Three kinds of morphologically distinct cell wall preparations were isolated from heat-killed Bifidobacterium infantis and examined for the relative antitumor efficacy with syngeneic Meth A fibrosarcoma in BALB/c mice. Ultrastructural examinations revealed that cell wall skeleton (CWS) did not retain morphologically recognizable cell wall structure but showed fibrous structure. By contrast, a new cell wall preparation, whole peptidoglycan (WPG), which was isolated from whole cells without being subjected to physically destructive methods, completely retained the intact cell wall structure. When WPG was disrupted by sonic treatment, it retained some degree of physical integrity of cell wall structure, as compared with CWS. The results of chemical analysis indicated that the three cell wall preparations had similar chemical properties. A single s.c. injection of either CWS, WPG, or sonicated WPG in a mixture with tumor cells resulted in a significant suppression of the tumor growth. They were of equally high activity. However, when WPG, sonicated WPG, or CWS was injected intralesionally five times into mice bearing 5-day-old tumors, the incidence of complete tumor regression was demonstrated to decrease in the order of 70, 40, and 20%, respectively. The in vitro cytotoxicity test excluded the possibility that the tumor cell destruction was the result of direct cytotoxicity of the cell wall preparations. From these findings, it was concluded that WPG was an active stimulator of host-mediated response at the tumor-growing sites.
Genetic deficiency of the alpha-gamma-subunit of the eighth complement component (C8 alpha-gamma) was found in a strain of the New Zealand White rabbit. The serum of this deficient rabbit lacked the immunochemical and functional alpha-gamma-subunit of C8. Mating tests indicate that the C8 alpha-gamma deficiency is transmitted as a simple autosomal recessive trait with the following physiologic characteristics. The body weight at the first week of life, mature weight, and litter size of the deficient animals were smaller than those of heterozygous and normal ones. In addition, survival rates for the first 3 mo of life of the deficient animals tended to be lower than those of heterozygous and normal littermates.
After monoassociation of Bifidobacterium longum with germ-free BALB/c mice (nu/+, heterozygous to nu gene), B. longum was recovered (10(2)-10(4) viable units per organ) from the mesenteric lymph nodes, liver and kidneys for 2 weeks post intragastric inoculation, but not after 4 weeks and later. Cessation of bacterial translocation was not observed in monoassociated nude (nu/nu) mice. Anti-B. longum IgA antibody was detected by ELISA using B. longum cell wall antigen in bile from Week 1 and in ileac wall extract from Week 8 post-association in both nu/+ and nu/nu mice. Total IgA levels in bile, ileac wall extract and caecal contents were also elevated in both mice after monoassociation. Cell-mediated immunity measured by the footpad test and macrophage migration inhibition test using B. longum protein fraction was detected in nu/+ mice in Week 4 and later, but not in nu/nu mice. Nu/nu mice reconstituted 4 weeks prior to monoassociation with lymphocytes from flora bearing nu/+ mice developed delayed footpad reactivity and bacterial translocation stopped after 4 weeks. Cell-mediated immunity rather than IgA antibody correlated well with the cessation of translocation.
The changes of prostatic tissue concentration of antibiotics by operative procedures have not been studied extensively. We measured the concentration of antibiotics in the rat prostatic tissues minced and agitated in the irrigation fluid, as a model of TUR-simulation. Our results suggest that TUR reduced the concentration of antibiotics in the prostatic tissues when compared to open prostatectomy.
The following results were obtained from the statistical studies and drug sensitivity tests of bacterial florae isolated from the urinary tract in 1981 and 1982. Of the 2,029 strains isolated from out-patients, 627 (30.9%) were gram positive cocci, 24.1% were E. coli, 9.4% were Proteus spp., 9.2% were Enterococci, 8.9% were S. epidermidis, 7.0 % were K. pneumoniae, 5.7% were P. aeruginosa and the rest were others. Of the 754 strains isolated from hospitalized patients, 276 (36.6%) were gram positive cocci, 16.8% were Enterococci, 11.4% were S. epidermidis, 11.1% were P. aeruginosa, 8.8% were Proteus spp, 7.2% were E.coli, 6.5% were K.pneumoniae and the rest were others. The number of Proteus spp., P. aeruginosa and Enterococci detected in the isolates from the out-patients and that of E.coli, K. pneumoniae and Proteus spp., detected in the isolates from the in-patients were smaller than in previous reports. More non-fermentative gram-negative bacilli and S. epidermidis were detected in the isolates from both patients were than in previous years. The major isolates i.e., 9 species, from out-patients were more susceptible to the antimicrobial agents tested. The susceptibility of TC and SBPC to major isolates was lower than in previous years. During the past two years, we have been routinely using on inpatients the so-called third generation cefem antibiotics to treat urinary tract infections. This might be why the number of isolates of gram positive cocci has increased especially isolates from inpatients.
4-Tertiary butyl catechol (TBC) causes depigmentation in humans and animals and stimulates formation of pheomelanosomes. In this study, we investigated the effects of noncytotoxic doses of TBC on glutathione S-transferase (GST) activity in the skin of Uscd strain mice and B16 murine melanoma cells in culture, in relation to changes in activities of glutathione reductase (GR) and gamma-glutamyl transpeptidase (GGT) reported to be involved in pheomelanogenesis. Occurrence of pheomelanosomes in skin melanocytes was demonstrated by electron microscopy and reduction (25%) of eumelanin content in melanoma cells was shown by spectrophotometry. Topical application of 1 M TBC-DMSO-acetone solution on the ear skin elevated GST activity about 27%, and activities of GGT and GR to 35% and 19%, respectively, within 1 week. Melanoma cells cultured in 10(-4) M TBC-containing medium for 2 h showed no changes in GST and GGT activities, but 12% increase of GR activity during the first 12 h. Activities of all 3 enzymes was elevated (11-17%) 24 h later. The elevation detected by 48 h was 25% for GST, 26% for GGT, and 14% for GR. The findings were interpreted to show that depigmentation produced by the antioxidant results from stimulated pheomelanogenesis through activation of glutathione-metabolizing enzymes and suppressed oxidation of eumelanin intermediates.

The toxicity of composite resin on rabbit dental pulp was investigated biochemically. A microsomal fraction of rabbit dental pulp was incubated with each of the components of composite resins, and the formation of peroxide was determined by the thiobarbituric acid reaction. Benzoyl peroxide (BPO), the most widely used catalyst, was the most effective on peroxidation, but monomers were not. Cations such as Cu2+ or Fe2+ were required for acceleration of this reaction. Authentic polyunsaturated fatty acids and phospholipids were extensively converted into their peroxides by BPO, but amino acids and carbohydrates were not. Among the active oxygens, hydroxyl radicals were thought to be responsible for BPO-dependent peroxidation. The results presented in this paper indicate that the lipid portion of the cells may be attacked by hydroxyl radicals produced by BPO and copper or iron. Therefore, BPO is considered to be the major factor responsible for the toxicity of composite resins.
The case of a patient with severe left main trunk disease is presented, in which spasm of the right coronary artery resulted in intraoperative myocardial ischemia and perioperative myocardial infarction. It is suggested that coronary spasm may be a cause of perioperative myocardial infarction, and has to be considered in case of unknown etiology.

Effects of 1-(2-ethoxyethyl)-2-(4-methyl-1-homopiperazinyl)benzimidazol e difumarate (KB-2413) on chemical mediators were investigated in vitro and in vivo. Antihistaminic activity of KB-2413 in vitro was about 2, 7 and 5 times more potent than those of chlorpheniramine, diphenhydramine and clemastine, respectively, and equipotent to that of ketotifen. This antihistaminic activity of KB-2413 was competitive antagonism to histamine. In the study preventing histamine-induced mortality in guinea pigs, the antihistaminic potency of KB-2413 (p.o.) was about 39, 780, 68 and 3 times more potent than those of chlorpheniramine, diphenhydramine, clemastine and ketotifen, respectively. In the study preventing histamine-induced increase in vascular permeability, the antihistaminic potency of KB-2413 (p.o.) was about 27 and 4 times greater than those of chlorpheniramine and ketotifen, respectively. The preventing activity of KB-2413 (i.v.) on histamine-induced increase in airway resistance was about 10 and 2 times more potent than those of chlorpheniramine and ketotifen, respectively. These results indicate that the antihistaminic potency of KB-2413 is stronger than that of chlorpheniramine and that of ketotifen which has the strongest antihistaminic activity of all reference drugs. On the other hand, KB-2413 had only very weak anticholinergic, antibradykinin and antiserotonin activity in the study in vitro or in vivo, so that KB-2413 had selective antihistaminic activity. Therapeutic index of KB-2413 calculated from the antihistaminic potency on histamine-induced mortality and acute toxicity in guinea pigs was about 110 times greater than that of chlorpheniramine.(ABSTRACT TRUNCATED AT 250 WORDS)
A study was made of the actual status of urinary tract infection induced by Serratia marcescens on a total of 88 patients who were admitted to our University Hospital in 1981 approximately 1982, and in whose urines not less than 10(4)/ml of S. marcescens were found. An investigation into the background factors of these patients revealed that indwelling catheter, operation for malignancy and use of antibiotics were the major factors responsible for the infection. S. marcescens was found to coexist with P. aeruginosa in not a few cases. The pathogens were highly susceptible to ST compound preparations, DKB and GM, and the pathogens in only 7 of the patients were tolerant to all of these antibiotics. None of the patients died of S. marcescens infection.
