Dynamic light scattering study of muscle F-actin.

By use of digital autocorrelation and fast Fourier methods, dynamic light-scattering studies of in vitro reconstituted muscle F-actin were made over a wide range of concentrations, 0.01-2 mg/ml F-actin. Measurements of correlation function [g1(t)]2 showed that a transition from a dilute to a semidilute regime for the Brownian motions of filaments occurred at around 0.3 mg/ml F-actin. Beyond this concentration, profiles of successively measured [g1(t)]2 showed very poor reproducibility. This resulted from the existence of very slow components, which could not be measured with a high statistical accuracy even for a measuring time of 3600 s/run. On the other hand, subtraction of these components automatically by an electronic circuit, [g-1(t)]2, or by computer processing, [g1(t)]2, resulted in a fairly good reproducibility of the profiles. The decay characteristics of [g1(t)]2 (and [g-1(t)]2) were very similar to those of [g1(t)]2 for dilute solutions. A theoretical model will be discussed which could account for the above situation. The time sequence [n(t,T)] of photoelectron counts at a sampling time T of light scattered from semidilute solutions of F-actin was stored on magnetic tapes, and both power spectra S(f) and correlation functions [g-1(t)]2 were computed by taking the ensemble average over many short records with duration 1024T. Since both S(f) and [g-1(t)]2 lacked frequency components lower than 1/(2048T) Hz, their profiles were highly reproducible. An analysis of S(f) confirmed our earlier results which had shown an apparent contradiction to later results by a correlation method. A comparison of S(f) and [g-1(t)]2 based on the same [n(t,T)] clarified the reasons why the bandwidth gamma of S(f) largely differed from the bandwidth gamma of [g1(t)]2 and [g-1(t)]2. The temperature dependence of gamma suggested that F-actin would be flexible and that the flexibility parameter would change with temperature.

5-HT immunoreactive hypothalamic neurons in rat and cat after 5-HTP administration.

Using immunohistochemistry for serotonin (5-HT) after administration of 5-hydroxytryptophan (5-HTP), we have demonstrated the existence of a particular group of 5-HT accumulating neurons in the posterior hypothalamus of the rat and cat. This cluster of neurons, which we termed "5-HT immunoreactive (IR) posterior hypothalamic magnocellular neurons," was characterized by: (1) diffuse 5-HT-IR staining; (2) rather large-sized cell bodies; (3) a marked activity of monoamine oxydase (MAO); (4) capacity of uptake and decarboxylation of 5-HTP; and (5) the absence of capacity of uptake and hydroxylation of tryptophan. These 5-HT-IR magnocellular neurons extended from the suprachiasmatic nucleus area to the caudal end of the hypothalamus. In the rat, the majority of these neurons was found in the tuberal and caudal magnocellular nuclei, whereas in the cat, it was found more widespread in the ventrolateral part of the posterior hypothalamus including the tuberomamillary nucleus. The characteristics of the 5-HT-IR magnocellular neurons which project directly both to the cerebral cortex and the pontine tegmentum are discussed in comparison with those of "APUD" cell series or "paraneurons," as well as with those of other 5-HT-IR neurons demonstrated in the hypothalamus after administration of 5-HTP or tryptophan.

Possible role of serotonin in the secretory activity of the subcommissural organ of the cat.

The possible role of serotonin (5-hydroxytryptamine, 5-HT) in the secretory activity of the subcommissural organ (SCO) was investigated in the cat by using 5-HT immunohistochemistry and Gomori's secretory staining technique. Administration of para-chlorophenylalanine, a potent inhibitor of 5-HT synthesis, was found to produce a marked decrease in both 5-HT-immunoreactive materials and secretory granules in the SCO. In contrast, administration of 5-hydroxytryptophan, an immediate precursor of 5-HT, to animals pretreated with para-chlorophenylalanine resulted in an increase in the amount of both secretory granules and 5-HT immunoreactivities in this circumventricular organ. These results strongly suggest that the secretory activity of the SCO of cats is regulated by 5-HT.

A light and electron microscopic immunohistochemical study of vasoactive intestinal polypeptide- and substance P-containing nerve fibers along the cerebral blood vessels: comparison with aminergic and cholinergic nerve fibers.

Vasoactive intestinal polypeptide (VIP)- and substance P-containing nerve fibers were observed in the cerebral blood vessels using an immunohistochemical technique. VIP-containing nerve fibers distributed in a spiral pattern, similar to that of muscle cells. Under electron microscopic observation, VIP-immunoreactive terminals lay close to a muscle cell in the inner layer of the adventitia. In contrast, substance P-containing nerve fibers showed a meshwork pattern in the outer layer of the adventitia. Using both acetylcholinesterase (AChE) staining and VIP immunohistochemistry, AChE-positive and VIP-immunoreactive nerve fibers revealed almost the same distribution in the same specimen. The present data suggest that VIP-containing nerve fibers may play a role in the smooth muscle control of the blood vessels, whereas substance P-containing nerve fibers may not take part in muscle control.

Morphological observations on frozen and thawed muscovy spermatozoa.

Fresh Muscovy drake spermatozoa were examined using a scanning electron microscope (SEM). The average lengths of the segments were: acrosome 1.8 micron, nucleus 10.9 micron, midpiece 3.6 micron and flagellum (exclusive of midpiece) 71 micron. Under the light microscope, the incidence of abnormal spermatozoa in Muscovy semen subjected to freezing and thawing (almost all with crooked necks) was about 5% higher than that in diluted unfrozen semen. In thawed semen, various abnormalities of the acrosome were observed under the SEM. It seemed that the most radical change was the complete separation of the acrosome from the apical part of the nucleus. The incidence of abnormal acrosomes was increased more than 20% by freezing and thawing. These results suggest that low fertility in thawed semen may be related to increases in the proportion of spermatozoa with crooked necks and acrosomal damage.

Comparative study of the effects of various cryoprotectants in preserving the morphology of frozen and thawed fowl spermatozoa.

The effects of various cryoprotectants (glycerol, dimethylsulphoxide (DMSO), methylformamide and ethylene glycol) of the same molarity on preserving the morphology of frozen and thawed fowl spermatozoa (especially midpiece and acrosome) were examined under the light (LM) and scanning electron microscope (SEM) to determine the most suitable. Under LM, the mean respective increases in sperm deformity in semen diluted with ethylene glycol, methylformamide, glycerol and DMSO were 4.5, 4.9, 5.0 and 6.5 percentage points. Under SEM, the mean respective increases in acrosomal deterioration in semen diluted with glycerol, ethylene glycol, DMSO and methylformamide were 6.6, 8.8, 13.1 and 31.2 percentage points. From these results, it appears that glycerol is superior to ethylene glycol DMSO and methylformamide as a cryoprotectant.

Histochemical localization of thiamine pyrophosphatase in the lomasomes in asci of Arthroderma vanbreuseghemii.

Thiamine pyrophosphatase localization in asci of Arthroderma vanbreuseghemii was investigated histochemically by electron microscopy. The reaction product of this enzyme was demonstrated in some paramural membranous structures that are believed to represent lomasomes. This observation suggests that at least a certain number of lomasomes might be involved in cell wall synthesis and thus behave in a manner similar to the Golgi apparatus in higher eukaryotic cells.

Renal allografts with glomerulonephritic change and proteinuria.

Eight cases of renal allografts with glomerulonephritic change and proteinuria were classified into three groups according to the morphological features of the glomerular lesions. Group I (3 cases): By light microscope, remarkable reduplication of glomerular basement membrane (GBM), widening of mesangial region, and slight increase in mesangial cells, were observed. Electron microscopy revealed thickening of subendothelial space by deposition of electron-lucent material, mesangial interposition, and dense deposits in various regions (mainly in the subendothelial space). Group II (3 cases): By light microscope, crescent formation and reduplication of GBM were observed, while by electron microscope, changes of GBM similar to group I, but less remarkable, were seen. Group III (2 cases): Light microscope revealed spike formation in one case, but not in the other. With an electron microscope, subepithelial dense deposits were observed in both cases. Thickening of subendothelial space by deposition of electron-lucent material was noted in one case, while thickening of lamina densa was observed in the other case. Morphological change caused by rejection was observed in all eight cases, with six cases showing massive proteinuria and the other two showing slight proteinuria.

A simple and sensitive method for determination of human urinary kallikrein activity (kininogenase activity), using human low molecular weight kininogen.

Human low molecular weight kininogen was partially purified and applied to the measurement of human glandular kallikrein as a substrate. The prepared human low molecular weight kininogen did not contain any significant amounts of kinin generating or destroying enzymes. When ethanol was added to the assay tube to stop the enzyme reaction, the substrate was almost completely removed from the incubation solution. Moreover, less than 1.25% ethanol had no effect on the kinin radioimmunoassay. These data suggest that the measurement of generated kinin can be done directly after the addition of ethanol. In this assay system, control tubes were unnecessary since the small volume of the urine samples (0.5 to 2.0 nl) contained negligible amounts of endogenous kinin. In a comparison of the availability as a substrate for human urinary kallikrein among human, dog and bovine low molecular weight kininogens, the enzyme activity was 5 or 100 times as high in the human substrate as in the dog and bovine substrates, suggesting that a human substrate is best for the human enzyme. A significant correlation was found between our previous method using bovine substrate and this method for human urinary kallikrein activity. In both methods, urinary kallikrein excretions were significantly lower in patients with essential hypertension and higher in those with primary aldosteronism, respectively. This simple, specific and sensitive kininogenase assay system seems to be very useful for investigating the physiological or pathophysiological role of the renal kallikrein-kinin system in hypertensive and renal diseases.

Cerebral blood flow and ICP patterns in patients with communicating hydrocephalus after aneurysm rupture.

Cerebral blood flow (CBF) was measured, the intracranial pressure (ICP) was continuously recorded, and the ventricular system size was evaluated on serial computerized tomography scans in 43 patients. These patients all had communicating hydrocephalus after subarachnoid hemorrhage (SAH) from rupture of an intracranial aneurysm. The studies were carried out both in the acute stage (within 7 days after SAH) and in the communicating hydrocephalus stage. In patients in the acute stage who had no ventricular dilatation, but who later developed communicating hydrocephalus, the mean CBF was reduced; lower CBF was associated with poorer clinical grades and a higher resting pressure range. Communicating hydrocephalus produced a significant decrease in CBF. The ICP tracing showed continuing plateau waves in conjunction with B-waves in patients in whom recordings were begun within 63 days after SAH. In general, patients with more dilated ventricular systems, with less frequent ICP irregularities, and with lower resting pressure ranges had a more marked decrease in CBF. A significant decrease in CBF was also found in patients with diffuse vasospasm in comparison to those without vasospasm. Patients with communicating hydrocephalus in whom ICP recordings were started more than 6 months after SAH showed no ICP irregularities. In these patients, a mean CBF of less than 25 ml/100 gm/min and a markedly low resting pressure range were observed. Shunting procedures were never effective in any of these patients. The results suggest that, in patients with communicating hydrocephalus, a mean CBF below this level may cause irreversible damage to the brain tissue in the terminal stage, and may inhibit the cerebral vasomotor reaction that participates in the development of ICP irregularities.

The OP-Rheometer system, a new device for analysis of viscosity and viscoelasticity of blood: description and clinical application.

OP-Rheometer is a rotational viscometer of concentric type, which was developed to measure blood viscosity at low shear rate (0.2-40 sec-1) and viscoelasticity at 0.1-3.0 Hz in the context of clinical medicine. This system consists of a mechanical unit, a control unit (console), a data processing unit and a printer. The dynamic or steady state flow data are automatically printed out. An outer cylinder (sample container) is connected to a torsion wire assembly supported by a pair of magnetic bearings. The angular displacement is detected by a pair of variable differential transformers. In clinical application of this rheometer, authours have confirmed that the viscosity and viscoelasticity of blood from diabetic patients are higher than for normal subjects.
