every week and continued for 6 weeks. The UGI examination showed partial response in stomach cancer, and liver metastasis disappeared on CT-scan after 2 courses. We performed total gastrectomy, lymphadenectomy, splenectomy and partial resection of liver. Histological effects showed Grade 1a on main tumor, Grade 2 on liver metastasis, and No. 3 lymphnode metastasis. The survival time was 476 days.

[Significance of proliferative breast lesions around breast cancers].

To clarify the relationships between benign proliferative diseases and cancers of the breast, 44 cases treated with breast conserving surgeries were analyzed histopathologically. Intraductal hyperplasia, sclerosing adenosis and adenosis were more frequently seen in cancerous than non-cancerous breasts, with a statistically significant difference. Additionally, histologies of some intraductal spread in carcinomas were identical to those of atypical hyperplasias. Further studies should be undertaken, and definitions of atypical hyperplasia, especially in cancerous breasts, should be critically redefined.

Effect of the oral administration of sepimostat mesilate on cerulein induced acute pancreatitis in rats.

The effect of a potent protease inhibitor, sepimostat mesilate (CAS 103926-82-5, FUT-187), on acute interstitial edematous pancreatitis induced by a supramaximal dose of cerulein, a cholecystokinin (CCK) analogue, was evaluated. The serum amylase activity increased 18-fold over normal control after the infusion of cerulein at 5 micrograms/kg/h for 6 h. The serum lipase activity showed a 235-fold increase. An elevated pancreatic water content, pancreatic interstitial edema, inflammatory infiltration and vacuolization of the acinar cells were found. Redistribution of cathepsin B shifted from the lysosomal pellet fraction to the zymogen granule pellet fraction was noted in the early stages. All these parameters of pancreatitis mentioned above were inhibited by FUT-187 pretreatment at doses of 30 to 300 mg/kg. These observations suggest that FUT-187 inhibits the redistribution of cathepsin B shift from the lysosomal fraction to the zymogen fraction in cerulein-induced acute pancreatitis and improves the parameters of acute pancreatitis.

Effects of linear polyacrylamide concentrations and applied voltages on the separation of oligonucleotides and DNA sequencing fragments by capillary electrophoresis.

Oligonucleotides and DNA sequencing fragments have been separated by capillary electrophoresis employing linear polyacrylamide (LPA) as a sieving matrix. A commercially available apparatus equipped with a laser-induced fluorescence (LIF) detection system has been utilized, but the capillary cartridge has been modified to position the capillaries without coiling. The performance of the separation, the relationship between resolution and analysis time, has been examined using poly(dT)16-500 by changing LPA concentration, capillary length, and electric field strength. It was found that, for large DNA fragments, the migration time interval between bands decreases linearly as DNA fragment size increases. This implies that there exists a maximum base number to be resolved, irrespective of the band width (we named the maximum base number Nmax). The higher value of Nmax is obtained when the applied field strength is lower, but this accompanies longer analysis time with a concomitant increase in band width. Simple experimental equations have been proposed to calculate resolution and migration times of DNA fragments separated in our system at given electrophoretic conditions. Using 9% T LPA and an electric field strength of 100 V/cm, single-base resolution of M13mp10 DNA fragments up to 520 nucleotides has been obtained.
