Pharmacodynamic actions of (S)-2-[4,5-dihydro-5-propyl-2-(3H)-furylidene]-1,3-cyclopentanedione (oudenone).

The pharmacodynamic actions of (S)-2-[4,5-dihydro-5-propyl-2(3H)-furylidene]-1,3-cyclopentanedione (oudenone) were studied in both anesthetized animals and isolated organs. Oudenone (10--40 mg/kg i.v.) induced an initial rise in blood pressure followed by a prolonged hypotension in the anesthetized rats. In unanesthetized spontaneously hypertensive rats (SHR), oudenone (5--200 mg/kg p.o.) caused a dose-related decrease in the systolic blood pressure. The initial pressor effect was diminished by pretreatments with phentolamine, guanethidine, hexamethonium and was abolished in the pithed rats. In addition, intracisternal administrations of oudenone (100--600 mug/kg) showed a marked increase in blood pressure in the anesthetized rats, suggesting that the pressor effect may be due to centrally mediated actions. Oudenone, given intra-arterially into the femoral artery (400--800 mug/kg), caused a long-lasting vasodilation in anesthetized dogs. At a relatively high dose (40 mg/kg i.v.), oudenone antagonized all pressor responses to autonomic agents and central vagus nerve stimulation in anesthetized rats and dogs, however, oudenone showed no anti-cholinergic,-histaminergic, beta-adrenergic and adrenergic neuron blocking properties.

An autopsied case of Eisenmenger syndrome complicated by recurrent thromboembolic phenomena in postpartal period.

Dyspnea, back pain, edema, and cyanosis developed suddenly in a 23-year-old woman during the last trimester of her first pregnancy. Although she had been noticed to have the enlarged heart and exertional shortness of breath to a slight degree, she had been apparently in good condition without any significant heart murmurs. Clinically, recurrent episodes of disseminated intravascular coagulation, including pulmonary thrombosis, were thought to be superimposed to Eisenmenger syndrome associated with toxemia of pregnancy. Anticoagulant and fibrinolytic treatments were tried, but their effectiveness was limited by hemorrhagic diathesis. She died of respiratory and circulatory failure after delivery of a moribund baby. Autopsy revealed Eisenmenger complex (a defect in the membranous portion of the interventricular septum and pulmonary vascular disease) and many fresh hemmorrhages in both lungs with a lot of new and organized thrombi. Fresh thrombi were also seen in the heart, the pancreas and the kidneys. The high peripartal mortality in Eisenmenger syndrome could be attributed to pulmonary thrombosis, which may be related to DIC, as well as to peripartal changes in circulatory function.

Human breast cancer serially transplantable in nude mice in ascites form.

Cell suspension of a human breast cancer cell line (Hattori line) was injected intraperitoneally into an athymic nude mouse to produce ascites form breast cancer (peritoneal carcinomatosis). Subsequent serail transfers of cancer cells in ascites were also successful in mice. All male and female nude mice injected 1 X 10(7) tumor cells died of accumulation of ascites after a latency period averaging 4 weeks, with one exception which died of a wasting disease. Multiple lung metastases were observed in some mice. The tumor cells retained cytological characteristics of the original cell line, and histology of the infiltrating tumor in the peritoneum and omentum was that of poorly differentiated adenocarcinoma. Differentiation not only toward acinar or duct lining cells but also toward myoepithelial cells was observed by histochemistry, immunohistochemistry, and electron microscopy.

Early cellular responses to diverse growth stimuli independent of protein and RNA synthesis.

Serum, elevated pH, excess Zn++, 9,10 dimethyl-1,2 dibenzanthracene (DMBA) and insulin accelerate the progress of growth-inhibited chick embryo cells into the S-period of DNA synthesis. A comparative study was made of their capacity to elicit other cellular responses within two hours after their application. All the agents studied stimulated the uptake of the glucose analogue 2-deoxy-D-glucose (2-dGlc). Elevated pH elicited a more striking increase than the other agents in the uptake of the amino acid analogue alpha-amino isobutyric acid (AIB). The application of subtoxic concentrations of Zn++ or DMBA did not stimulate the uptake of uridine by cells nor its incorporation into RNA when tested at 2 hours. However, it was found that the stimulation of uridine utilization did occur but was delayed several hours. Similarly, the accelerated onset of DNA synthesis was also delayed for several hours by these agents. Insulin acted like serum in stimulating the utilization of 2-dGlc, AIB and uridine. Serum and DMBA were particularly effective in stimulating the utilization of choline. It was concluded that the utilization of 2-dGlc, uridine and thymidine are affected similarly by all the agents, but that there may be differential effects in the utilization of AIB and choline. The inhibition of RNA synthesis by actinomycin D did not prevent the relative stimulation of 2-dGlc, AIB and choline utilization by serum and pH treatment. The inhibition of protein synthesis by cycloheximide did not prevent the relative stimulation of 2-dGlc and choline utilization by serum and pH treatment. It partially blocked the increased uptake of AIB and had erratic effects on the utilization of uridine. It was concluded that neither RNA nor protein synthesis is required for some, if not all, the early responses to growth stimuli measured here. The inhibited cell appears to be a poised system which carries out a programmed array of reactions characteristic of the cell type following perturbation by a variety of unrelated agents. In vivo specificity is provided by the physiological reagents available (i.e., hormones) and their capacity to interact with different cell types.

Hemodynamic and clinical significances of atrial fibrillation, pulmonary vascular resistance and left ventricular function in rheumatic mitral stenosis.

Seventy-four patients with rheumatic mitral stenosis were catheterized and hemodynamic and clinical significances of atrial fibrillation, pulmonary vascular resistance and left ventricular function were studied. These data were also compared to those in the 6 control cases. In addition to the correlation of mitral valve area to the functional classification of patients, significance of atrial fibrillation was also demonstrated. Patients with this arrhythmia had lower cardiac index than those with regular sinus rhythm by approximately 20%, throughout the range of mitral valve area observed. The lower average cardiac index was associated with a higher average left ventricular end-diastolic pressure in cases with atrial fibrillation than in cases without the arrhythmia, in the face of similar average heart rate and average mitral valve area; Average pulmonary vascular resistance correlated to the functional classification, but its systematic influence on the relation between mitral valve area and cardiac index was not observed. Abnormalities of left ventricular function were suggested frequently by various combinations of abnormal values in end-diastolic pressure, end-diastolic volume, ejection fraction, angiographically-measured circumferential fiber shortening velocity (Vcf), and pressure-derived maximal contractile element velocity (Vmax). Patients with enlarged left ventricle had significantly lower average cardiac index than those with normal ventricular size.

Left ventricular asynergy in mitral valve diseases. A cineangiographic study.

Mode of left ventricular contraction was studied by cine left ventriculography in 22 cases with mitral valve diseases and 7 control cases. In rheumatic mitral valve diseases, local hypokinesis of the left ventricular wall and poor contraction of the mitral annulus were observed frequently, at various location and to various extent. Transient asynergic motion of local wall was also seen either during systole or during diastole. Local hypokinesis was seen even in normal-sized left ventricles, and was associated with decreased cardiac pump performance, when its localization was extensive. Severe mitral regurgitation of rheumatic nature was always associated with poor contraction of the mitral annulus, although the reverse was not true.

Heterotransplantation of cultured human cancer cells and human cancer tissues into nude mice.

Twenty-seven out of 31 cultured human cancer cell lines transplanted subcutaneously into nude mice produced solid tumors. Direct transplantation of surgical materials proved less successful. In 35 attempts, only 1 of 6 gastric cancers, 2 of 3 liposarcomas, and 1 of 3 osteosarcomas were accepted. No positive tumor formation was noted after the inoculation of 3 nasopharyngeal carcinomas or 1 breast cancer. None of lymphomatous neoplasms and leukemias produced any tumor in 12 and 3 attempts, respectively. The histological and cytological characteristics of the tumors developed were studied with light and electron microscopes, in relation to the features in vitro and those of the parent tumors. Histological similarity of the tumors that developed in nude mice to their parent tumors were noteworthy. Serial transplantation was performed successfully in 9 cell lines and 2 tissues. Preservation of tissue construction ability and other differentiation abilities of in vitro cultured human cancer cells were ascertained in varying degrees.

Polynucleotides. XXII. Synthesis and properties of poly 7-deazainosinic acid.

Poly 7-deazainosinic acid has been prepared by the deamination and phosphorylation of tubercidin and the nucleoside diphosphate was polymerised using polynucleotide phosphorylase. The polymer has similar physical properties to poly(I), but has a low thermal stability in the double-stranded complex with poly(C). Poly(7-deaza I), in contrast, forms a more stable triple-stranded complex with poly(A) than 2 poly(I). poly(A), presumably due to the higher pK value.
