Interaction between sodium channels in mouse neuroblastoma cells.

Single sodium channels in mouse neuroblastoma cells (N1E 115) were studied in cell-attached patches. During a series of consecutive responses to depolarizing pulses, records with and without channel opening were seen to form clusters rather than appearing randomly. The probability of finding open channels on a record seemed to increase with increasing number of channel openings. The open times of channels became shorter with increasing closed time interval measured between consecutive channel openings. Overlapping openings showed a voltage-dependent open time, in contrast to single openings which had voltage-independent open time. On the basis of these observations interaction between neighbouring sodium channels is suggested.

Copper(II) and nickel(II) ternary complexes of L-dopa and related compounds.

The stability constants of the mixed ligand complexes of L-dopa, L-tyrosine, L-phenylalanine, and dopamine with copper(II) and nickel(II) ions and with 2,2'-bipyridyl and 1,10-phenanthroline were determined pH-metrically at 25 degrees C and an ionic strength of 0.2 mol/dm3 (KCl). Spectral studies were made to establish the binding mode of the ambidentate L-dopa in the ternary complexes. In contrast with the aromatic (N,N) donor atoms, the (O,O) binding mode of L-dopa is particularly favored in its ternary systems with copper(II) and nickel(II); thus, even at physiological pH there is a very considerable formation of (O,O)-bound mixed ligand complexes containing a free amino acid side-chain. Numerous binary transition metal-L-dopa complexes and the ternary complexes formed with various B ligands have been evaluated from a coordination chemistry aspect, with regard to the possibility of their therapeutic application in the treatment of Parkinson disease.

Neuromuscular transmission in the visceral muscle of locust oviduct.

Innervation of the locust oviduct has been investigated with morphological and electrophysiological methods. Using Co2+ and Ni2+ labelling technique, it was found that G7 N2B1 and B2a nerves innervate the oviduct musculature. Ultrastructurally two different terminals could be distinguished: (a) nerve endings containing mainly clear vesicles forming neuromuscular junctions with the muscle fibers; and (b) nerve terminals containing electron-dense granules which showed only "synaptoid" structures, but failed to form junctions with the muscle cells. The neuromuscular junctions proved to be functioning, since it was possible to record intracellularly miniature excitatory postsynaptic potentials and excitatory postsynaptic potentials from the muscle cells. The distribution of the amplitudes of the miniature excitatory postsynaptic potentials suggests a multiterminal innervation. Following electrical stimulation of N2B nerve, excitatory postsynaptic potentials similar to those appearing spontaneously could be evoked. After repetitive stimulation, facilitation or summation of excitatory postsynaptic potentials was observed. The results obtained show that locust oviduct muscle has a double, motor and modulatory innervation.

Ultrastructural damage of crural muscle in post-thrombotic syndrome.

The fine structure of crural muscles in 11 patients suffering from post-thrombotic syndrome was investigated. The contractile elements and the mitochondria were mainly involved in the pathological changes. Glycogen granules in increased number and strong lipid vacuolization were shown. The focal occurrence of damages has to be emphasized. Beside, ultrastructural changes are induced by hypoxia and by other factors. It can be established that limited functional capacity of the lower extremities in post-thrombotic syndrome is due not only to chronic haemodynamic insufficiency but to the serious fine-structural damage.

Wrist disarticulation of a deformed hand: appropriate prosthesis and the habilitation of a severely retarded young man.

A 24-year-old man, who was late attaining developmental milestones and tested in the severely mentally retarded range, had incurred third- and fourth-degree burns at 2.5 years that resulted in a deformed, anesthetic, and useless distal left upper limb. Following wrist disarticulation, he was fitted with a cosmetic prosthesis that he accepted enthusiastically. It permitted him to use his left upper limb as an effective assister. Attempts to wear and use a functional prosthesis were not successful because the bulbous stump was covered with abnormal skin that ulcerated whenever he wore the prosthesis. At present, he resides at home with his mother and has all self-care skills. He attends a day treatment program and helps his mother with household chores as well as neighborhood errands. The prognosis for continued functional improvement is good, even though he remains severely mentally retarded.

Sequence homology between potato spindle tuber viroid and U3B snRNA.

Sequence homology was found by computer analysis between potato spindle tuber viroid (PSTV) RNA and U3B snRNA of Novikoff hepatoma cells. This homology is colinear in arrangement, extends in length to 81% of the entire U3B snRNA molecule and is involved in the PSTV molecule unique sites which, if depicted in terms of the secondary structure of the circular PSTV molecule, reveal a conspicuous regularity in their location. A strong relation in primary structure between PSTV and U3B snRNA is demonstrated by statistical analysis.

Single Na channels in mouse neuroblastoma cell membrane. Indications for two open states.

Single sodium channel currents were studied in mouse neuroblastoma cells. Channel conductance (gamma) was 11.6 +/- 3.4 pS at 6-8 degrees C. Unitary current amplitudes and channel activity increased with increasing temperature. The Arrhenius plot of the conductance was linear between 5 and 35 degrees C with an activation enthalpy of 27.1 kJ/mole (Q10 = 1.28). Amplitude distributions were fitted by the sum of two Gaussian functions indicating the presence of two different single channel amplitudes: smaller i1 and larger i2. The relative probability of appearance of i1, which has a shorter mean open time, was higher during the early phase of depolarization (t less than 16 ms). The open time histograms, inactivating phase of macroscopic currents and delay time histograms were fitted by the sum of two exponentials. The distinct kinetic and steady-state parameters reflect two open states of sodium channels.

Angiography of the bronchial and pulmonary arteries in chronic non-specific lung diseases.

Examination of the bronchial artery system by means of contrast materials and pneumo-angiography has made it possible to determine the extent of the pathological process and to provide an answer to some of the problems raised by chronic non-specific diseases. When free passage is disturbed in the bronchi and alveolar ventilation is abolished, arterial blood will reach through the arterio-arterial anastomosis the branches of the pulmonary arteries, followed by a local hypertension in the affected region and a high oxygen saturation in the terminal branches of the pulmonary artery. In the case of bronchiectasis, the capillary circulation in the bronchial wall decreases because of the extension of the bronchial system which leads to the development of sclerosis and thus to a deterioration of the drainage function of the bronchus.
